  • 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), a heterocyclic amine, significantly inhibits nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated macrophages. The decrease in NO production was found to correlate well with a decrease in iNOS mRNA expression as demonstrated by Northern blot analysis.(omitted)

Compositions of Extractive Nitrogenous Constituents and Their Monthly Variation for Fresh Capsosiphon fulvescens

  • To elucidate the composition of extractive nitrogenous components in the fresh Capsosiphons fulvescens cultured off the southern coast of Korea, and to determine the monthly variation of these nitrogenous components, extract samples collected monthly from December to March at Jangheung-gun, Jeonnam Province were analyzed for total nitrogen, free and combined amino acids, ATP and related compounds, betaines, trimethylamine oxide (TMAO) and trimethylamine (TMA). The content of extractive nitrogen was 1,090~1,233 mg/100 g on dry basis. The number of 21~25 ninhydrin-positive substances was detected in the analysis of free amino acids, and their total amount was 3,710~4,788 mg/100 g on dry basis. Among them, free proline, asparagine, glutamic acid, alanine, taurine and glutamine were found to be abundant. The combined amino acids amounted to 1,573~2,121 mg/100 g in total and the total amount of ATP and related compound was 33.8~84.0 mg/100 g ($1.06{\sim}2.46\;{\mu}mol/g$) on dry basis. Betaine, glycinebetaine, $\beta$-alaninebetaine, $\gamma$-butyrobetaine, homarine and trigonelline were detected in most of samples. Levels of free and combined amino acids, ATP and related compounds fluctuated from sample to sample, with their contents higher in December and January and lower in March.

Effect of Various Pathological Conditions on Nitric Oxide Level and L-Citrulline Uptake in Motor Neuron-Like (NSC-34) Cell Lines

  • Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disorder that causes progressive paralysis. L-Citrulline is a nonessential neutral amino acid produced by L-arginine via nitric oxide synthase (NOS). According to previous studies, the pathogenesis of ALS entails glutamate toxicity, oxidative stress, protein misfolding, and neurofilament disruption. In addition, L-citrulline prevents neuronal cell death in brain ischemia; therefore, we investigated the change in the transport of L-citrulline under various pathological conditions in a cell line model of ALS. We examined the uptake of [14C]L-citrulline in wild-type (hSOD1wt/WT) and mutant NSC-34/ SOD1G93A (MT) cell lines. The cell viability was determined via MTT assay. A transport study was performed to determine the uptake of [14C]L-citrulline. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis was performed to determine the expression levels of rat large neutral amino acid transported 1 (rLAT1) in ALS cell lines. Nitric oxide (NO) assay was performed using Griess reagent. L-Citrulline had a restorative effect on glutamate induced cell death, and increased [14C]L-citrulline uptake and mRNA levels of the large neutral amino acid transporter (LAT1) in the glutamate-treated ALS disease model (MT). NO levels increased significantly when MT cells were pretreated with glutamate for 24 h and restored by co-treatment with L-citrulline. Co-treatment of MT cells with L-arginine, an NO donor, increased NO levels. NSC-34 cells exposed to high glucose conditions showed a significant increase in [14C]L-citrulline uptake and LAT1 mRNA expression levels, which were restored to normal levels upon co-treatment with unlabeled L-citrulline. In contrast, exposure of the MT cell line to tumor necrosis factor alpha, lipopolysaccharides, and hypertonic condition decreased the uptake significantly which was restored to the normal level by co-treating with unlabeled L-citrulline. L-Citrulline can restore NO levels and cellular uptake in ALS-affected cells with glutamate cytotoxicity, pro-inflammatory cytokines, or other pathological states, suggesting that L-citrulline supplementation in ALS may play a key role in providing neuroprotection.

