Preliminary Evidence for a Metallothionein-like Cd-binding Protein in the Kidney of the Antarctic Clam Laternula elliptica

  • A Cd-binding protein was identified in the renal cytosol of the Antarctic clam Laternula elliptica which naturally contains high concentrations of Cd. The Cd-binding protein showed similar characteristics of metallothionein (MT) in molecular weight (about 10-12 kDa) and low spectral absorbance at 280 nm with relatively high absorbance at 254nm. Results of immuno-histochemical staining suggested that the MT-like Cd-binding protein was mainly located in the epithelial cells of the kidney. The MT-like protein was a major ligand of cytosolic Cd as shown in the elution profiles of chromatography and may play an important role in Cd sequestration and accumulation in L. elliptica kidney. A considerable amount of Cd was also found to be associated with particulate fraction, indicating the sequestration to particulate fraction is as important as binding to the cytosolic MT-like protein in Cd accumulation in the kidney.

Biomass, Primary Nutrient and Carbon Stock in a Sub-Himalayan Forest of West Bengal, India

  • Quantitative information on biomass and available nutrients are essential for developing sustainable forest management strategies to regulate atmospheric carbon. An attempt was made at Chilapatta Reserve Forest in Duars region of West Bengal to quantify its above and below ground carbon along with available "N", "P" and "K" in the soil. Stratified random nested quadrats were marked for soil, biomass and litter sampling. Indirect or non-destructive procedures were employed for biomass estimation. The amount of these available nutrients and organic carbon quantified in soil indicates that the forest soil is high in organic carbon and available "K" and medium in phosphorus and nitrogen. The biomass, soil carbon and total carbon (soil C+C in plant biomass) in the forest was 1,995.98, 75.83 and $973.65Mg\;ha^{-1}$. More than 90% of the carbon accumulated in the forest was contributed by the trees. The annual litter production of the forest was $5.37Mg\;ha^{-1}$. Carbon accumulation is intricately linked with site quality factors. The estimated biomass of $1,995.98Mg{\cdot}ha^{-1}$ clearly indicates this. The site quality factor i.e. tropical moist deciduous with optimum availability of soil nutrients, heavy precipitation, high mean monthly relative humidity and optimum temperature range supported luxuriant growth which was realized as higher biomass accumulation and hence higher carbon accumulated.

A Study on the Removal of Heavy Metals by Microorganism in the Biological Wastewater Treatment (생물학적 폐수처리 공정에서의 미생물에 의한 중금속 제거에 관한 연구)

  • In this research, biological uptake of heavy metals(Cd(II), Cu(II), Zn(II)) was measured under various conditions ; pH, initial heavy metal concentration, temperature, contact time and the amount of biomass through batch test. From this research, it was found that heavy metals might be removed through adsorption and accumulation in activated sludge process. Heavy metals were highly concentrated by microbial floc in activated sludge. Also, the removal efficiency was reached up to 80~90% within and after 1 hour the increase of removal efficiency was minimal. The order of accumulation efficiency was Cu(II)>Zn(II)>Cd(II), and the bonding strength between heavy metals and microbial floc may be expressed in order of Cu(II)>Zn(II)>Cd(II).

Highly-sensitive Detection of Salvianolic Acid B using Alumina Microfibers-modified Electrode

  • Alumina microfibers with porous structures were prepared through hydrothermal reaction, and then used to modify the surface of carbon paste electrode (CPE). After modification with alumina microfibers, the electrochemical activity of CPE was found to be greatly improved. On the surface of alumina microfibers-modified CPE, the oxidation peak current of salvianolic acid B, a main bioactive compound in Danshen with anti-oxidative and anti-inflammatory effects, was remarkably increased compared with that on the bare CPE surface. The influences of pH value, amount of alumina microfibers and accumulation time were studied. Based on the strong signal amplification effects of alumina microfibers, a novel electrochemical method was developed for the detection of salvianolic acid B. The linear range was from 5 ${\mu}gL^{-1}$ to 0.3 mg $L^{-1}$, and the detection limit was 2 ${\mu}gL^{-1}$ (2.78 nM) after 1-min accumulation. The new method was successfully used to detect salvianolic acid B in ShuangDan oral liquid samples, and the recovery was over the range from 97.4% to 102.9%.

Probability of performance failure of storm sewer according to accumulation of debris (토사 적체에 따른 우수관의 성능불능확률)

  • Statistical distribution of annual maximum rainfall intensity of 18 cities in Korea was analyzed and applied to the reliability model which can calculate the probability of performance failure of storm sewer. After the analysis, it was found that distribution of annual maximum rainfall intensity of 18 cities in Korea is well matched with Gumbel distribution. Rational equation was used to estimate the load and Manning's equation was used to estimate the capacity in reliability function to calculate the probability of performance failure of storm sewer. Reliability analysis was performed by developed model applying to the real storm sewer. It was found that probability of performance failure is abruptly increased if the diameter is smaller than certain size. Therefore, cleaning the inside of storm sewer to maintain the original diameter can be one of the best ways to reduce the probability of performance failure. In the present study, probability of performance failure according to accumulation of debris in storm sewer was calculated. It was found that increasing the amount of debris seriously decrease the capacity of storm sewer and significantly increase the probability of performance failure.

