(-) Epigallocatechin gallate restores ethanol-induced alterations in hepatic detoxification system and prevents apoptosis

  • The present study was designed to estimate the protective effect of (-) epigallocatechin gallate (EGCG) on ethanol-induced liver injury in rats. Chronic ethanol administration (6 g/kg/day ${\times}$ 60 days) caused liver damage that was manifested by the elevation of markers of liver dysfunction - aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, bilirubin and ${\gamma}$-glutamyl transferase in plasma and reduction in liver glycogen. The activities of alcohol metabolizing enzymes such as alcohol dehydrogenase and aldehyde dehydrogenase were found to be altered in alcohol-treated group. Ethanol administration resulted in the induction of cytochrome p450 and cytochrome-$b_{5}$ activities and reduction of cytochrome-c reductase and glutathione-S-transferase, a phase II drug metabolizing enzyme. Further, ethanol reduced the viability of isolated hepatocytes (ex vivo) as assessed by trypan blue exclusion test and induced hepatocyte apoptosis as assessed by propidium iodide staining. Treatment of alcoholic rats with EGCG restored the levels of markers of liver injury and mitigated the alterations in alcohol metabolizing and drug metabolizing enzymes and cyt-c-reductase. Increased hepatocyte viability and reduced apoptotic nuclei were observed in alcohol + EGCG-treated rats. These findings suggest that EGCG acts as a hepatoprotective agent against alcoholic liver injury.

Hypoglycemic Effect and Hepatic Detoxification Activity of Extracts from Crataegus fructus and Morus alba L. in Alcohol-treated Rats

  • To consider potentially new sources which have hypoglycemic effect and accelerating alcohol detoxification, this study was designed to investigate the effect of Crataegus fructus and Morns alba L. in alcohol-treated rats. I compared the body weight, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alcohol dehydrogenase (ADH), and aldehyde dehydrogenase (ALDH) of rats administered both alcohol and extract of experimental plants to rats treated with alcohol alone. Administration of extracts of C. fructus and M alba, respectively, resulted in a significant reduction in the blood glucose level and the activities of ADH of liver compared to the control rats, and administration of extract of M. alba showed significantly lower on bodyweight gain in the rats than in other treated rats. In contrast, the activities of ALDH of liver were increased. The activities of AST and ALT between the only alcohol-treated rats and the alcohol and experimental plants-treated rats were no significant difference. The results suggest that C. fructus and M alba have a hypoglycemic effect, and reduce liver damage by accelerating acetaldehyde metabolism in alcoholic rats, so the combined effect of C. fructus and M alba may be considered as an alternative remedy for hangovers, alcohol-induced overweight and alcohol-induced diabetes.

알코올성 간손상을 유발한 마우스 모델에서의 새싹인삼 지상부 추출물의 간 기능 보호효과 및 지표성분 함량분석 (Protective effects of extracts from the aerial parts of hydroponically cultured ginseng on alcohol-induced liver damage in mice and quantitative analysis of major ginsenosides)

  • 알코올성 간손상을 유발한 마우스 모델에서 새싹인삼 지상부 추출물(HGE)의 간 보호 효과를 확인하였다. 알코올성 간손상 모델은 25% 알코올을 마우스에 5 g/kg의 농도로 경구 투여하여 이루어졌고, HGE를 투여한 군에는 알코올을 투여하기 3일전부터 경구투여를 시작하여 10일간 마우스 개체 당 4와 12 mg/kg의 농도로 경구투여 하였다. HGE를 투여한 그룹에서는 알코올만 처리한 대조군에 비해 AST 및 ALT 수치가 농도의존적으로 낮아진 것을 확인하였다. 또한, 간손상에 의해 증가된 LDH 수치는 대조군과 유사하게 감소하였다. 새싹인삼 지상부 추출물로 부터 ginsenoside F5, F3, F1, 및 F2의 정량분석 결과는 각각 2.5, 4.4, 1.4, 및 23.3 mg/g으로 확인하였다.

