Development of Near Isogenic Lines and Genetic Analysis for Alkali Digestibility of Rice Grain

  • In order to develop near isogenic lines (NILs) the heterozygous rice plants for alkali digestibility value(ADV) were selected and tested in every generation from $F_5$to $F_9$ of a cross, Seratus malam/Suwon 345. Finally several sets of NILs, which were six low ADV lines and four medium-high ADV lines, were selected among $F_{10}$ lines. No differences of the plant growth characteristics, amylose content and protein content of rice grain were found between low and medium-high ADV lines. Rice flour of low ADV lines showed longer gel length in gel consistency test than medium-high ADV lines, and also showed different gelatinization characteristics measured by rapid visco analyzer compared with medium-high ADV lines. The result of genetic analysis indicated that ADV-NILs developed were differed in a pair of major gene controlling ADV, and low ADV was dominant over medium-high ADV.

Varietal Variation of Alkali Digestion Value and Its Relationship with Gelatinization Temperature and Water Absorption Rate of Milled Rice Grain (쌀 알칼리붕괴반응의 품종간 변이와 호화온도 및 수분흡수율과의 관계)

  • Fifty rice varieties were tested for alkali digestibility of milled rice grain at four different KOH levels, and twenty-four varieties selected were tested again for alkali digestibility at different degrading times and KOH levels. Gelatinization rate at several heating times and heating temperatures, and water absorption rate at 21$^{\circ}C$ and 77$^{\circ}C$ water temperatures were observed using rice samples of twenty-four varieties to clarify the relationship between alkali digestibility response, gelatinization rate and water absorption rate. Varietal difference of ADV in Japonica and Tongil tye rices was biggest at KOH 1.2%, but it was better to test at KOH 1.2% and 1.4% levels to know the exact alkali digestibility response of rice varieties. Rice varieties tested could be classified into three groups, low, intermediate and high, based on their alkali digestibility response at four KOH levels, and most of Korean cultivated rice varieties were belonged to intermediate or high ADV group. Varietal variation was also found in alkali degrading response at different soaking times in alkali solution. Low ADV varietal group showed higher gelatinization temperature and needed longer heating time for complete gelatinization compared with intermediate or high ADV group. Same trends was found between intermediate and high ADV groups, but varietal variation in the same ADV group was also found in gelatinization temperature and heating time needed for complete gelatinization of rice grain. Water absorption rate of low ADV group was lower than intermediate or high ADV group both at 21$^{\circ}C$ and 77$^{\circ}C$ water temperatures, and intermediate ADV group showed lower absorption rate than high ADV group only in initial water absorption stage at 21$^{\circ}C$.

  • These studies were carried out to evaluate the feasibility of using Alkali Digestibility Value (ADV) of soybean seeds as a selection criterion for cooking quality of soybean grain with rice, and to find out an optimal testing condition, varietal differences in ADV and the effect of cultural environment on the ADV, and its mode of inheritance. The ADV test was achieved succesfully by soaking the soybean grain for 24 hours in 25 to 30% KOH solution at 25 to $30^{\circ}C$. Varietal differences in the ADV were conspicuous, but cultural conditions affected the ADV little. All collected lines for cooking with rice showed high ADV and all the small grain cultivars introduced from abroad had low ADV, while recommended cultivars showed great variation in ADV. The characteristic of low ADV in soybean grains was controlled by single dominant gene.

