식물에서 선택적 스플라이싱에 의한 스트레스 반응 조절 (Regulation of Abiotic Stress Response by Alternative Splicing in Plants)

  • Pre-mRNA의 스플라이싱은 진핵생물 유전자의 적절한 발현에 매우 중요한 역할을 한다. 선택적 스플라이싱은 스플라이싱 위치가 서로 다르게 인식될 때 발생하며 동일한 pre-mRNA로부터 둘 이상의 전사체와 단백질을 생성할 수 있다. 스플라이싱 위치의 결정은 스플라이소솜과 SR 단백질, hnRNP, CBP 등의 스플라이싱 인자에 의해 조절된다. 고온, 저온, 고염, 건조, 저산소 등 다양한 환경 스트레스 조건에서 식물의 많은 스트레스 반응 유전자에 대해 선택적 스플라이싱이 일어나는 것이 알려져 있으며, 이러한 선택적 스플라이싱은 식물이 환경 변화에 적응하기 위한 중요한 기작 중 하나로 여겨진다. 저온, 고온, 고염, 건조 스트레스 조건에서는 스플라이싱 인자의 발현이 변하거나 또는 정상 조건에서와는 다른 스플라이싱 활성을 가짐으로써 선택적 스플라이싱이 일어난다. 환경 스트레스 반응 유전자의 스플라이싱 이소형은 각각 환경 스트레스에 대해 서로 다른 반응을 보이는데 생성되는 조직이 서로 다르기도 하고, 일부 이소형은 넌센스-매개 분해에 의해 분해되기도 한다. 스플라이싱 이소형의 단백질은 환경 스트레스 조건에서 정상 조건과 비교하여 세포 내 위치가 다르기도 하고, 전사인자 또는 효소로서 다른 활성을 가지기도 한다. 이러한 다양한 연구에도 불구하고 식물의 환경 스트레스 반응에서 선택적 스플라이싱에 대한 연구는 일부 스트레스와 유전자에 국한 되어 있고, 아직 분자 기전이 제대로 밝혀지지 않은 부분이 많아 앞으로 더 많은 연구가 필요하다.

Screening methods for drought and salinity tolerance with transgenic rice seedlings

  • Abiotic stress is one of the major serious limiting factors in rice (Oryza sativa) and caused rice production losses. It is important to precisely screen valuable genetic resources for improving stress tolerance and understanding tolerance mechanism to abiotic stresses. Because there are differences of experiment designs for screening of tolerant plant in several studies related to abiotic stress, this study has performed to provide the rapid and efficiency screening method for selection of tolerance rice to drought and salinity stresses. Two week-old rice seedlings that reached about three leaf stage were treated with drought and salinity stresses and examined tolerant levels with tolerant and susceptible control varieties, and transgenic plants. To determine the optimum concentration for the selection of drought and salinity condition, tolerant, susceptible and wild-type plants were grown under three soil moisture contents (5, 10 and 20% water contents) and three NaCl concentrations (100, 200 and 250 mM) for 10 days at seedling stage. 200 mM NaCl concentration and 5% moisture content soil were determined as the optimum conditions, respectively. The described methodologies in this study are simple and efficiency and might help the selection of drought and salinity tolerance plants at the 3,4-leaf-seedling stage.

Arabidopsis Transcription Factor ANAC032 Enhances Salinity and Drought Tolerance

  • The plant-specific NAC transcription factors control various biological processes, including plant development and stress responses. We have isolated an ANAC032 gene, one of the NAC transcription factor family, which was highly activated by multi-abiotic stresses, including high salt and drought in Arabidopsis. Here, we generated transgenic plants constitutively expressing ANAC032 and its knockout to identify the functional roles of ANAC032 in Arabidopsis under abiotic stress responses. The ANAC032-overexpressing plants showed enhanced tolerance to salinity and drought stresses. The anac032 knockout mutants were observed no significant changes under the high salt and drought conditions. We also monitored the expression of high salt and drought stress-responsive genes in the ANAC032 transgenic plants and anac032 mutant. The ANAC032 overexpression upregulated the expression of stress-responsive genes, RD29A and ERD10, under the stresses. Thus, our data identify that transcription factor ANAC032 plays as an enhancer for salinity and drought tolerance through the upregulation of stress-responsive genes and provides useful genetic traits for generating multi-abiotic stress-tolerant forage crops.

