Biphenyl dimethyl dicarboxylate (DDB) is a hepatoprotectant, which is used as an adjuvant agent in a treatment for chronic hepatitis. Amantadine is an antiviral agent, which is utilized primarily in the treatment of influenza, but also, occasionally in the treatment of hepatitis C. In a previous study, we reported that DDB, coupled with amantadine, would exert an anti-HBV effect, via the induction of interferon-inducible gene expression in the HepG2 2.2.15 cell line. The primary objective of the present study was to determine whether or not DDB and/or amantadine exhibit anti-HBV properties, and what mechanisms of action might be involved in such properties. In our study, we were able to determine that DDB stimulates Jak/Stat signaling, and induces the expression of interferon alpha $(IFN-\alpha)$ stimulated genes, most notably 6-16 and ISG12. In addition, the antiviral effectors induced by $IFN-\alpha$, PKR, OAS, and MxA, were regulated in the presence of DDB at its optimal concentration $(250{\mu}g/mL)$, to a degree commensurate with the degree of induction associated with the $IFN-\alpha$ treated group. Finally, we determined that the replication of pregenomic RNA and HBeAg was inhibited by DDB treatment, and this inhibition was maximized when coupled with the administration of amantadine $(25{\mu}g/mL)$. In conclusion, the results of this study demonstrated clearly that DDB, as well as the combination of DDB/amantadine, directly inhibited $IFN-\alpha$ signaling-mediated replication of HBV in infected hepatocytes, and thus may represent a novel treatment for chronic hepatitis B, which would be characterized principally by its improved safety over other treatment strategies.
Yukwa was prepared with different amounts of curry powder (0, 5, 10, 15, and 20%) based on the amount of waxy rice flour used, and its physical, sensory, and antioxidant properties were measured. In terms of the textural properties, the hardness began to increase when ${\geq}15%$ curry powder content was used; however, the crispiness was unchanged by the amount of curry powder used. The expansion degree of yukwa significantly decreased when 20% curry powder was used. The internal microstructure was measured by scanning electron microscopy (SEM), which showed a more compact and homogeneous structure as the curry powder content increased. The use of increased curry powder in yukwa decreased the acid and hydroperoxide values, and this could have resulted from the antioxidant action of curcumin in the curry powder. Through sensory evaluation, yukwa containing 15% curry powder was chosen as the most acceptable by the elderly.

DBM-2198, a six-membered azasugar nucleotide (6-AZN)-containing phosphorothioate (P = S) oligonucleotide (AZPSON), was described in our previous publication [Lee et al. (2005)] with regard to its antiviral activity against a broad spectrum of HIV-1 variants. This report describes the mechanisms underlying the anti-HIV-1 properties of DBM-2198. The LTR-mediated reporter assay indicated that the anti-HIV-1 activity of DBM-2198 is attributed to an extracellular mode of action rather than intracellular sequence-specific antisense activity. Nevertheless, the antiviral properties of DBM-2198 and other AZPSONs were highly restricted to HIV-1. Unlike other P = S oligonucleotides, DBM-2198 caused no host cell activation upon administration to cultures. HIV-1 that was pre-incubated with DBM-2198 did not show any infectivity towards host cells whereas host cells pre-incubated with DBM-2198 remained susceptible to HIV-1 infection, suggesting that DBM-2198 acts on the virus particle rather than cell surface molecules in the inhibition of HIV-1 infection. Competition assays for binding to HIV-1 envelope protein with anti-gp120 and anti-V3 antibodies revealed that DBM-2198 acts on the viral attachment site of HIV-1 gp120, but not on the V3 region. This report provides a better understanding of the antiviral mechanism of DBM-2198 and may contribute to the development of a potential therapeutic drug against a broad spectrum of HIV-1 variants.
In Korea, the Air Quality Conservation Act and the Petroleum and Petroleum Substitute Fuel Business Act stipulate certain quality standards for fuels distributed in Korea, thereby striving to reduce vehicle performance and emissions. Domestic petroleum products import and produce all the crude oil from each oil refiner so that the quality of the petroleum product is different according to the characteristics of the crude oil. As a result, vehicles have been improved by using the physical properties calculated through the physical property measurement that has tried to improve the accuracy of the measurement of the energy consumption efficiency of the automobile by using standard fuel from abroad. In this study, the same test procedure and method as the test method of domestic gasoline vehicle emission are applied using four samples of gasoline and the latest gasoline vehicle which are actually distributed, and the performance evaluation is performed. The purpose of this study is to contribute to improvement of vehicle technology and fuel quality by collecting necessary basic data and obtaining data on the effect of differences in gasoline property on vehicle emissions. The results of the test showed that the emission of gases (NMOG, CO) from gasoline vehicles was the most influenced by the sulfur content, unlike the previous studies that the vehicles emission had the greatest influence on the distillation characteristics and the specific gravity of aromatic compounds. The catalytic reaction such as the poisoning action of the three-way catalyst which is the abatement device was interfered and the emission was increased. The distillation characteristics and specific gravity of aromatic compounds were found to affect the emission of vehicles. According to the physical properties of the fuel, the emission difference was 28.0% in the urban mode and 17.6 % in the highway mode.
