에탄올에 의해 유발된 간독성에 대한 모과 추출물의 보호효과 (Protective Effects of Chaenomeles sinensis Koehne Extract on Ethanol-induced Liver Damage in Rat)

  • 모과 에탄올 추출물이 체내 항산화계에 미치는 효과를 구명하기 위하여 in vivo에서 모과 에탄올 추출물이 알코올 투여로 유도된 간조직 손상의 보호효과를 검토한 결과 4주간의 체중증가율과 체중에 대한 간 무게 비율은 유의적인 차이가 나타나지 않았지만, ET군이 다른 실험군에 비하여 다소 낮은 경향이었다. 식이효율은 ET군이 ND군에 비하여 유의하게 감소되었으나 알코올과 모과 에탄올 추출물을 단독 혹은 병합투여한 ET-CSL군과 ET-CSH군은 ND군의 식이효율과 비슷한 수치를 나타내었다. 혈청 중 ALT, AST 및 ALP 활성은 알코올 투여한 ET군이 실험군들에 비하여 유의적으로 증가하였고, 알코올과 모과 에탄올 추출물을 병합 투여한 ET-CSL군과 ET-CSH군은 ET군에 비하여 저하되었다. 간조직 중 과산화지질 함량은 ET-CSL군과 ET-CSH군이 ET군에 비해 유의적으로 감소하였으며, ND군과는 비슷한 수치를 나타내었다. 간조직의 XO, SOD, catalase 및 GSH-Px활성은 알코올을 단독 투여한 ET군이 알코올과 모과 에탄올 추출물을 병합 투여한 ET-CSL군과 ET-CSH 군에 비하여 증가하였으며, 모과 에탄올 추출물 투여 용량이 증가할수록 이들 효소의 활성도 농도 의존적으로 감소하였다. 또한 ET-CSL군과 ET-CSH군은 ND군과 이들 효소활성이 유사하였다. 간조직의 GSH 함량은 ET군이 ND군에 비하여 유의하게 감소되었으며, 알코올 투여로 감소된 GSH 함량은 모과 에탄을 추출물 투여로 ND군에 근접하도록 증가시켰다. 이상의 결과 모과 에탄올 추출물은 알코올 투여로 증가된 유리기 해독계 효소인 SOD, catalase 및 GSH-Px 활성은 억제시키고, GSH 함량은 증가시켰으며 지질과산화물 함량을 저하시켰다. 따라서 모과는 항산화제로서의 활용성과 알코올 투여로 손상된 간조직의 보호에 도움을 줄 수 있을 것이라는 가능성을 제시해 주었다.성으로 Rg1 함량은 잎의 80% 메탄올 추출물에 $44.15{\mu}g/g$이었고, Re 함량은 잎의 80% 메탄올 추출물에 $48.78{\mu}g/g$으로 가장 많았으며, Rd 함량은 잎의 80% 메탄올 추출물에 $25.57{\mu}g/g$으로 가장 많았고, Rc 함량은 잎의 80% 메탄을 추출물에 $4.87{\mu}g/g$으로 가장 많았다. 항산화활성은 잎의 물 추출물이 83.82%이었으며, 잎의 80% 에탄올 추출물이 89.74%이었으며, 잎의 80% 메탄을 추출물이 88.37%의 활성을 나타내었다.의 순이었다. 7. 액비저장조 이용주체는 전체 농가의 59.0%인 240농가가 자체적으로 이용을 하고 있었으며, 23.1%인 94농가는 액비 유통업체에 위탁하여 이용하고 있었고, 5.2%인 21농가는 방치하고 있는 것으로 나타났다. 경종 농가가 부담하는 ha당 액비살포 비용은 미부담 62.7%, 1만원 이상 16.2%, $4{\sim}5$천원 9.8%, $6{\sim}7$천원 9.6%, $8{\sim}9$천원 1.7%로 나타났다. 8. 경종농가의 액비사용전 액비시비처방서 발급은 54.1%인 220농가만이 의뢰한 것으로 나타났고, 41.5%인 169농가는 의뢰하지 않은 것으로 나타났다. 액비사용에 대한 사전 교육은 62.4%인 254농가가 교육을 받은 것으로 나타났으며, 30.2%인 123농가는 받지 않은 것으로 나타났고, 교육 자체가 없어서 못 받았다는 농가도 3.9%인 16농가로 나타났다. 9. 액비사용시 문제점은 전체 농가중 절반 이상인 220농가(54.1%)가 악취로 인한 문제라고 하였으며, 22.1%인 90농가는 액비를 뿌릴 수 있는 장비가 부족하다고 하였고, 사용 불편 14.5%(59농가), 과다살포 3.4%(14농가), 비용과다

