Effect of Environmental pH on End Products, Fermentation Balances and Bioenergetic As-pects of Lactobacillus bulg-aricus in a Glucose-limited “pH Stat” Continuous Culture.

  • A glucose-limited “pH-stat” continuous culture study of Lactobacillus bulgaricus NLS-4 in an anaerobic condition showed the marked effects of environmental pH on end products, fermentation blances and bioenergetic aspects of the organism. Lactic acid was the major end product of fermentation with minor products, such as acetic acid, formic acid and ethanol throughout the pH range tested. In acidic conditions below pH 6.5, a typi-cal pattern of homofermentation was revealed whereas in alkaline conditions, the metabolic pattern was changed from homofermentation to heterofermentation and led to acquire much energy. This metabolic change was likely due to the pH-dependent lactate dehydrogenase activity. Molar growth yields (Yglc=35.5-44.4) and YATP, $18.5\pm2.5$ in average which was 80% higher than the value ever postulated seemed to be accounted for less requirement of maintenance energy of the organism in the culture conditions.

알칼리환원수 음용이 급성 염증성장질환 생쥐 모델에 미치는 영향 (The Effect of Mineral-induced Alkaline Reduced Water on the DSS-induced Acute Inflammatory Bowel Disease Mouse Model)

  • 알칼리환원수 (Alkaline Reduced Water: ARW)는 아시아를 비롯한 여러 나라에서 음용수로 사용되고 있으며 항산화효과와 장내이상발효 개선효과 등을 중심으로 안전성과 유효성의 검증이 이루어지고 있다. 본 연구에서는 알칼리환원수가 급성 염증성대 장질환 (Inflammatory Bowel Disease: IBD) 동물모델에 미치는 영향을 알아보기 위해, ARW를 2주 동안 급이 시킨 후 4% DDS 로 염증성 대장염을 유발시키고 대장의 길이와 조직병리학적 변화를 관찰하였다. 그 결과 대장의 길이와 육안적, 현미경적 질환점수의 수치가 대조군과 비교하여 유의한 차이를 나타내지 않았다. 이 결과는 DSS로 유도된 급성 염증성장질환 모델에서는 2주 동안의 ARW 급이가 개선효과를 나타내지 않았음을 말해준다. 그러나 ARW가 장내환경을 개선시키고 위장관의 질환을 개선시키는 효과가 있음을 고려해볼 때 급성 IBD 동물모델이 ARW의 효과를 증명 하기에 적합하지 않았거나 MARW의 급이기간이 장내환경을 개선할 만큼 충분히 길지 않았을 가능성을 추측해 볼 수 있다. 알칼리환원수의 장내이상발효 개선효과는 확인되고 있으나 그 기전은 아직까지 구체적으로 확인되어지지 않고 있다. 이번 연구결과에 의하면 인위적으로 유발시킨 급성 염증성 동물모델에서 ARW가 유의한 영향을 미치지 않았지만, 다른 장질환 모델을 이용한 효과 검증과 ARW의 장기 급이에 따른 효과, 그리고 ARW가 장내환경에 미치는 작용기전에 대한 연구가 더 깊이 있게 이루어져야 할 것으로 사료된다.

Preparation and Characterization of Antioxidant Peptides from Fermented Goat Placenta

  • The goat placenta was fermented by Bacillus subtilis and the optimal fermentation parameters of strongest antioxidant capacity of peptides were obtained using response surface methodology (RSM). The effects of fermentation time, initial pH value and glucose content on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity of the goat peptides were well fitted to a quadric equation with high determination coefficients. According to the data analysis of design expert, the strongest DPPH radical scavenging capacity value was obtained with the following conditions: content of glucose was 2.23%, initial pH value was 7.00 and fermentation time was 32.15 h. The DPPH radical scavenging capacity commonly referring antioxidant activity showed a concentration dependency and increased with increasing peptide concentration. The effects of temperature and pH were assessed to determine the stability of antioxidant peptides prepared from goat placenta. Antioxidant peptides showed good stabilities when temperature was lower than $70^{\circ}C$. However, the antioxidant peptides lost antioxidant activities rapidly under alkaline and excessive acid condition. Ultrafiltration technique was performed to separate fermentation broth with different Mw (molecular weight). It was found that peptides in the range of < 3 KDa mainly accounted for the antioxidant activities.

