A STUDY ON OSTEOBLAST-LIKE CELL RESPONSES TO SURFACE-MODIFIED TITANIUM

  • Statement of problem: The success of implants depends on intimate and direct contact of implant material on bone tissue and on functional relationship with soft tissue contact. Creation and maintenance of osseointegration depend on the understanding of the tissue's healing, repairing, and remodeling capacity and these capacities rely on cellular behavior. Altering the surface properties can modify cellular responses such as cell adhesion, cell motility, bone deposition, Therefore, various implant surface treatment methods are being developed for the improved bone cell responses. Purpose: The purpose of this study was to evaluate the responses of osteoblast-like cells to surface-modified titanium. Materials and Methods: The experiment was composed of four groups. Group 1 represented the electropolished surface. Group 2 surfaces were machined surface. Group 3 and Group 4 were anodized surfaces. Group 3 had low roughness and Group 4 had high roughness. Physicochemical properties and microstructures of the discs were examined and the responses of osteoblast-like cells to the discs were investigated. The microtopography was observed by SEM. The roughness was measured by three-dimension roughness measuring system. The microstructure was analyzed by XRD, AES. To evaluate cell responses to modified titanium surfaces, osteoblasts isolated from calvaria of neonatal rat were cultured. Cell count, morphology, total protein measurement and alkaline phosphatase activities of the cultures were examined. Results and Conclusion: The results were as follows 1. The four groups showed specific microtopography respectively. Anodized group showed grain structure with micropores. 2. Surface roughness values were, from the lowest to the highest, electropolished group, machined group, low roughness anodized group, and high roughness anodized group. 3. Highly roughened anodized group was found to have increased surface oxide thickness and surface crystallinity. 4. The morphology of cells, flattened or spherical, were different from each other. In the electropolished group and machined group, the cells were almost flattened. In two anodized groups, some cells were spherical and other cells were flattened. And the 14 day culture cells of all of the groups were nearly flattened due to confluency. 5. The number of attached cells was highest in low roughness anodized group. And the machined group had significantly lower cell count than any other groups(P<.05). 6. Total protein contents showed no difference among groups. 7. The level of alkaline phosphatase activities was higher in the anodized groups than electropolished and machined groups(P<.05).

  • Corneal endothelium is mono-inner cell layer of cornea and lay on Descmet's membrane which comprised of various proteins called extracellular matrix such as fibronectin, collagen, laminin, and proteoglycan, etc. In this study, we fabricated transparent poly(lactic-co-glycolic acid) (PLGA) film because PLGA is widely used for tissue engineering based on their properties. We investigated the behaviors of rabbit corneal endothelial cells (rCEnCs) on PLGA film surfaces coated with various cell-adhesive molecules like fibronectin, laminin, collagen type I and IV and FNC coating mix. The morphologic images, proliferation and adhesion assay, immunofluorescence for ZO-1 and $Na^+/K^+-ATPase$ and RT-PCR for expression of specific markers were conducted. These results showed that PLGA film plays a role as CEnC carriers in vitro and the cell-adhesive molecules give positive effects on the behaviors of rCEnC.

Study on Incineration Behavior of Heavy Oil Fly Ash for Valuable Metal Recovery (유가금속(有價金屬) 회수(回收)를 위한 중유회(重油灰)의 연소거동(燃燒擧動)에 관한 연구(硏究))

  • To design and construct a moving bed stoker incinerator for incineration treatment of the domestic oil fly ash, operating condition and moving bed area of incinerator were determined by performing incinerate experiment of the oil fly ash in the muffle furnace which simulates moving bed stoker incinerator in all conditions. Incineration process of the oil fly ash could be divided into 3 stages, every stage needs the appropriate operating condition for effective incineration. The optimum content of water in the heavy oil fly ash was found to be 20 wt% to prevent the ash from flying and reduce the volume. Science combustion rate of oil fly ash depends on the oxygen content, the incinerator must have a equipment to control the oxygen content in the combustion air. The optimum temperature was $750{\sim}800^{\circ}C$ in order to prevent adhesion to the stocker and evaporation of metal compounds of low melting point. Uniform combustion reaction and acceleration of combustion rate required agitation during the combustion of oil fly ash. The incineration rate was $12.53kg/m^2hr$ and the working area of moving bed incinerator was found to be $60m^2$ to incinerate 18 tons of oil fly ash per day.