Effect of Aging on the Xylene Metabolism in $CCl_4$-Induced Liver Damaged Rats

  • To investigate an effect of aging on the xylene metabolism in liver damaged animals, a study was conducted. 50% carbon tetrachloride ($CCl_4$) in olive oil (0.1 ml/100 g body weight) was intraperitoneally given to 5-week and 12-week rats 12 times every other day and then one dose of 50% xylene in olive oil (0.25 ml/100 g body weight) was intraperitoneally given to the rats, and after 24 hr, the animals were sacrificed. On the basis of the functional findings in rat liver, ie, serum levels of alanine aminotransferase activity, liver protein and malonedialdehyde contents, 5-week rats showed less liver damage than 12-week rats. The increasing rate of urinary methylhippuric acid concentration to the control was significantly higher in 5-week rats than 12-week rats in case of xylene treatment after induction of liver damage. On the other hand, liver damaged 5-week rats showed significant rise of hepatic cytochrome P45O content compared with the liver damaged 12-week rats by the xylene treatment. And increasing rate of hepatic alcohol or aldehyde dehydrogenase activities to each liver damaged animals was higher tendency in 5-week rats than 12-week rats by the xylene treatment. In conclusion, 5-week rat showed greater metabolic rate of xylene than 10-week rats in case of liver injury because 5-week rats led to a slight liver damaged compared with 12-week rats.

Mature silkworm powder reduces blood alcohol concentration and liver injury in ethanol-treated rats

  • Hangover due to alcohol consumption causes social and physical problems. There is a growing interest in edible insects worldwide. We have previously published a new technology to make hard mature silkworm, Bombyx mori, into edible form, steamed and freeze-dried mature silkworm larval powder (SMSP). In this study, AIN-76 or SMSP (0.1 and 1 g/kg rat body weight) containing diets in SD rats were pretreated for 2 weeks, and ethanol (3 g/kg rat body weight) was administered as an oral gavage and sacrificed after 3 hours. As a result, blood alcohol and aldehyde levels were significantly decreased in SMSP fed rats. In addition, liver injury markers, alanine aminotransferase (ALT) and alkaline phosphatase (ALP) were significantly decreased in SMSP group compared to ethanol group. $TNF-{\alpha}$, an inflammatory cytokine, and malondialdehyde (MDA), an oxidative stress marker, also showed a dose-dependent decrease in the group receiving SMSP. Conclusively, consumption of SMSP not only reduced hangover induced by ethanol, but also decreased liver damage, oxidative stress, and inflammatory response.

알코올성 지방간을 유발시킨 마우스에서 유산균 발효 마늘추출물의 간 보호 효과 (Hepatoprotective Effects of Lactic Acid-Fermented Garlic Extracts on Fatty Liver-Induced Mouse by Alcohol)

  • 본 연구는 Lactobacillus plantarum으로 발효시킨 유산균 발효 마늘추출물의 간 손상 보호 효과를 알코올성 지방간을 유발시킨 C57BL/6J 마우스를 사용하여 확인하고자 하였다. 유산균 발효 마늘추출물은 일반식이군에 비해 지방간 유도와 함께 증가한 간 무게를 용량 의존적으로 감소시켰다. 유산균 발효 마늘추출물 200 mg/kg 투여군에서 대표적 간 손상 관련 혈청효소인 ALT, AST 및 ALP의 활성이 낮았으며, 간 손상의 다른 중요한 지표들인 TBIL, DBIL 값도 현저히 낮았다. 또한 유산균 발효 마늘추출물은 간의 조직학적 관찰에서도 알코올로 유도된 간 지방의 축적을 농도 의존적으로 개선하였다. 간 조직 내 GSH 함량을 측정한 결과, 알코올에 의해 감소된 GSH가 유산균 발효 마늘추출물 투여에 의해 점차 일반식이군 수준으로 회복됨을 알 수 있었다. 이상의 연구 결과는 유산균 발효에 의해 마늘의 냄새와 맛을 개선시킨 유산균 발효 마늘추출물이 알코올성 지방간의 예방 및 개선을 위한 기능성식품용 상업적 소재로서 높은 실용화 가능성을 시사한다.

