하이브리도마 세포배양에서 암모늄 이온의 영향 및 고정화 흡착제에 의한 암모늄 이온의 동시제거 (Ammonium Ion Effects and Its In Situ Removal by Using Immobilized Adsorbent in Hybridoma Cell Culture)

  • The effects of ammonium ion on cell growth kinetics, monoclonal antibody productivity, and cell metabolism of hybridoma cells were investigated. The mouse-mouse hybridoma cell line VlIIH-8 producing mouse IgG2a was used as a model system. Ammonium ion showed an inhibitory effect on cell growth and monoclonal antibody production. New immobilized adsorbents were developed for the reduction of the inhibitory effect of ammonium ion. The ammonium ion selective zeolite, Phillipsite-Gismondine was entrapped in calcium alginate bead or in dialysis membrane and applied to the hybridoma cell culture system for the in situ removal of ammonium ion from culture media. The effects of ammonium the both serum supplemented and serum free media on the cell growth were studied by applying immobilized adsorbents of calcium alginate bead type. The results demonstrated a substantial enhancement in cell growth. Applying immobilized adsorbents of dialysis membrane type to serum supplemented media also resulted in the stimulation of cell growth, cell viability and monoclonal antibody production.

  • Methylobacterium organophilum을 이용하여 메탄 올로부터 고점성 다당류의 생산에 대한 질소원 이용의 통력학척 분석을 유가식 배양에서 수행하였다. 발효중 세포는 배지 중에 첨가된 질소원이 고갈된 후에도 계속 성장하였고, 에틸란은 질소원 고갈조건 하에서만 생성되였다 세포 성장은 배양초기에 첨가 한 질소원인 암모니아의 농도에 비례적으로 이루어 졌으나, 에릴란 생산은 첨가한 암모니아 농도가 $0.75g/\ell$ 이상에서는 오히려 현저하게 감소하였다. 초기 암모니아 농도가 높아질수록, 비세포중식 속도, 비산물생성 속도, 비기질소비 속도가 감소하였다. 높 은 암모니아 농도의 발효 속도들에 대한 저해작용을 감소시키기 위해, 질소원을 일정농도(암모니아 농도=$0.15g/\ell$) 이하로 유지시키면서 공급하는 유가식 배양법으로 메틸란의 농도를 $12.5g/\ell$ 까지 증가시킬 수있었다.

  • There are many reports on the effect of salts to the fermentation of the yeasts, but there are a few reports on the effect of the ammonium thiocyanate(NH$_4CNS$) on the fermentation of the yeasts(Saccharomyces brenerei-Hefe-Rasse X11, Saccharomyces formosensis No. 396 IAM and Saccharomyces cerevisiae IAM). A study has been made on the effects of the ammonium thiocyanate on the fermentation of the above yeasts. The results of the research are as belows: 1) The addition of 0. 00001 mol. of ammonium thiocyanate makes the fermentation of the yeast(Sacch. formosensis No. 39 IAM) facilitate, but has not influenced the another yeast (Sacch. brenerei-Hefe-Rasse XII). 2) The addition of 0. 001 mol. of ammonium thiocyanate makes fermentation of Rases XII the fastest, but the on other yeast (Sacch. formosensis No - 396 IAM) was accelerated by the addition of 0. 1 mol., and it seems to have abnormal fermentation by the addition of 0. 0001 mol. The addition of ammonium thiocyanate(0. 00001-0. 001 mol.) inhibited the fermentatirn of the yeast(Sacch. cerevisiae IAM), but the concentration of 0. 1 mol. does not interrupt the fermentation of Sacch. cerevisiae IAM and increased the fermentation. 3) The order of effects to the fermentation of each yeast is Sacch. brenerei-Hefe-Rasse XII, Sacch. formosensis No. 396 IAM, Sacch. cerevisiae IAM.

