가스상 TCE 처리를 위한 추출막 생물반응기의 수학적 모사

  • In this work, an extractive membrane bioreactor containing coulture broth of Burkholderia cepacia G4 PR1 constitutively expressing the TCE-degrading enzyme, tolune-ortho-monooxygenase(TOM), was used for the degradation of TCE. The membrane bioreactor operates by seperating the TCE-containing waste gas from the aerated biomedium, by which the air-stripping of TCE without degradation was overcome that could occur in conventional aerobic biological treatments of TCE-contaminated waste gases. This was achieved by a silicone rubber membrane which was coiled around a perspex draft tube. TCE from the gas phase diffuses across the silicone rubber membrane into microbial culture broth that was continuously fed from a separate aerobic CSTR. Therefore, TCE degradation occured without the TCE being directly exposed to the aerating gas stream. Of the TCE supplied to the membrane bioreactor, 72.6% was biodegraded during the operation of this system. To construct a mathematical model for this system, parameters describing microbial growth kinetics on TCE were determined using a CSTR bioreactor. Else parameters used for numerical simulation were determined from either indepedent experiments or values reported in the literature. The model was compared with the experimental data, and there was a good agreement between the predicted and the measured TCE concentrations in the system. To achieve a higher treatment efficiency, various operating conditions were simulated as well.

Isolation and Characteristics of Photosynthetic Bacterium, Erythrobacter longus SY-46 which Produces Bacterial Carotenoids (Bacterial Carotenoids를 생산하는 광합성세균 Erythrobacter longus SY-46의 분리 및 특성)

  • The aerobic photosynthetic bacterium, which produces bacterial carotenoids was isolated and identified from coastal marine environments. This bacterium was identified by 16S rDNA sequencing and designated as Erythrobacter longus SY-46. E. longus SY-46 was Gram negative and rod shape, and the optimal culture conditions were $25^{\circ}C$, pH 7.0, and 3.0% NaCl concentration, respectively. The carbon and nitrogen sources required for the optimal growth were lactose and tryptone, respectively. Fatty acid compositions of E. longus SY-46 were $C_{18:1}$(78.32%), v-linolenic acid($C_{18:3n9.12.15c}:3.83%$), margaric acid($C_{17:0}$: 3.38%), palmitic acid($C_{16:0}$: 3.07%), and docosahexaenoic acid($C_{22:6n3}$: 2.21%). In addition, E. longus SY-46 showed the characteristic absorption peaks of bacterial carotenoids(in the region of 450 to 480 nm) and bacteriochlorophyll(770 to 772 nm). Major carotenoids of E. longus SY-46 were polyhydroxylated xanthophylls such as fucoxanthin and zeaxanthin.

Characterization of Bacillus species occurring anaerobic denitrification in night soil treatment

  • Bacillus species predominantly outgrown in a night soil treatment system were isolated and characterized. Cell interactions took place among them and cell population changed under various culture conditions. Maximum removal of $NH_4\;^+-N$ and cell production occurred under the conditions of 30% DO and C/N ratio of 8. Additions of 0.8% peptone and 0.3% yeast extract to a basal medium influenced the growth of isolates and the removal of $NH_4\;^+-N$ in flask culture, and metal ions such as Ca, Fe and Mg also did. During the flask experiment of nitrogen removal under an aerobic condition, active nitrification by the isolates occurred largely in 1 h with the decrease of COD and alkalinity destructed was only 74.6% of theoretical value. From the nitrogen balance, the percentage of nitrogen lost in the flask culture was estimated to be 29.0%. This conversion of ammonia to $N_2$ under an aerobic condition was confirmed by GC analysis. The B3 process using the Bacillus species seemed to have some economic advantage.

Bacteriological Characteristic of Atrina pectinata and Ruditapes philippinarum under Non-refrigerated and Refrigerated Storage Conditions

  • In order to estimate the necessity of refrigerated storage of fresh seafood for short-term storage, and evaluate the effect of refrigerated storage on pen shell Atrina pectinata and clam Ruditapes philippinarum collected from Jang-su of Deukryang Bay and I-mok of Sunchen Bay in South Korea, the counts of coliform, Escherichia coli and total aerobic bacteria in A. pectinata and R. philippinarum under non-refrigerated $(28{\pm}1^{\circ}C)$ and refrigerated storage conditions $(4{\pm}1^{\circ}C)$ were determined. The results indicated that the storage at temperature of $4^{\circ}C$ possessed significant effects on inhibiting bacterial growth in live seafood. And refrigerated storage had different effect on A. pectinata and R. philippinarum. Different species and culture environments significantly influenced the initial and ultima bacteria counts. This study confirmed that refrigerated storage for short-term storage of live seafood was necessary, and indicated that the effect of refrigerated storage was influenced by comprehensive effectors.

Treatment of Food Processing Wastewater bearing Furfural by Candida utilis (Candida utilis를 이용한 furfural 함유 식품가공 폐수의 처리)

  • A yeast treatment process was applied to treat food processing organic wastewater containing inhibitory material to anaerobic bacteria. The wastewater contained high concentration of the furfural as a by-product from the food processing. Aerobic yeast (Candida utilis) was selected to remove organics in wastewater. The batch test showed that the wastewater had an inhibition to anaerobic bacteria. The optimum level of temperature for yeast treatment was ranged from 25 to 45$^{\circ}C$. The pH range from 4 to 8 was favorable to yeast growth. The continuous flow reactor was operated at various SRTs. The results were satisfactory with the reduction of COD up to 90% at SRT of more than 1 day. Through the kinetic study of the yeast, the remained COD concentration was mainly caused by the formation of soluble microbial product (SMP).