Transgenic expression of rice MYB102 (OsMYB102) delays leaf senescence and decreases abiotic stress tolerance in Arabidopsis thaliana

  • MYB-type transcription factors (TFs) play important roles in plant growth and development, and in the rapid responses to unfavorable environmental conditions. We recently reported the isolation and characterization of a rice (Oryza sativa) MYB TF, OsMYB102, which is involved in the regulation of leaf senescence by downregulating abscisic acid (ABA) biosynthesis and the downstream signaling response. Based on the similarities of their sequences and expression patterns, OsMYB102 appears to be a homolog of the Arabidopsis thaliana AtMYB44 TF. Since AtMYB44 is a key regulator of leaf senescence and abiotic stress responses, it is important to examine whether AtMYB44 homologs in other plants also act similarly. Here, we generated transgenic Arabidopsis plants expressing OsMYB102 (OsMYB102-OX). The OsMYB102-OX plants showed a delayed senescence phenotype during dark incubation and were more susceptible to salt and drought stresses, considerably similar to Arabidopsis plants overexpressing AtMYB44. Real-time quantitative PCR (RT-qPCR) revealed that, in addition to known senescence-associated genes, genes encoding the ABA catabolic enzymes AtCYP707A3 and AtCYP707A4 were also significantly upregulated in OsMYB102-OX, leading to a significant decrease in ABA accumulation. Furthermore, protoplast transient expression and chromatin immunoprecipitation assays revealed that OsMYB102 directly activated AtCYP707A3 expression. Based on our findings, it is probable that the regulatory functions of AtMYB44 homologs in plants are highly conserved and they have vital roles in leaf senescence and the abiotic stress responses.

Spatial Variability in Distribution, Abundance and Species Composition of the Subtidal Macronlgal Assemblages Found Along the Geumo Archipelago in the Central South Sea of Korea

  • Dense macroalgal assemblages are a common feature of the rocky subtidal habitats along the coast of Geumo Archipelago in the central South Sea of Korea, but are highly variable in space. This study addresses two questions concerning the algal assemblages: (1) how variable the distribution, abundance and species composition of the assemblages are in space, and (2) how closely the distribution, abundance and species composition of the assemblages are correlated to the spatial variation in abiotic factors. To answer these questions, we investigated 30 sites along the coast in autumn of 2003. The nonmetric multidimensional scaling analysis showed that there were strong differences in the composition and abundance of species in the assemblages among the sites. The similarity among the sites based on presence/absence data was approximately 51%, whereas the similarity based on abundance data was less than 37%, suggesting that the abundance of species contributed much to these differences. There were also strong differences in the number of species, abundance and vertical distribution of the assemblages along the coast. Multiple regression analyses revealed that the number of species, abundance and vertical distribution of the assemblages had a positive relationship with water depth, but less than 58% of total variation in these variables was explained by this abiotic factor. The results suggest that spatial (between habitats) variation is an important and consistent component of subtidal algal assemblages in Geumo Archipelago and should be explained before any differences between localities are assessed.

근권미생물에 의한 식물의 생물·환경적 복합 스트레스 내성 유도 (Induced Systemic Tolerance to Multiple Stresses Including Biotic and Abiotic Factors by Rhizobacteria)