  • 풀러렌($C_{70}$)을 3-picoline N-oxide, pyridine N-oxide hydrate, quinoline N-oxide, isoquinolne N-oxide의 여러가지 아민 N-옥시드를 사용하여 $25{\sim}43^{\circ}C$, 초음파 조건에서 반응시켜 풀러렌 산화물을 합성 하였다. 동일한 초음파 조건에서 여러가지 아민 N-옥시드에 대한 풀러렌 ($C_{70}$)의 반응성은 3-picoline N-oxide : pyridine N-oxide hydrate : quinoline N-oxide : isoquinoline N-oxide 처럼 동일하게 나타났다. MALDI -TOF MS, UV-vis 그리고 HPLC를 사용하여 생성된 풀러렌 산화물을 분석한 결과 [$C_{70}(O)n$] (n=1) 임을 알 수 있었다.

  • Chitin is a major component of the shells of crustacea such as crab. shrimp and crawfish. Renal dipeptidase (RDPase. EC 3.4.13.19), an ectoenzyme of renal proximal tubules. is covalently bound to outer leaflet of lipid bilayer via glycosylphosphatidylinositol (GPI)-anchor. The biological role of RDPase was suggested as the hydrolysis of dipeptide into free-amino acids before renal reabsorption. The underlying biochemical mechanism of decreased RDPase release was suggested as nitric oxide (NO) production. (omitted)

공진단(拱辰丹)의 성분 분석 및 갱산화(坑酸化) 작용에 미치는 영향 (An analysis of the Gongjindan's ingredients and its efficacy on anti-oxidation)

  • Objectives : For the congenital feeble, the Gongjindan is useful medicine. The experiment is to estimate the value of the Gongjindan as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : General ingredients' of the Gongjindan's extract were analyzed first and the quantitative analysis of a reducing sugar, a soluble protein, an amino acid and minerals was made. The Gongjindan, which is extracted, concentrated, and freeze drying with water, ethanol and chloroform, have got applied for the experiment. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained amino acid, in order of high amount, is Arginine, Alanine, Glutamic acid, Proline and the contained free amino acid is Glutamic acid, Leucine, Lysine, Phenylalanine. The derivative of free amino acid is ${\gamma}-Aminoisobutyric$ acid, Phosphoserine, Taurine. And the Gongjindan is containing 13 species of minerals in order of Ca>K>Na>Mg>Fe>AI>Mn. Then, to assure of the Gongjindan's capability of anti-oxidation, these following subjects-polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibition- are analyzed and show high activity especially the most in ethanol extracts. Conclusion : With this analysis of ingredients, the Gongjindan is containing many materials functioning as anti-oxidation, anti-aging, anti-fatigue, neurotransmitter and immune agent. Moreover, In every water, ethanol, chloroform extracts, the Gongjindan's capability of anti-oxidation is confirmed so that we can apply to patients' treatment clinically.

Extremely Low Frequency Magnetic Field Modulates the Level of Neurotransmitters

  • This study was aimed to observe that extremely low frequency magnetic field (ELF-MF) may be relevant to changes of major neurotransmitters in rat brain. After the exposure to ELF-MF (60 Hz, 2.0 mT) for 2 or 5 days, we measured the levels of biogenic amines and their metabolites, amino acid neurotransmitters and nitric oxide (NO) in the cortex, striatum, thalamus, cerebellum and hippocampus. The exposure of ELF-MF for 2 or 5 days produced significant differences in norepinephrine and vanillyl mandelic acid in the striatum, thalamus, cerebellum and hippocampus. Significant increases in the levels of serotonin and 5-hydroxyindoleacetic acid were also observed in the striatum, thalamus or hippocampus. ELF-MF significantly increased the concentration of dopamine in the thalamus. ELF-MF tended to increase the levels of amino acid neurotransmitters such as glutamine, glycine and ${\gamma}$-aminobutyric acid in the striatum and thalamus, whereas it decreased the levels in the cortex, cerebellum and hippocampus. ELF-MF significantly increased NO concentration in the striatum, thalamus and hippocampus. The present study has demonstrated that exposure to ELF-MFs may evoke the changes in the levels of biogenic amines, amino acid and NO in the brain although the extent and property vary with the brain areas. However, the mechanisms remain further to be characterized.