Analysis of Varietal Variation in Alkali Digestion of Milled Rice at Several Levels of Alkali Concentration (쌀의 KOH 농도별 붕괴양상에 따른 품종변이 해석)

  • To analyze and classify the varietal variation of alkali digestibility in detail, which is closely connected with the gelatinization temperature and physical characteristics of cooked rice, the patterns of alkali decomposition changed along the alkali concentration were investigated for thirty three Korean leading rice cultivars and new breeding lines(japonica : 25, Tongil-type:8) including five glutinous rice. Principal component analysis was used to condense the information and to classify rice materials according to decomposed reaction pattern at several levels of potassium hydroxide(KOH) concentration. Thirty three rice varieties were classified largely into four groups by the distribution on the plane of upper two principal component scores which contained above 92% of total informations. Group I was consisted of one variety, Dobongbyeo, which owned almost same strong resistance to alkali digestion at the range of 0.8% to 1.6% KOH solutions. Group II included three japonica and Tongil-type glutinous rice varieties, which revealed medium alkali digestion value(ADV) at 1.4% KOH solution and intermediate change in ADV from 0.8% to 1.6% KOH solutions. Most of Tongil-type and early-maturity japonica rice, which exhibited medium-high ADV at 1.4% of KOH concentration and large ADV difference between low and high alkali solutions, were contained in Group III. Group N included most of medium or medium-late-maturity japonica, which showed high ADV at 1.4% KOH and medium or intermediate-high ADV change between low and high alkali solutions. The 1st principal component indicated the average index of ADV through 0.8-1.6% KOH solutions and the 2nd principal component pointed out the factor related with ADV difference between low and high alkali solutions or regression coefficients of ADV change along with the KOH concentrations.

Physicochemical Properties of Non-glutinous, Dull, and Glutinous Rice Grain in Segregating Populations of Dull/Glutinous Crosses

  • Dull grains segregated from F$_3$ and F$_4$ of the crosses between two dull mutants and a glutinous cultivar were compared with non-glutinous and glutinous segregants for their physicochemical properties. Amylose content of dull rice grain segregated from the dull/glutinous cross showed the intermediate value between glutinous and non-glutinous rice grain, whether it is controlled by the recessive or dominant gene. Alkali digestibility value (ADV) of dull rice grain was lower than that of glutinous or non-glutinous rice. A positive correlation was found between ADV and amylose content of homozygous non-glutinous or dull F$_4$ grains, but a negative relationship was observed in glutinous grains. Protein content of dull grain was significantly higher than that of glutinous or non-glutinous grain segregated from the same cross, while those of glutinous and non-glutinous grains were not different. Among gelatinization characteristics, initial pasting temperature and peak viscosity of dull grains were higher than glutinous rice, and were not different with non-glutinous grain. Hot, cool and consistency viscosities of dull grain were intermediate between glutinous and non-glutinous rices. Dull grains showed the highest breakdown viscosity and the lowest setback viscosity among the three endosperm types.

Water Absorption and Alkali Digestibility by Seed Size in Local Black Soybean Collections (종실크기에 따른 수집 검정콩 수분흡수 및 알칼리 붕괴도)

  • Seed characteristics such as seed size, seed shape and cotyledon color are important in relation to processing and cooking of soybean. Seed shape, water absorption rate, and alkali digestibility were evaluated for 28 local black soybean collections. Water uptake rate was low in small seed size at low temperature(5, 2$0^{\circ}C$). However, higher water absorption rate was observed at high temperature(40, 6$0^{\circ}C$) regardless of seed size. When seeds soaked up water, it reached to water absorption equilibrium after 10 hrs below 2$0^{\circ}C$, within 10 hrs at 4$0^{\circ}C$, and around 4 hrs at 6$0^{\circ}C$, respectively. Significant positive correlations were observed between 100 seed weight and length, width and thickness of seed, respectively. Alkali digestibility value(ADV) was higher in large seed group than in small seed group. Among seed characteristics, significant positive correlation was shown between ADV and 100 seed weight, length, width, thickness and hilum length of seed, respectively. Soybean seed with green cotyledon showed higher ADV than that with yellow cotyledon.

  • A seven-parent half-diallel cross was analyzed for white center, white belly and alkali digestibility value(ADV). The analysis of variences were significant for all characters at 1% level. Both general and specific combining ability were significant for all characters. Non-additive genetic varience predominated for ADV(12hr. treatment) and additive effects were more important than non-additive effects for other characters. With the exception of ADV(12hr. treatment), which showed complete dominance, partial dominance in a negative direction was observed for the other characters. Heritability values were high in all characters. White belly was high positively correlated with white center and ADV.

