Juice Clarification with the Use of Polygalacturonase Produced by Ganoderma lucidum (Ganoderma lucidum이 생산하는 Polygalacturonase를 이용한 과즙청징)

  • Ganoderma lucidum produced the potent pectolytic enzymes for clarifying cloudy fruit juice. Among the purified polygalacturonases (endo- and exo-polygalacturonase), endo-polygalacturonase had a good effect on juice clarification. The optimum temperature and concentration of endo-polygalacturonase for the juice clarification were $40^{\circ}C$ and 4 unit/5 ml juice, respectively. The apple juice was almost completely clarified at $40^{\circ}C$ for 60 min. It was suggested that culture filtrate of Ganoderma lucidum or it's ammonium sulfate fraction should be used as a good source of pectolytic enzyme for juice clarification.

Purification and Characterization of Acc I Endonuclease (Acc I endonuclease의 정제와 효소적 특성에 관한 연구)

  • Acc I endonuclease has been isolated from 300g (wet weight) cells of Acinetobacter calcoaceticus. The cells were broken by using French press at 20, 000p.s.i. After ammonium sulfate fractionation, the enzyme was further purified by heparin agarose, DEAE-sephades, Affi.-gel Blue, phosphocellulose, and hydroxylapatite column chromatography. The purified Acc I endonudlease has a single polypeptide species and its subunit molecular weight was 45,000 ${\pm}$ 1,000 daltons as judged by 10% SDS-polyacrylamide gel electrophoresis. The isolated enzyme was essentially free of contaminating nucleases as judged by homochromatography by using a $^{32}P-labeled$ oligonucleotide. The enzyme showed maximum activity at pH values between 8.0 and 11.0 and in the presence of $MgCl_2$. Acc I endonuclease was maximally active in the absence of NaCl and was completely inhibited at 200 mM NaCl.

  • The endosperm protein, vicilin, of ginseng (Panax ginseng C.A. Meyer) was purified by ammonium sulfate precipitaion, gel permeation and ion exchange column chromatography. Vicilin is a glycoprotein composed of 2 subunits with molecular masses of 55,000 (large subunit) and 44,000 (small subunit). The anti-vicilin antibody was raised in rabbit, and purified by DEAE Affi-Gel Blue affinity chromatography. The endosperm cells of the seed were reacted with this anti-vicilin antibody and colloidal gold conjugated secondary antibody. Gold particles were labelled on the elaborating granules of Golgi complex, electron-dense granules and protein bodies in the endosperm cells. These results indicated that the vicilin, which was synthesized in rough endoplasmic reticulum and transported to Golgi, was elaborated in saccules of the Golgi and then transported into protein bodies by electron-dense granules.anules.

Enzymatic Properties of Serratia marcescens Pretense (Serratia marcescens Protease의 효소학적 특성)

  • Serratia marcescens ATCC 25419 protease was purified to homogeneity by ammonium sulfate treatment, and DEAE-cellulose anion exchange chromatography. The specific activity of the enzyme was increased 448-fold during purification with an overall yield of 43.0%. Metal reactivation on the purified protease from S. marcescens was studied. S. marcescens protease was a metalloenzyme to be completely inhibited its activity by EDTA and the enzyme outstandingly inhibited by Hg, Fe, Cu, but the activity was increased approximately 20% by Co. The reactivation of the apoenzyme was effective with Mn, Co, Zn in pH range from 6 to 8. Among metalloenzymes prepared to the addition of Mn, Co, Zn to restore the degree of activity of native enzyme, Zn-enzyme was similar to the native enzyme in respects with enzyme activity, alkali-inactivation, thermo-stability.

Statistical Optimization for Improved Production of Cyclosporin A in Solid-State Fermentation

  • This work evaluates the effect of different amino acids on production of Cyclosporin (CyA) production in solid-state fermentation that was previously optimized for different fermentation parameters by one factor at-a-time for the maximum production of CyA by Tolypocladium inflatum MTCC557. Based on the Plackett-Burman design, glycerol, ammonium sulfate, $FeCl_3$, and inoculum size were selected for further optimization by response surface methodology (RSM). After identifying effective nutrients, RSM was used to develop mathematical model equations, study responses, and establish the optimum concentrations of the key nutrients for higher CyA production. It was observed that supplementation of medium containing (% w/w) glycerol, 1.53; ammonium sulfate, 0.95; $FeCl_3$, 0.18; and inoculum size 6.4 ml/5g yielded a maximum of 7,106 mg/kg as compared with 6,480 mg CyA/kg substrate using one factor at-a-time. In the second step, the effect of amino acids on the production of CyA was studied. Addition of $_L$-valine and $_L$-leucine in combination after 20 h of fermentation resulted in maximum production of 8,166 mg/kg.

