Objectives : We investigated the effects of gambigyeongsinhwan(GGH)(1) on body weight and non-alcoholic fatty liver disease(NAFLD) examined whether blood total cholesterol, LDL-cholesterol, free fatty acid and triglyceride levels and hepatic lipid accumulation are inhibited by it in high fat diet-fed obese male mice. Methods : 8 weeks old, high fat diet-fed obese male mice were divided into 5 groups: C57BL/6N normal, control, GGH(1)-1, GGH(1)-2 and GGH(1)-3. After mice were treated with GGH(1) for 8 weeks, we measured body weight gain, food intake, feeding efficiency ratio, fat weight, plasma ALT, leptin and lipid levels. We also did histological analysis for liver and fat on the mice. Results : Compared with controls, GGH(1)-treated mice had lower body weight gain and adipose tissue weight, the magnitudes of which were prominent in GGH(1)-3. Compared with controls, GGH(1)-treated mice had lower feeding efficiency ratio and blood leptin level, the magnitudes of which was prominent in GGH(1)-3. Compared with controls, GGH(1)-treated mice had lower blood plasma total cholesterol, LDL-cholesterol, free fatty acid and triglyceride levels. Compared with controls, GGH(1)-3 treated mice had lower blood plasma ALT concentration. Consistent with their effects on body weight gain, the size of adipocytes were significantly decreased by GGH(1), whereas the adipocyte number per unit area was significantly increased, suggesting that GGH(1) decreased the number of large adipocytes. Hepatic lipid accumulation was decreased by GGH(1). Conclusions : In conclusion, these results suggest that GGH(1) exhibits anti-obesity effects through the modulation of feeding efficiency ratio and plasma obesity parameters. Moreover, it seems that GGH(1) also contributes to improve NAFLD through the regulation of plasma ALT and hepatic triglyceride accumulation.
Objectives This study is to investigate the effects and mechanisms of Gastrodia elata extract (GEE) on the high-fat diet-induced obesity model. Methods C57BL/6 mice were randomly assigned into 5 groups (n=10). Control group was fed normal diet (ND). Obesity group was fed 60% high fat diet (HFD). The other three groups were fed HFD with 100, 200, 500 mg/kg GEE. After five weeks, body weight, liver and epididymal fat weight, triglyceride concentration in liver and serum, sterol regulatory element-binding protein-1 (SREBP-1), acetyl-CoA carboxylase (ACC), fatty acid synthase, peroxisome proliferator-activated receptor 𝛾 (PPAR-𝛾), CCAAT/enhancer binding protein 𝛼 (C/EBP-𝛼) expression level, insulin concentration in serum were measured. Results The GEE (100, 200, and 500 mg/kg)-treated animals exhibited substantial decreases in body mass, liver weight and epididymal white adipose tissue collate to the HFD-fed group. GEE treatment also reduced hepatic and serum triglyceride level. Furthermore, GEE treatment significantly inhibited adipogenesis in the GEE group by reducing the protein expression of SREBP-1, ACC and the messenger RNA expression of PPAR𝛾, C/EBP-𝛼, which are adipocyte differentiation-related genes. Conclusions These research outcomes recommend that GEE is possibly valuable for the prevention of HFD-induced obesity via modification of various pathways related with adipogenesis and adipocyte differentiation.
Background: Hepatic lipid disorder impaired mitochondrial homeostasis and intracellular redox balance, triggering development of non-alcohol fatty liver disease (NAFLD), while effective therapeutic approach remains inadequate. Ginsenosides Rc has been reported to maintain glucose balance in adipose tissue, while its role in regulating lipid metabolism remain vacant. Thus, we investigated the function and mechanism of ginsenosides Rc in defending high fat diet (HFD)-induced NAFLD. Methods: Mice primary hepatocytes (MPHs) challenged with oleic acid & palmitic acid were used to test the effects of ginsenosides Rc on intracellular lipid metabolism. RNAseq and molecular docking study were performed to explore potential targets of ginsenosides Rc in defending lipid deposition. Wild type and liver specific sirtuin 6 (SIRT6, 50721) deficient mice on HFD for 12 weeks were subjected to different dose of ginsenosides Rc to determine the function and detailed mechanism in vivo. Results: We identified ginsenosides Rc as a novel SIRT6 activator via increasing its expression and deacetylase activity. Ginsenosides Rc defends OA&PA-induced lipid deposition in MPHs and protects mice against HFD-induced metabolic disorder in dosage dependent manner. Ginsenosides Rc (20mg/kg) injection improved glucose intolerance, insulin resistance, oxidative stress and inflammation response in HFD mice. Ginsenosides Rc treatment accelerates peroxisome proliferator activated receptor alpha (PPAR-α, 19013)-mediated fatty acid oxidation in vivo and in vitro. Hepatic specific SIRT6 deletion abolished ginsenoside Rc-derived protective effects against HFD-induced NAFLD. Conclusion: Ginsenosides Rc protects mice against HFD-induced hepatosteatosis by improving PPAR-α-mediated fatty acid oxidation and antioxidant capacity in a SIRT6 dependent manner, and providing a promising strategy for NAFLD.