Distribution of Cd, Cu and Zn in a Sewage Sludge-treated Calcareous Soil

  • The distributions of Cd, Cu, and Zn concentration in soil treated with one (1988) or two (1988 and 1993) applications of sewage sludge at rates of 0, 25, 50, and $100Mg\;ha^{-1}$ (dry weight basis) were determined to assess the accumulation and mobility of the heavy metals. The heavy metals accumulated almost entirely in 0 to 15 cm soil depths. Small amounts of the metals moved out of the tillage zone (0-15 cm depth) into the subsoil, but even at the high rate of sewage sludge, little movement of heavy metals occurred below 100 cm depth. The water-extractable Cd, Cu, and Zn concentrations were very low regardless of the rate of sewage sludge application. Availability of metals as determined by DTPA extraction showed the percentage of DTPA-extractable/total concentration increased with sewage sludge application. In the 0-15 cm depth of sewage sludge treated soil, the percentage of DTPA-extractable/total concentration was higher than 46% for Cd, but the value was less than 27% and 17% for Cu and Zn, respectively. The Cd, Cu, and Zn added to this calcareous clay soil by sewage sludge application were not very mobile, and the amount of plant available form was very small.

Cadmium Accumulation, Phosphorus Concentration and Growth Response of Cd-treated Ectomycorrhizal Poplar Cuttings

  • We investigated whether cadmium (Cd) toxicity affects phosphorus (P) concentration and growth of poplar, which might be related to the ectomycorrhizal associations. Populus ${\times}$tomentoglandulosa cuttings were treated with 0.1 mM and 0.4 mM $CdSO_4$ and inoculated with ectomycorrhizal fungus, Pisolithus tinctorius (Pt) and grown in autoclaved peat vermiculite mixture for five months under greenhouse conditions. Ectomycorrhizal plants showed significantly higher Cd concentration in leaves, stems and roots than in non-mycorrhizal plants. Likewise, P contents in leaves and roots of ectomycorrhizal plants were higher than those of non-mycorrhizal plants. Acid phosphatase activity in leaves of ectomycorrhizal plants, however, was significantly lower than that of non-mycorrhizal plants. 0.1 mM Cd significantly increased P content in leaves and stems of non-mycorrhizal plants. In spite of high P concentration, which is accompanied by lower acid phosphatase activity, plant growth was not improved by inoculation with P. tinctorius. Total plant dry weight was lower than the non-mycorrhizal counterpart. The results imply that this might be caused by the large amount of energy consumption to alleviate Cd toxicity resulted from high Cd accumulation in their tissues.

Histological Change and Collagen Formation on Laser Wounded Rat using 808 nm Diode Laser and $CO_2$ Laser

  • Lasers are necessity in our life related to the fields of medicine and cosmetic surgery. With 808 nm diode laser and $CO_2$ laser, we made some wounds on a dorsum of rat by laser irradiation. All of irradiations shows thermal effects on the whole region of skin tissues. They make wound damage depending on laser power and irradiation time. Because a collagen is plays an important role in tissue repair, we studied collagen accumulation in wound tissue. For wound healing, collagen accumulation was found in the near region of damage in epidermis and dermis layer of the rat skin. In case of the quantitative analysis of collagen in wound tissue, the amount of collagen in wound tissue by $CO_2$ laser irradiation is higher than that of 808 nm diode laser irradiation. And re-epithelialization was significantly faster in wound by $CO_2$ laser irradiation compared with that of 808 nm diode laser irradiation.

  • In many of the industrialized countries an increasing amount of infrastructure is ageing. This has become specifically critical to bridges which are a major asset with respect to keeping an economy alive. Life of this infrastructure is scattering but often little quantifiable information is known with respect to its damage condition. This article describes how a damage tolerance approach used in aviation today may even be applied to civil infrastructure in the sense that operational life can be applied in the context of modern life cycle management. This can be applied for steel structures as a complete process where much of the damage accumulation behavior is known and may even be adopted to concrete structures in principle, where much of the missing knowledge in damage accumulation has to be substituted by enhanced inspection. This enhanced and continuous inspection can be achieved through robotic systems in a first approach as well as built in sensors in the sense of structural health monitoring (SHM).

  • The effects of the different temperatures on the amylase activity and growth rate of the rye seedling grown in the solutions containing radioactive sulfur- 35 were studied. The amylase activity of the coleoptiles obtained from the seedlings grown in the solutions of S-35, at 14$^{\circ}C$, appeared to be strongly stimulated in comparison to the control, but the culture temperatures of 22$^{\circ}C$ and 3$0^{\circ}C$ showed the decrease in the amylase activity. The amylase activity of the grains treated with the low intensity of the ratioactive material didn't show clear changes, at any culture temperatures, but the amylase activity of the grains treated with the high intensity of S-35, 50$\mu$c, showed definite decline at the elevated culture temperature, 3$0^{\circ}C$. Similar effects was also found in the growth of the seedlings. However, we would consider the effects of the radioactive materials on the acticity of the amylase and the growth of the seedlings are resulted from the accumulation of the much amount of the radioactive materials, and this accumulation rate depends upon actually the elevation of the culture temperature.
