다제내성 Staphylococcus aureus에 항균활성을 나타내는 CNU30122 균주가 생산하는 항생물질 (An Antibiotic against Multidrug-resistant Staphylococcus aureus Produced by Strain CNU30122)

  • 다제내성 Staphylococcus aureus균주에 강력한 항균활성을 나타내는 CNU30122 균주를 선발하였다. 선발된 CNU30122균주의 배양액 추출물로부터 Diaion HP-20 column chromatograpy, ethylacetate 추출, silica gel 및 Sephadex LH-20 column chromatography, HPLC 등에 의해 백색분말의 순수한 화합물을 분리정제 하였다. 본 물질의 구조분석을 위하여 $^1H$$^{13}C\;NMR,\;DEPT,\;^1H-^1H\;COSY,\;^1H-^{13}C\;COSY$ 및 HMBC spectrum을 분석한 결과 본 물질은 분자량 516, 분자식 $C_{31}H_{48}O_6$의 tetracyclic triterpenoic acid 계열인 fusidic acid로 동정 되었다. 본화합물은 streptomycin, pecicillin, kanamycin, tetracycline 등의 약제에 내성균주인 Staphylococcus aureus R209 균주에 선택적으로 강한 항균활성을 나타내었다.

Cytotoxic Activities of Indigenous Plant Extracts in Cultured Human Cancer Cells

  • In continuous efforts for discovery of novel potent antitumor agents from natural products, fifty-seven methanolic extracts derived from indigenous Korean plants were primarily evaluated for in vitro cytotoxic activity in cultured human lung (A549) and colon (Col2) cancer cells. As a result, 16 plant extracts were found to be active against A549 cells and 15 extracts were active against Col2 cells in the criteria of $IC_{50}$<$50\;{\mu}g/ml$. In particular, the extracts of Calystegia soldanella $(IC_{50}$<$8.0\;{\mu}g/ml\;in\;A549;IC_{50}=27.4\;{\mu}g/ml\;in\;Col2)$, Heloniopsis orientalis $(IC_{50}=4.6\;{\mu}g/ml\;in\;A549; IC_{50}=4.5\;{\mu}g/ml\;in\;Col2)$, and Thuja koraiensis $(IC_{50}=1.2\;{\mu}g/ml\;in\;A549;IC_{50}=0.6\;{\mu}g/ml\;in\;Col2)$ showed a potent cytotoxic activity. Further study for the identification of active compounds from these lead extracts might be warranted.

Successful treatment with vedolizumab in an adolescent with Crohn disease who had developed active pulmonary tuberculosis while receiving infliximab

  • Vedolizumab (VDZ) has been approved for the treatment of inflammatory bowel diseases (IBDs) in patients aged ≥18 years. We report a case of a pediatric patient with Crohn disease (CD) who was successfully treated with VDZ. A 16-year-old female developed severe active pulmonary tuberculosis (TB) during treatment with infliximab (IFX). IFX was stopped, and TB treatment was started. After a 6-month regimen of standard TB medication, her pulmonary TB was cured; however, gastrointestinal symptoms developed. Due to the concern of the patient and parents regarding TB reactivation on restarting treatment with IFX, VDZ was started off-label. After the second dose of VDZ, the patient was in clinical remission and her remission was continuously sustained. Ileocolonoscopy at 1-year after VDZ initiation revealed endoscopic healing. Therapeutic drug monitoring conducted during VDZ treatment showed negative antibodies to VDZ. No serious adverse events occurred during the VDZ treatment. This is the first case report in Korea demonstrating the safe and effective use of VDZ treatment in a pediatric CD patient. In cases that require recommencement of treatment with biologics after recovery of active pulmonary TB caused by anti-tumor necrosis factor agents, VDZ may be a good option even in pediatric IBD.

Anti-Melanogenic Effect of Dendropanax Morbiferus and Its Active Components via Protein Kinas e A/Cyclic Adenos ine Monophosphate-Responsive Binding Protein-and p38 Mitogen-Activated Protein Kinase-Mediated Microphthalmia-Associated Transcription Factor Downregulation