Cancer cachexia는 지방조직과 근육계 조직의 손실에 따른 체중의 현격한 감소를 특징으로 하고 있어 궁극적으로는 암 치료제에 대한 반응을 낮출 뿐만 아니라, 삶의 양은 물론 질도 낮추게 되는 시급히 해결되어야 하는 미충족 의료수요중의 하나이다. 아직까지 임상에서는 수많은 노략에도 불구하고 일부 완화시킬 수 있는 약제가 있기는 하나, 전반적으로 해결이 가능한 약제나 치료 방법이 아직은 없는 실정이다. 그러므로 이를 해결할 수 있는 방법으로 동물모델이 필요한 질환이라 하겠다. 이러한 배경하에 연구자 등은 우선 동물모델을 수립하고 이를 기반으로 적절한 치료제를 개발하기 목적으로 본 연구에서는 C26 대장 선암 세포를 이용한 Cancer cachexia 동물모델을 수립하여 이 모델에서의 변화를 소개함으로써 향후 더 진보된 치료제 개발이나 병태생리를 연구하는데 도움을 주고자 본 연구를 시행하여 다음과 같은 결과를 얻을 수 있었다. C26 adenocarcinoma를 대퇴부 주입 후 시간 경과에 따라 몸무게의 변화가 현저하여 2주 이후에 유의한 몸무게의 감소, 식욕부진, 활동감소가 관찰되었고, 이때의 혈청 Cytokine 및 이를 조절하는 여러가지 전사인자의 변화가 선행되었고, 현저한 근육계의 근감소가 관찰되었으며, 실험동물은 3주에 40%가 사망하는 변화를 보였다. 연구자 등은 본 동물모델은 향후 새로운 치료약제 개발이나 Cancer cachexia 병태생리 연구에 매우 도움이 되는 수립하기 간편하며, 기저 분자생물학적 변화를 관찰할 수 있는 우수한 Cancer cachexia 모델이라 결론지을 수 있었다.
Rubus crataegifolius (RC)는 장미과에 속하는 전통적인 아시아 약용 식물이다. RC 열매는 항산화 작용을 통해 성인병을 예방하는 것으로 알려져 있다. 본 연구에서는 RC 열매 추출물(RCex)이 비만과 비알코올성 지방간 질환(NAFLD)에 미치는 영향을 동물 모델을 통해 평가하였다. 28마리의 수컷 C57BL/6J 마우스에 8주간 비만을 유도한 후, 추출물을 8주간 경구 투여하였다. 그룹 1은 일반 대조군으로 표준사료를 섭취하였다. 그룹 2는 HFD 대조군으로, 그룹 3에는 심바스타틴(6.5 mg/kg/일)을, 그룹 4에는 RCex (200 mg/kg)을 투여하였다. RCex투여는 실험 마우스의 체중, 지방 조직, 간 무게를 감소시켰으며, 또한 지질 대사(ALT, AST, TC, TG, LDL, HDL)를 포함한 생화학적 바이오마커를 개선하였다. AMPK의 활성화는 지방생성 유전자(LXR, SREBP-1c, FAS, ACC1)의 발현을 감소시켰으며, RCex에 의한 CPT 활성 증진 효과를 검증하였다. RCex는 또한 에너지 소비 및 신진대사와 관련된 호르몬(adiponectin 및 leptin)의 혈장 수준에도 영향을 미쳤다. 또한, RCex가 HFD로 유도된 비만 mice의 포도당 불내성을 개선했음을 확인 하였다. RCex는 AMPK의 인산화를 통해 지방산 산화 및 지방산 합성을 조절함으로써 항비만 및 항NAFLD 효과를 가짐을 처음으로 입증하였다. 이는 R. crataegifolius가 비만 및 관련 NAFLD 예방에 좋은 보충제가 될 수 있음을 시사한다.

