Study on prevention of adhesion formation by use of sodium carboxymethylcellulose and dextran 70 I. Adhesion formation by artificial injuries and its preventive effect of sodium carboxymethylcellulose and dextran 70 in rabbits (Sodium carboxymethylcellulose 및 dextran 70 을 이용한 유착형성 방지에 관한 연구 I. 토끼에서 인공창상에 의한 유착형성과 sodium carboxymethylcellulose 및 dextran 70의 유착방지 효과)

  • 복강장기의 유착을 방지하기 위하여 토끼의 공장에 인공창상을 일으키고 sodium carboxymethylcellulose (SCMC)와 dextran 70을 단일 혹은 합제로 사용하여 이들의 유착방지 효능을 조사하였고, 아울러 체중의 변화를 조사함으로써 이들을 사용하였을 때 일어날 수 있는 부작용을 검토하였다. 인공창상에 의한 유착형성 유발빈도와 정도를 알아보기 위하여 토끼를 전신마취시킨 다음 개복수술을 시행하여 공장의 장막에 2cm 폭으로 3곳에 abrasion또는 electrocautery를 실시하였던 바, 이러한 인공창상들이 유의성있는 높은 유착형성율(abrasion, 70%; electrocatuery, 72.7%)과 심한 유착정도를 일으켰으며(abrasion, 1.80; electrocatuery, 2.44), 체중의 감소를 가져왔다(abrasion, -2.5%; electrocautery; 9.9%). Abrasion보다는 electrocatuery에 의한 자극이 더욱 심한 유착정도 및 체중감소를 보였으며 심할 경우 폐사를 일으키기도 하였다. 이러한 유착을 효과적으로 방지하기 위하여 1, 2, 3%의 SCMC 및 6, 10%의 dextran 70 용액을 단일 또는 합제를 만들어 abrasion방법으로 공장에 인공창상을 일으킨 토끼의 복강내에 주입하고 수술 4주후에 복강을 열어 유착형성율을 조사하였던 바, 1% SCMC와 10% dextran 70의 합제(Synthetic soln)에서 가장 낮은 유착형성율(0%)을 보여 유착방지 효과가 가장 뚜렸하였다. 아울러 수술 4주일후 체중의 변화도 유의하게 일으키지 아니 하였다. 그러므로 유착방지제로서 synthetic solution을 사용하는 것이 가장 효과적이라고 사려된다.

Ursodeoxycholic Acid (UDCA) Exerts Anti- Atherogenic Effects by Inhibiting Endoplasmic Reticulum (ER) Stress Induced by Disturbed Flow

  • Disturbed blood flow with low-oscillatory shear stress (OSS) is a predominant atherogenic factor leading to dysfunctional endothelial cells (ECs). Recently, it was found that disturbed flow can directly induce endoplasmic reticulum (ER) stress in ECs, thereby playing a critical role in the development and progression of atherosclerosis. Ursodeoxycholic acid (UDCA), a naturally occurring bile acid, has long been used to treat chronic cholestatic liver disease and is known to alleviate endoplasmic reticulum (ER) stress at the cellular level. However, its role in atherosclerosis remains unexplored. In this study, we demonstrated the anti-atherogenic activity of UDCA via inhibition of disturbed flow-induced ER stress in atherosclerosis. UDCA effectively reduced ER stress, resulting in a reduction in expression of X-box binding protein-1 (XBP-1) and CEBP-homologous protein (CHOP) in ECs. UDCA also inhibits the disturbed flow-induced inflammatory responses such as increases in adhesion molecules, monocyte adhesion to ECs, and apoptosis of ECs. In a mouse model of disturbed flow-induced atherosclerosis, UDCA inhibits atheromatous plaque formation through the alleviation of ER stress and a decrease in adhesion molecules. Taken together, our results revealed that UDCA exerts anti-atherogenic activity in disturbed flow-induced atherosclerosis by inhibiting ER stress and the inflammatory response. This study suggests that UDCA may be a therapeutic agent for prevention or treatment of atherosclerosis.

