Olive flounder Paralichthys olivaceus production has increased gradually in recent years, but prices have fallen. Thus, the development of a variety of processed foods incorporating olive flounder would help to increase the income of fishermen. This study was conducted to investigate the best method for olive flounder ball processing. Clean olive flounder were divided into five portions. Olive flounder meat (100 g with added egg white 39 g) was chopped and then mixed with 10 mL fresh cream and ingredients. The dough was molded into the shape of a ball. The olive flounder balls were then processed by two different methods. In the first method, the flounder ball was boiled in water for 3 min then vacuum-packed in polyethylene film and stored at $-20^{\circ}C$ for 7 days. After 7 days, the ball was thawed and heated in a microwave for 2 min (Sample-1). In the second method, the ball was vacuum-packed in polyethylene film without boiling and then stored at $-20^{\circ}C$ for 7 days before thawing and boiling in water for 3 min (Sample-2). After heating, both types of olive flounder balls were evaluated. Various factors (including the viable bacterial count, chemical composition, pH, hardness, thiobarbituric acid level, salinity, and free amino acid content) were measured, and a sensory evaluation was conducted. Based on the results of the sensory and hardness evaluations, Sample-1 was deemed to be superior to Sample-2.

In this study, new development of natural ventilation window was accomplished to control environment of greenhouse with no use of farced ventilation during hot season. The ventilation effect of developed ventilation window was investigated in experimental greenhouse which was designed using side wall panel and folding type panel fur natural ventilation. Folding panel type ventilation window was designed to open upper part of the side wall and top of the roof using two hinges which are located bottom of the side wall and the roof panel to grab one side of each panels and guide the other side along with the guidance rail. Developed ventilation window has top ventilation part with maximum moving distance X=ι (1-cos$\theta$)=848.5 mm and side ventilation part with maximum moving distance Y=ι/2 $\times$sin$\theta$=1,184.4 mm at 45$^{\circ}$ of theoretical opening angle. It took 4.5 minutes to open roof vent fully and temperature at 1.2 and 0.8 m height decreased after 1 minute from starting opening and became equilibrium state maintaining 3-4$^{\circ}C$ difference after 2 minutes from complete opening. Air exchange rate was 15.2~39.3 h$^{-1}$ which was more than 10~15 h$^{-1}$ of continuous type and Venlo type greenhouse. The descent effect of temperature by ventilation windows was two times higher than Venlo type greenhouse.

Olive flounder (Parlichthys olivaceus) is a large carnivorous fish that live at coastal area and shallow seas in Korea. It was good texture and clean taste because of a high collagen content and low lipid content. More than 70% of olive flounder annual production was traded alive, consequently processing food product from olive flounder is rare to be towed. This study was conducted to investigate the best method of olive flounder cutlet processing. Clean fillet (headless, skinless and contain no viscera part) of olive flounder were divided into 5 portion. Every 100 g of olive flounder meat was wrapped with vinyl then flatten with meat hammer. Flatten fillet then was coated with wheat flour, and seasoned with salt and pepper. These were then coated with egg wash and bread crumbs. Two different method of processing was to make this olive flounder cutlet. Cutlet-1 was fried for 1 min in olive oil, then kept in polyethylene film vacuum packaging ($20{\times}30{\times}0.05mm$) and stored at $-20^{\circ}C$ for 7 days. After 7 days the cutlet was thawed and heat up in microwave for 2 min (Sample-1). The other proup is cutlet-2, which is directly stored in polyethylene film vacuum packaging at $-20^{\circ}C$ for 7 days then thawed and fried for 1 min in olive oil (Sample-2). The factors such as pH, TBA value, amino-N, free amino acid, chemical composition, color value (L, a, b), texture profile, sensory evaluation and viable bacterial count of the olive flounder cutlet (Sample-1, Sample-2) were measured. From the result of sensory evaluation, Sample-2 showed a little high scores than Sample-1. But there was no significant differences in color, odor, taste, texture and overall acceptance between Sample-1 and Sample-2 products.