  • Dendropanax morbiferus H. Lev has been reported to have some pharmacologic activities and also interested in functional cosmetics. We found that the water extract of D. morbiferus leaves significantly inhibited tyrosinase activity and melanin formation in α-melanocyte stimulating hormone (MSH)-induced B16-F10 cells. D. morbiferus reduced melanogenesis-related protein levels, such as microphthalmia? associated transcription factor (MITF), TRP-1, and TRP-2, without any cytotoxicity. Two active ingredients of D. morbiferus, (10E)-9,16-dihydroxyoctadeca-10,17-dien-12,14-diynoate (DMW-1) and (10E)-(?)-10,17-octadecadiene-12,14-diyne-1,9,16-triol (DMW-2) were identified by testing the anti-melanogenic effects and then by liquid chromatography-tandem mass spectrometry (LC/MS/MS) analysis. DMW-1 and DMW-2 significantly inhibited melanogenesis by the suppression of protein kinase A (PKA)/cyclic AMP (cAMP)-responsive binding protein (CREB) and p38 MAPK phosphorylation. DMW-1 showed a better inhibitory effect than DMW-2 in α-MSH-induced B16-F10 cells. D. morbiferus and its active component DMW-1 inhibited melanogenesis through the downregulation of cAMP, p-PKA/CREB, p-p38, MITF, TRP-1, TRP-2, and tyrosinase. These results indicate that D. morbiferus and DMW-1 may be useful ingredients for cosmetics and therapeutic agents for skin hyperpigmentation disorders.

광물 자원에서 유래된 원료 의약품 및 첨가제의 사례 연구 (Case Study of Pharmaceutical Ingredients Derived from Clay Minerals)

  • 약학 분야에서 점토광물은 점토광물 자체의 약리작용을 확인하고 원료의약품으로 활용하거나 희석제, 유화제, 점증제, 활택제 등 의약품 제형의 완성도를 높이는 첨가제로서 사용되고 있다. 벤토나이트(Bentonite), 카올린(Kaolin), 규산알루민산마그네슘(Magnesium aluminum silicate), 탤크(Talc) 등은 원료의약품 혹은 첨가제로서 활용 가능한 대표적인 점토 광물로 국내외 의약품 공정서에 수재되어 있고, 약학적 활용시 의약품등급으로 규제되고 있다. 본 논문에서는 공정서에 수재된 점토 광물을 중심으로 공정서의 규격을 확인하고, 점토광물의 특성 및 원료의약품 혹은 의약품 첨가제로서의 점토 광물에 대한 활용 사례를 소개하고자 한다. 결론적으로 점토광물을 제약산업에 활용하는 것은 점토광물의 고부가가치화를 위한 한 가지 방법이 될 수 있으며, 자원의 개발 및 활용이라는 측면에서 매우 유용할 것이라 사료된다.

고추 부위별추출물에 의한 종양세포의 세포사유도 - Hepatoma 세포와 MCE-7 세포 - (Induction of Cancer Cell Apoptosis by the Extract of Capsicum annuum L. var. angulosum Mill Sorted According to the Parts in Hepatoma Cells and MCF-7 Cells)

  • Under the active search for biologically active novel agents for cancer prevention and treatment, some agents have been found from plants which are easily available. Our previous research on them revealed that C. annuum L. var. angulosum Mill have high antiproliferating effect on cancer cells. However, it has not been known whether the anticancer efficacy is different according to each part of C. annuum L. var. angulosum Mill or whether it can be changed by timing of harvest or solvent for extraction. Thus we compared the efficacy of each part of C. annuum L. var. angulosum Mill and assessed how much difference in the efficacy can be made according to the time of harvest or solvents for extraction. We observed the morphologic change and apoptosis 48 hr after treatment with the extract of each part of C. annuum L. var. angulosum Mill in MCF-7 mammary gland adenocarcinoma cells and human hepatoma cells. We also counted cancer cells by trypan blue method and MTT method to check the cytotoxicity. The leaf extract showed the highest anticancer effect among all the parts of C. annuum L. var. angulosum Mill; 50% and 70% reduction in the number of cancer cells was observed at 25 $\mu\textrm{g}$/mι and 50 $\mu\textrm{g}$/mι, respectively. It was more than 2 times as potent as 5-fluorouracil (5-FU). We found chromosomal fragmentation, clumping, and destuction by PI staining, and DNA fragmentation by electrophoresis. In conclusion, this study suggests that leaf extraction using water as solvent has the highest antiproliferative and apoptotic activity in cancer cells compared with other parts of extraction.

