Purification ana properties of alkaline pretense produced by Bacillus sp. KCTC 1723

  • Alkaline protease which is an important enzyme used in detergents, leather tanning and food industry was produced by alkalophilic bacterium, Bacillus sp. KCTC 1723 isolated from soil. The maximum productivity of the enzyme in alkaline medium containing 1% sodium bicarbonate was obtained by incubating for 3 days at 37$^{\circ}C$. The optimum pH of the enzyme was 11.5 and calcium ion was effective on stabilization of the enzyme at high temperature. The enzyme was not inhibited by metal chelating agent such as El)TA but inhibited by diisopropyl fluorophosphate. Purification of the enzyme was carried out DEAE- and CM-cellulose column chromatographies and molecular weight of the purified enzyme was determined

A Modified Alkaline Hydrolysis of Total Ginsenosides Yielding Genuine Aglycones nad Prosapogenols

  • To improve the yield of genuine aglycones from glycosides, the conditions of alkaline hydrolysis were investigated, and a modified method was established. The modified method empolyed pyridine as an aprotic solvent. To complete the hydrolysis and obtain 20(S)-protopanaxadiol (1) and 20(S)-protopanaxatriol(2), which are the genuine aglycones of ginsenosides, total ginsenosides were refluxed with sodium methoxide in pyridine. Addition of methanol, a protic polar solvent to the reaction miuxture, led partial hydrolysis yielding a mixture of the genuine prosapogenols. Of the prosapogenols compound 3 and 6 characteristically possessed D-glucopyranosyl moiety attached at the sterically hindered C-20 hydroxyl group. 3 and 6 were not obtaijned by other hydrolysisw methods except by the soil bacterial hydrolysis.

  • 토양으로부터 alkaline protease 생성능이 강한 Streptomyces griseus HC-1141을 분리하였으며, 효소생산의 최적 배양조건은 0.5 casein, 0.05 ammonium chloride, 0.1 ferrous sulfate, 2.0의 lactose, pH 8.0에서 84시간 배양했을 때이다. 효소의 정제는 ammonium sulfate 침전, DEAE-cellulose ion exchange chromatography, Sephadex G-150 gel filtration, crystallization으로 하여 53.23배 정제할 수 있었으며 polyacrylamide gel 전기영동상 단일밴드를 나타내었다.

Reversed Effects of Phosphate Fertilizer on Reducing Phytoavailability of Cadmium in Mine Tailing Affected Soil

  • To reduce effectively cadmium (Cd) phytoavailability by phosphate in mine tailing affected soil, fused and super phosphate (FSP), a main phosphate fertilizer in South Korea, was selected as phosphate source and then applied at the rates of 0, 78, 390, and 780 $P_2O_5kg\;ha^{-1}$. FSP did not decrease Cd extractability and radish Cd uptake, but increased significantly. The effect of Cd supplement and soil negativity increase through FSP application was not significant. Soil pH decreased markedly with increasing FSP application, which increased significantly soil Cd extractability and radish Cd uptake. As a result, phosphate fertilizer for reducing Cd phtyoavailability in heavy metals contaminated soil should be carefully selected as alkaline type.

  • Soil properties which affect the retention of Pb(I) were investigated in the laboratory. It was determined, through selective removal, that organic matter and Fe-oxides are of lesser importance in influencing Pb retention than are soil clay minerals. The following trend : clays > organic matter > Fe-oxides represents the relative importance of each constituent in the adsorption of Pb by soils. The consistently greater Pb uptake by surface over subsurface samples was apparently due to differences in organic matter content, inasmuch as organic matter removal from both resulted in similar adsorption characteristics. All five soils stooled exhibited a pH-dependent trend of adsorption. The extent of Pb adsorption was least at low pH values(4~5), was maximum in the neutral pH range, and leveled off or diminished under more alkaline conditions. There was no strong correlation between Pb uptake and soil cation exchange capacity as routinely measured by the NH$_4$OAc method. A knowledge of clay mineralogy in conjunction with soil pH is suggested as being the most reliable guide to predicting Pb retention by soils.

Effect of cement dust on soil physico-chemical properties around cement plants in Jaintia Hills, Meghalaya

  • Investigation was carried out to assess the effect of cement dust deposition on the physico-chemical properties of soil near some cement plants in Jaintia Hills, Meghalaya. Soil samples were collected and analysed and compared with the control site. Comparison of various soil physico-chemical parameters revealed that cement dust emanating from cement plants has changed the soil quality in the surrounding areas of cement plants. The normal soil pH in the area is generally acidic. However, due to the continuous deposition of cement dust soil pH was found slightly alkaline near the cement plants. The higher values of soil parameters such as electrical conductivity and bulk density were also noticed near the cement plants. However, lower values of water holding capacity, soil moisture content, soil organic carbon and total nitrogen content were found compared to the control sites. The effect of cement dust deposition on soil is more in areas nearer to the cement plants. At present the changes may not be so serious but if this trend continues, soil properties of a vast area around the cement plants are likely to change leading to multiple effects on flora, fauna and socio-economy of the area.

