Effects of L-Carnitine and Nicotinic Acid on Sperm Characteristics in Miniature Pigs

  • This study investigated the effects of L-carnitine (LC) and nicotinic acid (NA) on sperm viability during liquid storage at $18^{\circ}C$ in miniature pigs. $10{\mu}M$ LC and 30 mM NA, combined LC and NA (LN) were treated in fresh semen for 3, 7, and 10 days. In results, sperm survival increased in NA- and LN-treated semen on 7 and 10 days (p<0.05), mitochondrial integrity of live sperm increased in LN-treated semen on 7 days (p<0.05), but not NA-treated semen. In addition, we examined the acrosome reaction of sperm in miniature pigs. LC and NA did not influence on acrosome reaction of boar sperm. In conclusion, LC and NA effectively maintained the viability and quality of sperm during long-term storage in miniature pigs, suggesting that the combined LN may be useful for improving the semen extender for long-term liquid storage in pigs.

The Role of Mercury in the Etiology of Sperm Dysfunction in Holstein Bulls

  • A large number of toxicological substances and pharmacological and physical agents can cause reproductive intervention at the cellular and molecular level. The present study was designed to assess the effect of mercury ($HgCl_2$) at 50 to $550{\mu}M$ concentration ranges, in vitro, on the sperm membrane and DNA integrity, viability, and acrosomal status of normal bull spermatozoa. The samples were processed for sperm analyses using semen-diluting fluid (PBS, pH 7.2). We recorded a sharp increase in the lipid peroxidation/LPO rate; the highest was at $550{\mu}M$ mercury concentration, indicating a deleterious effect of mercury on the sperm membrane intactness. There was also a strong negative correlation between LPO rate and % viable spermatozoa (R = 0.987, p<0.001). Data obtained from a comet assay technique revealed that mercury is capable of inducing DNA breaks in the sperm nuclei. Interestingly, 92% of DNA breaks were double-stranded. The correlation between LPO rate and % DNA breaks was 0.984. Performing the gelatin test indicates that mercury is able to alter the integrity of acrosomal membranes showing an abnormal acrosome reaction. In this regard, a strong link was found between LPO rate and % halos (R = 0.990, p<0.001). Collectively, mercury proved to be a potent oxidant in the category of environmental factors affecting bull spermatozoa. Hence, considering the wide spread use of mercury and its compounds, these metals should be regarded with more concern.

개 동결 정액의 생존성과 AI 후 임신율에 관한 연구 (Studies on Viability of Frozen Sperm and Pregnancy Rates after AI with Frozen-Thawed Canine Semen)

  • 본 연구는 개 채취 정액의 동결후의 생존성과 신선 및 동결 정자의 capacitation, acrosome reaction과 생존성을 조사하고, 아울러 신선 및 동결 정액을 자연 발정 또는 발정 유기 암캐 에게 인공수정 후 임신율을 조사하였다. 개 채취 정액의 동결 융해 후의 생존성은 $64.5{\pm}2.30%$로서 신선 정액의 생존성에 비해 유의하게 낮게 나타났다(p<0.05). 신선 및 동결 정액의 capacitation, acrosome reaction 및 생존성은 각각 $52.5{\pm}4.5%,\;9.5{\pm}0.6%,\;68.8{\pm}4.5%$$16.2{\pm}3.2%,\;3.2{\pm}0.5%,\;24.5{\pm}2.5%$로 나타났다. 신선 및 동결 정액을 자연 발정 또는 발정을 유기한 암캐에 인공수정했을 때 임신율은 각각 50.0% 및 33.3%로서 동결 정액을 이용했을 때 임신율이 신선 정액에 비해 낮은 임신율을 나타냈다.

Staphylococcus saprophyticus and Escherichia coli: Tracking from sperm fertility potential to assisted reproductive outcomes

  • Objective: Bacteriospermia and urogenital infections are common problems in male infertility. This study aimed to evaluate the effects of bacteriospermia on sperm parameters and clinical outcomes in semen samples infected with two common bacteria (Staphylococcus saprophyticus and Escherichia coli) in northern Iran. Methods: Microbiological tests were performed to isolate and identify organisms from 435 semen samples from infertile couples. Semen samples were assessed according to the World Health Organization criteria. The protamine status, chromatin structure, chromatin condensation, and acrosome reaction of sperm and assisted reproductive outcomes were determined in couples with different male infertility factors. Results: Among the total cases, the two most prevalent pathogens were considered: S. saprophyticus (38.2%) and E. coli (52.9%). In the semen samples infected with E. coli, the spontaneous acrosome reaction and abnormal chromatin condensation were more common (p<0.05). Significant increases in abnormal chromatin condensation and deprotamination were seen in the presence of S. saprophyticus. In washed semen, tight adhesion between the sperm midpiece and S. saprophyticus was observed. There was also a significant decrease in the fertilization rate using semen samples infected with S. saprophyticus and E. coli during in vitro fertilization cycles (p<0.001). In addition, the presence of S. saprophyticus and E. coli in semen samples was associated with a lower likelihood of clinical pregnancy in couples with various factors of male infertility. Conclusion: Poor results of assisted reproductive techniques may be correlated with semen samples infected with two common bacteria in northern Iran.

  • Testes from the allied rock wallaby(Petrogale assimilus) have been examined by the electron microscopy in thin sections in order to examine spermiogenesis and structure of spermatozoa. The spermiogenesis can be divided into nine stages: early Golgi, late Golgi, collapsing, nuclear protrusion, condensation and flattening, nuclear shaping, rotation, nuclear ring contraction, and maturation. The acrosome has been abruptly formed following the collapse of expanded acrosomal vesicle without the cap stage described in the eutheria. The flatly condensed nucleus rotates obliquely to the axis of the axial filament complex and the folded acrosome covers the anterior third of the dorsal nuclear surface forming a wide subacrosormal space as the nuclear ring has contracted. The Sertoli cell reaction and spur are prominent during the nuclear protrusion and rotation stages. A mature spermatozoon has S-shape head which has an extended part reaching to the vicinity of the middle piece.

포유류 수정과정에서 정자 침체기질의 기능 (Role of Acrosomal Matrix in Mammalian Fertilization)

  • Sperm competent for fertilization can become capacitated, bind to the zona pellucida (ZP) of an egg in a specific manner, and complete acrosomal exocytosis. Failure to carry out these functions results in infertility. Although the interactions between the ZP and the plasma mem-brane overlying the sperm acrosome have been considered important for sperm-egg recognition and signalling, recent results have prompted a reassessment of current paradigms concerning these interactions. In this review, we're going to discuss about the roles of the acrosomal matrix, the particulate component of the acrosomal contents, in fertilization. The general hypothesis is that acrosomal exocytosis leads to the exposure of acrosomal matrix proteins that become de-facto extracellular matrix (ECM) on the surface of the sperm head, and that the dynamic interactions of this newly-exposed sperm ECM with the egg ECM (the ZP) govern sperm-egg recognition and sperm penetration of the ZP Informations from these experiments may provide new ways to address the poor ZP binding of sperm from some human infertility patients and may offer new avenues for contraception through the disruption of purposeful sperm-ZP binding.
