Bacterial Community Structure and Diversity of the Zoysia japonica Soil Treated with Liquid Fertilizer Containing Amino Acids (아미노산 액비를 처리한 들잔디 토양 미생물 군집구조 및 다양성)

  • T-RFLP analysis and clone sequencing analysis based on bacterial 16S rDNA were conducted to assess bacterial community structure and diversity in Zoysia japonica soil treated with liquid fertilizer containing amino acids(LFcAA) after spray with herbicide. The results of T-RFLP (terminal restriction fragment length poly-morphism) analysis using restriction enzyme Hae III showed that the T-RFs of various size appeared evenly in the 32 clones of KD3 and 38 clones of KD4 respectively that had been treated with liquid fertilizer containing amino acid(LFcAA) compared to 23 clones of KD2 hat had not been treated with LFcAA. The microbial com- munity structure in KD2 appeared less diverse than those in KD3 and KD4. Analysis of partial sequences for 110 clones from KDI (control), KD2 (non-treated), KD3 (LFcAA 1X), KD4 (LFcAA 2X), respectively, revealed that most bacteria were related with uncultured bacteria in a 16S rDNA sequence similarity range of 91-99% through blast search. Otherwise, the other clones were members of proteobacteria, Acidobacteria, Act-inobacteria, Sphingobacteria and Planctomyces groups. Especially in KD4, members of Alpha Proteobacteria, Rhizobiales, Sphigomonadales, Caulobacterales, Gamma Proteobacteria, the genus Pseudomonas, Betapro-teobacteria, Nitrosomonadales and genus Nitrosospira appeared to be dominant. In addition, Acidobacteria group, Actinobacteria group, Planctomycetacia and Sphingobacteria were also shown. The microbial com-munity structure in Z. japonica soil sprayed with herbicide was affected by LFcAA.

  • A pot experiment was carried out using Seomjin-byeo and Samgang-byeo to investigate the effects of nitrogen and sulfur on the growth, yields, and sulfur containing amino acid content of rice. The yields and number of tillers at 50 days after transplanting were increased with the increase of the amounts of nitrogen and sulfur, but N/S ratio was decreased in both varieties. There were a positive correlations between yield and contents of nitrogen and sulfur in straw and brown rice, but a negative correlation between N/S ratio and yields at harvesting stage. Crude protein contents increased with the increase of amounts of nitrogen and sulfur. contents of amino acid containing sulfur in brown rice were positively correlated with the sulfur and nitrogen contents in rice straw.

  • A pot experiment was conducted to learn the effect of non-compounded and compounded (with super phosphate) liquid fertilizers made of amino acid fermentation by product and the time of their application on the emergences and yield of orchard grass. Results obtained as follows; 1. The Application of the liquid fertilizers, on seeding day of the orchard grass raised soil pH and salt concentration more than the application on the days before seeding and decreased the number of sprouts of the grass which also lead to a lower yield of the grass compared with the latter. 2. Liquid compound fertilizer, however, tended to reduce the harmful effect on sprouting orchard grass compared with non-compounded liquid fetilizer, particulary in soil of on-lime applied. 3. The application of liquid and liquid compound fetilizers on somewhat acidic bare land a quite ahead of seeding or transplanting crops may not only reduce the effect of the fertilizers, provided the land is free from washing by rain or irrigation water, but also provide better conditions for sprouting and early growth of plant seedings than the use at the pre-seeding or pro-transplanting time.

Production of liquid fertilizer from broken eggs and evaluation of its effect on lettuce growth

  • Eggs are likely to be used in agriculture because they can provide enough nutrients for crop growth. Statistics show that a large number of eggs are lost due to breakage before reaching the final consumer. The purpose of this study was to make a natural liquid fertilizer as a substitute for chemical fertilizers using broken eggs as a resource and to evaluate the efficiency of the formulated fertilizer. To make the liquid fertilizer, the broken eggs and distilled water were mixed at ratios of 6 : 4 and 4 : 6. Then, effective micro-organisms (EM) and sugar were added, and the mixture was fermented. The temperature and electrical conductivity (EC) increased gradually with the fermentation while the pH decreased. When evaluated following the seed germination index method of the compost, it was found that the fertilizer matured 10 days after the beginning of the experiment. The growth experiment was conducted with lettuce in which the fermented liquid fertilizer was compared with a commercial liquid fertilizer. The 6 : 4 treatment produced plants with the densest fresh shoot and roots weighing 41.6 and 4.6 g, respectively. The number of leaves (12.3 per plant) was also the highest for the 6 : 4 treatment. Soil analysis showed that the soil pH was improved, and the soil organic matter was increased in the fermented liquid fertilizer treatment.

