호 알칼리성 Alcaligenes sp.가 생산하는 ${\gamma}$-Polyglutamic Acid의 용액 특성 (Solution Properties of ${\gamma}$-Polyglutamic Acid Produced by Alkalophilic Alcaligenes sp.)

  • The high viscous ${\gamma}$-polyglutamic acid(${\gamma}$-PGA) from alkalophilic Alcaligenes sp. was purified and its solution property was investigated. The intrinsic viscosities for Na+ form and H+ form of ${\gamma}$-PGA were 31.1 and 0.38d$\ell$/g, respectively. The viscosity of H+-PGA was not influenced by pH or salts while that of Na+-PGA was influenced. The intrinsic viscosity of Na+-PGA solution decreased remarkably at the alkaline or acidic pH and showed the sharp decrease when salts were added. ${\gamma}$-PGA exhibited the property of the polyelectrolyte showing the .sharp decrease of intrinsic viscosity by the addition of NaCl, and intrinsic viscosity of dilute solution with the low concentration of NaCl was exponentially dependent on temperature and its temperature dependency increased with increasing NaCl concentration. The chain stiffness, coil overlap parameter and critical concentration of Na+-PGA were 0.08, 5.25 and 0.07g/d$\ell$, respectively.

Cloning of the gense coding for extracellular proteases from alkalophilic xanthomonas SP. JK311

  • The alkalophilic bacterium, Xanthomonas sp. JK311, producing extracellular proteases, was isolated from soil. Xanthomonas sp. JK311 produced five extracellular proteases that are all metalloproteases. Four of them were resistant against 1% SDS. Chromosomal DNA of the Xanthomonas sp. JK311 was digested with BamHI and cloned into PUC19. Among E. coli strain HB101 transformants, a clone secreting the proteases was screened through halo formation on skim-milk agar plate and by Southern blot analysis. It had the recombinant plasmid pXEP-1 containing the 7.5 kb-BamHI DNA fragment and produced three extacellular proteases. Their protease properties corresponded to those of Xanthomonas sp. JK311.

  • Some functional and rheological properties of the ${\gamma}$-polyglutamic acid(${\gamma}$-PGA) produced from alkalophilic Alcaligenes sp. were investigated. Viscosity synergism with thickening agents, capacities for gelling, entrapig of heavy metals, and flocculability of ${\gamma}$-PGA were not observed, but the relatively good compatability with various polyvalent metallic ins, excellent absorption ability and spinability were observed. The ${\gamma}$-PGA solution showed non-Newtonian flow behavior and exhibited pseudoplastic property with a yield stress at above 1% concentration. The values of flow index for 1% solution were in the range of 0.41∼0.75 showing shear rate dependency and the value of yield stress was 2.28 Pa. The value of consistency index was 0.868 Pa$.$Sn and was exponentially dependent on concentration.

  • The study was performed to investigated the excellent microbial anticaries substance which is more effective than the chlorohexidine in the dental caries treatment. For the screening of alkaliphilic microorganism, more than 1200 bacterial strains were isolated from sea soil sample. A typ-ical strain which produced the most excellent antimicrobial substance was selected. The strain was identified novel alkalophilic Bacillus sp. through the results of morphological, biochemical and chemotaxonomical characteristics and 16S rDNA sequencing and designated as Bacillus alkalophilshaggy JY-827.

Calcite Production by Bacillus amyloliquefaciens CMB01

  • The bio-mediated production of calcite crystals by calcinogenic bacteria has great applicable value for the restoration of deteriorated calcareous monuments, because of its high purity and coherency. An investigation of the conditions for calcite production by an alkalophilic Bacillus amyloliquefaciens CMB01 strain was made. Optimal calcite precipitation occurred when the bacterium was cultured at pH 8.0 and 30$^{\circ}C$, and in B4 medium that consisted of 0.4% yeast extract, 0.5% glucose, and 1.5% calcium acetate. Calcium ion of the bacterially induced calcite was analyzed by an inductively coupled plasma (ICP) spectrophotometer. Optical and scanning electron microscopy (SEM) of the calcite revealed a typical rombohedral polycrystalline structure.

Isolation of Novel Alkalophilic Bacillus alcalophilus subsp. YB380 and the Characteristics of Its Yeast Cell Wall Hydrolase

  • An alkalophilic mi.croorganism (strain YB380), which produces yeast cell wall hydrolase extracellulary, was isolated from Korean soil. The rod-shaped cells were 0.3~0.4 by 2~4${\mu}{\textrm}{m}$ long, motile, aerobic, gram-positive, and spore-forming. The color of the colony was light yellow. The temperature range for growth at pH 9.0 was 25 to $45{\circ}C, with optimum growth at $35{\circ}C. The pH range for growth at $35{\circ}C was 8 to 11 with an optimum pH of 9.0. Therefore, the strain YB380 is an obligate alkalophile. The 16S rRNA of strain YB380 has a 99% sequence similarity with that of Bacillus alcalophilus. On the basis of physiological properties, cell wall fatty acid composition, and phylogenetic analysis, we propose that the isolated strain is Bacillus alcalophilus. The yeast cell wall hydrolase from Bacillus alcalophilus subsp. YB380 has been purified and partially characterized. The molecular weight was estimated to be 27,000 daltons with an optimum temperature and pH of $60{\circ}C and 9.0, respectively. The N-terminal amino acid sequence of the enzyme was analyzed as Gln- Thr- Val- Pro- Trp- Gly- Ile- Asn- Arg- Val.

