  • Bacillus species predominantly outgrown in a night soil treatment system were isolated and characterized. Cell interactions took place among them and cell population changed under various culture conditions. Maximum removal of $NH_4\;^+-N$ and cell production occurred under the conditions of 30% DO and C/N ratio of 8. Additions of 0.8% peptone and 0.3% yeast extract to a basal medium influenced the growth of isolates and the removal of $NH_4\;^+-N$ in flask culture, and metal ions such as Ca, Fe and Mg also did. During the flask experiment of nitrogen removal under an aerobic condition, active nitrification by the isolates occurred largely in 1 h with the decrease of COD and alkalinity destructed was only 74.6% of theoretical value. From the nitrogen balance, the percentage of nitrogen lost in the flask culture was estimated to be 29.0%. This conversion of ammonia to $N_2$ under an aerobic condition was confirmed by GC analysis. The B3 process using the Bacillus species seemed to have some economic advantage.

  • MTBE and other gasoline additives contained in gasoline are known to be a refractory substance resistant to biodegradation. As a method of removing these substances, a research of method using native microbes of polluted soil was progressed and among these, bio-degradation possibility under aerobic condition was evaluated. All of the experiments were progressed based on batch experiment of lab scale and analyzed by GC-FID using HS-SPME technique. The result of bio-degradation experiment based on MTBE and other additives(ETBE, TAME) was observed below 1 mg/L, which initial concentration were 100 mg/L for each method. And through production of by-product and CO2, partial mineralization was confirmed. Degradation velocity of each additive was promptly represented in the order of TBA>ETBE>MTBE>TAME. Through this study, bio-degradation possibility of native microbes of oil polluted soil, MTBE and other gasoline additives was confirmed and it was considered that the result could be used for basic experiment data in removing oil pollutants of soil.

  • In this study, nitrous oxide ($N_2O$) emission was compared between the operations of two different sequencing batch reactors, conventional sequencing batch reactor (CNVSBR) and simultaneous nitrification and denitrification sequencing batch reactor (SND-SBR), using synthetic wastewater. The CNV-SBR consisted of anoxic (denitrification) and aerobic phases, whereas the SND-SBR consisted of a microaerobic (low dissolved oxygen concentration) phase, which was achieved by intermittent aeration for simultaneous nitrification and denitrification. The CNV-SBR emitted 3.9 mg of $N_2O$-N in the denitrification phase and 1.6 mg of $N_2O$-N in the nitrification phase, resulting in a total emission of 5.5mg from 432mg of $NH_4^+$-N input. In contrast, the SND-SBR emitted 26.2mg of $N_2O$-N under the microaerobic condition, which was about 5 times higher than the emission obtained with the CNV-SBR at the same $NH_4^+$-N input. From the $N_2O$ yield based on $NH_4^+$-N input, the microaerobic condition produced the highest yield (6.1%), followed by the anoxic (0.9%) and aerobic (0.4%) conditions. It is thought that an appropriate dissolved oxygen level is critical for reducing $N_2O$ emission during nitrification and denitrification at wastewater treatment plants.

  • No-till direct-sown rice-wheat relaying cropping system has major advantages such as labor and cost saving by eliminating tillage and preparation of seed bed and transplanting. In this system, rice sowing was done simultaneously wheat harvesting. A paddy field experiment was conducted to evaluate effects of no-till years on soil microbial changes and soil physico-chemical characteristics with rice growth and development. Chemical fertilizers and agricultrual chemicals was not applied in no-till system. As the year in no-till direct-sown system the air permeability was increased and after water submerging soluble nitrogen was released Aerobic microbial-n was highest in May and then decreased after water irrigation. The population of aerobic soil microorganisms were steeply decreased after water submerging Soil microorganisms was decreased with the increased the soil depth. A month was needed for the seedling establishment in a no-tillage rice-wheat cropping system. Increased cropping years improved leaf greenness and leaf area index(LAI). But stomatal conductance(Gc) was higher in conventional cultivation system than no-till system. Stomatal conductance at panicle initiation stage was increased higher in conventional condition of leaves but the difference between conventional and no-till system was increased at heading stage. In no-till 4 years condition rice grain yield was spikelet numbers per panicle.

  • This study was carried out to investigate the effect of Synechocystis sp. KACC 91007 when added to a pig slurry or pig liquid fertilizer (PLF) on germination index (GI) of Chinese cabbage. The preliminary experiment involved the screening of inoculant levels which were; 0.05, 0.1, 0.2, and $0.3\%$, respectively. The $0.05\%$ level of inoculant was selected based on low phytotoxicity and high GI.. The PLF underwent a 107 day aerobic and anoxic processing conditions. The T-N, T-P, $NH_4$, and $NO_3-N$ concentrations of the untreated pig slurry were; 2,873, 753, 1,441.6, and 16.48 ppm, respectively. Using aerobic processing treatment, the fertilizer value of the PLF was 3,672, 164, 183.87, and 21.97 ppm, respectively. In contrast, the fertilizer value of the PLF processed under anoxic condition was reduced to 1,261, 68, 161, and 16.87 ppm. The GI value of the untreated PLF under aerobic and anoxic processing condition was 83 and $40.4^{*}\%$, respectively. With the addition of the $0.05\%$ microbial inoculant, the GI improved by more than 40 and $50\%$ respectively, when the PLF was processed under anoxic and aerobic conditions. The above findings proved that the aerobic processing of PLF for 107 days was better than anoxic and yielded higher T-N which is a macro-nutrient fertilizer material. Consequently, the addition of $0.05\%$ microbial inoculant resulted to a higher GI of the Chinese cabbage specifically under aerobic processing condition.