The Effect of Source of Dietary Fiber and Starch on Ileal and Fecal Amino Acid Digestibility in Growing Pigs

  • Studies were carried out with a repeated $4{\times}4$ Latin square design with eight cannulated pigs fed four experimental diets to investigate the effect of dietary fiber and starch sources on apparent ileal and fecal amino acid digestibility. Each period lasted 15 d, with diet acclimation from d 1 to 7, feces collection for 48 h on d 8 to 9 and ileal sample collection for 12 h on d 13 to 15. The four experimental diets consisted mainly of cooked rice with the addition of protein sources (CON), partial replacement of cooked rice with either potato starch (PS), sugar beet pulp (SBP) or wheat bran (WB). Chromic oxide was used as an indigestible marker. With the exception of histidine, lysine and tryptophan, no differences were observed in the apparent ileal digestibility of amino acids between diets. The inclusion of potato starch did not affect the ileal and fecal amino acid digestibility. In comparison with diet CON, a decreased (p<0.05) ileal digestibility of histidine was found in pigs fed diet SBP, while the ileal digestibilities of histidine, lysine and tryptophan were decreased (p<0.05) by the inclusion of wheat bran. Inclusion of fiber sources (sugar beet pulp and wheat bran) caused a reduction (p<0.05) in the fecal amino acid digestibility and the net disappearance of amino acids in the large intestine. Of the indispensable amino acids, there was a 'net synthesis' for methionine in the large intestine of pigs when diets were supplemented with dietary fiber. The decrease in fecal amino acid digestibility with the addition of dietary fiber indicates an increase in the synthesis of bacterial protein in the large intestine.

A study on Determination Method of (N-2-hydroxy-ethyl)valine(HEV) in Hemoglobin Adducts for Biological Monitoring of Ethylene Oxide Exposure

  • Ethylene oxide is a genotoxic carcinogen with widespread uses as industrial chemical intermediate and gaseous sterilant. 2-hydroxyethylated N-terminal valine in Hb is a good biomarker for biological monitoring of ethylene oxide exposure, because of its stability. We studied the determination method of (N-2-hydroxy-ethyl)valine in hemoglobin adduct by using GC/MS. PFPITC and TBMS were used as appropriate derivatives. Ethylene oxide formed Hb adducts as (N-2-hydroxy-ethyl)valine(HEV) in mouse with ethylene oxide inhalation exposure. Standard HEV can be synthesized with 2-amino-ethanol and 2-bromo-3-methylbutyric acid. GC/MS can measured them after derivatization with pentafluorophenylisothiocianate(PFPITC) and N-(tertiary butyl dimethylsiiyl)-N-methyl-trifluoroacetamide(TBDMS-TFA) by using Edman procedure. Concentrations of Hb adduct were proportionally increased with exposure levels. They were 230${\pm}$35(nmol g$^{-1}$ globin) and 410${\pm}$72(nmol $g^{-1}$ globin) at 200ppm and 400ppm ethylene oxide inhalation exposure, respectively.

Easy Preparation of Nanosilver-Decorated Graphene Using Silver Carbamate by Microwave Irradiation and Their Properties

  • We have successfully decorated reduced graphene oxide (RGO) with silver nanoparticles (AgNPs) by microwaving silver alkylcarbamate for 13 seconds using 1-amino-4-methylpiperazine. Uniform AgNPs (20-40 nm) were effectively prepared, and 1-amino-4-methylpiperazine acted as a reaction medium, reducing agent, and stabilizer. Particle size and morphology were correlated with the silver alkylcarbamate concentration and microwave time. The graphene/AgNPs composites were characterized by Raman, X-ray diffraction, and scanning electron microscopy to confirm that the AgNPs were uniformly decorated onto the graphene. Measurements of the transparent conductive property at room temperature indicated that these graphene/AgNPs nanosheets with 55.45% transmittance were electrically continuous with a sheet resistance of approximately $43{\Omega}/{\Box}$.