A lab-scale batch test was conducted to develop capping materials to reduce the sediment phosphorus in the stagnant water zone of Gyeongancheon in Paldang Lake. The mean grain size(Mz) of sediment in the investigated area was 7.7 ${\phi}$, which is very fine, and the contents of organic carbon($C_{org}$) was 2.4%, which is very high. For the phosphorous release experiment to select the optimal capping material, sand layer, powder-gypsum($CaSO_4{\cdot}2H_2O$), granule-gypsum, complex layer(gypsum+sand) and the control were compared and evaluated in the 150 L reactor for 45 days. In case of the capping with the sand, it was found that the phosphorous from the sediment could be reduced by around 50%. However, it was found that this caused the reduction of the dissolved oxygen in the water column(by less than 3 mg/L) due to the resuspension of sediment and the organic matter decomposition that comes from the generation of $CH_4$ gas in the 1 cm of the sand layer. Therefore, it is likely that the sand layer has to be thickener in case of the sand capping. Powder-gypsum and granule-Gypsum reduced phosphorous release by more than 80%. However, the concentration of ${SO_4}^{2-}$ in the water column increased, making it difficult to apply it to the drinking water protection zone. We developed Fe-Gypsum and $SiO_2$-gypsum materials to reduce the solubility of ${SO_4}^{2-}$. Powder-Gypsum creates the interception film that does not have any aperture on the sediment layer when it is combined with the water. However phosphorous release caused by the generation of $CH_4$ gas may happen at a time when the gypsum layer has the crack. Capping through the complex layer(granule-Gypsum+sand(1 cm)) found to be suitable for the drinking water protection zone because it was effective to prevent phosphorus release. Moreover, this leads to the lower solubility from the concentration of ${SO_4}^{2-}$ into the water column than the powder-Gypsum and granule-Gypsum. The addition of gypsum($CaSO_4{\cdot}2H_2O$) into the sediment can reduce the progress of methanogensis because fast early diagenesis and sufficient supply of ${SO_4}^{2-}$ to the sediment, stimulate the SRB(sulfate reducing bacteria) highly.
Objectives: To investigate the effect of glycopeptide-preferring polypeptide GalNAc transferase 1 (ppGalNAc T1 ) targeted RNA interference (RNAi) on the growth and migration of human bladder carcinoma EJ cells in vitro and in vivo. Methods: DNA microarray assays were performed to determine ppGalNAc Ts(ppGalNAc T1-9) expression in human bladder cancer and normal bladder tissues. We transfected the EJ bladder cancer cell line with well-designed ppGalNAc T1 siRNA. Boyden chamber and Wound healing assays were used to investigate changes of shppGalNAc T1-EJ cell migration. Proliferation of shppGalNAc T1-EJ cells in vitro was assessed using [3H]-thymidine incorporation assay and soft agar colony formation assays. Subcutaneous bladder tumors in BALB/c nude mice were induced by inoculation of shppGalNAc T1-EJ cells and after inoculation diameters of tumors were measured every 5 days to determine gross tumor volumes. Results: ppGalNAc T1 mRNA in bladder cancer tissues was 11.2-fold higher than in normal bladder tissues. When ppGalNAc T1 expression in EJ cells was knocked down through transfection by pSUPER-shppGalNAc T1 vector, markedly reduced incorporation of [3H]-thymidine into DNA of EJ cells was observed at all time points compared with the empty vector transfected control cells. However, ppGalNAc T1 knockdown did not significantly inhibited cell migration (only 12.3%). Silenced ppGalNAc T1 expression significantly inhibited subcutaneous tumor growth compared with the control groups injected with empty vector transfected control cells. At the end of observation course (40 days), the inhibitory rate of cancerous growth for ppGalNAc T1 knockdown was 52.5%. Conclusion: ppGalNAc T1 might be a potential novel marker for human bladder cancer. Although ppGalNAc T1 knockdown caused no remarkable change in cell migration, silenced expression significantly inhibited proliferation and tumor growth of the bladder cancer EJ cell line.