  • This study was carried out to investigate the effects of moisture content, temperature, C/N ratio and pH of the sewage sludge mixed with hulle and sawdusts for making compost under aerobic condition and to improve the defect of the structure of experimental equipment heat lose and handling method. and obtained results were as follows 1.The temperature was reached 73$^{\circ}$ C around 50 hours fermentation in the condition of 0.8 L/min. of air and 60.4% of moisture content. and favorable moisture content of initial condition ranged from 50 to 65% 2.The temperature near bottom of the batch composter was decreased due to evaporate water vapor and lose the heat produced during aeration. and it is required to be improved. 3.The temperature in the batch composter from the center to the inside wall surface was gradually decreased. the temperatures of the points located in r=9cm and the wall surface were 4$^{\circ}$ C and 6$^{\circ}$ C respectively. and therefore it is required to be insulated. 4. The maximum C02 production was obtained as 7.3% per volume in the temperature of 63$^{\circ}$C at the moisture content of 60% 5.The temperature range of active microbes growth was found to be as 20$^{\circ}$C to 40$^{\circ}$C in the case of mesophiles and 50$^{\circ}$C to 65$^{\circ}$C in the case of thermophiles due to increase and decrease C02 production. 6.C/N ratio after decomposition was 1.3 to 2.6 smaller than that of initial one due to increase the amount of nitrogen. The more C/N ratio increased. the less the reaction velocity decresed. The optimum of it as found to be 30. 7.pH values after decomposition were slightly increased than that of initial ones. The reaction velocity was decreased at acid and alkall condition. Therefore it is neseseary to neutralize the medium to improve the reaction.

  • To treat wastewater efficiently by a one-step process of nitrogen removal, a new strain of $N_2-producing$ bacteria from $NH{_4}{^+}$ under an aerobic condition was isolated and identified. By 16S-rDNA analysis, the isolate was identified as Enterobacter asburiae with 96% similarity. The isolate shows that the capacity of $N_2$ production under an oxic condition was approximately three times higher than that under an anoxic condition. The optimal conditions (pH, temperature and C/N ratio) of the immobilized isolate for $N_2$ production were found to be 7.0, $30^{\circ}C$ and 5, respectively. Under all the optimum reaction conditions, the removal efficiency of $COD_{Cr}$ and TN reached 56.1 and 60.9%, respectively. The removal rates of $COD_{Cr}$ and TN were highest for the first 2.5 hrs (with the removal $COD_{Cr}$ ratios of 32.1), and afterwards the rates decreased as reaction proceeded. For application of the immobilized isolate to a practical process of ammonium removal, a continuous bioreactor system exhibited a satisfactory performance at HRT of 12.1 hr, in which the effluent concentrations of $NH{_4}{^+}-N$ was measured to be 15.4 mg/L with its removal efficiency of 56.0%. The maximum removal rate of $NH{_4}{^+}-N$ reached 1.6 mg $NH{_4}{^+}-N/L/hr$ at HRT of 12.1 hr (with N loading rate of 0.08 $Kg-N/m^3-carrier/d)$. As a result, the application of the immobilized isolate appears a viable alternative to the nitrification-denitrification processes.

  • A company, which is chicken butcherying-plant, was scheduled to increase up to twice output. We researched the way to increase up to twice the processing efficiency at the target biological treatment tank of A company. We performed for this study to obtain the reason why organic matter and nitrogen removal efficiency is increased when MLSS concentration is increased. It's performed at the target of pressure flotation water on SBR system. We performed the research which MLSS was 12,700 mg/L for 30hours (in aerobic condition for 25 hours and in anoxic condition for 5 hours). As a result, the nitrification was happened completely in aerobic condition within 25 hours. Denitrification efficiency was also over 90% when C/N ratio was over 3:1. After the experiment, we changed the concentration of MLSS 5,600 to 12,700 mg/L. In condition MLSS was about 11,000 mg/L and HRT were 30 hours meet the Effluent quality standard.

  • Using superoxide dismutase (SOD)-deficient mutants of Saccharomyces cerevisiae, the oxidative stresses induced by 0.1 mM of copper ion $(Cu^{++})$ was studied. In aerobic culture condition, yeasts lacking MnSOD (mitochondrial SOD) showed more significant growth retardation than CuZnSOD (cytoplasmic SOD)-deficient yeasts. However, not so big differences in growth pattern of those mutants compared withwild type were observed under anaerobic condition. It was found that, under aerobic condition, the supplementation of 0.1 mM copper ioh:(Cu") into culture medium caused the remarkable increase of CuZnSOD but not so significant change in MnSOD. It was also observed that catalase activities appeared to be relatively high in the presence of copper ion in spite of the remarkable reduction of glutathion peroxidase in CuZnSOD-deficient yeasts, but the slight increments of catalase and glutathion peroxidase were detected in MnSOD-deficient strains. It implies that the lack of cytoplasmic SOD could be compensated mainly by catalase. However, these phenomena resulted in the significantincrease of cellular lipid peroxides content in CuZnSOD-deficient yeasts and the slight increment of lipid peroxides in MNSOD-deficient cells. In anaerobic cultivation supplementing copper ion, the cellular enzyme activities of catalase and glutathion peroxidase in SOD-deficient yeasts were slightly increased without any significant changes of lipid peroxides in cell membrane. It suggests that a little amount of free radicals generated by copper ion under anaerobic condition could be sufficiently overcome by catalase as well as glutathion peroxidase.dase.
