Characterization of Chemical Composition in Poplar wood (Populus deltoides) by Suppression of CCoAOMT Gene Expression (CCoAOMT 유전자 발현 억제에 의한 현사시나무의 화학조성 변화)

  • In this study, chemical compositions - holocellulose, lignin and monomeric sugars - were characterized with two poplar wood cell walls, one of which was grown at normal condition (CPW) and the other was genetically modified by antisence suppression of CCoAOMT gene expression (ACPW). Milled wood lignins were isolated from CPW and ACPW and subjected to methoxyl group, DFRC, Py-GC/MS, GPC, $^{13}C$-NMR analysis, respectively. There were few differences in holocellulose contents in both cell walls, which were determined to 81.6% in CPW and to 82.3% in ACPW. However, lignin contents in ACPW was clearly decreased by the suppression of CCoAOMT gene expression. In CPW 21.7% of lignin contents was determined, while lignin contents in ACPW was lowered to 18.3%. The relative poor solubility of ACPW in alkali solution could be attributed to the reduction of lignin content. The glucose contents of CPW and ACPW were measured to 511.0 mg/g and 584.8 mg/g and xylose contents 217.8 mg/g and 187.5 mg/g, respectively, indicating that suppression of CCoAOMT gene expression could be also influenced to the formation of monomeric sugar compositions. In depth investigation for milled wood lignin (MWL) isolated from both samples revealed that the methoxyl contents at ACPW was decreased by 7% in comparison to that of CPW, which were indirectly evidenced by $^{13}C$-NMR spectra and Py-GC/MS. According to the data from Py-GC/MS S/G ratios of lignin in CPW and ACPW were determined to 0.59 and 0.44, respectively. As conclusive remark, the biosynthesis of syringyl unit could be further influenced by antisense suppression of CCoAOMT during phenylpropanoid pathway in the plant cell wall rather than that of guaiacyl unit.

Studies on the Quality of Korean Rice (한국쌀의 품질에 관한 연구)

  • The rice qualities including cooking and eating qualities were studied using recommended Korean rice varieties (20 of japonica and 3 of indies type; IR 667) which were grown at Suwon, Korea in 1971. As the result, followings were obtained. 1. Amylose contents of white rice were varied with the varieties 21.1 to 25.5% and the average was 23.0%. Three indica type varieties (IR 667) showed higher amylose contents than the other japonica type varieties except Mankyung. Among japonica type varieties, Palkum, Mankyung composed the group of the highest amyloes content and Kimmaje was the lowest. 2. Blue values were distributed in the range of 0.38 to 0.48 and the average was 0.42 IR 667 varieties showed the highest blue value among them. Among japonica type varieties, Jaegun showd the highest blue value and Sooseung, Shirogane showed successively lower values, Shin #2, Nongbaik, Palkweng, Suwon #82, Mankyung, Nonglim #25 and Nongkwang relatively lower blue values. 3. Alkali numbers were in the range of 6.0 to 7.4 and the average was 6.8. Much difference was not shown in alkali number between IR 667 group and the japonica varieties group. 4. Gelatinization temperature were ranged from 59.5 to 64.0 IR 667 varieties showed relatively higher gelatinization temperature than japonica type varieties. 5. Water uptake ratios were measured in the range of 2.67 to 2.92 and the average was 2.79. IR 667 varieties were belonged to the group of highest water uptake ratio. Among japonica type varieties Kimmaje, Suwon #82, Nonglim #29, Deungpan #5, Jaegun, Jinhung, were belonged to the group of relatively high water uptake ratio and Palkweng, Palkeum and Paldal to the relatively low water uptake ratio. 6. Expaned volums were ranged from 29.8 to 33.7 and the average was 31.8. IR 667 varieties showed higher expanded volumes than japonica type varieties. 7. Intensities of starch-iodine blue value of residual liquid indicated 0.35 to 0.58. Among them IR 667 varieties showed relatively high intensities. 8. The range of total solids in residual liquids was 0.605 to 0.810 and the average was 0.700 Much difference was not shown in total solids in residual liquid between IR 667 varieties and japonica varieties. 9. pH values of residual liquids were in the range of 6.3 to 7.3 and the average was 6.95. IR 667 varieties showed lower pH than japonira type varieties.