Effects of Antioxidant and Anti-inflammatory Activity of Allii Macrostemonis Bulbus Cheonghyeol Plus on the Inhibition of Atherosclerosis (해백청혈플러스(AMCP)의 항산화 및 항염증 작용을 통한 죽상동맥경화 억제 효과)

  • The purpose of this study was to investigate the antioxidant, anti-inflammatory and anti-cellular adhesion molecules effects of Allii Macrostemonis Bulbus, Artemisiae Capillaris Herba, Curcumae Radix, Crataegi Fructus, Salviae Militiorrhizae Radix complex extract(AMCP) on the inhibition of atherosclerosis in HUVEC. We measured DPPH radical scavenging activity and ABTS radical scavenging activity of AMCP to evaluate its antioxidant effect. And we also measured the expression level of NF-κB, IκBα, ERK, JNK, p38 proteins to evaluate its anti-inflammatory effect. Lastly, we measured the expression level of MCP-1, ICAM-1, VCAM-1 mRNA and their level to evaluate its anti-celluar adhesion molecules. AMCP did not show any cytotoxicity in HUVEC within the concentraion tested except for a concentration of 400 ㎍/㎖. AMCP increased the DPPH radical scavenging activitiy and ABTS radical scavenging activity in HUVEC as the concentration of AMCP rises. AMCP significantly reduced NF-κB, IκBα, JNK, ERK and p38 protein expression in HUVEC compared to control group. AMCP significantly reduced MCP-1, ICAM-1, VCAM-1 gene expresion in HUVEC compared to control group. AMCP significantly decreased the levels of MCP-1, ICAM-1, VCAM-1 in HUVEC compared to control group. These results suggest that AMCP has effects on antioxidation, anti-inflammation and anti-cellular adhesion molecule, which helps the treatment and prevention of dyslipidemia and atherosclerosis.

Efficacies of Sodium Carboxymethylcellulose, Oxidized Regenerated Cellulose and Vitamin E in Prevention Against Postoperative Adhesion Formation in Dogs (개에서 복강수술 후 유착방지에 대한 Sodium carboxymethylcellulose, Oxidized regenerated cellulose, Vitamin E의 효과)

  • This study was performed to compare the efficacy of the sodium carboxymethylcellulose (SCMC), oxidized regenerated cellulose (ORC), and their combination with vitamin E for the prevention of postoperative adhesions in the dog. Twenty five dogs were randomly divided into five groups ; non-treated group (Control Group), SCMC-treated group (SCMC Group), ORC-treated group (ORC Group), vitamin E and SCMC-treated group (SCMC + E Group), and vitamin E and ORC-treated group (ORC + E group). After laparotomy, 5 abrasions were made to induce intraperitoneal adhesions on the surface of the ileal serosa. The adhesions were occurred in serosa to mesentary (37.6%), serosa to serosa (24%), serosa to omentum (8.8%) and serosa to parietal peritoneum (3.2%). The incidences of adhesions were 92%, 84%, 64%, 56% and 68% in Control, SCMC, ORC, SCMC+E and ORC + E Group, respectively. The adhesion scores in SCMC+E Group were significantly lower than those in the other groups (p < 0.01). In conclusion, this study showed that oral supplements of vitamin E and intraperitoneal administration of 2% SCMC solution were effective on reducing intraperitoneal adhesions in the dog.

  • Using cement as a road subbase is economical, easily modified and supplemented and has excellent road pavement quality control. In addition, cement adheres well to sandy soils without adhesion, and it plays a role of permanently preserving adhesion in viscous soils with adhesion, so it can be widely applied as stable treatment with the advantages of increased strength, reduced compressibility. and improved durability. However, while cement is excellent in terms of strength for a road subbase, the material properties mean that it is difficult to maintain and reinforce when cracks or fractures occur due to dry shrinkage, and the pH increases in the ground due to hexavalent chromium eluting from cement. which can cause environmental problems such as groundwater contamination. Therefore, this study evaluates the usability of alternatives in the road base layer such as environmentally cementitious stabilizer and on-site soil generated from the site. We intend to reduce the environmental damage and evaluate the durability. To evaluate the applicability of the site, Environmental stability test and freeze-thaw test and wetting-drying test was conducted to evaluate the strength characteristics of alternative materials on the road through the limited performance evaluation of environmentally cementitious stabilizer. The test ranges were set at mixing ratios of 10%, 20%, and 30% and ages of three days, and 28 days old to evaluate the early strength and reference strength according to the mixing ratio of the environmentally cementitious stabilizer.