Chronic Alcohol Consumption Results in Greater Damage to the Pancreas Than to the Liver in the Rats

  • Alcohol consumption increases the risk of type 2 diabetes. However, its effects on prediabetes or early diabetes have not been studied. We investigated endoplasmic reticulum (ER) stress in the pancreas and liver resulting from chronic alcohol consumption in the prediabetes and early stages of diabetes. We separated Otsuka Long-Evans Tokushima Fatty (OLETF) rats, a type-2 diabetic animal model, into two groups based on diabetic stage: prediabetes and early diabetes were defined as occurrence between the ages of 11 to 16 weeks and 17 to 22 weeks, respectively. The experimental group received an ethanol-containing liquid diet for 6 weeks. An intraperitoneal glucose tolerance test was conducted after 16 and 22 weeks for the prediabetic and early diabetes groups, respectively. There were no significant differences in body weight between the control and ethanol groups. Fasting and 120-min glucose levels were lower and higher, respectively, in the ethanol group than in the control group. In prediabetes rats, alcohol induced significant expression of ER stress markers in the pancreas; however, alcohol did not affect the liver. In early diabetes rats, alcohol significantly increased most ER stress-marker levels in both the pancreas and liver. These results indicate that chronic alcohol consumption increased the risk of diabetes in prediabetic and early diabetic OLETF rats; the pancreas was more susceptible to damage than was the liver in the early diabetic stages, and the adaptive and proapoptotic pathway of ER stress may play key roles in the development and progression of diabetes affected by chronic alcohol ingestion.

Effects of Extrahepatic Cholestasis on Hepatic $\alpha$-D-Mannosidase Activity in Chronic Ethanol Intoxicated Rats

  • Hepatic subcellular $\alpha$-D-mannosidases activities and its Km and Vmax values were determined in chronic ethanol intoxicated rats with extrahepatic cholestasis induced by common bile duct ligation to manifest the biochemical background of alcohol drinking hazard under the hepatobiliary disease. In case of extrahepatic cholestasis, chronic ethanol intoxication in animals led to the increased activities of liver Golgi and microsomal $\alpha$-D-mannosidase as well as the Vmax values of these enzymes. However, the difference of Km values on hepatic subcellular enzymes were not found between the experimental groups. Therefore, the results indicate that the liver Golgi and microsomal $\alpha$-D-mannosidase may be more induced in chronic ethanol intoxication animals in case of cholestasis. Accordingly, the resulting data supported the fact that alcoholic drinks may led to enhancement of the hepatobiliary liver damage.

Cytoprotective effect of polysaccharide isolated from different mushrooms against 7-ketocholesterol induced damage in mouse liver cell line (BNL CL. 2)

  • Cytoprotective ability of polysaccharides isolated from different edible mushrooms was investigated on the 7-ketocholesterol-induced damaged cell line. Polysaccharide extracts from six different edible mushrooms-Flammulina velutipes, Peurotus ostreatus, Lentinus edodes, Agrocybe aegerita, Agaricus blazei, and Cordyceps militaris- were prepared by hot water extraction and alcohol precipitation. Cytoprotective ability was evaluated by measuring the viable cells of the normal embryonic liver cell line (BNL CL. 2) in the presence of 7-ketocholesterol. At $80\;{\mu}g/mL$ of 7-ketocholesterol, cytotoxicity was very high with a loss of 98% of viable cells after 20 h of incubation. With the addition of $200\;{\mu}g/mL$ of each polysaccharide isolate to the cell line containing $80\;{\mu}g/mL$ of 7-ketocholesterol, polysaccharide isolates from both Flammulina velutipes and Peurotus ostreatus could significantly inhibit the 7-ketochoelsterol-induced cytotoxicity in the cells. But other polysaccharide isolates were not effective in inhibiting cell damage caused by the oxLDL-induced cytotoxicity.

