금속 프탈로시아닌을 이용한 아세트알데히드의 촉매연소 (Catalytic Combustion of Acetaldehyde by Metal Phthalocyanines)

  • Catalytic combustion of acetaldehyde has been investigated as a representative of unpleasant odor by its reaction with metal-phthalocyanines(PC). The experiment was conducted at the reaction temperature of 200~41$0^{\circ}C$ and the concentratio of acetaldehyde in air at the range of 0.07~0.94 mole% The pretreated metal-PC has been characterized by UV-VIS and XRD analysis. According to this study catalytic activity of metal -PC was improved by air pretreatment at 45$0^{\circ}C$ for 1hr. Under this pretreatment condition Co-PC and Cu($\alpha$)-PC were destroyed and new metal oxides were formed such as Co3O4 and CuO respectively. However Zn-PC retained its basic structure even afte air pretreatment. The order of catalytic activity on acetaldehyde combustion was summarized as follows : Zn-PC$\alpha$)-PC. It was found that the complete combustin of acetaldehyde with Cu($\alpha$)-PC was accomplished at its concentrations below 0.2mole% (32$0^{\circ}C$) and 0.6 mole%(35$0^{\circ}C$) in air.

  • The changes of the contents of acetaldheyde and alcohol in Chungdo Bansi was determined during the removal of astringency by treatment of $CO_2$. On the other hand, the difference of acetaldehyde and alcohol contents were determined to comparedits contents in the part of Chungdo Bansi and Buju, respectively. It showed that the variation of acetaldehyde and alchohol contents was higher 10 times non-astringent persimmon than astringent one, and acetaldehyde and alcohol contents in the core of non-astringent persimmon and sweep one were the higher than the other parts. It was assumed that acetaldehyde and ethanol contents had intimately relation to removal of astringency.

  • Alcoholism has been associated with folate deficiency in humans and laboratory animals. Previous study showed that ethanol feeding reduces the dehydrogenase and hydrolase activity of 10-formyltetrahydrofolate dehydrogenase (FDH) in rat liver. Hepatic ethanol metabolism generates acetaldehyde and acetate. The mechanisms by which ethanol and its metabolites produce toxicity within the liver cells are unknown. We purified FDH from rat liver and investigated the effect of ethanol, acetaldehyde and acetate on the enzyme in vitro. Hepatic FDH activity was not reduced by ethanol or acetate directly. However, acetaldehyde was observed to reduce the dehydrogenase activity of FDH in a dose- and time-dependent manner with an apparent $IC_{50}$ of 4 mM, while the hydrolase activity of FDH was not affected by acetaldehyde in vitro. These results suggest that the inhibition of hepatic FDH dehydrogenase activity induced by acetadehyde may play a role in ethanol toxicity.

Impaired Osteogenesis in Human Induced Pluripotent Stem Cells with Acetaldehyde Dehydrogenase 2 Mutations

  • Acetaldehyde dehydrogenase 2 (ALDH2) is the second enzyme involved in the breakdown of acetaldehyde into acetic acid during the process of alcohol metabolism. Roughly 40% of East Asians carry one or two ALDH2*2 alleles, and the presence of ALDH2 genetic mutations in individuals may affect the bone remodeling cycle owing to accumulation of acetaldehyde in the body. In this study, we investigated the effects of ALDH2 mutations on bone remodeling. In this study, we examined the effects of ALDH2 polymorphisms on in vitro osteogensis using human induced pluripotent stem cells (hiPSCs). We differentiated wild-type (ALDH2*1/*1-) and ALDH2*1/*2-genotyped hiPSCs into osteoblasts (OBs) and confirmed their OB characteristics. Acetaldehyde was administered to confirm the impact caused by the mutation during OB differentiation. Calcium deposits formed during osteogenesis were significantly decreased in ALDH2*1/*2 OBs. The expression of osteogenic markers were also decreased in acetaldehyde-treated OBs differentiated from the ALDH2*1/*2 hiPSCs. Furthermore, the impact of ALDH2 polymorphism and acetaldehyde-induced stress on inflammatory factors such as 4-hydroxynonenal and tumor necrosis factor α was confirmed. Our findings suggest that individuals with ALDH2 deficiency may face challenges in acetaldehyde breakdown, rendering them susceptible to disturbances in normal bone remodeling therefore, caution should be exercised regarding alcohol consumption. In this proof-of-concept study, we were able to suggest these findings as a result of a disease-in-a-dish concept using hiPSCs derived from individuals bearing a certain mutation. This study also shows the potential of patient-derived hiPSCs for disease modeling with a specific condition.

자동차 폐촉매의 물리 화학적 특성 (Physical and Chemical Characteristics of Waste Automotive Catalysts)

  • 본 연구는 자동차 폐촉매의 재활용 가능성을 검토하기 위하여 촉매의 물리화학적 특성 및 acetaldehyde의 연소활성에 관하여 조사하였다. 자동차 폐촉매의 물리화학적 특성을 조사하기 위하여 EA(Elemental analysis), ICP-AES (Inductively coupled plasma-atomic emission spectrophotometer), XRD(X-ray diffraction) 분석을 수행하였다. Carbon 침적량은 촉매의 전반부가 후반부보다 많았으며, 주행거리에 따라 증가하는 경향을 보였다. Pt, Pd, Rh 함유량은 제작사에 따라 구성성분의 차이를 보였다. 모델 VOC로서 acetaldehyde를 선택하였으며, 상압유통식 반응장치를 이용하여 촉매의 연소활성을 조사하였다. 자동차 폐촉매의 후반부가 전반부보다 촉매 활성이 우수하였다. 또한 주행거리의 증가에 따라 촉매활성은 감소하는 경향을 나타내며, 주행거리와 촉매활성은 매우 우수한 상관성을 보였다. 자동차 폐촉매의 acetaldehyde 연소활성은 매우 우수하였으며, 소규모의 배출원 제어에 활용 가능한 것으로 평가된다.

