In Vitro법에 의한 식이섬유의 포도당 및 담즙산 흡수 지연 효과 (Retarding Effect of Dietary Fibers on the Glucose and Bile Acid Movement Across a Dialysis Membrane in Vitro)

  • In order to anticipate the physiological function of dietary fibers, glucose and bile acid retarding effects were experimented by using in vitro methods based on dialysis for commercial fibers and dietary fiber residue of food samples. The glucose retarding effect in commercial fibers increased in the order of alginic acid, guar gum, CM-cellulose, citrus pectin > apple pectin > $\alpha$-cellulose and the effect in food fiber residues increased in the order of sea mustard > Korean cabbage, apple > rice bran, barley, soybean, and tangerine. The bile acid retarding effect in commercial fibers increased in the order of citrus pectin, guar gum > CM-cellulose, alginic acid > apple pectin > $\alpha$-cellulose and the effect in food fiber residues increased in the order of barley, rice bran > sea mustard > tangerine > Korean cabbage, soybean > apple. The higher the retarding effect of glucose movement through the dialysis membrane, the more effective the control of the human blood glucose level. As the retarding effect of bile acid movement across the dialysis membrane increased, the human serum cholesterol level correspondingly reduced. Consequently these in vitro methods can be used as a preceding test before undertaking animal and human experiments to predict the physioloical effects of fiber residues from diverse food samples as well as commercially refined fibers.

액화셀룰로오스의 제조 및 GC-MS에 의한 그 성분 분석 (Preparation of Liquefied Cellulose and Analysis of Its Components by GC-MS Spectrometry)

  • α-Cellulose를 페놀 및 황산과 1: 6.2 : 0.05(g/g㎖)의 비율로 혼합하여 질소기류 하에서 180℃에서 60분의 액화 처리한 후 변호된 셀룰로오스의 성분분석을 GC-MS에 의하여 실시한 결과는 다음과 같다. 셀룰로오스의 액화율이 98.8%로 높은 것으로 보아 상당히 저분자화 된 것을 알 수 있었다. 셀룰로오스 액화물 중의 13.6%인 페놀함유량은 용매로 투입된 페놀의 양보다 매우 적은데 이는 액화 생성된 화합물의 구조 중에 hydroxyphenly기나 phenyl기가 존재하는 것에서 셀룰로오스는 액화시 산 촉매에 의하여 C-C, C-O 결합의 일부 개열함과 동시에 페놀과 반응하기 때문이라 판단된다. 약 54%인 12개 셀룰로오스 액화물의 구조를 분석하였으며, 그 중 양적으로 많은 양이 검출된 물질은 phenol핵이 주종을 이루는 2,2\`-methylenebisphenol, 4,4\`-methylene-bisphenol, 3-methyl-4-hydroxyphenyl-2\`-hydroxyphenylmethane, 1-methoxy-4-(2-phenylethenyl) benzene, (e)-2,4\` dihydroxystilbene, 1-phenyl-1-(4\`-hydroxy) phenyl methanol, p-isopropylphenol 등이 검출되었다. 황산 촉매에 의하여 셀룰로오스는 액화과정에서 탈수반응 후 열분해 되어 셀룰로오스링이 끊어져 생성된 중간체들이 용매인 phenol과 치넌자 치환반응을 일으켜 phenol 화합물들을 많이 생성한다고 판단된다.

알로에 베라 유래 식이섬유의 In Vitro 및 In Vivo 생리기능 특성 (In Vitro and In Vivo Physiological Characteristics of Dietary Fiber from By-product of Aloe vera Gel Processing)

  • A fiber fraction (Aloe cellulose), the by-product obtained from Aloe vera gel processing was freeze dried and investigated for in vitro glucose/ bile acid retarding effects of powdered sample (100 mesh) comparing with commercial $\alpha$-cellulose as a reference sample. We also examined the effectiveness of physiological functionality such as the antiobesity and anti-constipation on Sprague-Dawley (SD) rat. The Aloe cellulose powders during in vitro dialysis experiment for 2 hours exhibited the glucose and bile acid retarding index of 20.32-35.2% and 53.13-28.30%, respectively. Especially, freeze dried aloe cellulose showed the 2.5 and 1.2-6 times higher effect on in vitro glucose and bile acid retardation than those of $\alpha$-cellulose. These relatively good retarding effects on glucose and bile acid diffusion suggest a potential of preventing from diabetes and arteriosclerosis of some extent. Also, the results from animal experiments on SD rats fed a high-fat diet for 4 weeks suggested that Aloe cellulose might be used as a novel dietary fiber showing an effective anti-obesity and anti-constipation effect.

  • To observe the changes of serum proteins according to the process of Babesia gibsoni(B gibsoni) infection, the babesia protozoa($10^8/kg$) were inoculated into the cephalic vein of healthy dogs. The serum proteins of experimentally infected dogs were separated by using cellulose acetate electrophoresis. The results obtained were as follows; 1. Cellulose acetate electrophoresis was fractionated to total 6 of bands such as, albumin, ${\alpha}_1$, ${\alpha}_2$, ${\beta}_1$, ${\beta}_2$ and $\gamma$-globulin. 2. The concentration of total protein was shown a decreasing tendency after B gibsoni infection. Albumin and A/G ratio were lowered through all periods of the infection, but they were not significant changes. 3. The level of ${\alpha}_1$-globulin was significantly(p<0.05) incresed in early stage of the infection. 4. The levels of ${\alpha}_2$ and total $\alpha$-globulin were shown highly significant decreases (p<0.01) through all periods of the infection. 5. The levels of ${\beta}_1$ and total $\beta$-globulin had highly significant changes (p<0.01) that was increased in early stage of infection and decreased later. 6. The level of $\gamma$-globulin was seen to be constantly increased through all periods of infection. It was a highly significant change (p<0.01). 7. Plasma protein: fibrinogen (PP:F) ratio was shown a temporally significant increase (p<0.05) following the decrease in early infection.

