극세 공극 직물 침구가 먼지 양과 집먼지진드기 Allergen에 미치는 효과에 대한 연구 (A Study for the Effects of Fine Gap Fabric Bedding on the Levels of Dust and House Dust Mite Allergens)

  • 집먼지진드기에 의한 호흡기 알레르기 질환의 예방 및 치료를 위해 고려할 수 있는 가장 효율적인 방법 가운데 하나는, 항원이 통과할 수 없는 특수 재질의 커버로 직접 침구류를 씌우는 것이다. 따라서 본 연구에서는 특수 커버 사용 후 먼지 양의 감소와 집먼지진드기 allergen 노출에의 감소 정도를 측정하여 특수 커버의 효과를 검증하였다. 본 연구 결과 우리나라의 일반적인 생활환경에서는 특수 커버의 사용이 집먼지진드기 allergen에 대한 노출을 완전히 막아 주지는 못하였다. 그러나 노출되는 양은 감소시킬 수 있었으며 먼지 양이 감소하는 효과도 있었다.

돼지고기 중 알레르기 유발성분의 동정 (Identification of Allergens in Pork Meat)

  • The pork meat has been reported as one of the food occurring allergic reactions predominantly to korean. To identify the potential food allergens in pork meat, sera were collected from 25 allergic patients to the pork meat and 10 allergic patients not to pork meat as well as 5 normal subjects after skin prick test and open food challenge test. Crude extracts were prepared by blending raw pork meat in phosphate buffered saline (pH 7.0) and the heat treatment on crude extracts was carried to characterize sensibility of the allergens to heat. ELISA was performed to determine specific IgE antibody levels of allergic patients to pork meat, and resulted in twofold higher mean value than that of tolerated patients. Extracted proteins from pork meat was separated with SDS-PAGE followed by immunoblotting using sera from pork sensitive patients and control subjects, respectively. The IgE binding response to pork meat by immunobots correlated with quantitative specific IgE value of each person. Immunoblots showed four prominent IgE-binding bands (66, 60, 50, 44 kDa) in crude extract, but two bands of those (60, 44 kDa) were heat-labile. These results suggest that most prominent allergens from pork meat are four components(66, 60, 50, 44 kDa) in korean and the heat treatment on allergen is additional parameter to characterize allergen.

집먼지진드기에 대한 침엽수 정유의 기피효과 (Repellent effect of essential oils from coniferous trees against the house dust mites (Dermatophagoides farinae and D. pteronyssinus))

  • The avoidance of the allergen of the house dust mite is one of the challenges to reduce and treat the symptoms of allergic diseases. Accordingly, various acaricidal agents are being used to kill the mite, but just killing it leaves the remains of it, which still act as the allergen. Therefore expelling the mite is thought as best policy to avoid the mite allergen. For this, some materials have been applied to repellent agents against the mite. Among them, a material with natural origin, known as the phytoncide, is being used for its repellent activity, as well as for its benefits for health. In this experiment, essential oils extracted from Korean white pine (Pinus koreaiensis S. et Z.) and hinoki cypress (Chamaecyparis obtusa), which are widely used as the source of phytoncide products, are studied for demonstration of the repellent effect against the house dust mites, Dermatophagoides farinae and D. pteronyssinus. Direct contact method was used to evaluate the repellent effect (%). And the results suggest the oils have a significant effect enough to be used as a source of repellent agent. For the repellent effect, the most effective concentration was $0.5{\mu}l/cm^2$ at 45 minutes both in the white pine and the hinoki oil.

Effect of Zedoariae rhizoma on Bronchial Inflammation and Allergic Asthma in Mice

  • There are detailed descriptions of the clinical experiences and prescriptions of asthma in traditional Korean medicine. Zedoariae rhizoma is one of the Korean herbal medicines used to treat bronchial asthma and allergic rhinitis for centuries. However, the therapeutic mechanisms of this medication are still far from clear, In this study, a house-dust-mite (Dermatophagoides pteronyssinus [Der p])-sensitized murine model of asthma was used to evaluate the immunomodulatory effect of Zedoariae rhizoma on the allergen-induced airway inflammation in asthma. Three different protocols were designed to evaluate the treatment and/or long-term prophylacitic effect of Zedoariae rhizoma in Der p-sensitized mice. Cellular infiltration and T-cell subsets in the bronchoalveolar lavage fluid (BALF)of allergen-challenged mice were analyzed. Intrapulmonary lymphocytes were also isolated to evaluate their response to allergen stimulation. When Zedoariae rhizoma was administered to the sensitized mice before AC (groups A and C), it suppressed airway inflammation by decreasing the number of total cells and eosinophil infiltration in the BALF, and downregulated the allergen- or mitogen-induced intrapulmonary lymphocyte response of sensitized mice as compared to those of controls. This immunomodulatory effect of Zedoariae rhizoma may be exerted through the regulation of T-cell subsets by elevation or activation of the CD8+ and double-negative T-cell population in the lung. However, the administration of Zedoariae rhizoma to sensitized mice 24 h after AC (group B) did not have the same inhibitory effect on the airway inflammation as Zedoariae rhizoma given before AC. Thus, the administration of Zedoariae rhizoma before AC has the immunomodulatory effect of reducing bronchial inflammation in the allergen-sensitized mice. On the other hand, to determine the potentiality of prophylactic and/or therapeutic approaches using a traditional herbal medicine, Zedoariae rhizoma, for the control of allergic disease, we examined the effects of oral administration of Zedoariae rhizoma on a murine model of asthma allergic responses. When oral administration of Zedoariae rhizoma was begun at the induction phase immediately after OVA sensitization, eosinophilia and Th2-type cytokine production in the airway were reduced in OVA-sensitized mice following OVA inhalation. These results suggest that the oral administration of Zedoariae rhizoma dichotomously modulates allergic inflammation in murine model for asthma, thus offering a different approach for the treatment of allergic disorders.

  • To examine pollen preparations which are being sold as a health food, one of these pollen tablets and one of the capsules were selected and investigated by microscopy. It was shown that the pollen coat which can act as a potent allergen was not removed and that the tablets contained other impurities. The results suggest that they are not safe as a food and that they may cause systemic allergy to certain persons. On the contrary, the pollens of the capsule preparation did not have the pollen coat and, therefore, have a less risk of causing allergy.

  • Histone deacetylase (HDAC) inhibition has been demonstrated to change the expressions of a restricted set of cellular genes, T cells have an essential role in the pathogenesis of allergen-induced airway inflammation [1]. In recent studies, it has been demonstrated that treatment with HDAC inhibitors induces a T cell-suppressive effect [2]. The purpose of this study was to determine whether treatment with trichostatin A (TSA), a representative HDAC inhibitor, would reduce the allergen-induced airway inflammation in a mouse asthma model.   • Using genetic engineering, the Vitreoscilla (bacterial) hemoglobin gene (vgb) was integrated stably into the chromosomes of and Burkholderia cepacia. Similar to previous results, the wild type VHb improved growth for Burkholderia cepacia and degradation of benzoic acid under both normal and low aeration conditions. The stable expression of VHb enhanced these parameters. The results demonstrate the possibility that the positive effects provided by VHb may be augmented by protein engineering.