  • 토양으로부터 분리한 호알카리성 세균 Bacillus sp. No. 1911 파쇄하여 유안투석, DEAE-cellulose column chromatography, Sephadex G-150 gel filtration으로 포도당 이성화효소를 정제하여 효소적 특성을 조사하였다. 이 효소는 Sephadex G-200 gel filtration에서 분자량은 170,000으로 측정되었고 SDS-polyacrylamide 전기영동에서 43,000의 단일 band를 얻어, 이 효소는 4개의 동일한 subunit로 구성되었음을 나타내었다. 이 효소는 pH7.5 및 $65^{\circ}C$에서 최적활성을 나타내었으며 pH7.5에서 7$0^{\circ}C$ 까지 안정하였고 Co$^{++}$ 존재하에 6$0^{\circ}C$에서 30분간 가열하였을 때 pH 6-9까지는 비교적 안정되었다.

  • 일반 토양과 부엽토, 퇴비로부터 alkaline cellulase 생성이 우수한 균주를 분리한 후 형태적, 배양학적 및 생화학적 동정을 실시한 결과 호알칼리성 Bacillus sp. HSH-810 균주인 것으로 판명되었다. 분리균 Bacillus sp. HSH-810의 생육과 효소 활성은 $30^{\circ}C$와 pH 10.0에서 가장 높은 것으로 나타났다. 본 균주는 배지 중에 탄소원과 질소원, 무기염으로 1.0%의 CMC와 0.5%의 peptone, 0.02%의 $CaC1_2$, $CoC1_2$를 사용하였을 경우 최대의 alkaline cellulase생산성을 나타내었다. 효소의 최적 활성 pH와 온도는 각각 10.5와 $50^{\circ}C$였다. 이 효소는 pH 6.0-13.0과 $50^{\circ}C$의 온도에서 매우 안정하였다. 효소액에 계면활성제를 첨가하여 효소 활성을 측정한 결과, 효소액은 sodium-$\alpha$-olefin sulfonate (AOS)와 sodium dodecyl sulfonate (SDS), Tween 20, Tween 80에 대하여 안정성 이 높은 특징을 보였으나 0.1%의 linear alkyl-benzene sulfonate (LAS)를 첨가한 경우에는 효소 활성이 심하게 저해되었다.

호알칼리성 Bacillus sp.의 Lactose 투과와 이용성 (Transport and Utilization of Lactose by Alkalophilic Bacillus sp.)

  • To study the reduced growth and synthesis, proeviously reported, of ${\beta}$-galactosidase of alkalophilic Bacillus sp. YS-309 at the higher lactose concentration of 0.5% (w/v) in the medium, lactose transport and utilization were examined. The results showed that lactose transport was influenced by the addition of four kinds of antibiotics, and tetracycline stimulated most but not valinomycin. PEP-potentials of the cells grown on lactose was estimated lower than the cells on glucose and on galactose. Thus, the transport of lactose was independent of intracellular PEP and phosphorylation reactions, and was thought to be uptaked directly or oxidized in part in the transport process. In the other hand, once lactose was uptaked into the cells, it was hydrolyzed by ${\beta}$-glactosidase to glucose and galactose. The former was metabolized fast but the latter was accumulated. Galactose and lactose were not utilized until glucose was mostly depleted in the medium. The ${\beta}$-galactosidase synthesis decreased in the presence of glucose over 0.2% and galactose over 0.05 to 0.1%, respectively. In conclusion, it was considered for glucose as a repressor and galactose as a inducer for ${\beta}$-galactosidase synthesis even though the mechanisms were not elucidated. Catabolite repression of glucose on the enzyme synthesis was not relieved by the addition of exogeneous cAMP.

  • 호알칼리성 Bacillus sp. YC-335가 생산하는 CGTase의 효소학적 특성 및 작용반응을 살펴보았다. ${\alpha}-CD,\;{\beta}-CD$${\gamma}-CD$로부터 glucosyl residues를 설탕으로 전이시키는 반응에 대한 효소의 최대 반응속도, Vmax 값은 각각 $16.13,\;21.8,\;9.8{\mu}moles glucose/min/mg\;protein$이었으며 Km 값은 각각 1.68, 0.33, 0.37 mM이었다. 효소의 전분 가수분해활성은 여러 당류에 의해 촉진되었으며 특히 전분 가수분해 산물인 maltose와 glucose에 의한 효과가 가장 좋았다. 이 효소는 ${\beta}CD$에 의해 효소의 전분 분해활성이 저해되었으며 비경쟁적 저해형식을 보였다. 또한 전분으로부터 효소작용에 의해 생성된 산물을 총당량법 및 HPLC 분석을 통해 조사한 결과 이 효소는 cyclization 작용 뿐만 아니라 transglycosylation 작용과 disproportionation 작용을 가지는 것으로 확인하였다.