지표공기중(地表空氣中)의 ${\alpha}$ 방사능측정(放射能測定) 및 해석(解析) (Measurement and Assessment of Alpha Activity in Ground Air)

  • CA 80-15 및 LR 115-1 cellulose nitrate 비적검출기를 사용하여 지표공기중의 ${\alpha}$방사능을 측정 해석하였는바 이 검출기의 검출효율은 $0.1{\mu}Ci$$^{241}Am\;{\alpha}$선을 사용하여 결정하였다. 지표에서 방출되는 라돈과 그 자핵종들에 의한 ${\alpha}$방사능을 검출하기 위하여 두가지의 라돈컵을 사용하였으며 이들을 각각 지름이 15cm, 깊이가 45cm인 이웃한 두개의 땅구덩이를 만들어 그 안에 설치하여 일정기간 ${\alpha}$방사능에 노출하였다. 두 라돈컵중 하나는 구덩이 지표면에 거꾸로 밀착시켜 밀폐공간이 되게 하고 다른 하나는 바닥에서 일정한 거리를 띄우고 컵 윗면에 구멍을 뚫어 공기의 소통이 가능하도록 하여 그 검출결과를 비교 검토하였다. ${\alpha}$입자에 피폭된 비적검출기는 부식처리한 후 현미경으로 판독하고 통계분석 하였으며 그 결과를 지표공기 단위부피당 ${\alpha}$방사능농도로 환산 평가하였다.

건성안검사용 셀룰로즈 스트립 개발 (Development of Cellulose Strip for Dry Eye Inspection)

  • 건성안 진단에 있어서 가장 많이 사용되는 방법 중 하나이나, 부정확한 결과로 신뢰도가 떨어지는 쉬르머 검사법을 대체할 수 있는 눈물양 측정 도구를 개발하고자 하였다. 이를 위해 다양한 흡수제에 대한 흡수성 시험을 통하여 알파 셀룰로즈 펄프를 흡수제로 선택하였다. 2차례에 걸쳐 시제품을 제작하여 평가하였으며, 최종적으로 알파 셀룰로즈 펄프를 폴리우레탄에 접착한 스트립 형태의 눈물양 측정 도구를 제작하여 사용성 평가를 실시하였다. 그 결과 해외에서 최근에 개발된 눈물양 측정 도구인 Strip Meniscometry와 유의한 상관성을 갖는 것을 확인할 수 있었으며, 측정시간 및 측정의 정확성 측면에서 쉬르머 검사법의 대체 검사법으로 유효한 것으로 판단되었다.

Effects of electron beam treatment on cotton linter for the preparation of nanofibrillated cellulose

  • Nanofibrillated cellulose (NFC) was prepared from cotton linter after electron beam irradiation to investigate its effects on the manufacturing efficiency of the NFC preparation and the property changes by the treatment. Mechanical device (Super Masscolloider) was used to prepare the NFC and its passing frequency for each NFC preparation was recorded. More electron beam irradiation resulted in less passing frequency. Alpha cellulose content, molecular weight, crystallinity index, and thermal decomposition behavior of each treatment were lowered by electron beam treatment (10 and 100 kGy) and grinding process. NFC films were prepared to investigate their mechanical properties. There were little changes in tensile properties of the NFC films.

Overproduction of Cellulose in Acetobacter xylinum KCCM 10100 Defective in GDP-Mannosyltransferase

  • GDP-mannosyltransferase (GMT) is an enzyme responsible for the addition of a mannose to glucose ($\alpha$[1$\rightarrow$3]) during biosynthesis of the water-soluble branched polysaccharide acetan in Acefobacter species. In an effort to obtain a cellulose-overproducing bacterium, a mutant defective in GMT of Acetobacter xylinum KCCM 10100 was constructed by single crossover homologous recombination using part of the aceA gene encoding GMT amplified by polymerase chain reaction. The GMT-disrupted mutant produced 23% more cellulose, but 16% less water-soluble polysaccharide than those of the wild-type strain. Analysis of the sugar composition by gel permeation chromatography revealed that water-soluble polysaccharides produced by the GMT-defective mutant contained no mannose molecule.

  • This research was studied dissolving pulp for rayon production, which is not KP, SP methods but steam explosion and oxygen alkali treatment. The variation of characterization in wood cellulose after steam explosion and oxygen alkali treatment this follows. In the oxygen alkali treatment of exploded pine wood, yield of dissolving pulp was lower than steam-exploded oak wood, and we have obtained dissolving pulp that high purity cellulose, but degree of polymerization was inclined to decrease less than 200. In the case of steam-exploded oak wood, degree of polymerization was higher than steam-exploded pine wood but purity of cellulose was lower than its.

  • Various grades of hydroxyethyl cellulose (HEC, MS 0.65-3.20) were prepared throngh reaction of the knit-cotton-waste with ethylene oxide(EO). The knit-cotton-waste was composed of 98% of $\alpha$-cellulose and 2% of other components, and the cellulose was highly pure. The molar ratio of EO to knit-cotton-waste and that of NaOH to knit-cotton-waste, and the agitation speed were the important factors determining the molar substitution(MS) during the preparation of HEC. The MS of HEC was remarkably increased with increasing molar ratio of EO' to knit-cotton-waste. When the molar ratio of EO to knit-cotton-waste was 3.5, that of NaOH to knit-cotton-waste was 1.25, and agitation speed was 450rpm, it was possible to prepare HEC of MS 2.5. The structure and crystallinity of HECs prepared were determined by FT-IR and X-ray diffraction.