축산폐수의 혐기성 고정법에 있어서 암모니아성 질소의 영향 (Effect of ammonium nitrogen in anaerobic biofilter using live-stock-wastewater)

  • In this research, the synthetic livestock wastewater was prepared to study the characteristics of organic matter removal, the change of VFA production, and the amount of gas production with respect to the change of ammonium nitrogen concentration in the waste using anaerobic fixed bed process, which is an anaerobic biofilm process. The HRT and operation temperature were 1 day and $35{\pm}1^{\circ}C$, respectively. Also, the characteristics of organic matter removal and the inhibitory effect on microorganism in the anaerobic process were studied on the organic loading and ammonium nitrogen concentration. The results obtained were as follows: For COD loading of $10kg/m^3$-day and five levels of ammonium nitrogen concentration ranging from 1,000 to 5,000 mg/L, organic removal efficiencies were about 81, 74, 67, 58, and 51%, and gas productions were 3,860, 3,520, 3,240, 3,020, and 2,790 ml/l-day, respectively. Average methane contents in the gas produced on COD loading of $10kg/m^3$-day was about 76%. Throughout the whole period of experiment, remaining VFA (as COD base) in the effluent was over 90% of remaining COD. This result indicated the inhibitory effect of high concentration of ammonium nitrogen through the facts that accumulated VFA was almost COD and organic removal efficiency decreased also with the increase of ammonium nitrogen. Especially, that implys which high concentration of ammonium nitrogen not only inhibits methane forming bacteria, but also acid forming bacteria.

  • It is very important to have a good kinetic model which considers the effects of both ammonium and glucose for the control and optimization of the poly-${\beta}$-hydroxybutyrate (PHB) fermentation. A kinetic model for the growth of Alcaligenes eutrophus and the biosynthesis of PHB under both ammonium and glucose limitation was proposed. Growth rate of residual biomass was expressed as a function of concentrations of residual biomass, glucose and ammonium having glucose inhibition. PHB production rate was expressed as a function of concentrations of residual biomass, glucose, ammonium and PHB content having ammonium and product inhibitions. Novel approaches were made to estimate the parameters in the model equations which considered two limiting substrates. Model parameters were evaluated by graphical and simplex methods. The proposed kinetic model fitted the data very well.

  • This study was carried out to search for growth and quality -of turfgrass by Nitrogen source (Urea Ammonium sulphate) and organic matter, and to obtain methods of pr6per turfgrass management by the results. This experiment was conducted in turf field from 1988 to 1989. The results of this experiment was summarized as follows; 1.Without application of organic matter, turfgrass growth between urea and ammonium sulphate was not significant, however ammonium sulphate resulted in superior to urea in total dry weight when organic matter was applied. 2.The addition of organic matter showed significant effect on color rating and visible quality, and ammonium sulphate was higher quality than urea. 3.Shoot density increased, particularly in ammonium sulphate treatment, when organic matter was applied. 4.Rhizome and stolon were shown significant effect in urea treatment by the application of organic matter. 5.Surface soil pH was allowed to become slightly acid by the ammonium sulphate treatments, but soil acidity remained mostly unaffected when organic matter was supplied. 6.Total nitrogen of log tissue and organic matter content increased when organic matter w-as applied.

  • Wild-type 곰팡이인 Tolypoc/adium inflatum의 고정상배양을 통해 탄소원, 질소원, 아미노산 등이 peptide 항생제 계통 의 면역억제제인 cyclosporin A(CyA) 생합성에 미치는 영향을 조사하였다. 질소 원으로서 ammonium sulfate가 첨가된 경우에, f fructose 또는maltose 등을 탄소원으로 이용하는 배 지가 glucose가 첨가된 배지에 비해 월등히 높은 C CyA 생산성을 보였으나. ammonium sulfate의 부 재시에는 탄소원들의 종류에 관계없이 CyA 생산성 이 매우 낮게 나타났다. 질소원의 경우는 무기 질소 원인 ammonium sulfate가 CyA 생산에 가장 훌륭 한 성분으로 작용했으며, 그밖에 ammonium phos p phate, ammonium citrate 역시 CyA 생산을 어느 정도 증가시키는 효과를 보였다. 최척 ammonIum s sulfate의 농도는 109/L로 밝혀 졌으며 배양초기 에 a ammonium sulfate릎 첨가해 주는 경우가 배양중간 에 첨가하는 경우보다 CyA 생산에 더 효율적인 것 으로 나타났다. 아미노산의 경우 L-valine에 의한 C CyA 생합성 증진 효과가 가장 투렷하게 나타났다. 최적 L-valine 농도는 109/L이였으며 L-valine이 배양 초반부터 배지 중에 존재하는 것이 CyA 생합 성에 가장 유리한 것으로 나타났다.