Wind and temperature have been shown to be the critical sources causing changes in the modal properties of large-scale bridges. While the individual effects of wind and temperature on modal variability have been widely studied, the investigation about the effects of multiple environmental factors on structural modal properties was scarcely reported. This paper addresses the modeling of the simultaneous effects of wind and temperature on the modal frequencies of an instrumented cable-stayed bridge. Making use of the long-term monitoring data from anemometers, temperature sensors and accelerometers, a neural network model is formulated to correlate the modal frequency of each vibration mode with wind speed and temperature simultaneously. Research efforts have been made on enhancing the prediction capability of the neural network model through optimal selection of the number of hidden nodes and an analysis of relative strength of effect (RSE) for input reconstruction. The generalization performance of the formulated model is verified with a set of new testing data that have not been used in formulating the model. It is shown that using the significant components of wind speeds and temperatures rather than the whole measurement components as input to neural network can enhance the prediction capability. For the fundamental mode of the bridge investigated, wind and temperature together apply an overall negative action on the modal frequency, and the change in wind condition contributes less to the modal variability than the change in temperature.
The antiparkinsonian agent l-deprenyl, a selective monoamine oxidase (MAO)-B inhibitor, is metabolized in part to l-methamphetamine and l-amphetamine. l< /I>-Deprenyl was evaluated for amphetamine and methamphetamine-like discriminative stimulus effects in rats and its mechanism of action was investigated. Rats were trained under a 5-response, fixed ratio schedule of stimulus-shock termination or a 10-response. Fixed-ratio schedule of food-presentation which discriminate between d-amphetamine (1mg/kg, i.p.) and saline or d-methamphetamine (1mg/kg, i.p.) and saline in a two-lever, operant conditioning procedure. Full generalization was obtained to d-amphetamine (1~3mg/kg). d-methamphetamine (1~3mg/kg) and l-deprenyl (17~30mg/kg) under both the food presentation and stimulus shock termination schedule. l-Deprenyl has dose-dependent amphetamine-and methamphetamine-like discriminative stimulus properties in rats only at doses of 17 and 30mg/kg. Reversible MAO-B inhibitor, RO 16-6491 didn`t show any amphetamine-like discriminative properties. Aromatic amino acid decarboxylase inhibitor, NSD 1015 decreased % responding of l-deprenyl in the methamphetamine-trained rats under the stimulus-shock termination schedule. SKF-525A produced partial inhibition of methamphetamine-like discriminative effects of l-deprenyl under the food presentation schedule. These results suggest that l-deprenyl has no abuse liability at the therapeutic range but there needs some caution at high doses and furthermore, drug discrimination studies under the food presentation and shock termination schedule are useful for the assessment of abuse liability of psychostimulants.
Background: Essential oils are of great interest for their analgesic and anti-inflammatory properties. We aimed to study the content of the essential oil of the Origanum vulgare of the Armenian highlands (OVA) in different periods of vegetation and to investigate its antinociceptive and anti-inflammatory effects in mice (in vivo) and cytotoxic action in cultured cells (in vitro). OVA essential oil was extracted from fresh plant material by hydro-distillation. Methods: For OVA essential oil contents determination the gas chromatography-mass spectrometry method was used. Formalin and hot plate tests and analysis of cell viability using the methyl-thiazolyl-tetrazolium (MTT) assay were used. Results: The maximal content of β-caryophyllene and β-caryophyllene oxide in OVA essential oil was revealed in the period of blossoming (8.18% and 13.36%, correspondently). In the formalin test, 4% OVA essential oil solution (3.5 mg/mouse) exerts significant antinociceptive and anti-inflammatory effects (P = 0.003). MTT assay shows approximately 60% cytotoxicity in HeLa and Vero cells for 2.0 µL/mL OVA essential oil in media. Conclusions: The wild oregano herb of Armenian highlands, harvested in the blossoming period, may be considered as a valuable source for developing pain-relieving preparations.
