Lysosomal Enzyme Inhibiting Activity of Alkaloidal Fraction from Tylophora indica Leaves in Arthritic rats

  • Ethanolic extract (50% v/v) and alkaloid fraction of Tylophora indica leaves were examined for lysosomal enzyme inhibitory activity in adjuvant-induced arthritic rats. The alkaloid fraction showed statistically significant inhibition of arthritic lesions (p < 0.05) from day 18, (p < 0.025) from day 20 and (p < 0.001) from day 21 onwards in the adjuvant-induced arthritis, which was comparable to the response of standard drug Indomethacin. The ethanolic extract was less significant than the alkaloidal fraction in inhibition of arthritis. Alkaloid fraction showed significant (p < 0.001) inhibitory effect on the lysosomal enzyme activities in adjuvantinduced arthritic rats. It also significantly prevented decrease in collagen levels and synovial damage observed during arthritis and also inhibited increase in urinary excretion levels of collagen degradation products like hydroxyproline, hexosamine, hexuronic acid, etc. Both ethanolic extract as well as the alkaloid fraction, however, did not show any significant activity in normal nonarthritic rats. The ethanolic extract and the alkaloid fraction may thus be able to inhibit the progress of inflammation and inhibit the destructive activity of lysosomal enzymes on structural macromolecules like collagen etc. in the synovial capsule in joints during arthritic states. They may thus prevent synovial damage observed during arthritis.

The antinociceptive and anti-inflammatory effect of water-soluble fraction of bee venom on rheumatoid arthritis in rats

  • We recently demonstrated that bee venom (BV) injection into acupoint (i.e. Zusanli) produced more potent anti-inflammatory and antinociciptive effect in Freunds adjuvant induced rheumatoid arthritis (RA) model as compared with that of non-acupoint injection(i.e back). However, the precise components underlying BV-induced antinociceptive and/or anti-inflammatory effects have not been fully understood. Therefore, we further investigated the anti-arthritic effect of BV after extracting the whole BV according to solubility (water soluble: BVA, ethylacetate soluble: BVE). Subcutaneous BVA treatment (0.9 mg/kg/day) into Zusanli acupoint was found to dramatically inhibit paw edema and radiological change (i.e. new bone proliferation and soft tissue swelling) caused by Freunds adjuvant injection. In addition, the increase of serum interleukin-6 by RA induction was normalized by the BVA treatment as similar with that of non-arthritic animals. On the other hand, BVA therapy significantly reduced arthritis induced nociceptive behaviors (i.e., nociceptive score for mechanical hyperalgesia and thermal hyperalgesia). Furthermore, BVA treatment significantly suppressed adjuvant induced Fos expression in the lumbar spinal cord at 3 weeks post-adjuvant injection. However, BVE treatment (0.05 mg/kg/day) has not any anti-inflammatory and anti-nociceptive effect on RA. Based on the present results, we demonstrated that BVA might be a effective fraction in whole BV for long-term treatment of RA-induced pain and inflammation. However, it is clear necessary that further fraction study about BVA was required for elucidating an effective component of BVA.

마전자약침(馬錢子藥鍼)이 Adjuvant 관절염(關節炎)에 미치는 영향(影響)에 관한 실험적(實驗的) 연구(硏究) (Effect of Strychni Semen Aqua - acupuncture in Rats with Arthritis induced by Freund's Complete Adjuvant)

  • Objectives and Mtthods : In order to study on the Effect of Strychni Semen aqua-acupuncture on the arthritis induced by injecting Freund's complete adjuvant to the right posterior planta of rat's foot, rats were divided 5 groups equally, one group was not treated(Normal) and the other groups were induced arthritis by injecting Freund's complete adjuvant(Control, ST, SS, SST). Control was not treated after injecting of Freund's complete adjuvant. ST was 0.2cc Saline-injected Group at Chok-Samni(ST36) either days during 2 weeks after injecting of Freund's complete adjuvant. SS was 0.2cc Strychni Semen-subcutaneous injected group either days during 2 weeks after injecting of Freund's complete adjuvant. SST was 0.2cc Strychni Semen-injected group at Chok-Samni(ST36) either days during 2 week after injecting of Freund's complete adjuvant. And then effect of Strychni Semen aqua-acupuncture on the edema, numbers of blood WBC, platelet, hematocrit, contented quantities of total protein, albumin in rats with F.C.A were measured. Results : The following results have been obtained. 1. In effect of Strychni Semen aqua-acupuncture on plantar edema, SS showed significant decrease compared with control. 2. In effect of Strychni Semen aqua-acupuncture on WBC, SS and SST showed significant decrease compated with control. 3. In effect of Strychni Semen aqua-acupuncture on platelet, SS and SST did not show significant decrease. 4. In effect of Strychni Semen aqua-acupuncture on hematocrit, SST showed significant increase compared with control. 5. In effect of Strychni Semen aqus-acupuncture on the contents quantity of total protein and albumin, SS showed significant increase compared with control: Conclusions : Accoring to this result, Strychni Semen aqua-acupunture inserted into Chok-Samn i(ST36) appeared to have effect on Adjuvant arthritis of rats. Therefore it is oxpected that Strychni Semen aqua-acupunture may be available clinically.