Effect of probiotics Lactobacillus paracasei GKS6, L. plantarum GKM3, and L. rhamnosus GKLC1 on alleviating alcohol-induced alcoholic liver disease in a mouse model

  • BACKGROUND/OBJECTIVES: Heavy alcohol consumption causes the development of alcoholic liver disease (ALD), a neglected but important public health problem. Many studies have pointed out that probiotics could improve gut health, which is also considered to be a cause of ALD. Therefore, this study screened the probiotics, Lactobacillus casei GKC1 (GKC1), L. fermentum GKF3 (GKF3), Bifidobacterium lactis GKK2 (GKK2), L. rhamnosus GKLC1 (GKLC1), L. paracasei GKS6 (GKS6), and L. plantarum GKM3 (GKM3), for their potential benefits in alleviating ALD for applications to disease prevention. SUBJECTS/METHODS: C57BL/6N mice were divided into 8 groups (n = 6 in each): normal control, positive control (alcohol-diet fed), and treatments of feeding probiotics GKC1, GKF3, GKK2, GKLC1, GKS6, and GKM3 under an oral dose 0.82 g/kg B.W. per day by oral gavage. The experiment was conducted for 8 weeks, and the concentrations of alanine aminotransferase (ALT), aspartate aminotransferase, triglyceride (TG), and total cholesterol (TC) in mice were measured. The glutathione (GSH), catalase (CAT), and histology were analyzed after sacrifice. RESULTS: The results showed a decrease in the serum ALT, liver TG, and liver TC levels in the GKS6, GKM3, and GKLC1 groups compared to the positive control. In addition, the decreasing GSH and CAT levels were inhibited in the GKS6 and GKM3 groups. The histopathological results showed that all probiotics could reduce the accumulation of liver fat. Furthermore, there was a significant difference in GKLC1 with lower stomach damage compared to the alcohol-fed mice without any addition of probiotics. CONCLUSIONS: GKLC1, GKS6, and GKM3 can be used as supplements for alleviating the development of ALD.

밭미나리 발효액의 항산화 활성과 흰쥐에서 알코올성 간 손상 보호효과 (Antioxidant Activity of Dropwort (Oenanthe javanica DC) Fermented Extract and its Hepatoprotective Effect against Alcohol in Rats)

  • Antioxidant activity of dropwort fermented extract (DFE) was measured according to fermentation period, and liver protective effects were examined using Sprague-Dawley rats. Total polyphenol and flavonoid contents as well as DPPH and ABTS radical scavenging activities increased up to 60~80 days and then decreased slightly. Proper fermentation time for DFE was more than 60 days and less than 80 days. Administration of alcohol to rats for 10 days at 10 mL/kg/day raised serum AST, ALT, total cholesterol, and triglyceride (TG) levels, which were then lowered by DFE and sugar liquid with the same soluble solids. While sugar liquid increased the blood lipid profile, especially TG levels, DFE had no effect due to its antioxidant activity. When TBARS content of the DFE group in liver tissue significantly decreased in a concentration-dependent manner compared to that of the ALH group (p<0.05). Liver damage was recovered by DFE treatment and was confirmed by hamatoxylin-eosin staining. These results suggest that DFE has a protective effect against alcohol-induced hepatotoxicity in SD rats.

The hepato-protective effect of eupatilin on an alcoholic liver disease model of rats

  • Eupatilin is known to possess anti-apoptotic, anti-oxidative, and anti-inflammatory properties. We report here that eupatilin has a protective effect on the ethanol-induced injury in rats. Sprague-Dawley rats were divided into 6 groups: control, vehicle, silymarin, eupatilin 10 mg/kg, eupatilin 30 mg/kg, and eupatilin 100 mg/kg. Plasma levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were analyzed to determine the extent of liver damage. Total cholesterol (TC) and triglycerides (TG) were analyzed to determine the level of liver steatosis. Malondialdehyde level, superoxide dismutase (SOD) activity, and glutathione (GSH) level were analyzed to determine the extent of oxidative stress. Tumor necrosis factor (TNF)-α and interleukin (IL)-1β were quantified to verify the degree of inflammation. Based on our findings, chronic alcohol treatment significantly changed the serum indexes and liver indicators of the model rats, which were significantly improved by eupatilin treatment. Rats in the eupatilin-treatment group showed reduced levels of AST, ALT, TG, TC, TNF-α, and IL-1β, increased SOD activity and GSH levels, and improved overall physiology compared to the alcoholic liver disease model rats. H&E staining also verified the eupatilin-mediated improvement in liver injury. In conclusion, eupatilin inhibits alcohol-induced liver injury via its antioxidant and anti-inflammatory effects.
