찹쌀가루와 메주가루의 배합비율(무게비)을 달리한 고추장(찹쌀가루 : 메주가루 = 1:1, 1:2, 1:3, 2:1, 3:1)의 숙성 중 품질 변화를 평가하기 위해 12주 동안 숙성시키면서 고추장의 이화학적 특성, 효소 활성 및 레올로지 특성을 연구하였다. 숙성 중 고추장의 수분함량은 $47.4{\sim}47.9%$에서 점차 증가하여 8주에는 최대 수분함량을 보였다. 환원당 함량은 $2{\sim}4$주에 가장 높았으며 이후에서는 점차 감소하는 경향을 보였다. 아미노태 질소 함량은 지속적으로 증가하였다. ${\alpha}-amylase$와 ${\beta}-amylase$ 활성은 숙성 $2{\sim}4$주에 최대 역가를 나타냈다. Acidic protease와 neutral protease는 각각 숙성 2주와 4주에 가장 높은 활성을 보였다. Power law 모델에 의한 유동성 측정에서 고추장은 낮은 유동성지수(flow behavior index, $n=0.21{\sim}0.36$)를 가진 전단담화(shearing-thinning)거동을 보여주었으며, 점조도 지수(consistency index, K)와 겉보기점도 $({\eta}_{a,10})$는 숙성 2주에 급격히 증가하고 이후에는 감소하였다. Casson 모델식에 의한 항복응력$({\sigma}_{oc})$은 숙성 2주에 급격히 증가하고 이후에는 감소하였다. 동적 점탄성 측정에서는 저장 탄성률 (G#)이 손실탄성률 (G') 보다 높은 수치를 나타내었고 주파수(frequency, ${\omega}$)가 증가함에 따라 증가하는 경향을 보여주었다. 또한 숙성기간이 경과함에 따라 (G#)과 (G@)이 증가하는 경향을 나타냈다.
This study was carried out to obtain basic information necessary for aggregation and in-vitro culture of mouse embryos by treating phytohemagglutinin-p (PHA-P). The 4-, 8-cell and morula embryos were obtained from female mice of albino BALE/C, CBA and C57BL strains, those were injected 5 i.u pregenant mare serum gonadotrophin and 5 i.u human chorionic gonadotrophin to superovulation. The zona pellucidia was removed by placing the embryos in Acidic Tyrode solution containing 1.0% protease or/and 5 ug/ml PHA-P. The pairs of zona free embryos were subjected to aggregation by glassneedle in BMOC-3 containing 5 ug/ml PHA-P. The aggregation embryos were cultured in Brinster's mouse ova culture-3(BMOC-3) medium under the gas phase of 5% $CO_2$ in air $37^{\circ}C$ for 13 to 50 hours. The results obtained in this study are summarised as follows : 1. When 4-, 8-cell and morula embryos were zona-freed in acidic Tyrode solution containing 1.0% protease or/and 5 ug/ml PHA-P, and cultured in vitro to blastocysts, the 4- and 8-cell embryos showed slightly less development rates than the morula one did, and solution of 5 ug/ml PHA-P brought some higher development rate than negative control. 2. As 2, 5 or 10 ug/ml PHA-P was added to the solution to aggregate 4-, 8-cell or morula embryos, 2 ug/ml solution represented slightly lower aggregation rate than the higher levels solutions, and 4- and 8-cell embryos showed higher rates than morula one did (P<.05). 3. In respect to the development rates of aggregated embryos to morula no significant difference was found among PHA-P levels and between 4-and 8-cell embryos. With respect to those of aggregated embryos to blastocysts the different levels of PHA-P showed similar results, however, the 4- and 8-cell embryos represented higher rates than the morula one did (P<.05). 4. The mean time necessary for development of aggregated 4-, 8-cell and morula embryos to blastocysts were 38.5-40, 26-27 and 19-20hrs. Respectively in solution for aggregation. 5. The aggregation rates of embryos were 34-94%, when treated protease or/and PHA-P. Supplementation of 5 ug/ml PHA-P to the solution for aggregation showed a trend demonstrating higher aggregation rate compared to negative control, although no significance was found. However, 4- and 8-cell embryos represented significantly higher aggregation rates than the morula one did (P<.05). 6. The development rates of 4- and 8-cell embryos to morula were 52.7-84.7 and 73.8-87.2%, respectively, showing no significant difference between two cell stages. However, the aggregation rates of embryos treated with solution containing PHA-P were higher than negative control (P<.05). 7. The development rates of 4- and 8-cell and morula embryos to blastocysts were 41.7-77.7 78.7-83.0 and 0-19.2%, respectively. The rates of 4-cell embryos treated with PHA-P were significant higher than the negative control (P<.05). The 8-cell and morula embryos also showed more rates when treated PHA-P.