Silver nanoparticles (AgNPs) have potential applications in medicine, photocatalysis, agriculture, and cosmetic fields due to their unique physicochemical properties and strong antimicrobial activity. Here, AgNPs were synthesized using actinobacterial SL19 strain, isolated from acidic forest soil in Poland, and confirmed by UV-vis and FTIR spectroscopy, TEM, and zeta potential analysis. The AgNPs were polydispersed, stable, spherical, and small, with an average size of 23 nm. The FTIR study revealed the presence of bonds characteristic of proteins that cover nanoparticles. These proteins were then studied by using liquid chromatography with tandem mass spectrometry (LC-MS/MS) and identified with the highest similarity to hypothetical protein and porin with molecular masses equal to 41 and 38 kDa, respectively. Our AgNPs exhibited remarkable antibacterial activity against Escherichia coli and Pseudomonas aeruginosa. The combined, synergistic action of these synthesized AgNPs with commercial antibiotics (ampicillin, kanamycin, streptomycin, and tetracycline) enabled dose reductions in both components and increased their antimicrobial efficacy, especially in the case of streptomycin and tetracycline. Furthermore, the in vitro activity of the AgNPs on human cancer cell lines (MCF-7, A375, A549, and HepG2) showed cancer-specific sensitivity, while the genotoxic activity was evaluated by Ames assay, which revealed a lack of mutagenicity on the part of nanoparticles in Salmonella Typhimurium TA98 strain. We also studied the impact of the AgNPs on the catalytic and photocatalytic degradation of methyl orange (MO). The decomposition of MO was observed by a decrease in intensity of absorbance within time. The results of our study proved the easy, fast, and efficient synthesis of AgNPs using acidophilic actinomycete SL19 strain and demonstrated the remarkable potential of these AgNPs as anticancer and antibacterial agents. However, the properties and activity of such particles can vary by biosynthesized batch.
Nanofluids are dispersions of particles smaller than 100 nm (nanoparticles) in base fluids. They exhibit high thermal conductivity and are mainly applied in cooling applications. Nanolubricants use nanoparticles in base oils as lubricant additives, and have recently started gathering increased attention owing to their potential to improve the tribological and thermal performances of various machinery. Nanolubricants reduce friction and wear, mainly by the action of nanoparticles; however, only a few studies have considered the rheological properties of lubricants. In this study, we adopt a parallel slider bearing model that does not generate geometrical wedge effects, and conduct thermohydrodynamic (THD) analyses to evaluate the effect of higher thermal conductivity and viscosity, which are the main rheological properties of nanolubricants, on the lubrication performances. We use a commercial computational fluid dynamics code, FLUENT, to numerically analyze the continuity, Navier-Stokes, energy equations with temperature-viscosity-density relations, and thermal conductivity and viscosity models of the nanolubricant. The results show the temperature and pressure distributions, load-carrying capacity (LCC), and friction force for three film-temperature boundary conditions (FTBCs). The effects of the higher thermal conductivity and viscosity of the nanolubricant on the LCC and friction force differ significantly, according to the FTBC. The thermal conductivity increases with temperature, improving the cooling performance, reducing LCC, and slightly increasing the friction. The increase in viscosity increases both the LCC and friction. The analysis method in this study can be applied to develop nanolubricants that can improve the tribological and cooling performances of various equipment; however, additional research is required on this topic.

Purpose: Research comparing and evaluating the properties of various dental materials is an important topic in the field of dentistry. This study aims to evaluate the fluoride release and microhardness properties of various fluoride-containing restorative materials used in dental treatments. Materials and Methods: Thirty specimens of four restorative materials were prepared (5 mm × 2 mm, cylindrical): alkasite-based material (Cention N, CN), Self-cure glass Ionomer (Riva self cure, RS), Resin-modified glass ionomer cement (Fuji LC II, FL) and composite resin (Filtek Z350XT, FZ). Fluoride release measurements were conducted on 25 specimens at intervals of 1, 2, 6, 13, 20, and 27 days with a fluoride electrode connected to the pH/ISE Meter (Orion Star A214, Thermo Scientific, USA) and cumulative fluoride release was calculated. Vickers microhardness measurements were performed on five specimens from each material with microhardness tester (DM2D, AFFRI, Italy). Results: As a result of measuring the amount of fluoride release over 27 days, the amount of fluoride release in CN showed a gradual increase, while the amount of fluoride release in RS, FL, and FZ gradually decreased. The cumulative fluoride release amount for 28 days was significantly higher in CN and FL than in RS (P < 0.05). FZ and RS demonstrated significantly higher microhardness compared to CN and FL (P < 0.05). FZ and RS showed similar microhardness, and FL showed the lowest microhardness. Conclusion: Cention N (CN) exhibited superior fluoride release compared to Glass Ionomer Cement (RS), making it a promising option for preventing secondary caries. However, it displayed a lower microhardness than the composite resin (FZ), indicating potential limitations in terms of mechanical strength. Therefore, if an anti-caries action is required, Cention N may be considered first; however, it appears to be difficult to use in posterior permanent teeth.