  • 식물은 재배기간 동안 세균, 진균, 바이러스 등의 생물 스트레스뿐만 아니라 고온, 염, 건조 등 다양한 환경 스트레스에도 노출되어 왔다. 최근에는 기후 이상현상으로 인하여 환경 스트레스의 빈도 및 강도가 불규칙적으로 증가하고 있으며 이로 인해 병원균의 생장과 영향도 변화하여 생물과 환경의 복합 스트레스가 식물 재배에 큰 영향을 주고 있다. 유용미생물을 이용한 식물의 저항성 유도는 다양한 생물과 환경 스트레스로부터 식물을 보호하는 데 도움을 주며, 이러한 스트레스에 대한 피해를 감소시킬 수 있는 가능성을 열어 주었다. 본 리뷰에서는 식물의 생물과 환경 스트레스에 대한 피해를 감소시키는 데 영향을 주는 미생물의 결정인자에 대해 기술하였으며 미생물 결정인자에 의해 유도되는 식물 신호전달 체계 변화에 대해 기술하였다. 또한 복합 스트레스 경감을 위한 미생물의 역할과 연구 방향에 대해 기술하였다. 이 리뷰를 통해 변화하는 환경에 대비하기 위해서 다양한 방안을 마련하고 있는 농민들에게 도움이 되기를 바라며, 실제 유용미생물 연구가 식물 재배 중 발생할 수 있는 다양한 스트레스에 따른 농가 피해를 감소시킬 효과적 대응 방안으로 이어지길 바란다.

A Molecular Switch for the Induction of Resveratrol Biosynthesis in Grapes

  • Resveratrol has been reported to possess a variety of biological and pharmaceutical activities. Regardless of its beneficial effects on health, the amount of resveratrol in grapes is very low. In order to induce the resveratrol biosynthesis, the promoter region of a genomic fragment encoding the resveratrol synthase was isolated and a molecular switch was identified which provides us with defining biotic or abiotic inducers that transcriptionally up-regulate the gene expression involved in the resveratrol biosynthesis. We could successfully increase the amount of resveratrol in grapes up to 3-fold by using these environmental factors.

Soybean Improvement for Drought, Salt and Flooding Tolerance

  • Drought, salinity and flooding are three important abiotic factors limiting soybean production worldwide. Irrigation, soil reclamation, and drainage systems are not generally available or economically feasible for soybean production. Therefore, productive soybean varieties with tolerance are a cost effective means for reducing yield losses due to these factors. Genetic variability for higher tolerance to drought, salt and flooding is important. However, only a small portion of nearly 200,000 world soybean accessions have been screened to find genotypes with tolerance for use in breeding programs. Evaluation for tolerance to drought, salinity and flooding is difficult due to lack of faster, cost effective, repeatable screening methods. Soybean strains with higher tolerance to the above stresses have been identified. Crosses with lines with drought, salt and flooding tolerance through conventional breeding has made a significant contribution to improving tolerance to abiotic stress in soybean. Molecular markers associated with tolerance to drought, salt and flooding will allow faster, reliable screening for these traits. Germplasm resources, genome sequence information and various genomic tools are available for soybean. Integration of genomic tools coupled with well-designed breeding strategies and effective uses of these resources will help to develop soybean varieties with higher tolerance to drought, salt and flooding.

Composition and Abundance of Wood-Boring Beetles Inhabited by Pine Trees

  • Plants are consumed by a myriad of organisms that compete for resources. Direct interactions among multiple plant-feeding organisms in a single host can range for each species from positive to negative. Wood-boring beetle faces a number of biotic and abiotic constraints that interfere with the good prospects from the tree. Biotic factors, including arthropod pests and diseases, and abiotic factors, such as drought and water-logging, are the major constraints affecting the species. The present study aimed to provide basic data for analyzing forest health, identify the kinds of wood-boring beetles in the central part of Korea. Our second goal was to analyze the species composition and diversity of regional communities and to examine. A total of 10,461 individual wood-boring beetles belonging to 8 families and 50 species attracted to trap trees in the pine forests were recorded during the study period on study sites. The results of the analysis of collected species showed that the community structure on all study sites was similar. Seasonal occurrences of dominant wood-boring beetles (5 species) from each study site showed the highest number of all species, except for Siphalinus gigas in May, followed by a gradual decline, and the largest number of Siphalinus gigas appeared in June. The similarity index of species composition was relatively high, ranging from 0.75 to 0.90 for each study site.