The changes of microflora and enzyme activities in Alaska pollack sikhae were evaluated in 3 different temperature conditions, 5$^{\circ}C$, 2$0^{\circ}C$ and alternating temperature (stored at 5$^{\circ}C$ after 10 days of fermentation at 2$0^{\circ}C$), respectively. The number of proteolytic bacteria and 2 lactic acid bacteria including Lactobacillus sp. and Pediococcus sp. increased rapidly up to 10 days and composed major portion of total viable cell (TVC) in sikhae fermented at 2$0^{\circ}C$, whereas those of TVC were occupied by Lactobacillus sp., Pediococcus sp. and yeast after 10 days of fermentation. The major species of microflora in sikhae fermented at alternating temperature were, composed of Lactobacillus, Pediococcus and Streptococcus after 10 days of fermentation. Especially, Leuconostoc sp. was kept up to 27 days at 5$^{\circ}C$ than other temperature conditions (16 days). The activities of protease and lipase in acidic region (pH 3.0) were higher at 2$0^{\circ}C$ than at 5$^{\circ}C$ due to sensitivity of temperature, although those of protease and lipase in neutral region (pH 7.0) were not found any differences in both temperatures. Changing temperature condition from 2$0^{\circ}C$ to 5$^{\circ}C$ in alternating temperature inactivated protease activity, whereas lipase activity was still maintained during fermentation.
Changes in microflora and enzyme activities of three kinds of kochujang were investigated during 6 months of fermentation. Three different kinds of kochujang were prepared using Aspergillus oryzae, Aspergillus oryzae plus Bacillus licheniformis and Aspergillus oryzae, Bacillus licheniformis plus Saccharomyces rouxii. The pH of kochujang showed a slight decrease during fermentation. The number of mold and bacteria increased up to 30 days of fermentation and then decreased rapidly thereafter and the numbers of yeast increased after 30 days of fermentation. The viable cell counts of anaerobic bacteria increased remarkably up to 30 days of fermentation and then decreased to 60 days of fermentation. The activities of ${\alpha}-$, ${\beta}-\;amylase$ and protease were the highest in kochujang prepared with Aspergillus oryzae plus Bacillus licheniformis.

The antimicrobial substance produced by Lactobacillus bulgaricus was inactivated by protease. It showed inhibitory activity against Staphylococcus aureus ATCC6538, Streptococcus agalactiae ATCC14364, some Gram-positive and Gram-negative bacteria, and characteristics of a bacteriocin. The optimal temperature and culture time for the production of bacteriocin were $30^{\circ}C$ and 10 h, respectively, in the culture of L. bulgaricus. The bacteriocin production started in the exponential phase and reached a maximum at the early stationary phase. Using Staph. aureus ATCC6538 and Strep. agalactiae ATCC14364, known as common bovine mastitis pathogens, as indicator strains for determination of the bacteriocin activity, the antimicrobial activity of the bacteriocin was found to be stable in acidic and neutral pH's (2- 7) even at lOOT, whereas it was lost at high pH (10- 11) and $100^{\circ}C$. The mode of action for the antimicrobial activity was bacteriocidal, and the molecular weight determined by SDS-PAGE and overlay method was 14 kDa.
Enzyme activities, production of killer toxin and some functionality of forty seven yeasts isolated from traditional Meju were investigated in culture broth and cell free extracts. Activities of $\alpha$-galactosidase, invertase and inulinase were detected in cell free extracts of 38 strains, 43 strains and 45 strains, respectively and acidic and neutral protease activities also were detected in culture broth of all the strains, $\beta$-Galactosidase activity was detected in cell free extracts of OE-20 and S-14 strains. Killer toxins were produced by OE-12, S-8 (Candida spp.), OE-19 (Zygosaccharomyces spp.) and S-3 (Saccharomyces spp.). Culture broth of OE-23 and S-9 showed 61.3% and 59.2% of antioxidant activity to $\alpha$, $\alpha$-diphenyl-$\beta$-picrylhydrazyl(DPPH), but nitrite-scavenging ability as well as inhibition of tyrosinase and polyphenol oxidase were not appeared in all the strains.