암모늄염으로 처리된 느티나무의 난연성 시험 (Flame Retardancy of Zelkova Sarrata Treated with Ammonium Salts)

  • This study was performed to test the flame retardancy of zelkova sarrata-based materials by the treatment of ammonium salts. Zelkova sarrata plate was soaked by the treatment with three 20 wt% ammonium salt solutions consisting ammonium chloride (AMSL), monoammonium phosphate (MAPP), and diammonium phosphate (DAPP), respectively, at the room temperature. After the drying specimen treated with chemicals, combustion properties were examined by the cone calorimeter (ISO 5660-1). When the ammonium salts were used as the retardant for zelkova sarrata, the flame retardancy improved due to the treated ammonium salts in the virgin zelkova sarrata. However the specimen shows increasing CO over virgin zelkova sarrata and It is supposed that toxicities depend on extents. Also, the specimen with ammonium salts showed the higher total smoke release (TSR) than that of virgin plate. Of specimens treated with ammonium salts the ammonium chloride handled the test side was considered a improved inhibitory effect of combustion.

암모니아성 질소가 미생물연료전지에서 전류 발생에 미치는 영향 (Effect of ammonium on the current generation in the microbial fuel cell)

  • These studies carried out to know the effect of ammonium on the current generation in the microbial fuel cells (MFCs). MFCs used in the study were enriched with anaerobic digestion sludge and operated for 3 years using artificial wastewater (AWW). When the current was stably generated, ammonium ion with $27.0{\pm}0.0$, $51.5{\pm}0.0$, $103.5{\pm}0.0mg/L$ with acetate fed into the anode compartment. The current values under condition included ammonium were changed from its initial $6.30{\pm}0.06$ to $6.28{\pm}0.36$, $5.95{\pm}0.61$, $5.64{\pm}0.38mA$, respectively. The current value was slightly decreased to $5.64{\pm}0.38mA$ compared to $6.30{\pm}0.06mA$ generated from MFC without ammonium ion in the AWW. But After 3days operating under ammonium concentration with $103.5{\pm}0.0mg/L$, the current was unstably generated when artificial wastewater without ammonium was fed again. MFC enriched with AWW without ammonium ion was inhibited by high concentration of ammonium. At this time, the ammonium was removed 5.27~16.41 mg per day under all conditions.

  • CHO 세포외 성장 및 생산성을 저해하는 엄포늄 이온의 동시 제거를 위해 PhJlhpsite~Gismondine 합성 zeolite가 alginate, cellulose acetate. dialysis membrane에 고정화된 흡착제가 개발되었다. 고정화 흡착체에 의한 암모늄 이온의 제거 효율 및 비에 따른 세표성장의 증진효과를 비교한 결과 최적의 고정화 흡착제로 membrane type이 선정되었다. 암모늄 이온의 제거 효파를 더욱더 명확하게 하고 고멸도의 세포 배양을 모사하가 위하여 8mM의 ammonium chloride를 첨가하고 membrane type 고정화 흡착제를 투여하여 암모늄 이온의 동시제거 효과를 조사한 결과 최대 세포 농도가 3배 이상 증가하였으며 생존율 역시 증가하였고 40%정도의 tPA 생산성 향상올 보여 주었다­. 이러한 증진 효과는 암모늄 이온의 고농도 시스템일수록 커졌다. 고정화 흡착제의 최적 투며 시기를 조사해본 결과 ammonium chlonde를 첨가하지 않았을 때 membrane type 고 정화 흡착제의 최적 투여 시기는 배양 시작 후 48시간이었고, 8mM의 ammonium chloride를 침가한 경우의 최적 투여 시기는 배양 시작 후 72시간이었다.