On the Properties of TLCP/PBT Blends Prepared by In Situ Polymerization in PBT Solution (In situ 중합에 의해서 제조된 TLCP/PBT 블렌드의 특성 연구)

  • A new thermotropic liquid crystalline polymer(TLCP) containing a triad aromatic ester type mesogenic unit and butylene terephthalate unit(BT) in the main chain was synthesized by polycondensation reaction. The TLCP synthesized showed nematic mesophasic behavior and its transition temperature from solid to mesophase was $260^{\circ}C$. The TLCP/PBT blends were prepared by in-situ polymerization in PBT solution and characterized by differential scanning calorimeter(DSC), thermogavimetric analyzer(TGA), scanning electron microscope(SEM), x-ray diffractometer(XRD), and dynamic mechanical thermal analyze, (DMTA). The blends showed well dispersed TLCP phases with domain sizes $0.05{\sim}0.2{\mu}m$ in the PBT matrix. As the increasing TLCP content from 5 to 20 wt%, ${\Delta}Hm$ values of pure PBT in the blend were increased because TLCP acts as a nucleating agent in the PBT matrix. The mechanical properties of the blends depended on the TLCP contents because the TLCP acted effectively as a reinforcing material in the PBT matrix. The blends showed good interfacial adhesion between the TLCP phase and PBT matrix.The blends prepared by in-situ polymerization showed higher mechanical properties and well dispersed TLCP domains than those of the blends prepared by melt blending.

Evaluation of Fluidity Over Time and Mechanical Properties of Cement-based Composite Materials for 3D Printing (3D 프린팅용 시멘트계 복합재료의 경시변화 및 역학적 특성평가)

  • This study evaluated changes in fluidity and rheological properties over time for 3D printed composite materials, and evaluated compressive strength and splitting tensile strength properties for laminated and molded specimens. The composite material for 3D printing starts to change rapidly after 30 minutes of extrusion, and the viscosity of the material tends to be maintained up to 90 minutes, but it was confirmed that construction within 60 minutes after mixing is effective. The compressive strength of the laminated test specimen showed equivalent or better performance at all ages compared to the molded test specimen. In the stress-strain curve of the laminated specimen, the initial slope was similar to that of the molded specimen, but the descending slope was on average 1.9 times higher than that of the molded specimen, indicating relatively brittle behavior. The splitting tensile strength of the P-V laminated specimen was about 6% lower than that of the molded specimen. It is judged that this is because the interfacial adhesion force against the vertical load is affected by the pattern direction of the laminated test specimen.

Homogenization of Plastic Behavior of Metallic Particle/Epoxy Composite Adhesive for Cold Spray Deposition (저온 분사 공정을 위한 금속입자/에폭시 복합재료 접착제의 소성 거동의 균질화 기법 연구)

  • A combination of a metallic mesh and an adhesive layer of metallic particle/epoxy composite was introduced as an intermediate layer to enhance the adhesion between cold-sprayed particles and fiber-reinforced composites (FRCs). Aluminum was considered for both the metallic particles in the adhesive and the metallic mesh. To predict the mechanical characteristics of the intermediate bond layer under a high strain rate, the properties of the adhesive layer needed to be calculated or measured. Therefore, in this study, the Al particle/epoxy adhesive was homogenized by using a rule of mixture. To verify the homogenization, the penetration depth, and the thickness decrease after the cold spray deposition from the undeformed surface, was monitored with FE analysis and compared with experimental observation. The comparison displayed that the penetration depth was comparable to the diameters of one cold spray particle, and thus the homogenization approach can be reasonable for the prediction of the stress level of particulate polymer composite interlayer under a high strain rate for cold spray processing.