  • Six commercial rice varieties were cultivated at different locations in 1987 and 1988. Rice samples harvested from 8 to 20 locations for a variety each year were used to measure or observe grain appearance, amylose content and alkali digestibility, cooked rice texture using rheometer, gelatinization and viscosity of rice flour using amylograph, and eating Quality of cooked rice by sensory evaluation. Relationship between Quality characteristics showing large locational variation were analized to approach the long-term objective, rice grain Quality standardization. Percent white-center and white-belly grain of the same variety showed great variation between locations, but did not affect on 1000-grain weight, amylose content and alkali digestibility, amylogram and rheogram characteristics, and eating Quality of cooked rice. Positive correlationship were obtained between maximum viscosity and break down, and maximum viscosity and alkali digestibility. Set back was correlated negatively with maximum viscosity, break down and alkali digestibility. Rice samples having significantly lower values of maximum viscosity and break down and higher set back value at the same time showed higher ADV and lower texture palatability index (TPI) and viscousness/hardness ratio (Vi/H), and lower sensory evaluation score of cooked rice compared with those of rices having typically higher values of maximum viscosity and break down and lower set back value. When TPI, Vi/H and overall sensory eating score of cooked rices measured were divided into three categories, high, medium and low using deviation from locational mean value of a variety, many of rice samples having high TPI or high Vi/H showed medium or high in overall sensory eating score of cooked rice.

  • This study was carried out to clarify the changes in dry matter weight and physicochemical property of job's tears grain during ripening period. Four varieties of job's tears were seeded at April 20, May 10, and May 30, andgrains were harvested at every 7 days from heading to maturity, The whole seed weight of thick hull varieties, Kimje and Aewon, was higher than that of improved thin hull varieties, Suwon 3 and Suwon 6, from heading to maturity, However, the dehulled grain weight of thin hull variety were higher than that of thick hullvariety from 35 days after heading. The first heading spikelet showed lower 100-grain weight com-pared with the spikelets headed at full blooming stage among the same plant. The increased rate of de hulled grain weight during maturing period was higher in thin hull variety than thick hull one, later seeding date than early seeding, and intermediate headed spikelet than early head-ed one, respectively. As maturing proceeds the protein content of de hulled grain was decreased, alkali digestibility value(ADV) showed a rising trend, and little change was found in gel consistency of milled grain flour. And same trends were found in protein content, ADV and gel consistency as the seeding date was delayed. Among amylogram properties of job's tears flour the maximum, minimum, and final viscosity as well as breakdown were decreased, and set back was increased in later seeding plots

Changes of Seed Chemical Component during Grain Filling in Job′s Tears (율무의 등숙에 따른 종실 성분함량 변화)

  • The reports on the chemical composition of developing job's tears seeds is very insufficient by this time. The purpose of this study was to obtain the fundamental information on chemical component of the developing job's tears seed, including starch, sugar, oil, protein, fatty acid, Alkali Digestibility Value(ADV), 100-grain weight. Two job's tears varieties in different by maturity types were analyzed for those components from 10 days after heading to maturity, with 5-day intervals. Total sugar content in seeds followed a pattern of linear increase after heading, but it started to decreased around 15 days after heading. On the other hand, starch content continued to increased until maturity and the highest increasing occurred between 15 and 20 days after heading. The protein content of seeds was decreased, while oil content was increased through the ripening process. Starch content showed a positive correlation with 100-grain weight, ADV, oil and germination percent, but it had negative correlation between total sugar and protein contents in ripening process. Maximum starch content, 100 grain weight, and germination percent obtained by quadratic equation were attained 38 to 43 days after heading. So that the physiological maturity of job's tears seeds was considered to be 40 days after heading.