Dose Related Effects of Poloxamer/Sodium Alginate Mixture in Prevention of Postoperative Adhesion Formation in Dogs (개에서 Poloxamer/Sodium Alginate 혼합물의 용량에 따른 복강 유착방지 효과)

  • This study was performed to determine the minimum dose of Poloxamer/Sodium alginate (PX/SA) mixture on preventing intraperitoneal adhesions to evaluate organ toxicity. Twenty five healthy adult mongrel dogs (weighing 4.68${\pm}$1.67 kg) were divided into five experimental groups composed of five dogs respectively; negative control group (NC, non-treated), positive control group (PC, 2% carboxymethyl chitosan solution treated), and experiment 1 group (E1, 0.25 ml PX/SA mixture of abraded area), experiment 2 group (E2, 0.5 ml PX/SA mixture of abraded area), experiment 3 group (E3, 1.0 ml PX/SA mixture of abraded area). Venous blood specimens were collected from all experimental animals for hematologic and biochemical analysis: WBC, fibrinogen, AST, ALT, ALP, BUN and creatinine. The anti-adhesion effect was evaluated using a serosa abrasion model. The denuded ileum was coated with PX/SA mixture, carboxymethyl chitosan solution or neither. The tensile strength of the adhesion site was evaluated with a tensiometer. For histopathological examination, tissue samples of the liver and kidney were collected from all dogs. According to the results, the frequency and tensile strength values for adhesion separation in PX/SA group were significantly lower than those in negative control group (p < 0.05). In E2 group, the tensile strength was significantly decreased in consideration of PX/SA dose. The values of AST, ALT, ALP, BUN and creatinine of the control and the experimental groups showed no statistical differences. No obvious microscopic differences were noted among tissue sections obtained from all groups. The results suggest that PX/SA mixture may be effective on reducing peritoneal adhesion formation in dog and that 0.5 ml PX/SA mixture of abraded area is most effective dose. Moreover, PX/SA mixture was considered not to have toxicity for the liver and the kidney.

Therapeutic Effect of Prednisolone in Tuberculous Pleurisy -A prospective study for the prevention of the pleural adhesion- (결핵성 흉막염에서 프레드니솔론의 치료효과 -흉막유착 발생에 대한 전향적 연구-)

  • Background: The routine application of the combined regimen of corticosteroid-antituberculosis therapy to the tuberculous pleurisy remains controversial. Steroid therapy to tuberculous pleurisy could be effective on the acceleration of absorption of pleural effusion and symptom improvement, but there has been debate about the effect of prednisolone on the prevention of pleural adhesion. So we studied the efficacy of combined regimen of prednisolone-antituberculosis therapy on the absorption of pleural effusion and prevention of pleural adhesion. Method: A prospective, randomized study was performed in 82 patients, 50 patients(non-steroid group) were treated with only antituberculosis regimen for 6 months and in 32 patients(steroid group) prednisolone(30mg/day) were administered in addition to antituberculosis regimen for one months and tapered for another month. The amount of pleural effusion was compared at the beginning of treatment, 2nd month, 6th month and final visit with chest X-ray findings which were graded from grade 0(complete absorption) to grade 6(near total haziness). Results: The amount of pleural effusion of steroid group at 2nd month, 6th month and final visit was lesser than that of non-steroid group(P<0.05). The incidence of the complete absorption of the pleural effusion was 3/32(9.4%) in steroid group, 1/50(2%) in non-steroid group at 2nd month after treatment; and 12/32(37.5%) in steroid group, 6/50(12%) in non-steroid group at 6th month after treatment(P<0.05). At final observation, the incidence of residual pleural thickening was 15/32(47%) in steroid group and 37/50(74%) in non-steroid group(P<0.05). No serious side effects were noted during the treatment with prednisolone. Conclusion: The administration of prednisolone in conjunction with antituberculosis chemotherapy improved the absorption of pleural effusion and decreased the residual pleural thickening.