Properties of Liquid Crystalline Polyester/Poly(ethylene 2,6-naphthalate) Blend Fibers (액정 폴리에스테르/PEN 블렌드 섬유의 성질)

  • A thermotropic liquid crystalline polymer(TLCP) which has flexible butylene/hexylene spacers in the main chain and a triad aromatic ester type mesogenic unit containing a naphthyl group was prepared by solution polycondensation. The in-situ composites based on poly(ethylene 2,6-naphthalate) (PEN) and a thermotropic liquid crystalline polymer(TLCP) were prepared and melt spun at different TLCP contents and different draw ratios to produce monofilaments. Blends of the TLCP with PEN were investigated in terms of thermal, mechanical properties and morphology. The TLCP synthesized showed nematic mesophasic behavior and its transition temperature to isotropic melt from mesophase was 249℃. The blends showed well dispersed TLCP phases in the PEN matrix without macroscopic phase separation. Inclusion of TLCP in the blends decreased the cold crystallization temperature of PEN in the blend, therefore, the TLCP acts as a nucleating agent in the blend and showed good interfacial adhesion between the dispersed LCP phases and PEN matrix with domain sizes 40~50 nm in diameter and well developed fibrillation in the monofilaments. The TLCP acted effectively as a reinforcing material in the PEN matrix at the 10wt% level, it led to an increase of initial modulus up to 270% and tensile strength by 235%, while the elongation rate increasing with higher draw ratios.

Correlation Analysis between Damage of Expansion Joints and Response of Deck in RC Slab Bridges (RC 슬래브교의 신축이음 손상과 바닥판 응답과의 상관관계 분석)

  • RC slab bridges account for the largest portion of deteriorated bridges in Korea. However, most RC slabs are not included in the first and second classes of bridges, which are subject to bridge safety management and maintenance. The highest damaged components in highway bridges are the subsidiary facilities including expansion joints and bearings. In particular, leakage through expansion joints causes deterioration and cracks of concrete and exposure of reinforced bars. Therefore, this study analyzed the effect of adhesion damage at expansion joints on the response of the deck in RC slab bridges. When the spacing between the expansion joints at both ends was closely adhered, cracks occurred in the concrete at both ends of the deck due to the resistance rigidity at the expansion joints. Based on the response results, the correlation analysis between displacements in the longitudinal direction of the expansion joint and concrete stress at both ends of the deck for each damage scenario was performed to investigate the effect of the occurrence of damage on the bridge behavior. When expansion joint devices at both sides were damaged, the correlation between displacement and stress showed a low correlation of 0.18 when the vehicles proceeded along all the lanes. Compared with those in the intact state, the deflections of the deck in the damaged case at both sides showed a low correlation of 0.34 to 0.53 while the vehicle passed and 0.17 to 0.43 after the vehicle passed. This means that the occurrence of cracks in the ends of concrete changed the behavior of the deck. Therefore, data-deriven damage detection could be developed to manage the damage to expansion joints that cause damage and deterioration of the deck.

A Study on the Optimal Process Parameters for Recycling of Electric Arc Furnace Dust (EAFD) by Rotary Kiln (Rotary Kiln에 의한 전기로 제강분진(EAFD)의 재활용을 위한 최적의 공정변수에 관한 연구)

  • As a recycling technology for recovering zinc contained in large amounts in electric arc furnace dust (EAFD), the most commercialized technology in the world is the Wealz Kiln Process. The Wealz Kiln Process is a process in which components such as Zn and Pb in EAFD are reduced/volatile (endothermic reaction) in high-temperature Kiln and then re-oxidized (exothermic reaction) in the gas phase and recovered in the form of Crude zinc oxide (60wt%Zn) in the Bag Filter installed at the rear end of Kiln. In this study, an experimental Wealz kiln was produced to investigate the optimal process variable value for practical application to the recycling process of large-scale kiln on a commercial scale. Additionally, Pellets containing EAFD, reducing agents, and limestone were continuously loaded into Kiln, and the amount of input, heating temperature, and residence time were examined to obtain the optimal crude zinc oxide recovery rate. In addition, the optimal manufacturing conditions of Pellets (drum tilt angle, moisture addition, mixing time, etc.) were also investigated. In addition, referring to the SiO2-CaO-FeO ternary system diagram, the formation behavior of a low melting point compound, a reaction product inside Kiln according to the change in the basicity of Pellet, and the reactivity (adhesion) with the castable constructed on the inner wall of Kiln were investigated. In addition, in order to quantitatively investigate the possibility of using anthracite as a substitute for Coke, a reducing agent, changes in the temperature distribution inside Kiln, where oxidation/reduction reactions occur due to an increase in the amount of anthracite, the quality of Crude zinc oxide, and the behavior of tar in anthracite were also investigated.
