토끼에서 ACAT 억제에 의한 3,4-다이하이드록시 하이드로시나믹산의 동맥경화 완화 효과 (Attenuation of Atherosclerosis by 3,4-Dihydroxy-Hydrocinnamic Acid in Rabbits by Partial Inhibition of ACAT)

  • 폴리페놀 성분은 심혈관질환에서 좋은 효과를 나타낸다고 보고되고 있다. 폴리페놀성 화합물인 3,4-다이하이드록시 하이드로시나믹산은 항산화 활성과 항암 활성을 나타낸다고 보고되었다. 이 연구의 목적은 3,4-다이하이드록시 하이드로시나믹산이 항동맥 경화 효과를 나타내는지를 뉴질랜드 흰 토끼에서 평가하는 것이다. 8주동안 고콜레스테롤 식이를 급여한 대조그룹 토끼의 광범위한 동맥 부위에서 동맥경화 초기병변이 형성되었다. 반면에 고콜레스테롤 식이를 급여하면서 3,4-다이하이드록시 하이드로시나믹산을 투여한 토끼에서는 대조 그룹의 토끼에 비해 동맥경화 병변 형성이 감소하였고, 병변 내로 침윤한 대식세포의 양도 감소하였다. 이러한 3,4-다이하이드록시 하이드로시나믹산의 효과에서 전신적으로나 국부적으로 독성이 관찰되지 않았다. 간의 아실-코엔자임 A: 콜레스테롤 아실트렌스페라제 활성이 고콜레스테롤 식이를 급여하면서 3,4-다이하이드록시 하이드로시나믹산을 투여한 토끼에서 대조 그룹의 토끼에 비해 22% 감소하였다. 이러한 연구 결과는 3,4-다이하이드록시 하이드로시나믹산이 토끼에서 아실-코엔자임 A: 콜레스테롤 아실트렌스페라제를 억제함으로써 항동맥경화 효과를 나타낸다는 것을 증명해 준다.

  • Twigs from llex macropoda were extracted with MeOH, and the concentrated extracts were partitioned with $CH_2Cl_2$, EtOAc, n-BuOH, and $H_2O$. Repeated column chromatography of the $CH_2Cl_2$ fraction ultimately resulted in the isolation of two compounds, via activity-guided fractionation, using ACAT inhibitory activity measurements. According to the physico-chemical data, the chemical structures of these isolated compounds were identified as lupeol (1) and betulin (2). Compounds 1 and 2 were shown to inhibit the activity of hACAT-1 and hACAT-2 in a dose-dependent manner, and compounds 1 and 2 inhibited hACAT-1 with $IC_{50}$ values of 48 and $83{\mu}M$, respectively.

  • Study of Acyl-CoA:cholesterol acyltransferase (ACAT) inhibitors from medicinal plants, we found strong inhibitory activity of ACAT enzyme from rat liver microsome by the CHC1$_3$ extract of Piper longum. Bioactivity-guided fractionation led to the isolation of Guineensine (1), its structure was elucidated by spectroscopic (IR, UV, MS and NMR) means. It inhibited ACAT activity in a dose-dependent manner with IC$\sub$50/ values of 1.2 $\mu\textrm{g}$/ml on in vitro assay using rat liver microsome.

상추 (Lactuca sativa L.) callus로부터 ACAT 억제 활성물질, phytol의 생산 (Production of Phytol, an ACAT Inhibitor, from Callus Culture of Lettuce (Lactuca sativa L.))

  • 상추의 기내 callus 배양으로부터 ACAT (Acyl-CoA: Cholesterol acyltransferase) 억제 활성물질로 알려진 phytol 의 생산가능성이 연구되었다. Callus는 0.5mg.L$^{-1}$ NAA가 첨가된 MS 배지에 4주간 치상된 자엽 절편체로부터 유도되었으며, 다양한 수준의 chlorophyll을 함유하고 있었다. Phytol이 chlorophyll분자를 구성하는 물질로 알려져 있음에 도 불구하고, phytol의 추출량은 녹색의 자엽에서 보다 callus 에서 높게 나타났다. GC 분석결과, phytol의 추출량은 callus의 chlorophyll 함량과 비례하지 않는 것으로 나타났으며, callus의 chlorophyll 함량수준은 callus의 형성능과 관련이 있는 것으로 사료되었다. 본 연구의 결과는 phytol의 기내생산에 있어서 상추의 callus 배양의 잠재성을 제시한다 하겠다.