Studies on the Production and Characteristics of Glucose Isomerase from Steptomyces sp. GI 32. (Streptomyces GI 32 방선균의 Glucose Isomerase 생산과 효소특성)

  • Steptomyces sp. GI 32 with high production of glucose isomerase was isolated from soil. The maximum enzyme production was observed in the culture medium containing 1% sorbitol, 0.6% tryptone, 0.4% yeast extract, 1mM Fe$_2$(SO$_4$)$_3$ with initial pH 7.0 when the cell was cultured at 35$^{\circ}C$ about 18 hours with shaking. The enzyme was partially purified by ammonium sulfate fractionation and DEAE cellulose chromatography. The enzyme was also appeared to be relatively thermostable, and no apopreciable inactivation was observed after incubation at 7$0^{\circ}C$ for 1 hour. The optimal pH and temperature of the enzyme were pH 8.0 and 7$0^{\circ}C$, respectively.

Effect of Larva Gut Enzyme on Pollen (벌 유충창자의 효소가 화분에 미치는 영향)

  • This study was conducted to confirm amino acids from the filterate of pollen treated by purified enzyme of bee larva gut for improving pollen food. The larva gut enzyme were purified by ammonium sulfate, ethanol and Sephadex gel filtration. The filterate of pollen treated with enzyme purified by ammonium sulfate showed the highest amount of amino acids. The amount of amino acids of the filterate from which the pollen was fermented with natural contaminants is 5.5 times as much as the pollen which was not fermented. The filterate of pollen treated with enzyme of bee larva gut contained eighteen different amino acids as follows : aspartates, glutamate, serine, glycine, histidine, arginine, threonine, alanine, proline , tyrosine, valine, methionine, cysteine, isoleucine, leucine, phenylalanine, lysine and trytophane.

Biochemical Characterization of Lectin Isolated from Cherry Tomato Fruit (방울토마토 열매로부터 분리된 lectin의 생화학적 특성)

  • Biochemical characterization of lectin isolated from fruit of cherry tomato through neutral saline extraction, ammonium sulfate precipitation, and affinity chromatography on Sephadex G-200 was studied. The lectin was agglutinated by trypsin-treated human ABO erythrocytes, and the most pronounced activity of agglutination was observed at B type erythrocyte. The analysis of the lectin by SDS-PAGE showed the high intensity band with molecular weights of 10.7 kDa. The optimal temperature and thermal stability of the lectin was $40^{\circ}C$ and $40-60^{\circ}C$, respectively. The maximal pH of this lectin was pH 7.2.

  • Effects of Cetyl Trimethyl Ammonium Bromide (CTAB), which is a kind of cationic surfactants, and Sodium Dodecyl Sulfate (SDS), which is a kind of anionic surfactants on the anodic formation of nanoporous niobium oxide were compared. The addition of SDS could protect the surface from dissolution for long time, leading to the formation of niobium oxide with a double thickness (~400 nm) compared to that prepared without surfactant, whereas dissolution seriously occurred in the solution containing CTAB. The different behaviors were attributed to the interaction between the surfactants with positive (or negative) charge and positively charged niobium oxide.

Studies on the Amylase of Rhizopus(III) (Rhizopus의 아밀라제에 관한 연구 3)

  • In order to clarify the best cultural conditions of Rhizopus niveus the effects of aeration, pH and various nutrients, such as different carbon and nitrogen sources, vitamins, and growth substances, on the mycelial growth were studied through liquid culture, and amylase activities of the fungus at different cultural periods were measured. Soluble starch, xylose and galactose are excellent sources of carbon for growth of the fungus. Sorbose and lactose are utilized slightly for growth. peptone, ammonium sulfate and alanine are excellent nitrogen sources for growth, tyrptophane nad potassium nitrate are utilized slightly for growth and sodium nitrite is not utilized. Thiamine nad gibberellin are excellent growth substances for the fungal growth, and biotin, nicotinamide and indole acetic acid (IAA) are also effective. Rhizopus niveus grows better at rotatory culture than at stationary culture and earlier growth of the fungus increases remarkably at rotatory culture. Optimum pH than at pH3. Growth increases linerly with an increase of soluble starch content up to 100g per liter medium, but 5 grams of ammonium sulfate per liter is the optimum nitrogen concentration for growth, if Pfeffer's medium is employed. Amylase activities of Rhizopus at different cultural periods showed that the maximum amylase production takes place after the cell population has reached its peak in the culture. Dextrinogenic amylase production has reached maximum at stationary phase, and maximum saccharogenic maylase production takes place in the pahse of negative gorwth acceleration.