Chemical properties of liquid swine manure for fermentation step in public livestock recycling center

  • The nutrients in livestock manure produced during fermentation processes in public livestock recycling centers are used as fertilizers. However, the large amounts of swine manure produced in intensive livestock farms can be a nonpoint source of pollution. In this experiment, we investigated the chemical properties, inorganic components, and heavy metal contents in 101 samples of liquid swine manure collected from 28 public livestock recycling centers throughout the nation. Results showed that the average pH of the samples was alkaline (pH range 5.18 to 9.54), and their maximum EC was $53.2dS\;m^{-1}$. The amounts of total nitrogen and total phosphorus were in the range of 1000 - 2000 and $200-800mg\;L^{-1}$ while potassium, which constituted 47% of the total inorganic ions recovered from the liquid swine manure, amounted to $1500mg\;L^{-1}$. The most distinctive heavy metals recovered from the liquid swine manure were copper and zinc although the amounts of both heavy metals were much lesser than those of the standards as livestock liquid fertilizer set by the Rural Development Administration. On the other hand, the amount of nitrogen decreased rapidly with an increasing fermentation period from immature to mature, assumed to be lost as volatile compounds, such as ammonia, which are the major odor components during the fermentation process.

Effects of Brown Rice Extract Treated with Lactobacillus sakei Wikim001 on Osteoblast Differentiation and Osteoclast Formation

  • Phytic acid (myo-inositol hexakisphosphate) or phytate is considered an anti-nutrient due to the formation of precipitated complexes that strongly reduces the absorption of essential dietary minerals. In this study, brown rice with reduced phytate was made by inoculation with Lactobacillus sakei Wikim001 having high phytase activity. The effects of brown rice extract treated with L. sakei Wikim001 (BR-WK) on osteoblast differentiation and osteoclast formation were investigated. The proliferation of SaOS-2 cells was measured by the MTT assay. Treatment with BR-WK increased cell proliferation by 136% at a concentration of $100{\mu}g/mL$. The Alkaline phosphate activity in SaOS-2 cells was 129% higher when BR-WK was processed at a concentration of $100{\mu}g/mL$. The proliferation of bone marrow macrophages decreased by nearly 60% in response to treatment with BR-WK. In addition, BR-WK reduced the number of tartrate-resistant acid phosphatase-positive ($TRAP^+$) multinucleated cells from bone marrow macrophages. These results indicate that BR-WK stimulates bone formation through its positive action on osteoblast differentiation and function and furthermore, decreases osteoclast differentiation.

New Finding and Optimal Production of a Novel Extracellular Alkaline Lipase from Yarrowia lipolytica NRRL Y-2178

  • Lipases are industrially useful versatile enzymes that catalyze numerous different reactions including hydrolysis of triglycerides, transesterification, and chiral synthesis of esters under natural conditions. Although lipases from various sources have been widely used in industrial applications, such as in food, chemical, pharmaceutical, and detergent industries, there are still substantial current interests in developing new microbial lipases, specifically those functioning in abnormal conditions. We screened 17 lipase-producing yeast strains, which were prescreened for substrate specificity of lipase from more than 500 yeast strains from the Agricultural Research Service Culture Collection (Peoria, IL, U.S.A.), and selected Yarrowia lipolytica NRRL Y-2178 as a best lipase producer. This report presents new finding and optimal production of a novel extracellular alkaline lipase from Y. lipolytica NRRL Y-2178. Optimal culture conditions for lipase production by Y. lipolytica NRRL Y-2178 were 72 h incubation time, $27.5^{\circ}C$, pH 9.0. Glycerol and glucose were efficiently used as the most efficient carbon sources, and a combination of yeast extract and peptone was a good nitrogen source for lipase production by Y. lipolytica NRRL Y-2178. These results suggested that Y. lipolytica NRRL Y-2178 shows good industrial potential as a new alkaline lipase producer.

구멍갈파래의 알칼리 과산화수소 전처리 및 효소 가수분해 특성 (Enzymatic Hydrolysis of Pre-treated Ulva pertusa with Alkaline Peroxide)

  • 해조류는 잠재력이 있는 풍부한 발효기질이다. 해조류의 효소가수분해를 상업용 셀룰라아제 및 비스코자임과 함께 알칼리 과산화수소 전처리에 의해서 검토하였다. 본 연구에서 사용된 해조류는 구멍갈파래이며, 전처리 평가는 효소가수분해 후 얻어진 글루코오스의 수율로 나타내었다. 알칼리 과산화수소는 과산화수소 1 wt%에 수산화나트륨을 1~1.75 wt% 범위로 혼합하였다. $60^{\circ}C$에서 3 h 동안 전처리한 결과 5 wt% 과산화수소를 사용했을 때에 가장 높은 글루코오스 전환율을 보였다. 알칼리 과산화수소로 전처리한 후 필요한 효소의 양은 전처리하지 않은 구멍갈파래에 비하여 상당히 절감되었으며, 아울러 효소 가수분해동안 얻어진 글루코오스의 양은 증가되었다.