Microarray Analysis of the Hypoxia-induced Gene Expression Profile in Malignant C6 Glioma Cells

  • Hypoxia is commonly featured during glioma growth and plays an important role in the processes underlying tumor progression to increasing malignancy. Here we compared the gene expression profiles of rat C6 malignant glioma cells under normoxic and hypoxic conditions by cDNA microarray analysis. Compared to normoxic culture conditions, 180 genes were up-regulated and 67 genes were down-regulated under hypoxia mimicked by $CoCl_2$ treatment. These differentially expressed genes were involved in mutiple biological functions including development and differentiation, immune and stress response, metabolic process, and cellular physiological response. It was found that hypoxia significantly regulated genes involved in regulation of glycolysis and cell differentiation, as well as intracellular signalling pathways related to Notch and focal adhesion, which are closely associated with tumor malignant growth. These results should facilitate investigation of the role of hypoxia in the glioma development and exploration of therapeutic targets for inhibition of glioma growth.

  • Prostate cancer, with a lifetime prevalence of one in six men, is the second cause of malignancy-related death and the most prevalent cancer in men in many countries. Nowadays, prostate cancer diagnosis is often based on the use of biomarkers, especially prostate-specific antigen (PSA) which can result in enhanced detection at earlier stage and decreasing in the number of metastatic patients. However, because of the low specificity of PSA, unnecessary biopsies and mistaken diagnoses frequently occur. Prostate cancer has various features so prognosis following diagnosis is greatly variable. There is a requirement for new prognostic biomarkers, particularly to differentiate between inactive and aggressive forms of disease, to improve clinical management of prostate cancer. Research continues into finding additional markers that may allow this goal to be attained. We here selected a group of candidate biomarkers including PSA, PSA velocity, percentage free PSA, $TGF{\beta}1$, AMACR, chromogranin A, IL-6, IGFBPs, PSCA, biomarkers related to cell cycle regulation, apoptosis, PTEN, androgen receptor, cellular adhesion and angiogenesis, and also prognostic biomarkers with Genomic tests for discussion. This provides an outline of biomarkers that are presently of prognostic interest in prostate cancer investigation.

Effect of Vitamin E Against the Cytotoxicity of Reactive Oxygen Species on Vascular Endothelial Cells

  • Reactive oxygen species (ROS) is one of the main pathological factors in endothelial disorder. For example, an atherosclerosis is induced by the dysfunction of vascular endothelial cells. The dysfunction of vascular endothelial cells cascades to secrete intercellular adhesion molecule (ICAM)-l substance by ROS. Therefore, The ROS is regraded as an important factor of the injury of vascular endothelial cells and inducement of atherosclerosis. Oxygen radical scavengers playa key role to prevention of many diseases mediated by oxidative stress of ROS. In this study, the toxic effect of ROS on vascular endothelial cells and the effect of antioxidant, vitamin E on bovine pulmonary vascular endothelial cell line (BPVEC) treated with hydrogen peroxide were examined by the colorimetric assay. ROS decreased remarkably cell viability according to the dose- and time-dependent manners. In protective effect of vitamin E on BPVEC treated with hydrogen peroxide, vitamin E increased remarkably cell viability compared with control after BPVEC were treated with $15{\mu}M$ hydrogen peroxide for 6 hours. From these results, it is suggested that ROS has cytotoxicity on cultured BPVEC and oxygen radical scavenger such as vitamin E is very effective in prevention of oxidative stress-induced cytotoxicity.