Objective: We aimed to develop and validate a deep learning system for fully automated segmentation of abdominal muscle and fat areas on computed tomography (CT) images. Materials and Methods: A fully convolutional network-based segmentation system was developed using a training dataset of 883 CT scans from 467 subjects. Axial CT images obtained at the inferior endplate level of the 3rd lumbar vertebra were used for the analysis. Manually drawn segmentation maps of the skeletal muscle, visceral fat, and subcutaneous fat were created to serve as ground truth data. The performance of the fully convolutional network-based segmentation system was evaluated using the Dice similarity coefficient and cross-sectional area error, for both a separate internal validation dataset (426 CT scans from 308 subjects) and an external validation dataset (171 CT scans from 171 subjects from two outside hospitals). Results: The mean Dice similarity coefficients for muscle, subcutaneous fat, and visceral fat were high for both the internal (0.96, 0.97, and 0.97, respectively) and external (0.97, 0.97, and 0.97, respectively) validation datasets, while the mean cross-sectional area errors for muscle, subcutaneous fat, and visceral fat were low for both internal (2.1%, 3.8%, and 1.8%, respectively) and external (2.7%, 4.6%, and 2.3%, respectively) validation datasets. Conclusion: The fully convolutional network-based segmentation system exhibited high performance and accuracy in the automatic segmentation of abdominal muscle and fat on CT images.
회충증은 Ascaris lumbricoides에 의하여 유발되는 장 질환이다. 대부분의 회충증 환자는 무증상이지만 장내 유충이 많은 경우에는 장 폐색, 폐쇄성 황달, 담관염, 담낭염, 췌장염과 같은 합병증이 생길 수 있다. 저자들은 컴퓨터단층촬영(이하 CT)에서 구불창자간막에 혈종과 활동성 출혈이 발생하여 회충증으로 진단된 74세 남자의 사례를 경험하였다. 이후에 환자가 시행한 CT에서는 이와 더불어 구불창자의 천공 발생이 확인되었다. 복강경을 통한 저위전방절제술이 시행되었고 구불창자간막에 큰 혈종이 있었으며 장간막 지방 조직에서 현미경적으로 회충이 확인되었다. 이러한 회충증의 드문 임상적, 영상의학적 소견은 기생충이 장관의 내강 안에서 복강 내로 이동하는 과정에서 발생한 순차적인 합병증들을 보여준다.
대두 이소플라본은 갱년기 증상에 대한 유익한 효과 때문에 폐경여성들의 관심을 끌고 있다. 본 연구는 난소절제 쥐에서 대두 이소플라본인 제니스테인과 수영운동의 동시처리(Gen+SE)가 제니스테인 단독처리(Gen)와 수영운동 단독처리(SE)에 비해 비만과 간 기능 개선에 유익한 시너지 효과를 나타낼 것인지를 조사하였다. 난소절제 쥐는 무작위로 대조군(Con), Gen, SE 및 Gen+SE으로 나누어 8주 동안 고지방식이를 섭취하였다. 몸무게, 백색지방조직의 무게, 간 조직의 지질추적 및 혈청 속 ALT와 AST 수치를 조사한 결과, Con에 비해 Gen과 SE는 감소하였고 Gen+SE는 Gen과 SE에 비해 더 효과적으로 감소하였다. 간 조직에서 염증성 사이토카인인 IL-6와 TNF-𝛼 유전자의 발현은 Con에 비해 Gen과 SE 모두에서 감소하였고, Gen+SE는 Gen과 SE에 비해 더욱 감소하였다. 그러나 adiponectin 유전자의 발현은 반대의 결과가 나타났다. 간 조직에서 지방산 산화와 관련된 유전자의 발현은 Con에 비해 Gen과 SE에서 증가하였고 Gen+SE는 Gen과 SE에 비해 더 증가하였다. 따라서 본 연구는 대두 이소플라본과 수영운동의 상호작용은 난소절제 비만 쥐에서 비만 조절과 저하된 간 기능 개선에 매우 효과적이며, 이것은 난소절제 쥐에서 간의 지방산 산화를 촉진함으로써 발생한 것이라는 것을 제안한다.