볏짚 발효사료 제조시 과산화수소 전처리 효과 (The Effects of Hydrogen Peroxide Pretreatment on Rice Straw Fermentation for Feed)

  • 볏짚의 사료가치를 증진시키기 위하여 과산화수소에 의한 전처리와 섬유소 및 리그닌분해력이 강한 Pleurotus florida 균주로 발효사료를 제조하고 화학성분, 결정도 및 in vitro 소화율을 조사하여 사료적 가치를 검토하였다. P. florida에 의한 볏짚발효시 hemicellulose, cellulose 및 lignin은 각각 22.5%, 11.4%, 28.1% 감소하였고, $H_2O_2$$H_2O_2(pH\;11.5)$ 전처리 후 발효시에는 4% 이하의 저농도에서 감소가 심하였으며 발효기간이 경과할수록 cellulose, lignin 등이 감소하였고 아미노산함량은 무처리 볏짚에 비하여 배양 30일 후에 약 28.9% 증가하였으며 4% $H_2O_2$ 전처리 후 발효시는 35.1% 증가하였다. 볏짚 섬유소의 결정도는 $H_2O_2$ 처리 및 P. florida에 의한 발효처리로 감소하였고 $H_2O_2(pH\;11.5)$ 처리시 증가하였으며 $H_2O_2(pH\;11.5)$ 처리 후 수세시의 잔류볏짚의 결정도는 더욱 증가하였고 P. florida에 의한 볏짚배양 30일 후 in vitro 소화율은 17.1% 증가하였으며, $H_2O_2$ 전처리 후 발효시킨 볏짚은 6% 이하의 저농도에서 소화율 상승효과가 있었다.

탁주료중의 단백질분해효소에 관한 연구 (Studies on the proteinase in Takjoo mashes during the process of brewing)

  • The mash of Takjoo, Korean flour wine, is fermented through two brewing processes ; the primary brewing process to saccharify and the main one to produce ethyl alcohol. The activities of acid proteinase (pH3), weak acid proteinase (pH 6), and alkaline proteinase (Ph 80 on the processes are determined with time by the Folin phenol method as a strength of casein digestion. Hydrogen ion concentration, the content of total organic acids, protein, free amino acids and oligopeptides, which effect the activities of proteinase, are also measured. The results are briefly summarized as follows : 1. In general, the activities of acid proteinase and weak acid proteinase in the mesh of primary brewing process are stronger than those in main brewing process. 2. The activities of acid proteinase are remarkably stronger than those of weak acid proteinase in both processes. It reveals that they decrease slowly through the fermentation. Activities of alkaline proteinase are weaker than others. 3. As the raw materials are mixtured, the total amount of organic acids is equivalent to 0.150 mg/ml acetic acid in the mesh of primary brewing process and 0.02 mg/ml acetic acid in the main one. They increase gradually with time. 4. Hydrogen ion concnetration shows 3.9 in the mesh of main brewing process and 3.28 in the primary one. They increase to the maximum in 60-72 hrs., and decrease since 108 hrs. 5. The content of crude protein shows 66.90mg/ml in the mesh of main brewing process, while shows 64.29mg/ml in the mesh of primary one. they decrease slowly with time. it seems that a small content of crude protein, as a substrate, converts into amino acids and soluble nitrogen compounds by proteinase. 6. The content of free amino acids and oligopeptides shows 0.36 mg/ml in the mesh of primary brewing process and 0.24mg/ml in the main brewing process. It is evident that the reason they increase continuously through the fermentation is the effect of proteinase. 7. According to the results, the strong activities of proteinase in primary brewing process has been derived from the decrease of hydrogen ion concentration due to the production of organic acids.

역미셀을 이용한 Bacillus licheniformis 발효액으로 부터 알카리성 단백질 분해효소의 추출에 관한 연구 (A Study on the Extraction of Alkaline Protease from Bacillus licheniformis Fermentation Broth using Reverse Micelle)

  • 역미셀을 이용하여 박테리아 (Bacillus licheniformis)의 발효액으로부터 알카리성 단백질 분해효소를 분리할 때 여러가지 인자들, 즉 이온강도, pH, 계면활성제의 농도 등이 분리효율에 미치는 영향에 대하여 다음과 같은 결과를 얻었다. 이온강도를 높여 주기 위하여 KCI을 사용하여 조절하였으며, 발효액의 이온강도를 높여줄수록 추출되는 효소의 양은 감소하였으며, 역추출 용액의 이온강도를 높여줄수록 역추출되는 효소의 양은 증가하였다. 계면활성제로는 sodium-di-2-ethylhexyt sulfosuce-inate (Aerosol-OT또는 AOT)를 사용하였으며, 유기상내의 계면활성제 농도를 높여줄수록 더 많은 양의 효소가 추출됨을 알 수 있다. pH는 IN HCI과 0.1N NaOH를 사용하여 조절하였고, 발효액의 pH가 5.3이었을 때 가장 많은 양의 변성되지 않은 효소가 추출되었으며, 역추출 용액의 pH가 7.5일 때 가장 많은 양의 변성되지 않은 효소가 역추출됨을 알 수 있다.