  • Rhodotorula sp. Y -55 효모의 중식특성을 etha nol, acetic acid 또는 acetaldehyde 상에서 batch culture로 조사하였다. 기질농도가 중가함에 따라 중식수율이 감소하고 유도기가 연장되었다. 비성장속도(${\mu}$)는 지수기에서 ethanol과 acetaldehyde의 초기농도에 따라 변화하였으나 acetic acid는 이와 상관이 없었으며 그 값은 $O.107h^{-1}$로 나타났다. 최대 비성장속도는 20g/L의 ethanol에서 $O.270h^{-1}$, O.2g/L의 acetaldehyde에서 $O.041h^{-1}$이었다. 또 최대 중식수융은 109/L의 ethanol에서 32.6%, 20g/L의 a acetic acid에서 25.6%와 O.2g/L의 acetaldehyde 에서 45%였다. 세포내 조단백질과 핵산 함량은 e­t thanol상에서 각각 41.5wt%와 4.9wt %였고 acetic acid상에서는 각각 40.2wt %와 4.7wt%로 나타났다.

청간해주탕이 에탄올 매개성 cytokine 발현에 미치는 영향 (The Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Ethanol-mediated Cytokine Expression)

  • Object : This study was designed to investigate the effects of Chungganhaeju-tang (Qingganjiejiu-tang) on cytotoxicity, growth inhibition, apoptosis and expression of cytokine in damaged HepG2 cells. Method : Toxicity on HepG2 cell induced by ethanol and acetaldehyde was measured for viability, cell growth, DNA replication and generation of apoptosis and cytokine. The recovery of the cell activity by Chungganhaeju-tang was estimated for the measured parameters using PCR with different cycle numbers, DNA gel-electrophoresis, and densitometric analysis, Results : Chungganhaeju-tang improves the recovery of HepG2 cells damaged by ethanol or acetaldehyde. The suppressed DNA synthesis of the cell damaged by ethanol or acetaldehyde is improved by Chungganhaeju-tang. A liver-protection effect was shown by the reduction of apoptosis and $TNF-{\alpha},{\;}IL-1{\beta}$ expressions that are induced by ethanol or acetaldehyde. Conclusion : The result indicates that Chungganhaeju-tang reduces toxicity induced by ethanol or acetaldehyde and recovers damaged liver function.

알데히드 탈수소 효소 활성에 미치는 글루타치온의 영향 (Effect of Glutathione on Aldehyde Dehydrogenase Activity)

  • It is known that alcoholics have significantly lower mitochondrial aldehyde dehydrogenase (ALDH)s'activity than do normal subjects or nonalcoholics with liver disease. However, there are only few reports that explain the reasons behind this reduction of ALDHs'activities. In this study, ALDH activity is inhibited by acetaldehyde, a substrate for ALDH However, the addition of glutathione (GSH) protected ALDH activities against the inhibitory effects of acetaldehyde in vitro. Furthermore, when GSH depletion is induced using diethyl maleate (DEM) in rats by 24% in cytosol and 43% in mitochondria, ALDH activities were also depressed by 31% and 63%, respectively compared to non-treated rats without significant reductions in other hepatic enzymes. These results suggest that ALDHs'activities are closely related to the concentration of acetaldehyde and/or cellular GSH contents . Therefore in alcoholic liver disease, increased productions of acetaldehyde and decreased contents of mitochondrial GSH may involved in the depression of ALDHs'activities.

  • An acetaldehyde, a representative food waste odor, was decomposed using a hybrid system comprised of a non-thermal plasma and catalyst at an ambient temperature under high humidity. A five wt.% Mn was impregnated on two differently structured microporous zeolites, namely Beta and ZSM-5, with a different molar ratio of SiO2/Al2O3. Under high humidity conditions, the acetaldehyde degradation was higher in zeolites with the high ratio of SiO2/Al2O3. Among studied catalysts, a five wt.% Mn/Beta (SiO2/Al2O3 = 300) showed the highest acetaldehyde removal activity owing to its high hydrophobicity and reducibility. During long term stability test using the same catalyst for 110 hours, the acetaldehyde removal activity was relatively well-maintained.

Effects of Plant Vinegar Extract on the Reduction of Blood Concentration of Alcohol and Acetaldehyde in Alcohol Administrated Rats

  • Excessive drinking causes 'alcohol hangover' within 8-16 hours. The cause of 'hangover' has not been elucidated exactly until now, but it is reported that it is caused by the creation of blood ethanol and acetaldehyde as ethanol metabolites. In this study vinegar extract of wood (VE) or OC-1, to which the powder extract of green tea leaves extract is added, was administered to the rats 30 minutes before the oral administration of ethanol (3 g/kg) and the blood ethanol and acetaldehyde concentration was measured in order to evaluate the efficacy of the beverage material for detoxification. As a result, the blood ethanol concentration in the group of the VE-1(vinegar crude extract) and VE-2 (double diluted solution) is statistically lower (P,0.05) than the exclusive alcohol administered control group. The blood acetaldehyde concentration of all groups of VE and OC-2, which is the double dilution of OC-1, is statistically low after 7 hours following ethanol administration. Especially, the AUC value of OC-2 group is statistically low compared to the control group. Accordingly, it indicates the conclusion that VE and OC-1, reducing the blood ethanol and acetaldehyde concentration which are two leading factors of 'hangover' after drinking, and worthwhile to be developed as beverage materials to eliminate 'hangover'.