  • Strain BCNU 5011 was isolated from forest soil samples collected in the Taebaek mountain in the Gangwon province, Korea. The biochemical, physiological and 16S rRNA sequence analysis strongly indicated that this isolate was most closely related to Paenibacillus polymyxa. A maximum production level of antimicrobial substances of Paenibacillus sp. BCNU 5011 was achieved under aerobic incubation at $30^{\circ}C$ for 3 days in SST broth.Paenibacillus sp. BCNU 5011 showed a broad spectrum of activity against Gram positive and Gram negative bacteria, including methicllinresistant Staphylococcus aureus (MRSA). Paenibacillus sp. BCNU 5011 was also shown to inhibit the growth of different potential human pathogenic bacteria and fungi in vitro. Peptide extract showed better antimicrobial activity than solvent extracts. But active antimicrobial compounds might be included in both peptide extract and solvent extracts. Further separation, purification and identification of active principles leads project to develop antimicrobial agents and anti-MRSA agents.

전착제 가용이 6-Benzylamino Purine의 사과묘목 덧가지 발생촉진에 미치는 영향 (Effects of Surface-active Agents to 6-Benzylamino Purine on Feathering of Maiden Apple Trees)

  • 사과묘목의 덧가지 발생을 촉진하기 위하여 BAP 처리에 대한 전착제의 가용효과를 확인하기 위하여 Spreader 500ppm, Cover 500ppm, Spreadersticker 250ppm, Siloxane 350ppm, Tween 201%를 각각 BAP 400ppm에 가용하여 3회 처리한 다음 BAP 단용처리구와 덧가지 발생을 비교하였다. Tween 20을 제외하면 공시한 전착제는 모두 가용효과가 없었다. Tween 20의 적정 가용농도를 구명한 결과 0.2, 0.5, 0.8, 1.0% 모두 BAP 400ppm 단용 4회 처리에 비해 덧가지 발생이 현저히 많아 가용효과가 인정되었으나 농도간에는 뚜렷한 차이를 나타내지 않았다.

한국영화의 초국가적 수용: 영화리뷰를 중심으로 (Transnational Reception of Korean Film: Analyses of Film Reviews)

  • This paper is based on the view that film should be conceived as a form of cultural practice whose meaning is always in the process of being produced within diverse socio-cultural contexts and aims to examine the ways in which the meaning of Korean film is (re)mediated or received in diverse cultural contexts outside the country. In this paper, we employ two theoretical grounds. Firstly, it positions itself in line with the audience studies within the field of cultural studies where the audience is conceived as active agents who produce the meaning of a popular culture text. The recruitment of the theoretical propositions from the audience studies enables recognition of the significance of the reception in film practice which recently seems to be oriented on production and distribution. Secondly, we conceive transnationality of film as that which is being produced in the process of transaction between the film and the audience, that is to say, transnationality is a form of discourse that emerges upon cultural interaction. The empirical work involves examination of a set of reviews of four films--Chihwaseon, Oldboy, Thirt, Poety--that have been published in daily newspapers and some popular film magazines in the U. S., the U. K. and France. Through the analysis of the film reviews, we identify four interpretive schemes or rather discourses recruited via which the Korean films are approached and understood: auteurism, formalism, universal themes, emotional response. We propose that these four kinds of discourse provide a common ground for the audience from different cultural backgrounds to understand Korean film. Furthermore, we also suggest that transnationality of Korean cinema needs to be reconsidered in terms of the reception as the audience from different socio-cultural backgrounds should be understood as active agents who are capable of engaging in Korean cultural texts such as film in their own way producing various meanings and these are also constituent of the meaning of the cultural texts.

이동에이전트 기반 능동규칙을 이용한 분산형 절전제어시스템 (Distributed Power Saving Control System Using Mobile Agent Based Active Rules)

  • 본 논문에서 제안하는 이동에이전트를 이용한 분산형 절전제어 방법은 다수의 센서와 이동에이전트를 이용하여 능동적 지능적으로 조명장치의 제어(on/off 및 디밍 조절 등)를 가능하게 하여 에너지 절감에 효과적이며, 다양한 능동규칙들의 적용에 의한 원격감시 및 제어기능 추가가 용이하므로 효율적 절전제어시스템 설계 및 구축에 따른 비용절감 등의 효과를 유도한다. 또한, 이동에이전트 기반의 센서네트워크 미들웨어의 기능을 통하여 상황정보나 사용자의 감성 등을 규칙화하여 실시간으로 이벤트 처리 및 제어함으로써 센싱 데이터의 효용성을 향상시킨다. 본 논문의 결과는 다양한 능동적 센서네트워크 응용들의 개발에 있어 제안한 이동에이전트를 이용한 제어시스템의 활용가능성을 제시한다.
