Urinary albumin excretion rate and puberty in non-diabetic children and adolescents

  • Slightly elevated urinary albumin excretion rate (microalhuminuria) is a marker of early diabectic nephropathy, but it is unclear if the established definition of microalbuminuria ($20-200{\mu}g/min$) is correct for children and adolescents. We investigated th.: albumin excretion rate, albumin/creatinine ratio and urinary albumin concentration in 150 healthy schoolchildren and adolescents to (a) obtain a reterence value for albumin excretion rate, (b) relate albumin excretion to pubertal stages and (c) evaluate albumin/creatinine ratio and morning albumin concentration as screening methods for elevated albumin excretion rate. Albumin concentration was measured by immunoturbidimetry in timed overnight urine samples. The albumin excretion showed a skewed distribution (geometric mean $3.2{\mu}g/min$, 95 percentile ($15.1{\mu}g/min$). In girls. a peak in the albumin excretion rate was found at the pubertal stage 4 (Tanner) and in boys at stage 5. Albumin/creatinine ratio of 2.5 mg/mmol as a screening level for elevated albumin excretion ($15{\mu}g/min$) showed a high positive (0.88) and negative (0.99) predictive value.

  • In attempt to develop a drug delivery system using serum albumin microspheres, bovine serum albumin microspheres containing antitumar agent. Cytarabine, were prepared. The shape, surface characteristics, size distribution, behavior of in vivo distribution, drug release behavior, and degradation of albumin microsphers in animal liver issue homogenate and proteolytic enzyme were investigated. The shape of albumin microspheres was spherical and the surface was smooth and compact. The size distribution of the albumin microspheres was effected by dispertion forces during emulsification and albumin concentration. Distribution of albumin microspheres after imtravenous administration in rabbit was achieved immediately. In vitro, albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin concentration ratio and size distribution. After drug release test, the morphology of albumin microspheres was not changed. Albumin microsphere matrix was degraded by the animal liver issue homogenate and proteolytic enzyme.   • Bovine serum albumin microspheres containing cytarabine were prepared using cross-linking agent, formaldehyde. The shape and the size distribution of them were observed. The shape of them was spherical and the susrface was compact and smooth. The size distribution of them was affected by dispersion forces during emulsfication. The release of cytarabine from albumin microspheres was dependent upon cross-linking time, amount of cross-linking agent and drug/albumin ratio. However, the difference of drug release by the dispersion forces was not great. After release test, the shape of albumin microspheres was nonspherical and the albumin matrix seemed to be a little relaxed. The degradation tests of albumin microspheres by the proteolytic enzyme showed that albumin microspheres were progressively digested according to the cross-linking degree.

  • In ethanol fermentations with Saccharomyces sake, phosphatidylcholine-egg albumin as a supplement in fermentation media was much more effective in enhancing ethanol production than linoleic acid-ergosterol. It came from the differences in alcohol-tolerance between egg albumin and ergosterol. The egg albumin was supposed to function as a nutrient rather than to form protective layers around the cells against ethanol.

  • Over the years, nanoparticle drug delivery systems have demonstrated versatile potentials in biological, medical and pharmaceutical applications. In the pharmaceutical industry nanotechnology research has mainly focused on providing controlled drug release, targeting their delivery to specific organs, and developing parenteral formulations for poorly water soluble drugs to improve their bioavailability. Achievement in polymer industry has generated numerous polymers applicable to designing nanoparticles. From viewpoints of product development, a nanocarrier material should meet requirements for biodegradability, biocompatibility, availability, and regulatory approval crieteria. Albumin is indeed a material that fulfills such requirements. Also, the commercialization of a first albumin-bound paclitaxel nanoparticle product (Abraxane$^{TM}$) has sparked renewed interests in the application of albumin in the development of nanoparticle formulations. This paper reviews the intrinsic properties of albumin, its suitability as a nanocarrier material, and albumin-based parenteral formulation approaches. Particularly discussed in detail are albumin-based particulate injectables such as Abraxane$^{TM}$. Information on key roles of albumin in the nab$^{TM}$ technology and representative manufacturing processes of albumin particulate products are provided. It is likely that albumin-based particulate technology would extend its applications in delivering drugs, polypeptides, proteins, vaccines, nucleic acids, and genes.

