Greenhouse gas emissions are one of the critical factors that drive change in rice cropping systems. Within this changing system, less water irrigation and chemical fertilizer are seriously considered, as well combining precision farming technologies with irrigation control. Water and phosphorus (P) fertilizer are two of the most critical inputs in rice cultivation. Due to the lack of water availability in the system, P fertilizer is not available, especially in acidic soil conditions. Moreover, the various types of abiotic stresses, such as drought, high temperature, salinity, submergence, and limited fertilizer result in significant yield loss in the system. Even in the late stage of growth, the waves caused by diseases and insects make the field more unfruitful. Therefore, agronomists and breeders need to identify the secondary phenotypes to estimate the yield loss of when stress appears. The prediction will be clearer if we have a set of markers tagging the causal variation and the associated precise phenotype indices. Although there have been various studies for abiotic stress tolerance, we still lack functional molecular markers and phenotype indices. This is due to the underlying challenges caused by environmental factors in highly unpredictable regional and yearly environmental conditions in the field system. Pupl (phosphorus uptake 1) is still known as the first QTL associated with phosphorus uptake and have been validated in different field crops. Interestingly, some pyramiding lines of Pupl and other QTLs for other stress tolerances showed preferable phenotypes in the yield. Precise physiological studies with the help of genomics are on-going and some results will be discussed.

The GASA protein (Gibberellic acid-stimulated Arabidopsis) are family of small cysteine-rich peptides found in plants. These GASA gene family mainly involved in biotic/abiotic stress responses and plant development. Despite being present in a wide plant species, their action and functions still remain unclear. In this study, using the in-silico analysis method we identified 41 GASA genes in peanuts (Arachis hypogaea L.). Based on the phylogenetic analysis 41 GASA genes are classified in the four major clusters and subclades. Mainly, clusters IV and III comprise the majority of GASA genes 15 and 11 genes respectively, followed by cluster I and cluster II with 9 and 6 genes respectively. Additionally, based on in-silico analysis we predicted the post-transcriptional and post-translational changes of GASA proteins under abiotic stresses such as drought and salt stress would aid our understanding of the regulatory mechanisms. Hence, a further study is planned to evaluate the expression of these GASA genes under stress in different plant tissues to elucidate the possible functional role of GASA genes in peanut plants. These findings might offer insightful data for peanut advancement.