Adjuvant로 유발된 관절염의 흰 쥐에서 초음파의 임상반응 (Clinical Responses of Ultrasound on Adjuvant-Induced Arthritis in Rats)

  • The purpose of this study was to assess the effects of ultrasound on adjuvant-induced arthritis in rats. Adjuvant arthritis was induced in 26 female Sprauge-Dawley rats by the subcutaneous injection of a single dose of .1 mL of Complete Freund's Adjuvant (CFA) (1 mg of Mycobacterium Butyricum suspended in .1 $m{\ell}$ paraffin oil) into the right hind paw. After confirming inflammatory edema and arthritis in the paw 2 weeks later, the arthritic rats were divided into 3 groups, i.e., a control group, a pulsed ultrasound group (Group A), and a continuous ultrasound group (Group B) with 8 rats placed in each group. The rats in Group A were treated with pulsed ultrasound at 1 MHz frequency with .5 $W/cm^2$ intensity in 1 : 4 mode for 3 minutes. The rats in Group B were treated with continuous ultrasound at 1 MHz frequency with 2 $W/cm^2$ intensity in the continuous mode for 3 minutes. The ultrasound treatment was done in the left and right ankles for 2 weeks. Clinical, radiographic and histopathologic findings were then evaluated before and after treatment and yielded the following results. 1. No significant difference was present in body weight between the control group and the treated groups. 2. A statistically significant decrease in the edema of the paw was seen in the rats in Group A that was treated with pulsed ultrasound by 26~29 days after the treatment started (p<.05). 3. According to radiological examination, Group A showed the lowest score in arthritis scale which means it showed a tendency to suppress arthritic inflammation of the left and right hind paws. However, no statistically significant difference was present in the score between the control group, Group A and Group B. 4. According to histopathologic findings, the degree of infiltration by inflammatory cells and hypertrophy of the synovium were less in Group A compared with the control group and Group B. The results of the study show that rats that were treated with the pulsed ultrasound effectively suppressed adjuvant arthritis. However, more effort is needed to objectively prove the effectiveness of ultrasound by developing more sensitive testing methods that could quantitatively evaluate the treatment effects of acute rheumatoid arthritis and by trying out different ultrasound treatment methods.

Suppressive Effects of Propolis in Rat Adjuvant Arthritis

  • The effects of ethanolic extract (EEP) of propolis on chronic inflammation were evaluated using rat adjuvant arthritis. In the chronic inflammatory animal model, the arthritis index was suppressed by EEP treatments (50 mg/kg/day and 100 gm/kg/day, p.o.). Moreover, physical weakness, induced by the chronic disease state, was dose-dependently improved in the EEP-treated groups. It s analgesic effect, assessed using the tail-flick test, was comparable to prednisolone (2.5 mg/kg/day, p.o.) and acetyl salicylic acid (100 mg/kg/day, p.o.). In carrageenan rat hind paw edema, which was conducted to test the effects of subfractions of EEP, the petroleum ether sub-fraction (100 mg/kg, p.o.) showed an inhibitor effect on the paw edema whereas EEP (200 mg/kg, p.o.) showed a significant anti-inflammatory effect at 3 and 4 hrs after carrageenan injection. From these results, we conclude that the ethanolic extract of propolis had a profound anti-inflammatory effects on both chronic and acute inflammations.

Antiinflammatory and Analgesic Effects of Higenamine, a Component of Aconiti Tuber

  • The antiinflammatory and analgesic activities of higenamine were evaluated by measuring edema volume and pain threshold in adjuvant arthritic rats and acetic acid-induced writhing test in mice. Higenamine, with consecutive oral administrations at doses of 10 and 50 mg/kg/day, showed significant antiedemic effect and elevation of pain threshold during the secondary lesion of adjuvant arthritis. Higenamine also showed a significant inhibition of acetic acid-induced writhing syndrome with a single oral administration (200 mg/kg). From these results, it is postulated that higenamine might possess both of centrally and peripherally mediated analgesic properties.

