The Effect of Salting Conditions on Formation of Cholesterol and Cholesterol Oxides During Gulbi Processing and Storage (염장조건이 굴비 가공 중 콜레스테롤 및 콜레스테롤 산화물 생성에 미치는 영향)

  • Cholesterol and cholesterol oxide products (COPs) of Gulbi (Pseudosciaena manchurica) processed and stored at different salting times of 5 hours and 5 days were analyzed. The content of cholesterol was $133.4{\pm}5.20\;mg/100\;g$ in the fresh sample. Cholesterol content was decreased during salting and storage; its contents were $130.3{\pm}2.95\;mg/100\;g$ and $87.2{\pm}3.49\;mg/100\;g$ in 5 hours and 5 days salting samples, respectively. The content of 7-ketocholesterol in 5 hours and 5 days salting samples were $75.2{\pm}2.70\;{\mu}g/100\;g$ and $82.4{\pm}3.30\;{\mu}g/100\;g$. The 7-ketocholesterol content of salting sample for 5 hours had no significantly difference for 2 days sun drying, but it was dramatically increased during 5 days sun drying and then increased during storage days. $7{\alpha}-$ and $7{\beta}-hydroxycholesterol$ were not detected in fresh sample, but increased during Gulbi processing and storage. The $7{\alpha}-hydroxycholesterol$ was increased during Gulbi processing and storage and a higher level of $658.1{\pm}6.20\;{\mu}g/100\;g$ was detected in 5 hours salting sample than in 5 days salting sample after 21 days storage. The $7{\beta}-hydroxycholesterol$ also showed similar tendency with $7{\alpha}-hydroxycholesterol$; it was largely increased between 7 and 14 days storage in 5 days salting sample.

The Heading Response and Regional Adaptability of Rice Varieties under the Temperature and Day-Length Conditions of Major Rice Production Areas in North Korea (북한 주요 벼 재배지역의 기온과 일장 환경에서 품종의 출수 반응과 지역 적응성 분석)

  • The heading responses of rice varieties that originated from South Korea, North Korea, and northern China were examined under the temperature and day-length conditions of 13 major rice production areas in North Korea. Kenjiandao3 and Nongdae3 originated from China, Olbyeo1, Olbyeo2 and Sonbong9 from North Korea, and Joun from South Korea demonstrated the earliest heading stage depending on the regional environment. Out of 40 rice varieties, 34 reached the heading stage within the regional safe marginal heading date (SMHD) under Haeju and Sariwon environmental conditions, while 16 to 17 varieties reached the heading stage under Wonsan, Changjon, Supung, and Yongyon environmental conditions. Some middle and mid-late maturing varieties that originated from South Korea reached the heading stage within the SMHD under the temperature and day-length conditions of Kaesong, Haeju, Sariwon, Nampo, and Pyongyang that are located in the west-southern plain. The majority of early maturing varieties, but not the middle or mid-late ones, reached the heading stage within the SMHD under the environmental conditions of Singye, Anju, Kusong, and Sinuiju. Only a few early maturing varieties demonstrated the heading stage within the SMHD under Yongyon, Changjon, and Wonsan environments. The number of days to heading was highly positively correlated among all regions; however, it was not consistent among the rice varieties. The 40 rice varieties that had been tested were classified into seven groups according to their heading responses to the temperature and day-length variations of the 13 regional conditions at 65% similarity level in cluster analysis.

Contamination level and congener profiles of PCBs, Co-PCBs and PCDD/DFs in transformer insulation oil samples (변압기 절연유 중 PCBs, Co-PCBs 및 PCDD/PCDFs 오염수준 및 이성체 분포)

  • The levels of total PCBs, Co-PCBs and PCDD/Fs in the transformer insulation oil samples obtained using GC/ECD and HRGC/HRMS were ranged from N.D. to 77.3 ppm, from 0.0863 to 2.49 ppm and from N.D. to 0.00241 ppm, respectively. In terms of WHO-TEQ values, Co-PCBs and PCDD/Fs were ranged from 23.3 to 600 pgTEQ/g and from N.D. to 128 pgTEQ/g, respectively (${\Sigma}Co$-PCBs+PCDD/Fs concentration was calculated 24.4~728 pgTEQ/g). Although, the contribution of PCDD/Fs was below 12% in total TEQ concentration, it is suggested contamination of PCDD/Fs in transformer insulation oils. Among 10 samples, 4 samples showed higher concentration than 2 ppm (specific waste criterion of Korea) and Aroclor 1242, 1248, 1254 and 1260 was detected in samples as a single or mixture of Aroclor. It was shown reliable relationship between concentration of Co-PCBs and those of PCDD/Fs (p<0.003), however, was not shown between production year of transformer and concentration of PCBs. The distribution pattern of Co-PCB congeners showed that the ratios of mono-ortho substituted congeners were higher than non-ortho substituted congeners. Among that, PCB-118 congener was predominant. In addition, the OCDD congener was predominated in PCDD/Fs congeners as above 53%. Moreover, the congener pattern of Co-PCBs was similar to that of Aroclor as well as ambient air, which suggested that PCBs volatilization from transformer insulation oil affected the pattern of Co-PCBs in ambient air.

