4-Substituted-kynurenic Acid Derivatives:A Novel Class of NMDA Receptor Glycine Site Antagonists

  • A series of 4-substituted-kynurenic acid derivatives possessing several different substituents at C4-position which are consisted of both a flexible propyloxy chain and an adjunct several type of carbonyl groups has been synthesized and evaluated for their in vitro antagonist activity at the glycine site on the NMDA receptor. Of them, N-benzoylthiourea 15c and N-phenylthiourea 15a were found to have the best in vitro binding affinity with $IC_{50}$ of 3.95 and $6.04{\mu}M$, respectively. On the other hand, in compounds 12a-c and 13 the displacement of a thiourea group to an amide or a carbamate caused a significant decrease of the in vitro binding affinity. In the SAR study of the 4-substituted kynurenic acid derivatives, it was realized that the terminal substitution pattern on a flexible C4-propyloxy chain of kynurenic acid nucleus significantly influences on the binding affinity for glycine site; the binding affinity to the NMDA receptor might be increased by the introduction of a suitable electron rich substituent at C4 of kynurenic acid nucleus.

$^{13}C$nmr Studies on the Interaction of Lithium Salts to Formamide, Acetamide and Propionamide Systems (Formamide, Acetamide 및 Propionamide계에 대한 리튬염의 상호작용에 관한 $^{13}C$ nmr 연구)

  • $^{13}C$resonances of carbonyl and various alkyl groups in amides are found to shift down-field on the interaction with lithium salts and it is shown that lithium ion binds directly to the carbonyl group in amides. The magnitudes of the $^{13}C$ chemical shifts of various amides depend not only on the size of alkyl groups in amides but also on the interaction with anion. The change of $^{13}C$chemical shift of amide in LiCl is smaller than that in$LiClO_4$ due to the difference of the charge density of the anion.

Synthesis, Characterization, and Electrochemical Behavior of Viologen-Functionalized Poly(Amidoamine) Dendrimers

  • Amineterminated, ethylenediamine core polyamidoamine starburst dendrimers of generation 2 (G2), generation 4 (G4) and generation 6 (G6) have been successfully surface-modified via an amide coupling reaction with 4-ethyl, 4'-(3-propionic) bipyridinium cation and the electrochemical behavior of the resulting dendrimers were investigated in aqueous potassium chloride electrolyte solutions. The 1-[3-(dimethylamino)propyl]-3-ethylcarbodiimide hydrochloride/N-hydroxysuccinimide-mediated reaction resulted in 25-39% end-group functionalization. The water-soluble 4-ethyl, 4'-(3-propylamide) bipyridinium dibromide dendrimers (G2-V2+, G4-V2+ and G6-V2+) were characterized by $^1H$ NMR and UV-Vis spectroscopic methods. The cyclic voltammetric and chronoamperometric experiments were performed to determine the diffusion coefficient and the number of electrons transferred in the process of the first reduction of the viologen-functionalized dendrimers. Adsorption of viologen-functionalized dendrimers at electrode surface was evidenced in the voltammograms. Experimentally determined diffusion coefficients were in good agreement with the values expected from the Stokes-Einstein relation, while the number of electrons transferred concurred with the extent of functionalization determined by $^1H$ NMR and UV-Vis spectra.

Docking and Quantitative Structure Activity Relationship studies of Acyl Guanidines as β-Secretase (BACE1) Inhibitor

  • ${\beta}$-Secretase (beta-amyloid converting enzyme 1 [BACE1]) is involved in the first and rate-limiting step of ${\beta}$-amyloid ($A{\beta}$) peptides production, which leads to the pathogenesis of Alzheimer's disease(AD). Therefore, inhibition of BACE1 activity has become an efficient approach for the treatment of AD. Ligand-based and docking-based 3D-quantitative structure-activity relationship (3D-QSAR) studies of acyl guanidine analogues were performed with comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) to obtain insights for designing novel potent BACE1 inhibitors. We obtained highly reliable and predictive CoMSIA models with a cross-validated $q^2$ value of 0.725 and a predictive coefficient $r{^2}_{pred}$ value of 0.956. CoMSIA contour maps showed the structural requirements for potent activity. 3D-QSAR analysis suggested that an acyl guanidine and an amide group in the $R_6$ substituent would be important moieties for potent activity. Moreover, the introduction of small hydrophobic groups in the phenyl ring and hydrogen bond donor groups in 3,5-dichlorophenyl ring could increase biological activity.