Petrochemical Study on the Micrographic Granite in the Wando Area (완도지역(莞島地域)에 분포하는 미문상화강암(微文象花崗岩)에 대한 암석화학적(岩石化學的) 연구(硏究))

  • Petrochemical study on the micrographic granite distributed in the Wando area, the southernmost part of the Yeongdong-Kwangju depression is performed to investigate the petrogenesis and differentiation processes of the granitic magma. Polarized light microscopy for modal analyses, electron probe microanalyses of feldspars and biotite, inductively coupled plasma analyses for major and trace element contents were adopted in the study. The lithology of the study area consists of Precambrian metasediments, Mesozoic volcanic and sedimentary rocks, and micrographic granite which intrude into the former. The micrographic granite in the Wando area are distributed in the shape of a cauldron. Modal and nonnative mineral analyses of the micrographic granite fall in the area of granite and granodiorite. The chemical composition indicates that the micrographic granite is I-type and magnetite series. The micrographic granite is characterized by more than 90% of micrographic texture in volume percent. Feldspars in the micrographic granite is alkali feldspars (Or, 45~93) and plagioclases (albite to oligoclase). The biotite has a intermediate composition between phlogopite and annite solid solution. The results of the petrochemical studies indicate that the granitic magma of calc-alkaline source materials reactivated in a compressional environment at the continental margin, and then was differentiated by fractional crystallization. The micrographic granite intruded into a shallow level of the crust (5~7 km) in the late Cretaceous.

  • The granitic rocks in the study area are divided into the schist and gneiss complex, Yongdok pluton, Yonghae pluton and Onjong pluton by their texture, fabric and relationship to the adjacent rocks in the field, Schist and gneiss complex occurs as xenolith or roof pendant in the Yongdok, Yonghae and Onjong plutons. The Yongdok pluton occurs in association with pegmatite and aplite in many places of its pluton. In the field it is obviously clarified that the Yongdok pluton is unconformably overlay by the Cretaceous sedimentary rocks. The Yonghae and Onjong plutons are gradationally changed each other, and these plutons truncate both the Yongdok pluton and the Cretaceous sedimentary rocks. Petrographically, the Yongdok pluton consists of granodiorite and granite with minor quartz monzonite. The Yonghae pluton is composed of diorite, quartz diorite, tonalite, and granodiorite. The Onjong pluton also ranges granodiorite to granite. Both the Yongdok and Yonghae-Onjong plutons are different in the constituent minerals, such as alkali feld~par, myrmekite, mica, sphene and mafic minerals. This suggests that each pluton might have been different crystallization sequence and characteristically different gological history during the crystallization period. Iron/magnesium ratio in biotite and hornblende from both the Yongdok and Yonghae-Onjong plutons gradually decrease as the differentiation index increasing in the whole rock. The decrease of this ratio strongly depend on the increase of opaque mineral contents. From the results of chemistry in the whole rocks and some mafic minerals, it is suggest that the granite plutons of the two different geological ages would have been suffered the environment of high oxygen fugacity in the process of magmatic emplacement and during the crystallization period.