Streptomyces sp. YSA-130이 생산하는 Alkaline Protease의 정제 및 특성 (Purification and Properties of Alkaline Protease from Streptomyce sp. YSA-130)

  • 토양으로부터 분리한 Streptomyces sp. YSA-130으로부터 활성이 좋은 결정화된 alkaline protease를 분리하였다. Alkaline protease 생산의 최적 배양조건은 2.0% soluble starch, 1.0% soytone, 0.3% $K_2$HPO$_4$, 0.02% MgSO$_4$.7$H_2O$, 0.8% $Na_2$CO$_3$ 3$0^{\circ}C$, pH 10.5에서 72 시간 배양하였을 때 였다. Alkaline protease이 정제는 (NH$_4$)$_2$SO$_4$. 분별침전, 투석, DEAE cellulose column chromatography, Sephadex G-75 gel filtration, crystallization으로 하였으며, 그 결과 비활성도 14,290unit/mg, 정제도 23.8 배였고, 수율은 20.0% 이었다. Alkaline protease의 반응 최적온도와 pH는 6$0^{\circ}C$ 와 11.5이었으며, 효소의 pH 안정성은 5.5-12.0에서 안정하였고, 온도 안정성은 5$0^{\circ}C$까지 안정하였으며, $Ca^{++}$ ion 첨가시 6$0^{\circ}C$까지 안정성이 증가하였다. Alkaline protease의 분자량은 30,000이었으며 금속이온, EDTA, 환원제는 활성에 영향이 없었고 DFP에 의해 저해되었다. 계면활성제에 저항성이 크고 $H_2O$$_2$에 대한 잔존활성은 60% 을 유지하였다.

Production and Characterization of an Alkaline Protease from Bacillus licheniformis MH31

  • A alkalophilic strain, Bacillus licheniformis MH31 producing an alkaline protease was isolated from mine soil of Boryeong in Korea. Production of a high level of alkaline protease was achieved 42 h after incubation when the bacterium was grown at pH 9.0 and $35^{\circ}C$ in Horikoshi medium supplemented with 0.5%(w/v) starch and 1%(w/v) skim milk as carbon and nitrogen source, respectively. The molecular weight of partially purified enzyme was estimated to be 30 kDa by SDS-PAGE and its optimum pH was pH 10. The enzyme showed optimum temperature at $50^{\circ}C$, and was stable up to $60^{\circ}C$ after 1 h incubation. The protease was strongly inhibited by 1 mM of PMSF which was known well as strong inhibitor of serine proteases, but almost not inhibited by 5 mM of EDTA and 1,10-phenanthroline. When the protein hydrolysis products of 1% skim milk by partially purified protease was compared with available commercial proteases using HPLC analysis, most of hydrolysis products were detected below molecular weight of 10,000 and the hydrolysis ratio of purified enzyme was 24.8% lower than those(above 32%) of commercial proteases.

알칼리성 Prottease를 생산하는 Xanthomonas sp. YL-37의 분리 및 조효소의 성질 (Production and Characterization of ans Alkaline Protease from an Isolate,Xanthomonas sp.YL-37)

  • A bacterial strain, which showed the high protease activity at low temperature and the high tolerance for the surfactant, was isolated from soil and identified as Xanthomonas sp. YL-37. The optimal temperature, initial pH, and cultivation time for the production of the alkaline protease by Xanthomonas sp. YL-37 were 20$\circC , 11.0, and 84 hours, respectively. In the jar fermenter culture of Xanthomonas sp. YL-37, the alkaline protease activity was about 15,000 DU/ml/-broth after cultivating for 108 hours. The optimal pH and temperature for the protease activity were 70$\circC and 11.0, respectively. The protease was relatively stable at the pH range of 7.0~12.0 and at the temperatures below 50$\circC . The protease activity at 20$\circC was about the level of 40% of its activity at 70$\circC . The enzyme was suggested as a serine protease because the enzyme activity was inhibited by phenylmethane sulfonyl fluoride, a serine modifier.

방선균 일주에서 포자형성과 호알칼리성 단백질 분해효소의 생합성과의 관계성 (Relationship between Sporulation and Synthesis of Alkaline Protease in Streptomyces sp.)

  • 성장속도가 빠르고포지형성이 우수한 방사균 일주를 토양에서 분리한 뒤 분리균의 특성을 조사한 결과 세포외 단백질 분해 효소가 중성과 알카리성의 두 종류가 생성되었으며 $\beta$-lactamase도 생성함을 알았다. 이 균주를 acriflarin 또는 NTG로서 처리하여 얻은 변이주는 포자의 형성, $\beta$-lactamase의 생성 및 protease의 생합성 능력이 소실 또는 크게 저하되었다. 일단계의 변이주 취득에서 동시에 형질의 변화가 다양하게 나타난 원인을 규명한 결과 호알카리성 protease의 생합성이 크게 저하되었음을 알았다. 따라서 방선균에서 포자형성과 호알카리성 protease의 활성이 일정한 연관성이 있을 것으로 판단되었다.