Meju for doenjang and kochujang was prepared as a model at Sunchang areaand monitored with major changes. Kochujang meju was prepared on September 12 and doenjang meju on November 12, 1995. Kochjang meju was found to be naturally fermented at 80~90% RH, 15~2$0^{\circ}C$ and doenjang meju was at 80~90% RH and 0~5$^{\circ}C$. The shapes of kochujang meju and doenjang meju were doughnut-type and rectangular, respectively. Weight losses during fermentation were 48% and 28%, respectively. The pH drop and acip production of kochujang meju were negligible. However, pH of doenjang meju decreased from 6.29 to 5.88 and acidity increased from 0.08 to 0.23% as lactic acid. Protein in meju was found to be rapidly solubilized during the early stage of fermentation. Soluble protein cotents of kochujang meju after 7 days and 60 days were 8.23%, respectively. The doenjang mejus were 2.15% after 20 days and 5.72% after 60 days. Soluble suger content increased with the fermentation time. The soluble sugar content was higher in kochjang meju. Acidic protease was highly produced during meju fermentation. $\alpha$-Amylase and $\beta$-amylase were detected in the kochujang meju, of which glutinous rice consisted, but negligible in doenjang meju. Lipase was detected in kochujang meju, but was, negligible in doenjang meju. Microbial population increased drastically after 7 days of fermentation in kochujang meju and 20 days of fermentation in doenjang meju.
A microbial cryoprotectant formulation using a W/O/W multiple emulsion system was developed. The psychrotolerant microorganism, B4, isolated from soil in South Korea, was observed by the drop freezing method, in which the microorganism sample inhibited ice nucleation activity. The antifreeze activity was eliminated when the microorganism sample was treated with protease, indicating that the antifreeze activity was due to the presence of antifreeze protein. The result of the l6S rDNA sequencing indicated the B4 strain was most closely related to a species of the genus Bacillus. Culture broth of B4 strain (Bacillus sp.) and rapeseed oil containing 1 % polyglycerine polyricinolate (PGPR) were used as core and wall material, respectively. The most stable W/O emulsion was prepared at a core/oil ratio of 1:2. The highest W/O/W emulsion stability was achieved when the primary emulsion to external aqueous phase containing 0.5% caster oil polyoxyethylene ether $(COG25^{TM})$ ratio was 1:1. Microcrystalline cellulose showed better W/O/W emulsion stability than other polymer types. The viability of cells in a W/O/W emulsion was higher than free cells during storage at $37^\circ{C}$. An acidic pH and UV exposure decreased the viability of free cells, but cells in W/O/W emulsion were more stable under these conditions.
Changes in microorganisms, enzyme activities and major components of two types of Doenjang prepared with spring Meju and autumn Meju and Kochujang were investigated during 4 months of fermentation. The viable cell counts of aerobic bacteria in Doenjang and Kochujang were increased up to 60 days of fermentation, but viable cell counts of anaerobic bacteria did not show remarkable changes during fermentation. Viable cell count of yeast showed a rapid increase up to 15 days of fermentation in Doenjang and 60 days in Kochujang. It was found that $\alpha$-amylase activity of autumn Meju Doenjang and glucoamylase activity of Kochujang were higher than the other. Acidic and neutral protease showed the highest activity during 15-30 days of fermentation. The pH of Doenjang was increased up to pH 7.0 until 60 days of fermentation, but pH of Kochujang gradually decreased during fermentation. Moisture content of spring Meju Doenjang and Kochujang decreased to 40% during ferme- ntation and reducing sugar content of Kochujang increased up to 15 days of fermentation, but decreased after that.
Mucor-rennin, the crystalline milk-clotting enzyme, isolated from Mucor pusillus var. Lindt, has an acid protease activity. The optimum pH for the digestion of k-casein is 4.5, while that for hemoglobin digestion is 4.0. The skim milk solution was easily clotted acidic solution than alkalin solution, and the milk clotting activated by Ca ion. The enzyme was heat stable against heat from pH 4.0 to 6.0 but was more stable at pH 5.0. The activity of the enzyme at pH 5.0 did not decrease at 30 C for 15 days and the activity was not effected by sodium propionate and salicilic acid. Therefore, the enzyme of liguid type could store for a long time and could be transported from Erzyme production Co. to Manufacture of cheese Co. by adding the antiesptic and by adjusting pH to 5.0.