A Study on the Literature Review of Acorn in Korea (도토리에 대한 국내의 연구 동향)

  • This study was surveyed and compiled the contribution about acorn in korea .the physico-chemical properties of acorn and acorn starch. preparation of acorn starch and elimination of the acorn tannin, the rheological properties of acorn starch gel. Nutrition of acorn and antioxidative activity of tannin in acorn extract. The result obtained were as follow. 1. The chemical compositions of acorn were water 6.5∼13.7%, crude ash 1.9∼3.4%, crude fat 1.1∼5.0%, crude protein 5.8∼7.8%, crude fiber 2.1∼3.6%, N-free extract 71∼77.5%, total tannin 4.6∼9.3%, Ca 92.7∼460.9mg%, p 80.0∼740.9mg%, Na 66.2∼93.9 mg%, and K 867.9∼983.1mg%. 2. The acorn tannin was extracted with water, acetone, and ethanol. The generation method was wash SE settling method with water. 3. The shape of acorn starch granule was rounded triangular and some elliptical, rasing power 12.4∼12.5, Blue value 0.43∼0.47, Alkali number 10.8∼11.3, Amount of Arnylose 28.8∼30.50% Tannin contents on the initial go tim-zation temperature of acorn starch were not influenced but maximum and cooling viscosity o the acorn starch were decreased. 4. Nutritional effect and diet absorption rate were not influence, by rice and 20% T-A(elimination of tannin from the acorn) mixed diet, and the content of lipid in s rum were not influenced by rice 40% and T-A (elimination of tannin from the acorn) mixed diet. 5. jallic acid, digallic acid, gallotannin were contained in acorn powder extract. The main antioxidative : stlvity was speculated due to the gallic acid.

Efficacy of Alkali-treated Sugarcane Fiber for Improving Physicochemical and Textural Properties of Meat Emulsions with Different Fat Levels

  • The objective of this study was to evaluate the efficacy of alkaline-treated sugarcane bagasse fiber on physicochemical and textural properties of meat emulsion with different fat levels. Crude sugarcane bagasse fiber (CSF) was treated with calcium hydroxide ($Ca(OH_2)$) to obtain alkaline-treated sugarcane bagasse fiber (ASF). The two types of sugarcane bagasse fiber (CSF and ASF) were incorporated at 2% levels in pork meat emulsions prepared with 5%, 10% and 20% fat levels. Alkaline-treatment markedly increased acid detergent fiber content (p=0.002), but significantly decreased protein, fat, ash and other carbohydrate contents. ASF exhibited significantly higher water-binding capacity, but lower oil-binding and emulsifying capacities than CSF. Meat emulsions formulated with 10% fat and 2% sugarcane bagasse fiber had equivalent cooking loss and textural properties to control meat emulsion (20% fat without sugarcane bagasse fiber). The two types of sugarcane bagasse fiber had similar impacts on proximate composition, cooking yield and texture of meat emulsion at the same fat level, respectively (p>0.05). Our results confirm that sugarcane bagasse fiber could be a functional food ingredient for improving physicochemical and textural properties of meat emulsion, at 2% addition level. Further, the altered functional properties of alkaline-treated sugarcane bagasse fiber had no impacts on physicochemical and textural properties of meat emulsions, regardless of fat level at 5%, 10% and 20%.

A Study on Adsorption of Heavy Metal Ions Using Chitosan and Chitosan Derivative (Chitosan 및 Chitosan유도체를 이용한 중금속 이온 흡착에 관한 연구)

  • We have synthesized the water-insoluble chitosan derivative, N-dithiocarboxy chitosan sodium salt, through the reaction of chitosan with carbon disulfide in the presence of alkali metal hydroxide, Chitosan itself has been prepared using chitin, one of the most abundant compounds in nature, as a starting material. To elucidate this natural polymer the capacity of adsorbing heavy metal ions, we have performed adsorption experiments using chitosan derivatives of various average molecular weights with different contents of sulfur. The effect of pH, adsorption time and temperature on adsorption efficiency was also studied. The adsorbent derived from chitosan of average molecular weight ranging $5,700{\sim}20,000$ was shown to have the highest capacity of adsorbing heavy metal ions. Adsorbing efficiency was increased as the reaction time was increased and as the reaction temperature range of $25{\sim}45^{\circ}C$. The adsorption capacity at various pH, however, appeared to vary depending on the heavy metal ions studied.