The use of plant growth-promoting rhizobacteria is economically viable and environmentally safe for mitigating various plant stresses. Abiotic stresses such as flood and drought are a serious threat to modern agriculture. In the present study, the indole-3-acetic acid-producing rhizobacterium R. sphaeroides KE149 was selected, and its effects on the growth of adzuki bean plants under flood stress (FS) and drought stress (DS) were investigated. IAA quantification of bacterial pure culture revealed that KE149 produced a significant amount of IAA. Moreover, KE149 inoculation notably decreased stress-responsive endogenous abscisic acid and jasmonic acid and increased salicylic acid in plants under DS and FS. KE149 inoculation also increased proline under DS and methionine under FS. In addition, KE149 inoculation significantly increased the levels of calcium (Ca), magnesium (Mg), and potassium (K) while lowering the sodium (Na) content in the plant shoot under stress. KE149-treated plants had markedly greater root length, shoot length, stem diameter, biomass, and higher chlorophyll content under both normal and stressed conditions. These results suggest that KE149 could be an efficient biofertilizer for mitigating water stress.
Osmotic stress is one of major limiting factors in crop production. In particular, seasonal drought often causes the secondary disease in the field, resulting in severe reduction in both quality and productivity. Recent efforts have revealed that many genes encoding protein kinases play important roles in osmotic stress signal transduction pathways. Previously, the AtSIK (Arabidopsis thaliana Stress Inducible Kinase) mutants have shown to enhance tolerance to abiotic stresses, accompanying with higher expression of abiotic stress-related genes than did the wild-type plants. In this study, we have transformed potato (cv. Taedong Valley) with the AtSIK expression cassette. Both PCR and RT-PCR using AtSIK-specific primers showed stable integration and expression of the AtSIK gene in individual transgenic lines, respectively. Foliar application of herbicide ($Basta^{(R)}$) at commercial application rate (0.3% (v/v)) revealed another evidence of stable gene introduction of T-DNA which includes the bar gene for herbicide resistance. Overexpression of the AtSIK gene under dual CaMV35S promoter increased sensitivity to salt stress (300 mM NaCl), which was demonstrated by the reduction rate of chlorophyll contents in leaves of transgenic potato lines. These results suggest that possible increase of osmotic tolerance in potato plants may be achieved by antisense expression of AtSIK gene.
To withstand ever-changing environmental stresses, plants are equipped with phytohormone-mediated stress resistance mechanisms. Salt stress triggers abscisic acid (ABA) signaling, which enhances stress tolerance at the expense of growth. ABA is thought to inhibit the action of growth-promoting hormones, including brassinosteroids (BRs). However, the regulatory mechanisms that coordinate ABA and BR activity remain to be discovered. We noticed that ABA-treated seedlings exhibited small, round leaves and short roots, a phenotype that is characteristic of the BR signaling mutant, brassinosteroid insensitive1-9 (bri1-9). To identify genes that are antagonistically regulated by ABA and BRs, we examined published Arabidopsis microarray data sets. Of the list of genes identified, those upregulated by ABA but downregulated by BRs were enriched with a BRRE motif in their promoter sequences. After validating the microarray data using quantitative RT-PCR, we focused on RD26, which is induced by salt stress. Histochemical analysis of transgenic Arabidopsis plants expressing RD26pro:GUS revealed that the induction of GUS expression after NaCl treatment was suppressed by co-treatment with BRs, but enhanced by co-treatment with propiconazole, a BR biosynthetic inhibitor. Similarly, treatment with bikinin, an inhibitor of BIN2 kinase, not only inhibited RD26 expression, but also reduced the survival rate of the plant following exposure to salt stress. Our results suggest that ABA and BRs act antagonistically on their target genes at or after the BIN2 step in BR signaling pathways, and suggest a mechanism by which plants fine-tune their growth, particularly when stress responses and growth compete for resources.

High salinity is a common stress condition that adversely affects plant growth and crop production. In response to various environmental stresses, plants activate a number of defense genes that function to increase the tolerance. To isolate Arabidopsis genes that are involved in abiotic stress responses, we carried out genetic screening using various mutant lines. Among them, the blh8 ($\b{B}$EL1-$\b{L}$ike $\b{H}$omeodomain $\underline{8}$) mutant specifically shows chlorotic phenotypes to ionic (specifically, $Na^+$ and $K^+$) stresses, but no differences in root growth. In addition, BLH8 is related to plant development and abiotic stress as predicted by a Graphical Gaussian Model (GGM) network program. It implies that BLH8 functions as a putative transcription factor related to abiotic stress responses. Collectively, our results show that gene network analysis is a useful tool for isolating genes involved in stress adaptation in plants.
We have investigated the effects of abiotic and biotic stresses on leaf senescence using transgenic tobacco plants, in which cellular contents of polyamines were increased by introducing the genes of polyamine and ethylene biosynthesis in sense or antisense orientation. These transgenic plants showed accumulations of polyamines at higher levels than were found in wild-type. Stress-induced senescence was attenuated in transgenic plants cpmpared with wild-type plants, in terms of total chlorphyll loss and phenotypic changes after oxidative stress of hydrogen peroxide($H_2O_2$), high salinity, acid stress (pH3.0), ABA and fungal pathogen(phytophothora parasitica pv.Nicotianae). Transcripts for antioxidant enzyme, glutathionine-S-transferase and catalase, were also more abundant in transgenic plants than wild-type plants. These result suggested that higher expression of those genes caused a broad-spectrum resistance to abiotic stress/biotic stress. These phenomena indicate that polyamines may play an important role in contributing to the antioxidant defense function in plants. Our findings suggest that facilitate the improvement of stress tolerance of crop plants.