The Effect of Wood Extract as a Water-Soluble Fertilizer in the Growth of Lactuca sativa

  • Recently, due to environmental and toxicity issues, there has been increasing attention on research regarding natural products that can reduce the use of chemical fertilizers. Wood extracts derived from the biorefining process contain various fertilizer ingredients. HPLC analysis revealed that wood extract contains approximately 5.2% hemicellulosic sugar. The growth of lettuce (Lactuca sativa) upon treatment with wood extract (extract obtained from steam-exploded pine) or water-soluble fertilizers containing different nutrients was analyzed in this study. After two weeks, the growth characteristics of lettuce as affected by wood extract or water-soluble fertilizers were significantly different. The effect of water-soluble fertilizers containing ascorbic acid, magnesium sulfate, citric acid, potassium nitrate, amino acids, or seaweed extract was less desirable than that of wood extracts regarding plant height (18.6 cm), number of leaves (10), leaf length (14.1 cm), shoot fresh wight (9.8 g/plant), root fresh weight (0.8 g/plant) and shoot dry weight (0.6 g/plant). The plant height, number of leaves, leaf length, shoot fresh wight, root fresh weight, shoot dry weight of water-soluble fertilizers containing wood extract were significantly different compared to the control (plant height :13.5 cm, number of leaves : 7, leaf length : 9.4 cm, shoot fresh wight : 5.3 g/plant, root fresh weight : 0.7 g/plant, shoot dry weight : 0.4 g/plant, root dry weight : 0.07 g/plant). From these results, it was concluded that wood extract can be used as a potential water-soluble fertilizer to increase the yield of leafy vegetables.

  • In an effort to determine the bio-synthesis in the soybean as investigate to the variance of each substance: Glutamic acid, Aspartic acid and its amides during the growth of younger soybean plants. 1. The variance-curve of Gultamic acid and Aspartic acid as the acidic amino acids in the cotyledons was appeared the peak the first half period at Glutamic acid and the latter half at Aspartic acid in the growth of soybeans, and was received the symmetrical impression centering around the stage of adult leaf-development. But, in the embryonic organ, it appears the peak at both part, in the developmental stage of adult leaf and also appears near phenomena of increase and decrease in the variation-curve of metabolites. 2. It's amides-Gultamine and Asparagine-appears the peak at the developmental stage of adult leaf in the both cotyledons and embryonic organ, and rapid increase in the cotyledons were very impressed compare with the decrease at fallen stage of cotyledons in the embryonic organs. 3. In the relation of variance at Glutamic acid and Aspartic acid, both substance were discovered the fact of translocation from cotyledon to embryonic organ, and Glutamic acid could supposed that bear the charges of outrider substance in other amino acid as the Glutamic acid-self and major basic function for receiving the ammonia as the nitrogen contain constituent of plant. In the case of Glutamine, formation-mechanism of ammonia which develops due to its hydrolysis in the latter period of soybean growth, suggested that was forfeit its function till instance of fallen cotyledons. 4. In the relation the Aspartie acid and Asparagine, Aspartic acid which begins to decrease from seed-state was supposed that bear sufficiently the charge of outrider substance in the formation of Asparagine other than translocated to embryonic organ from cotyledon. And, formation-theory of Aspartic acid which suppose as formational substance from Kreb's cycle were recognized from latter period of soybean growth, and then, rapid accumulation of Asparagine's amounts were supposed that adapt to two theory: Theory which consider to transformation as Asparagine state for pressing to less than noxious weight the concentration of ammonia developing from the cells, and was formate and accumulate as ammonia or carbohydrates containing excess in the cotyledons.