Effects of alkali solutions on corrosion durability of geopolymer concrete

  • This paper presents chloride induced corrosion durability of reinforcing steel in geopolymer concretes containing different contents of sodium silicate ($Na_2SiO_3$) and molarities of NaOH solutions. Seven series of mixes are considered in this study. The first series is ordinary Portland cement (OPC) concrete and is considered as the control mix. The rest six series are geopolymer concretes containing 14 and 16 molar NaOH and $Na_2SiO_3$ to NaOH ratios of 2.5, 3.0 and 3.5. In each series three lollypop specimens of 100 mm in diameter and 200 mm in length, each having one 12 mm diameter steel bar are considered for chloride induced corrosion study. The specimens are subjected to cyclic wetting and drying regime for two months. In wet cycle the specimens are immersed in water containing 3.5% (by wt.) NaCl salt for 4 days, while in dry cycle the specimens are placed in open air for three days. The corrosion activity is monitored by measuring the copper/copper sulphate ($Cu/CuSO_4$) half-cell potential according to ASTM C-876. The chloride penetration depth and sorptivity of all seven concretes are also measured. Results show that the geopolymer concretes exhibited better corrosion resistance than OPC concrete. The higher the amount of $Na_2SiO_3$ and higher the concentration of NaOH solutions the better the corrosion resistance of geopolymer concrete is. Similar behaviour is also observed in sorptivity and chloride penetration depth measurements. Generally, the geopolymer concretes exhibited lower sorptivity and chloride penetration depth than that of OPC concrete. Correlation between the sorptivity and the chloride penetration of geopolymer concretes is established. Correlations are also established between 28 days compressive strength and sorptivity and between 28 days compressive strength and chloride penetration of geopolymer concretes.

The effect of pH, adsorption time and temperature on adsorption efficiency was also studied. The adsorbent derived from water-soluble chitosan of average molecular weight ranging $9,000{\sim}120,000$ was shown to have the highest capacity of adsorbing heavy metal ions. On the whole, adsorbing efficiency was increased as the reaction time goes longer and also increased as the reaction temperture goes higer in temperture range of $15^{\circ}C{\sim}45^{\circ}C$. The adsorption capacity at various pH, however, was appeared to vary depending on the heavy metal ions studied Judging from these finding, water-soluble N-dithiocarboxy chitosan sodium salt, a derivative of a biodegradable nature polymer, is believed to be a potential adsorbent for heavy metal ions since it not only is shown to lower the concentration of heavy metal ions to below the drainage quality standard, but also it would not cause acidification and hardening of soil which is one of the detrimental effects of synthetic macromolecular adsorbents present.

A Comparison Study on Physicochemical Properties of Two Small Red Bean ( Black and Red ) Starches and Gels (거두와 적두전분의 이화학적 특성 및 Gel 특성에 관한 연구)

  • This study has been carried out in order to investigate the physicochemical properties of two small red bean starches. Some of rheological properties of the starch gels were also studied by experiments of various starch concentrations. Water binding capacity of black bean starch was 172.3% and that of red bean starch was 199.0%. Black bean starch had lower swelling power than red bean starch, but the solubility of the black bean starch was higher. When the temperature increased from 60$^{\circ}C$ to 70$^{\circ}C$, the transmittance of two starches rapidly increased. The gelatinized temperature in DSC for black bean was 66.2$^{\circ}C$ and that for red bean was 66.0$^{\circ}C$. Black bean and red bean starches had the blue vlaues of 0.55 and 0.56 and the alkali numbers of 4.40 and 4.13. The molecular weight of amylose was 40,000 and 33,611. The amylose contents of two starches were same at 52%. Brabender Amylographs of two small red bean starch pastes showed C pattern, which is stable. The results of compression test pointed out that TPA parameters varied with the change of storage time, and black bean starch gels had the higher TPA value. The retrogradation study by glucoamylase digestion method revealed that red bean starch gels were more easily retrogradated than black bean. X-ray diffraction patterns of two small red bean starches were A pattern, and diffraction peaks disappeared with gelatinization of starches.