Sweet 봉약침과 저준위 레이저침이 Complete Freund's Adjuvant로 유발된 관절염 백서 모델에 미치는 영향 (Effects of Sweet Bee Venom Pharmacopuncture and Low Level Laser Acupuncture on a Rat Model with Complete Freund's Adjuvant-induced Arthritis)

  • Purpose : This research was conducted to investigate the effect of sweet bee venom pharmacopuncture and low level laser acupuncture on paw edema, pain index, anti-inflammatory factor, AST, ALT and complete blood cell count of a rat model with Complete Freund's Adjuvant-induced arthritis. Methods : Five experimental groups were formed with each consisting of six rats: normal group, control group, sweet bee venom pharmacopuncture group, lower level laser acupuncture group, and sweet bee venom pharmacopuncture, lower level laser acupuncture group. The experimental model of arthritis was induced by two injections of Freund's adjuvant into the left knee joint of Sprague Dawley(SD) rats. The second injection of Freund's adjuvant was given ten days after the first one. Ten days later, sweet bee venom pharmacopuncture and low level laser acupuncture were administered separately or together by assigned groups at $GB_{34}$ and $GB_{39}$ of rats twice a week for a total of six times. Thereafter, edema rate, pain index, tumor necrosis factor-${\alpha}$, interleukin-6, aspartate aminortansferase, alanine aminotransferase and complete blood cell count were measured. Results : We noticed synergic effects of sweet bee venom pharmacopuncture and low level laser acupuncture according to the results of the paw edema and Von Frey pain index. The sweet bee venom pharmacopuncture(BVA) and sweet bee venom pharmacopuncture+ low level laser acupuncture(BVA+LLA) groups experienced a more significant effect when compared with the control group. Conclusions : These results suggest that Sweet Bee Venom Pharmacopuncture and low level laser acupuncture at GB34 and GB39 have a significant anti-inflammatory effect on Freund's adjuvant arthritis in rats.

Adjuvant로 유발된 관절염의 흰쥐에서 초음파와 고전압 맥동 전류 치료의 효과 (Effects of Ultrasound and High-Voltage Pulsed Current on Adjuvant-Induced Arthritis in Rats)

  • The purpose of this study was to assess the effects of High-Voltage Pulsed Current (HVPC) and ultrasound on adjuvant-induced arthritis in rats. Adjuvant arthritis was induced in female Sprauge-Dawley rats by the subcutaneous injection of a single dose of $.1m{\ell}$ of Complete Freund's Adjuvant (CFA) (1 mg of Mycobacterium Butyricum suspended in $.1m{\ell}$ paraffin oil) into the right hind paw. A randomized, parallel-groups design of 24 subjects was used. All rats were randomly assigned to control (n=8), ultrasound (n=8), and HVPC (n=8) were compared with those of injured rats. The rats in the pulsed ultrasound group were treated at 1 MHz frequency with $.5W/cm^2$ intensity in 1:4 mode for 5 minutes per day. The rats in the HVPC group were treated at 120 pulses per second and $50{\mu}s$ phase duration, 20 mA intensity for 30 min per day. Treatment was done in the left and right hind limb for 2 weeks. We evaluated clinical, radiographic, hematologic and histopathologic findings before and after treatment and obtained the following results. 1. Edema of the right hind paw was more significantly reduced in the ultrasound and HVPC groups than the control group on days 9, 12, and 14 (p<.05). Edema of the left hind paw was more significantly reduced in ultrasound and HVPC groups than the control group on days 12, 14 (p<.05). 2. WBC counts of the ultrasound and HVPC groups as compared with the control group were becoming remarkably decreased after the treatment. 3. In radiologic findings, arthritis formation was seen according to the score of arthritis, which was the highest in the control group, upon the observation of radiographs of the left and right hind paws. However, no statistically significant difference was present in the score within three groups. 4. In the histopathologic findings, ultrasound and HVPC groups had effectively suppressed erosions of articular cartilage and inflammatory cell infiltrations. Therefore, the results of the study show that rats that were treated with the ultrasound and HVPC effectively suppressed adjuvant arthritis. However, no statistically significant difference was present between the ultrasound group and the HVPC group.

백굴채ㆍ목과ㆍ위영선 혼합 약침액이 관절염 백선의 염증과 간에 미치는 영향 (Effects of Herba ChelidoniiㆍClematis Florida ThunbㆍChaenomelis Fructus Complex Herbal Acupuncture on Anti-Inflamation and Liver in Rats with Arthritis induced Freund's Complete Adjuvant)

  • Objective : In order to study the effects of Herba ChelidoniiㆍClematis Florida ThunbㆍChaenomelis Fructus complex herbal acupuncture on anti-inflammation and liver in rats with arthritis induced Freund's complete adjuvant. Method : We performed several experimental items : that is edema value, numbers of WBC. total protein, total bilirrubin, SGOT, SGPT. Result: The change in the right plantar edema shows that Control group was reduced by 0% but, ST group ,Sample A group and Sample B group were reduced by 287.2%, 343.0%, 272.5% respectively. In the WBC count, ST group, Sample A group, Sample B group showed a decrease with statistical significance as compared with control group. In the total protein, Sample B group showed a decrease with statistical significance as compared with control group. In the total bilirrubin, SGOT, SGPT, None showed noxious variation with statistical significance as compared with control group. Conclusion : As a result of this experiment, it is concluded that Herba ChelidoniiㆍClematis Florida ThunbㆍChaenomelis Fructus complex herbal acupuncture showed that therapeutic effect of anti-inflammation in adjuvant arthritis and non-toxic effect of liver.