  • This study was carried out to investigate in influence of inoculation B. sp. and lime types on nutrient content of peanut seed, and the correlation between nutritient contents in seed and plant. 1. Inoculated B. sp., content of crude protein, phenylalanine, isoleucine, cystine, histidine, aspartic acid, serine, and alanine in seed were increased significantly, but that of crude fat was decreased. 2. The content of crude fat was the highest without lime, glutamic acid was the highest with calcium sulfate, valine, phenylalanine, isoleucine, leucine, lysine, aspartic acid, serine, glycine and alanine with slaked lime, and histidine with calcium carbonate. 3. The content of crude protein in seed was negatively correlated with that of crude fat. The contents of chlorophyll, allantoin, ammonia, free amino acid, T-N, $P_2O_5$, and CaO in leaf at 100 days after sowing were positively correlated with the content of crude protein and various kinds of amino acid in seed, but negatively with the content of crude fat. The content of nitrate in leaf at 100 days after sowing showed the result of contrary tendency.

Effect of Nitrogen Split Application on Growth and Quality of Okro Tea Leaves(Camellia sinensis var. sinensis) (질소 분시횟수가 옥로차엽의 생육과 품질에 미치는 영향)

  • The split application of a nitrogen fertilizer was applied at the levels to 3, 4, 5, 6 and 7 time. The results of yield and quality of Okro tea are summarized as follows: Among five treatments, the growth and yield were somewhat more excellent in the five time split treatments compared with other treatments. The chemical components such as total nitrogen, total amino acid and vitamin C were somewhat higher in the five time split treatment compared with other treatment. However, the content of components such as tannin, caffeine, chlorophyll were not different among plots of $4{\sim}7$ time split application except three time split application manuring. The content of fatty acid was producted $2,888{\sim}2,933mg/100g$. Especially, the content of oleic acid, linoleic acid and linolenic acid was the higheat in the five time split application. Sixteen kinds of free amino acid including theanine as a good taste component of Okro tea were isolated and measured by HPLC. Theanine in the five time split application was higher with value of $72{\sim}513mg/100g$ than in other treatment. In scoring test, tea appearance and interal qualities of 3 time split application become lower than any other treatment as 80.2 point, while five time application had the highest with 84.7 point. Conquently, those results meant that five time split application was optimum split application of fertilizer for high yield and good quality of Okro tea.

  • In a series of studies on the chemical compositions and properties of organic matter from peats, volcanic ash soils and paddy rice soils, a comparative study on the composition and the fractional distribution of nitrogenous organic matters were undertaken for the elucidation on their characteristics. The results obtained are summerized as follows. 1. Gimpo and Peongtaeg peats showed higher content of organic matter, more than 60%, than Gimje and Yeongdong peats, and there is a great difference between the two groups in total-N, CEC and ash content. 2. The content of Lignin, Hollocellulose and Hemicellulose in organic matter was 12-25%, 15-31% and 7-14% respectively, and Gimpo peats were lots of Lignin content compared to others. 3. Sixty seven to eighty eight percent of the total-N in the soil was hydrolysable by 6 N-HCl and they were decreased in the order of volcanic ash soils (86.4%), mineral paddy soils (77.2%) and peats (72.3%). 4. The fractionations of acid hydrolysable organic-N were ${\alpha}$-amino acid-N(25-45%), unidentified-N (12-50%), amino sugar-N (1-7%) and ammonium-N (12-25%). 5. Almost same pattern in the amino acid composition of the acid hydrolysates of peat and soil organic matter was showed in the thin-layer chromatogram.

  • Humic acids were extracted from straw of wheat and rye at three different stages of decomposition. Contents and distribution of amino acids in the hydrolysates of humic acids were examined and the results obtained can be summarized as the following: 1. Contents and distribution of amino acids in the hydrolysates of humic acids differ from plant to plant and from one stage of decomposition to another. 2. Neutral amino acids as a group take the largest portion of the total amino acids in humic acid hydrolysates followed by the acidic and the basic. 3. The total amount of amino acids in decomposed wheat straw at the 90 days of humification was greater than that in the case of rye straw. 4. Contents of amino acids other than arginine, histidine and tyrosine were increased in the case of wheat straw, while only the contents of lysine, phenylalanine, tyrosine and methionine were observed to increase in the case of rye straw. 5. Exceptionally high contents of phenylalanine and tyrosine were measured in the hydrolysate from rye straw taken at the end of experimental period. 6. No amount of arginine was detected in any hydrolysate of humic acids from decomposed plant residues.