  • Red rice Yeasts는 홍국 또는 붉은 코지로 불리며 오래전부터 아시아에서 기능성 식품 친 전통의학에서 사용되고 있다. 홍국쌀에는 여러가지 생리 활성 물질을 포함하여 기능적인 2차대사산물과 6가지 유형의 색소를 생성한다. 간 질환의 주요한 원인인 알코올로 인한 알코올성 간 질환(ALD)은 단순 지방증 또는 염증으로 섬유증 및 간경변에 이르기까지 다양할 수 있으며, 장 누수 및 전신 내독소혈증을 유발하여 장장벽을 파괴하고 장내 미생물 변화를 증가시킨다. 본 연구는 홍국쌀(Monascus purpureus; RYR) 에탄올 추출물이 과량의 에탄올로 유발된 간 및 장 손상 마우스모델에서 간 및 장에 미치는 영향을 조사하였다. RYR 추출물을 10일 동안 200 mg/kg 체중/일의 농도로 C57/BL/6N 마우스에 경구 투여한 후 과량의 알코올 (5g/kg/용량)을 12시간 간격으로 3회 처리하였다. 과량의 알코올 노출은 혈장의 내독소, AST 및 ALT 활성과 간 트리글리세리드 수치를 유의적으로 증가시켰지만 RYR 처리는 이들 수치를 감소시켰다. 또한, RYR 전처리는 과량의 알코올 유발 장누수 및 간 손상에서 알코올 유발 산화 인자 단백질 및 세포사멸 인자를 유의하게 감소시켰다. 이러한 결과는 RYR이 알코올로 인한 급성 장누수 및 간 손상을 예방할 수 있음을 시사한다.

Effect of a Static Magnetic Field on Susceptibility to Ethanol-Induced Hepatic Dysfunction in Rats

  • To determine whether alcohol-treated rat liver cells can be protected by a static magnetic field (SMF), we analyzed the blood chemistry and histology of hepatic tissue removed from alcohol-exposed rats that had been exposed to a static magnetic field. The rats were exposed to a 0.3 tesla (3,000 gauss) magnetic field (MF) for 24 hr daily for 5 weeks with appropriate controls. Glutamic pyruvic transaminase activity and the triglyceride levels in animals exposed to the north (N) or south (S) pole of the MF decreased significantly (p<0.01 and p<0.05, respectively) compared with negative control animals with alcohol exposure. A histological examination of hepatic tissue revealed a moderate to severe accumulation of fat vacuoles of various sizes in the cytoplasm of the hepatocytes of animals in the negative control group throughout the study; whereas in groups exposed to the MF poles, fewer fat vacuoles were seen compared with the negative control group. Electron microscopic observations showed that exposure to the N or S pole protected organelles, including the nucleus, from damage during exposure to this toxic agent, as indicated by the fact that the nucleus and the mithochondria virtually retained their shape throughout this study. These results suggest that exposure to a SMF could be an excellent way of protecting against alcohol-induced damage to the rat liver cell.

Pretreatment of Albino Rats with Methanolic Fruit Extract of $Randia$ $Dumetorum$ (L.) Protects against Alcohol Induced Liver Damage

  • Alcohol abuse and its medical and social consequences are a major health problem in many areas of the world. The present study was conducted to evaluate the protective effect of methanolic fruit extract of $Randia$ $Dumetorum$ (L.) on alcohol-induced liver damage in rats. Rats were divided into five different groups (n=6), group I served as a control, group II received ethanol (3 ml/100 g/day p.o.), group III served as standard group and received silymarin (50 mg/kg p.o.), group IV and V served as extract treatment groups and received 50 & 100 mg/kg methanolic extract of $R.$ $dumetorum$. All the treatment protocols followed 30 days and after rats were sacrificed blood and liver were used for biochemical and histological studies, respectively. The activities serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), direct bilirubin (DB), total bilirubin (TB) and lipid peroxidation were statistically increased in rats exposed to alcohol while total protein and glutathione decreased compared to control rats. Treatment with $R.$ $dumetorum$ significantly decreased the elevated levels of ALT, AST, TG, DB, TB and lipid peroxidation compared to the group exposed to alcohol only. $R.$ $dumetorum$ significantly resulted in increased levels of total protein and reduced glutathione compared to the group that received alcohol only. Histology of the liver section of the animals treated with $R.$ $dumetorum$ improved the hepatotoxicity caused by alcohol. Hence the study concluded that $R.$ $dumetorum$ has potential hepatoprotective activity.

Molecular Links between Alcohol and Tobacco Induced DNA Damage, Gene Polymorphisms and Patho-physiological Consequences: A Systematic Review of Hepatic Carcinogenesis

  • Chronic alcohol and tobacco abuse plays a crucial role in the development of different liver associated disorders. Intake promotes the generation of reactive oxygen species within hepatic cells exposing their DNA to continuous oxidative stress which finally leads to DNA damage. However in response to such damage an entangled protective repair machinery comprising different repair proteins like ATM, ATR, H2AX, MRN complex becomes activated. Under abnormal conditions the excessive reactive oxygen species generation results in genetic predisposition of various genes (as ADH, ALDH, CYP2E1, GSTT1, GSTP1 and GSTM1) involved in xenobiotic metabolic pathways, associated with susceptibility to different liver related diseases such as fibrosis, cirrhosis and hepatocellular carcinoma. There is increasing evidence that the inflammatory process is inherently associated with many different cancer types, including hepatocellular carcinomas. The generated reactive oxygen species can also activate or repress epigenetic elements such as chromatin remodeling, non-coding RNAs (micro-RNAs), DNA (de) methylation and histone modification that affect gene expression, hence leading to various disorders. The present review provides comprehensive knowledge of different molecular mechanisms involved in gene polymorphism and their possible association with alcohol and tobacco consumption. The article also showcases the necessity of identifying novel diagnostic biomarkers for early cancer risk assessment among alcohol and tobacco users.