The effects of black garlic (Allium satvium) extracts on lipid metabolism in rats fed a high fat diet

  • BACKGROUD/OBEJECTIVES: The mechanism of how black garlic effects lipid metabolism remains unsolved. Therefore, the objectives of this study were to determine the effects of black garlic on lipid profiles and the expression of related genes in rats fed a high fat diet. MATERIALS/METHODS: Thirty-two male Sqrague-Dawley rats aged 4 weeks were randomly divided into four groups (n=8) and fed the following diets for 5 weeks: normal food diet, (NF); a high-fat diet (HF); and a high-fat diet + 0.5% or 1.5% black garlic extract (HFBG0.5 or HFBG1.5). Body weights and blood biochemical parameters, including lipid profiles, and expressions of genes related to lipid metabolism were determined. RESULTS: Significant differences were observed in the final weights between the HFBG1.5 and HF groups. All blood biochemical parameters measured in the HFBG1.5 group showed significantly lower values than those in the HF group. Significant improvements of the plasama lipid profiles as well as fecal excretions of total lipids and triglyceride (TG) were also observed in the HFBG1.5 group, when compared to the HF diet group. There were significant differences in the levels of mRNA of sterol regulatory element binding protein-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), and glucose-6-phosphate dehydrogenase (G6PDH) in the HFBG1.5 group compared to the HF group. In addition, the hepatic expression of (HMG-CoA) reductase and Acyl-CoA cholesterol acyltransferase (ACAT) mRNA was also significantly lower than the HF group. CONCLUSIONS: Consumption of black garlic extract lowers SREBP-1C mRNA expression, which causes downregulation of lipid and cholestrol metahbolism. As a result, the blood levels of total lipids, TG, and cholesterol were decreased.

곤충자원으로부터 항동맥경화 활성물질 탐색 (Screening of Anti-atherogenic Substances from Insect Resources)

  • Lipoprotein-associated phospholipase $A_2\;(Lp-PLA_2)$ is a potential biomarker of coronary heart disease and plays an important proinflammatory role in the progression of atherosclerosis. Also acyl-CoA: cholesterol acyltransferase (ACAT) and oxidized low-density lipoprotein (LDL) playa key role in atherosclerosis, respectively. And so, the inhibitory activities of the methanol extracts of 42 insect resources were examined on $Lp-PLA_2$, ACAT, and LDL oxidation for screening of anti-atherogenic substances. Among them, the methanol extracts of Eurydema rugosa significantly inhibited all of upper three activities. Several kinds of tested insects having high inhibitory effect with the methanol extracts were extracted with n-hexane, ethyl acetate, and acetone, and their inhibitory activities were tested.

Study on the hypochlolesterolemic and antioxidative effects of tyramine derivatives from the root bark of Lycium chenese Miller

  • The aim of the present study was to investigate the hypocholesterolemic effect and potential of tyramine derivatives from Lycii Cortex Radicis (LCR), the root bark of lycium (Lycium chenese Miller) in reducing lipid peroxidation. The activities of enzymes, hepatic 3-hydroxy 3-methylglutaryl (HMG) CoA reductase and acyl-CoA:cholesterol acyltransferase (ACAT) and LDL oxidation were measured in vitro and animal experiments were also performed by feeding LCR extracts to rats. The test compounds employed for in vitro study were trans-N-p-coumaroyltyramine (CT) and trans-N-feruloyltyramine (FT), LCR components, N-(p-coumaroyl)serotonin (CS) and N-feruloylserotonin (FS) from safflower seeds, ferulic acid (FA) and 10-gingerol. It was observed that FT and FS at the concentration of 1.2 mg/mL inhibited liver microsomal HMG CoA reductase activity by ~40%, but no inhibition of activity was seen in the cases of CT, CS, FA and 10-gingerol. Whereas, ACAT activity was inhibited ~50% by FT and CT, 34-43% by FS and CS and ~80% by 10-gingerol at the concentration of 1 mg/mL. A significant delay in LDL oxidation was induced by CT, FT, and 10-gingerol. For the animal experiment, five groups of Sprague-Dawley male rats were fed high fat diets containing no test material (HF-control), 1 and 2% of LCR ethanol extract (LCR1 and LCR2), and 1% of extracts from safflower seed (Sat) and ginger (Gin). The results indicated that total cholesterol level was significantly lower in Saf, LCR2 and Gin groups, and HDL cholesterol level was lower only in Gin group when compared with HF-control group; while there was no difference in the serum triglyceride levels among the five experimental groups. The level of liver cholesterol was significantly lower in LCR1 and LCR2 groups than HF-control Serum levels of TBARS were significantly lower only in LCR2 group when compared with HF-control group. From the observed results, we concluded that LCR can be utilized as a hypocholesterolemic ingredient in combination with ginger, especially for functional foods.