Heterogeneity of Mammalian Plasma Albumin (포유류 혈장알부민의 이질성)

  • Plasma albumin was purified from the fresh bovine blood using a minor modification of the polyethyleneglycol and ethanol procedure. The resulting protein solution was tested for its purity by both electrophoretic and immunochemical methods and found to contain only the albumin molecules. Each of the four thiol reagents, maleate, iodoacetate, iodoacetamide and glutathione, was incubated with the purified plasma albumin. The electrophoresis on cellulose acetate of those complexes in various buffers with different component and pH demonstrated that the albumin-glutathione complex was separated into two zones in all buffers used except the barbital and sodium acetate buffers, that the complexes of albumin-iodoacetate and albumin-iosoacetamide also into two zones only in pH 4.8 citrate buffer and in pH 4.8 succinate buffer and that the new zone had more positive net charge compared to the native protein in any case. These results might suggest a possibility that the electrophoretic albumin fraction is composed of at least two molecular species with different conformation.

  • In order to examine the interaction of albumin with the sperm during capacitation in mouse, proteins of cauda epididymal sperm were extracted under various conditions and analyzed with SDS-PAGE. Sperm surface labeling patterms were also examined using fluorochroin~conjugated wheat germ agglutinin (WGA) and bovine serum albumin (BSA). Albumin was detached from the sperm surface during the incubation and seemed to be constituted the major protein components of the conditioned media in which sperm incubated for 90 mm. Detachment of albumin from the sperm was not affected by the Ca2+ in the medium. WGA-FITC labeling confirmed that Triton X-100 permeabilired plasma membrane overlaying the apical segment of sperm head and detached plasma membrane associated proteins having negatively charged glycoconjugates. BSA-FITC labeling of epididymal sperm occurred on the apical segment of periacrosoinal region and postacrosomal region of the head. BSA-FITC labeling was not observed in periacrosoinal region of the sperm treated with Ca2+-ionophore ~3187 (10 MM)~ whereas the postacrosome region of acrosome-reacted sperm was still labeled after the AR. These results suggest that albumin bound to the surface of epididymal sperm is detached during the capacitation process, and it might be involved In physiological change of sperm plasma membrane accompanying the capacitation.

  • Simple coacervation of bovine serum albumin was studied to prepare biodegradable microacapsule. Albumin microcapsules were prepared by using acrylonitrilestyrene polymer (M-80) resin beads and sulfamethoxydiazine as the core materials. The albumin to beads ratio was found to be 1:2.3 and the albumin to sulfamethoxydiazine ratio to be 1:2.9. The 50% release times (T$_{50%}$) of sulfamethoxydiazine and microencapsulated sulfamethoxydiazine were 6 min. and 73 min., respectively. The surface appearance of the microcapsule collected after release experiment was not different from those of the original microcapsule. In addition to slowing release of drug the microencapsulation process masked characteristic bitter taste of sulfamethoxydiazine.

  • This study was trying to find what physical changes occurred to albumin when it reacted with estrogen and other ligands. Each concentration of human serum albumin with 100$\mu$l estradiol reacted at the highest binding capacity of 280nm. In addition, 1 hr of reaction time showed the highest binding rate. Conformational changes in human serum albumin with dietylstillbesterol and N-ethyl-maleimide produced strong binding capacities. The changes were immediate and they did not increase or decrease over time. Effects of human serum albumin with estriol induced no interaction each other. The binding capacity of human serum albumin with vitamin D$_2$was lower than estradiol. and the highest binding rate showed 1 hr of reaction time. Vitamin D$_2$ was very similar to the binding capacity of estradiol.