헛개나무 열매를 주성분으로 제조한 새로운 처방이 알코올 분해 및 간 손상에 미치는 영향 (Effect of New Remedies Mainly Comprised of Hovenia dulcis Thunb on Alcohol Degradation and Liver Protection in Sprague Dawley Male Rats)

  • 본 연구에서는 헛개나무열매를 주성분으로 제조한 두 종류의 혼합물들(NHDT-1과 NHDT-2)이 알코올 및 아세트알데히드의 분해를 촉진시켜 숙취의 발생을 저하시킬 수 있는지 여부와 만성적으로 알코올을 복용시켰을 때 간손상 및 지방간의 발생을 억제할 수 있는지 여부를 조사하였다. NHDT-1은 헛개나무열매에 진피, 창출 및 감초를 첨가하여 제조하였고, NHDT-2는 NHDT-1에 홍삼을 첨가하여 제조하였다. NHDT-1과 NHDT-2의 조성 차이는 홍삼임에도 불구하고 백서에게 투여하였을 때 알코올 및 아세트알데히드의 분해와 간손상을 억제하는 작용에는 차이가 있었다. 홍삼이 함유되어있지 않은 NHDT-1은 급성 알코올 실험에서 알코올 및 아세트알데히드 분해를 촉진시켜 혈청농도를 저하시켰으나, 홍삼이 함유된 NHDT-2는 혈청 알코올과 아세트알데히드 농도를 저하시키지 못했다. 급성실험과는 반대로 장기간 알코올을 투여한 실험에서는 NHDT-1은 NHDT-2에 비해 알코올로 인한 간의 손상과 지방간 발생을 억제하지 못했다. 결론적으로 간기능이 좋은 상태에서 일시적으로 복용하는 경우에는 NHDT-1이 알코올과 아세트알데히드의 분해를 촉진하여 숙취해소에 효과적이지만, 간이 좋지 않은 상태에서는 장기간동안 NHDT-2를 복용하여 간의 기능을 향상시키는 것이 음주 후 숙취제거에 효과적일 것으로 사료된다.

Involvement of Hepatic Innate Immunity in Alcoholic Liver Disease

  • Excessive alcohol consumption is one of the critical causative factors leading to alcoholic liver disease (ALD). ALD is characterized by a wide spectrum of liver damage, ranging from simple uncomplicated liver steatosis (fatty liver) to steatohepatitis and liver fibrosis/cirrhosis. It has been believed that the obvious underlying cause for ALD is due to hepatocyte death induced by alcohol itself. However, recent sparkling studies have shown that diverse immune responses contribute to ALD because liver is enriched with numerous immune cells. Especially, a line of evidence has suggested that innate immune cells such as Kupffer cells and natural killer (NK)/NKT cells are significantly involved in the pathogenesis of ALD via production of pro-inflammatory cytokines and other mediators. Indeed, more interestingly, hepatic stellate cells (HSCs), known as a major cell inducing liver steatosis and fibrosis, can be killed by liver NK cells, which could be suppressed by chronic alcohol consumption. In this review, with the view of liver as predominant innate immune organ, we describe the pathogenesis of ALD in which what roles of innate immune cells are and how they are interacting with HSCs.

Citrus junos Fractions Decrease Alcohol-induced Liver Damage and Influence Lipid Metabolism in Alcohol-fed Rats

  • The effect of treatment with Citrus junos fractions (citron 3W, citron 3H, citron 4W and citron 4H) upon rat hepatocytes exposed to alcohol was investigated. We compared the serum biochemistry of rats administered both alcohol and Citrus junos fractions to control rats treated with alcohol alone. The effects of Alanine amino transferase (ALT) were significantly lower in the citron 3H extract group compared with the negative control group (p<0.05) and other experimental groups were not significantly low but a little low compared to negative control group. The levels of triglyceride (TG) were significantly low in all experimental groups compared with negative control group. Especially triglyceride level of citron 3H was lowest near to normal control group. The concentration of total cholesterol was significantly high in negative groups compared with normal control group but in all experimental groups, the concentration of total cholesterol was similar to that of negative control group. Total cholesterol of the citron 4W group was somewhat low compared with negative control group. In contrast, activities of alcohol dehydrogenase (ADH) were significantly higher in all experimental groups compared with the negative control (p<0.05) group. These data suggest that Citrus junos fractions may represents an excellent candidate for protection of rat hepatocytes from alcohol-mediated damage.