In this study, geochemical composition, CFCs (Chlorofluorocarbons), ${\delta}^{18}O$, ${\delta}D$, ${\delta}^{13}C$ isotopes and noble gases isotopes (He, Ne) were analyzed to determine their recharge age, source of $CO_2$ gas and noble gases of carbonated hot spring water and carbonated-water samples collected in the Seoqwipo of the Jeju. The pH of the carbonated waters ranges from 6.21 to 6.84, and the high electrical conductivity range ($1,928{\sim}4,720{\mu}S/cm$). Their chemical composition is classified as $Mg(Ca,\;Na)-HCO_3$ and $Na(Ca,\;Mg)-HCO_3$ types. As a result of the calculation of groundwater age using CFCs concentrations as an environmental tracer, the carbonated water and groundwater were estimated to be about 47.5~57.2 years and about 30.3~49.5 years, respectively. The ${\delta}^{13}C$ values of carbonated water range from -1.77 to -7.27‰, and are plotted on thr deep-seated field or the mixing field of the deep-seated and inorganic origin. Noble gases isotopic ($^3He/^4He$, $^4He/^{20}Ne$) ratio shows that helium gas of carbonated hot waters comes from deep-seated magma origin.
Background: Elevated testicular temperature disrupts spermatogenesis and causes infertility. In the present study, the protective effect of enzymatically biotransformed Panax ginseng Meyer by pectinase (GINST) against chronic intermittent heat stress-induced testicular damage in rats was investigated. Methods: Male Sprague-Dawley rats (4 wk old, 60-70 g) were divided into four groups: normal control (NC), heat-stress control (HC), heat-stress plus GINST-100 mg/kg (HG100), and heat-stress plus GINST-200 mg/kg (HG200) treatment groups. Each dose of GINST (100 mg/kg and 200 mg/kg) was mixed separately with a regular pellet diet and was administered orally for 24 wk. For inducing heat stress, rats in the NC group were maintained at $25^{\circ}C$, whereas rats in the HC, HG100, and HG200 groups were exposed to $32{\pm}1^{\circ}C$ for 2 h daily for 6 mo. At week 25, the testes and serum from each animal were analyzed for various parameters. Results: Significant (p < 0.01) changes in the sperm kinematic values and blood chemistry panels were observed in the HC group. Furthermore, spermatogenesis-related molecules, sex hormone receptors, and selected antioxidant enzyme expression levels were also altered in the HC group compared to those in the NC group. GINST (HS100 and HS200) administration significantly (p < 0.05) restored these changes when compared with the HC group. For most of the parameters tested, the HG200 group exhibited potent effects compared with those exhibited by the HG100 group. Conclusion: GINST may be categorized as an important medicinal herb and a potential therapeutic for the treatment of male subfertility or infertility caused by hyperthermia.
Fish sauce is a traditional Korean fermented seafood and has been used as a condiment since long time ago. Northern sand lance sauce was manufactured with koji, enzyme, and squid viscera of fermentation accelerating agents, and ripened at $15^{\circ}C$. Moisture contents of all samples decreased gradually as fermentation progressed. Crude protein contents increased rapidly up to 1 month-fermentation and then increased slightly up to 5 month. Northern sand lance sauce with squid viscera was the highest in the contents of protein and lipid. The ash content increased gradually during the fermentation periods, while pHs decreased. TMA contents increased up to 3 month-ripening and then decreased slightly. Amino nitrogen content of Northern sand lance sauce with squid viscera increased as fermentation progressed and was the highest among all samples. VBN contents increased up to 5 month-ripening and that of Northern sand lance sauce with squid viscera was the higher than others.

Background: Panax ginseng Meyer polysaccharides exhibit various biological functions, like antagonizing galectin-3-mediated cell adhesion and migration. Galectin-8 (Gal-8), with its linker-joined N- and C-terminal carbohydrate recognition domains (CRDs), is also crucial to these biological processes, and thus plays a role in various pathological disorders. Yet the effect of ginseng-derived polysaccharides in modulating Gal-8 function has remained unclear. Methods: P. ginseng-derived pectin was chromatographically isolated and enzymatically digested to obtain a series of polysaccharides. Biolayer Interferometry (BLI) quantified their binding affinity to Gal-8, and their inhibitory effects on Gal-8 was assessed by hemagglutination, cell migration and T-cell apoptosis. Results: Our ginseng-derived pectin polysaccharides consist mostly of rhamnogalacturonan-I (RG-I) and homogalacturonan (HG). BLI shows that Gal-8 binding rests primarily in RG-I and its β-1,4-galactan side chains, with sub-micromolar KD values. Both N- and C-terminal Gal-8 CRDs bind RG-I, with binding correlated with Gal-8-mediated function. Conclusion: P. ginseng RG-I pectin β-1,4-galactan side chains are crucial to binding Gal-8 and antagonizing its function. This study enhances our understanding of galectin-sugar interactions, information that may be used in the development of pharmaceutical agents targeting Gal-8.

This is a basic research for improving soft magnetic property of Fe based nano crystalline alloy powder core. The main study is done around characteristics of permeability, core loss, and DC bias depending on amount of insulation coating agent and particle size. First, $Fe_{73.5}Si_{13.5}B_9Nb_3Cu_1$ amorphous alloy ribbon was fabricated by using the planar flow casting (PFC) device. Then, heat treatment and ball milling were done to obtain alloy powder. The amount of polyether imide (PEI) added to it was varied by 0.5, 1.0, 2.0, 2.5 wt% to have compression molding into $16ton/cm^2$. After going through crystalline heat treatment, the made toroidal nano crystalline powder core ($OD12.7mm^*ID7.62mm^*H4.75mm$) had smaller permeability as amount of insulation coating agent decreases. However, it was found out that core loss and DC bias characteristics have been improved. The reason for this results were expected to be because green density of power core decreases as amorphous alloy powder particles become smaller as amount of alloy powder insulation coating agent increases, it was determined that 1 wt% of insulation coating agent is appropriate. Also, for powder core made based on alloy powder size with amount of insulation coating agent fixed at 1 wt%, effective permeability and core loss were outstanding as particle size became bigger. However, characteristics of DC bias became worse as applied DC field increases. This is expected to be due to insulation effect, residual pores, or molding density of powder core resulting from thickness of coating on surface of alloy powder.