Surface Modification of Polypropylene Fiber by Plasma Discharge (방전처리에 의한 Polypropylene섬유의 표면개질)

  • Polypropylene(PP) films were treated with plasma glow discharge to produce peroxy radicals on the surfaces. The peroxy radicals formed on the PP film surfaces were subsequently used for the graft polymerization of acrylic acid and acrylamide in an aqueous solution by heating, respectively. Introduction of acrylic acid and acrylamide on the PP film could be confirmed by the observation of carbonyl and primary amine absorptions based on carboxylic acid and amide, respectively. And introduction of functional group could be confirmed by weight analysis and ESCA. The water contact angle(90$^{\circ}$) of PP film was constant, irrespective of elapsed time, while plasma-treated and functional monomer-grafted PP films were slowly increased with elapsed time, showing the rearrangement of surface polar groups in air condition. The water contact angle$(90^\circ)$ of PP film was decreased by the plasma treatment$(56^\circ)$ and further decreased by the grafting of acrylic acid$(34^\circ)$ and acrylamide$(37^\circ)$, indicating increased hydrophilicity of the modified surfaces. The water contact angle of plasma-treated PP film increased a little as time elapsing. The half-life periods of surface voltage on acrylic acid-(31sec) and acrylamide-grafted PP(42sec) were significantly decreased when compared to those on PP(950sec) and plasma-treated PP film(241sec). In the experiments using acid, basic and disperse dyes, absorbance and $\Delta{E}$ values of functional monomer-grafted PP films were significantly increased than that of oxygen plasma-treated one.

The new Copolymers which contain Flexible Spacers in the Main Chain. (주 사슬에 유연 격자를 포함한 새로운 공중합체)

  • The new copolymers which contain flexible spacers in main chain were prepared and their properties were studied. the copolymers were synthesized in the melt polymerization melted from diacetoxynaphthalene isomers, ${\alpha},{\omega}bis(4-carboxyphenoxy)$ alkanes and p-acetoxy benzoic acid in the molar ratio of 1:1:2 and characterized for the effects of structure of naphthalenediol isomer moieties and the spacers on thermal crystalline and liquid crystalline properties of the resulting polymers. Diacetoxy naphthalene isomers were used 1. 5-,1. 6-,2. 6-and 2. 7-disubstituted ones and the spacers was either tetramethylene or decamethylene. The polymer was characterized by elemental and spectropic analysis, differential scanning calorimetry (DSC) on a polaring microscope, wide-angle X-ray diffractometry and thermogravimetry (TGA). There glass transition temperatures (Tg) and melting temperature (Tm) on the structure of the naphthalenediol isomer moieties and the length of the spacers. The presence of spacers was found to the helpful in crystallization of 1. 6-naphthalenediol copolyesters. The presence of amide group in the backbone chain of polymer was found to increase transition temperature, crystallization, thermal stability of polymers but to decreas solubility.