  • The most efficient method for reducing the organic content of dilute liquid waste is by aerobic-biological treatment. Basically, the organisms responsible for treatment possess the ability to decompose complex organic compounds and to use the energy so liberated for their bodily functions: reproduction, growth, locomotion and so on. That part of organic matter used to produce energy is converted to the essentially stable end products of carbon dioxide, water and ammonia, while the remainder is converted to new cells which can be settled and thus removed from the liquid before the waste is discharged to the receiving body water. Oxygen must be supplied continuously during the aerobic process. In the field of sewage treatment the Warburg respirometer is used mainly for the measurement of the oxygen uptake of samples. In this experiment the Warburg constant volume respirometer was used to determine the oxygen uptake by bacteria in the presence of various glucose concentrations. The rate of oxygen uptake by the bacteria was expressed as the respiratory quotient. The result indicated that the oxygen uptake was proportional to the glucose concentration.   • The combined SHARON (Single reactor system for High ammonium Removal Over Nitrite)-ANAMMOX (Anaerobic ammonium oxidation) reactor were operated in mesophilic condition ($35^{\circ}C$). In this study, microbial granulation and characteristics of SHARON and ANAMMOX sludges were investigated using settling test, Scanning Electron Microscopy (SEM) and Fluorescence In Situ Hybridization (FISH). In SHARON reactor, Aerobic granulation with diameter of 1.5~2.5 mm was accomplished but aerobic granulation was weaker than anaerobic granular sludge. Initial seed sludge of ANAMMOX reactor was used as attached media for biofilm growth. ANAMMOX sludge was more compact and rounder rather than seed sludge. Though ANAMMOX sludge has high activity, it has lower settling ability than the seed granule. The color of ANAMMOX sludge was changed from dark to redish brown granular with diameter of 1~2 mm. In FISH of ANAMMOX sludge, high fraction of Candidatus B. stuttgartiensis which paid great role of nitrogen conversion was detected. Also, FISH results reveals that ANAMMOX bacteria inhabit at inner parts near surface, having advantages in utilization of substrates and protection from oxygen inhibition.

Effects of Garlic Extracts on the Aerobic Bacteria Isolated from Kimchi (김치에서 분리한 호기성 세균의 생육에 대한 마늘의 영향)

  • Twenty-one aerobic bacteria were isolated from Kimchi and the influences of aqueous extract of garlic on the isolates were studied. The bacteria were 11 Bacillus spp., 2 Staphylococcus spp., a Micrococcus sp., a Flavobacterium sp., an Enterobacteriaceae, and 4 Vibrionaceae. In nutrient broth growth of the bacteria was inhibited by the extract. The antibacterial effects of the garlic were different from each other depending upon the bacteria. The results revealed that the inhibitory effects were due to the bacteriocidal action of the garlic extracts.

Optimum Conditions for the Production of Tetramethylpyrazine Flavor Compound by Aerobic Fed-batch Culture of Lactococcus lactis subsp. lactis biovar. diacetylactis FC1

  • Optimum conditions for the production of acetoin and ammonia as the precursors of tetramethylpyrazine(TMP) were determined using Lactococcus lactis subsp. lactis biovar. diacetylactis FC1 in a modified Lactose-citrate broth containing galactose, citrate, and arginine. The cell growth and the productivity of acetoin and ammonia were remarkably increased in an aerobic culture with 10 $\mu M$ of hematin. For the optimum conditions of acetoin and ammonia production, the concentration of citrate and arginine were adjusted to 156 mM and 50 mM after 18 hr cultivation, and citrate and galactose to 156 mM and 50 mM after 36 hr cultivation, respectively. In these conditions, acetoin and ammonia were produced to the final concentration of 127 mM and 195 mM, which were the highest concentations, respectively. The optimum conditions of the TMP production were also determined as follows; 4 hours at 121, pH 8.3, and the maximal yield of TMP under these conditions was 0.81 g/l.

Changes in Physicochemical Properties of Packaged Backsulgi during Storage (포장한 백설기의 저장과정 중 이화학적 변화)

  • Packaged Backsulgi cooked by steam/convection oven and then rapidly chilled was examined by research of microbiological test and sensory evaluation while storing them at the temperatures of 3$^{\circ}C$ and 3$0^{\circ}C$ for 0, 2, 4, 6, 10 days . The pH and reducing sugar content were seemed to change little at 3$^{\circ}C$. However the pH was rapidly reduced until 4 days and then decreased a little at 3$0^{\circ}C$, the reducing sugar content was inclosed little by little. In the microbiological test, any microbial growth in total aerobic, psychrophilic, anaerobic, spore forming bacteria, yeast and molds was not observed until 10 days at 3$^{\circ}C$, but microbial changes of aerobic, psychrophilic and anaerobic bacteria increased to 6 logCFU/g until 10 days at 3$0^{\circ}C$. However microbial changes of them decreased from 6 logCFU/g to 5 logCFU/g. As a result of the sensory evaluation, appearance, taste, color, softness, chewiness and overall Quality were significantly decreased during storage times(p<0.05), but scores of taste and overall quality on 6th days were 7.38${\pm}$1.06, 7.00${\pm}$0.93. Therefore we concluded that there was no problem about stability of storage 6 days at 3$^{\circ}C$.