In this study, we compared growth pattern, floc yield, Exo-polysaccharides (EPS) production, Poly-${\beta}$-hydroxybutyrate (PHB) accumulation, resistance to osmotic and acid stress in Methylobacterium strains CBMB20, CBMB27, CBMB35, and CBMB110. Modified high C:N ratio medium denoted as HCN-AMS medium was used with a C:N ratio of 30:1. The HCN-AMS medium favored increased growth in all the studied strains. All Methylobacterium strains tested positive for EPS production and showed positive fluorescence with calcoflour stain. Elevated levels of EPS production from 4.2 to 75.0% was observed in HCN-AMS medium. Accumulation of PHB in HCN-AMS medium increased by 3.8, 36.7, and 12.0% in strains CBMB27, CBMB35, and CBMB110 respectively. Among the abiotic stresses, osmotic stress-induced growth inhibition of Methylobacterium strains was found to be lowered when grown in HCN-AMS medium. Likewise, growth inhibition due to acid stress at pH 5.0 was lower for strains grown in HCN-AMS medium compared to growth in AMS medium. Enhanced survivability under stress conditions may be attributed to the high EPS and PHB production at increased carbon concentration in the growth medium.

Zoysia japonica Steud.(Zj) is a typical warm-season Korean lawn grass, which is used in many places such as river banks, roadside and soccer fields in Korea. Recently, it has also been used in school yards and the Saemangeum reclaimed land to reduce water pollution. Although the cultivated area of turfgrass is steadily increasing worldwide, it grows fast requiring frequent mowing and is difficult to grow in shady areas and the cold region. Therefore this study aims searching for useful gene(s) to develop abiotic stress tolerant and dwarf zoysiagrass. We isolated Ycf4 gene based on the sequence from Oryza sativa Japonica through RT-PCR and RACE PCR. Ultimately, open reading frame (ORF) of ZjYcf4 was 558bp long, encoding a protein of 186 amino acid residues. NCBI blast results showed that the ZjYcf4 protein is evolutionarily closely related to Ycf4 protein from Zoysia macrantha and Setaria italica (100% and 98%, respectively). To determine whether ZjYcf4 was involved in environmental stress in wild-type zoysiagrass, expression patterns of the gene were analyzed by real-time PCR under salt, cold and dark conditions. They were analyzed after each stress treatment for 3 hours. In salt and cold stresses, the expression was higher compared to control (3-fold and 1.5-fold, respectively), although there was a 1.6-fold decrease in expression under dark stress treatment. As reported previously, we suggest that ZjYcf4 gene affects abiotic stress such as salt, cold and dark.
Calcium ($Ca^{2+}$) plays pivotal roles as an intracellular second messenger in response to a variety of stimuli, including light, abiotic. and biotic stresses and hormones. $Ca^{2+}$ sensor is $Ca^{2+}$-binding protein known to function in transducing signals by activating specific targets and pathways. Among $Ca^{2+}$-binding proteins, calmodulin (CaM) has been well reported to regulate the activity of down-stream target proteins in plants and animals. Especially plants possess multiple CaM genes and many CaM target proteins, including unique protein kinases and transcription factors. Thus, plants are possible to perceive different signals from their surroundings and adapt to the changing environment. However, the function of most of CaM or CaM-related proteins have been remained uncharacterized and unknown. Hence, a better understanding of the function of these proteins will help in deciphering their roles in plant growth, development and response to environmental stimuli. This review focuses on $Ca^{2+}$-CaM messenger system, CaM-associated proteins and their role in responses to external stimuli of both abiotic and biotic stresses in plants.