Effect of Alkali Surface Modification on Adhesion Strength between Electroless-Plated Cu and Polyimide Films (알카리 표면개질 처리가 무전해 구리 도금피막과 폴리이미드 필름의 접합력에 미치는 효과)

  • The effects of the alkali surface modification process on the adhesion strength between electroless-plated Cu and polyimide films were investigated. The polyimide surfaces were effectively modified by alkali surface treatments from the hydrophobic to the hydrophilic states, and it was confirmed by the results of the contact angle measurement. The surface roughness increased by alkali surface treatments and the adhesion strength was proportional to the surface roughness. The adhesion strength of Cu/polyimide interface treated by KOH + EDA (Ethylenediamine) was 874 gf/cm which is better than that treated by KOH and KOH + $KMnO_4$. The results of XPS spectra revealed that the alkali treatment formed oxygen functional groups such as carboxyl and amide groups on the polyimide films which is closely related to the interfacial bonding mechanism between electroless-plated Cu and polyimide films. It could be suggested that the species and contents of functional group on polyimide films, surface roughness and contact angle were related with the adhesion strength of Cu/polyimide in combination.

Adamantyl-based N-arylamide as a Novel Series of Androgen Receptor Antagonists (아다만틸을 기반한 N-아릴아미드 신규 안드로겐 수용체 길항제)

  • A novel series of adamantyl derivatives of N-aryl amides as androgen receptor antagonists were synthesized and their anti-androgenic activities were evaluated. The N-aryl amides containing adamantyl derivatives had more activity than those lacking adamantyl substitutions. The synergistic effect of bulkiness of the adamantyl group and modulation of electron density on the aromatic ring by pendant groups was crucial to the potent antagonism. Molecular modeling studies were performed to elucidate the interactions between ligands and receptors.

ENHANCED ADHESION STRENGTH OF Cu/polyimide AND Cu/Al/polyimide BY ION BEAM MIXING

  • the Cu/polyimide system is known to be the best candidate for a multilevel interconnection system due to the low resistance of Cu and to the low dielectric constant of polyimide respectively. Ion beam mixing of Cu(40nm)/polyimide was carried out at room temperature with 80 keV Ar+ and N2+ form $1.5\times$1015 to 15$\times$1015 ions/cm2. The quantitative adhesion strength was measured by a standard scratch test. X-ray photoelectron spectroscopy and x-ray emission spectrocopy are employed to investigate the chemical bonds and the interlayer compound formation of the films Cu/Al/polyimide showed more adhesion strength than Cu/polyimide after ion beam mixing and N2+ ions are more effective in the adhesion enhancement than Ar+ with the same sample geometry. The XES results shows the formation of interlayer compound of CuAl2O4 which can reflect more adhesive Cu/Al/polyimide which has not been reported previously. The latter results is understood by the fact that N2+ ions produce more pyridinelike moiety, amide group and tertiary amine moiety whcih are known as adhesion promotors.

Establishment of discrimination system using multivariate analysis of FT-IR spectroscopy data from different species of artichoke (Cynara cardunculus var. scolymus L.) (FT-IR 스펙트럼 데이터 기반 다변량통계분석기법을 이용한 아티초크의 대사체 수준 품종 분류)

  • To determine whether FT-IR spectral analysis based on multivariate analysis for whole cell extracts can be used to discriminate between artichoke (Cynara cardunculus var. scolymus L.) plants at the metabolic level, leaves of ten artichoke plants were subjected to Fourier transform infrared(FT-IR) spectroscopy. FT-IR spectral data from leaves were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). FT-IR spectra confirmed typical spectral differences between the frequency regions of 1,700-1,500, 1,500-1,300 and $1,100-950cm^{-1}$, respectively. These spectral regions reflect the quantitative and qualitative variations of amide I, II from amino acids and proteins ($1,700-1,500cm^{-1}$), phosphodiester groups from nucleic acid and phospholipid ($1,500-1,300cm^{-1}$) and carbohydrate compounds ($1,100-950cm^{-1}$). PCA revealed separate clusters that corresponded to their species relationship. Thus, PCA could be used to distinguish between artichoke species with different metabolite contents. PLS-DA showed similar species classification of artichoke. Furthermore these metabolic discrimination systems could be used for the rapid selection and classification of useful artichoke cultivars.