  • The study was conducted to investigate the effect of dietary energy and protein levels on milk production in Saanen (Capra hircus) dairy goats fed the diets with the different ratio of forage and concentrate. Four lactating goats in mid lactation ($57.5{\pm}10.8\;kg$ BW) were used in a $4{\times}4$ Latin square with 21-d periods. Animals were randomly allocated to one of the four groups; 10.59 MJ ME/kg and 11.90% CP (TJ), 10.14 MJ ME/kg and 12.73% CP (T2), 9.45 MJ ME/kg and 15.19% CP (T3), 8.75 MJ ME/kg and 16.60% CP DM (T4). Also ratio of forage to concentrate was 49: 51 (Tl), 59: 41 (T2), 74: 26 (T3) and 90: 10 (T4). Intakes of forage and concentrate were linearly increased (p<0.05) with their increasing energy levels in the diets.   • To increase the anti-carcinogens phenylethylisothiocyanate (PEITC), myrosinase (MYR), and glutathione S-transferase (GST), genes related to PEITC pathway were isolated and the gene expressions were regulated by Agrobacterium transformation. Isolated cDNAs, MYR, and GST genes were 1,647 bp and 624 bp, respectively, and the protein expression was confirmed through pET system. Thereafter, we constructed a sense-oriented over-expressing myrosinase (pBMY) and RNAi down-regulated GST (pJJGST) binary vectors for the Chinese cabbage transformation. After the transformation, thirteen over-expressing transgenic Chinese cabbage plants (IMS) with pBMY and five down-regulated ones (IGA) with pJJGST were selected by PCR analysis. Selected $T_0$ transgenic plants were generated to $T_1$ plants by self-pollination. Based on the Southern blot analysis on these $T_1$ transgenic plants, 1-4 copies of T-DNA were transferred to Chinese cabbage genome. Thereafter, RNA expression level of myrosinase gene or GST gene was analyzed through real-time RT PCR of IMS, IGA, and non-transgenic inbred lines. In case of IMS lines, myrosinase gene was increased 1.03-4.25 fold and, in IGA lines, GST gene was decreased by 26.42-42.22 fold compared to non-transgenic ones, respectively. Analysis of PEITC concentrations using GC-MS it showed that some IMS lines and some IGA lines increased concentrations of PEITC up to 4.86 fold and up to 3.89 fold respectively compared to wild type. Finally in this study IMS 1, 3, 5, 12, and 15 and IGA 1, 2, and 4 were selected as developed transgenic lines with increasing quantities of anti-carcinogen PEITC.

Quantification of seleno proteins in Korean blood serum using solid phase extraction and affinity chromatography-inductively coupled plasma/mass spectrometry (고체상 추출과 친화 크로마토 그라피-유도결합 플라즈마 질량분석법을 이용한 한국인 혈청에서의 셀레노 단백질 정량)

  • Interferences were removed using anion exchange solid phase extraction (AE SPE) in quantification of selenoproteins in Korean human blood serum with affinity high performance liquid chromatography (AF HPLC)-inductively coupled plasma/mass spectrometry (ICP/MS). The average selenium level obtained for healthy Koreans was $94.3{\pm}2.3ngg^{-1}$ using isotope dilution method. AE SPE was coupled to AF column to separate 3 selenoproteins, glutathione peroxidase GPx, selenoprotein SelP, and selenoalbumin SeAlb. Post column isotope dilution was employed to quantify the proteins. The certified reference material of human blood serum BCR-637 was analyzed to give total selenoprotein concentration of $85.4{\pm}3.4ngg^{-1}$, which agreed well with the reference value of $81{\pm}7ngg^{-1}$. The pooled concentration of GPx, SelP, and SeAlb from healthy Koreans (n=20) was $12.1{\pm}1.4ngg^{-1}$, $57.2{\pm}2.0ngg^{-1}$, and $20.0{\pm}1.9ngg^{-1}$, respectively. The sum of selenoproteins is $89.3ngg^{-1}$, which is about the same as the total selenium concentration of $94.3ngg^{-1}$. The fact suggests that selenium in blood serum is mostly consisted of selenoproteins. After the removal of interference, GPx showed a significant decrease (more than 50%) from $25.0ngg^{-1}$ to $12.1ngg^{-1}$. It was identified that the interference in blood serum was mostly from GPx and the use of AE SPE was proven to be efficient in eliminating Cl and Br that cause interference to GPx.

Determination of Shelf-life of Black Mini Tomato Based on Maturity and Storage Temperature (흑색 방울토마토의 숙기 및 저장온도에 따른 상품성 유지기간 구명)

  • Black mini tomato 'Hei-G' fruits were harvested at different stages of maturity (immature-mature green and mature-black red) and stored at different temperatures (8, 12, and $20^{\circ}C$) to investigate the quality and lycopene content during storage. Weight loss increased dramatically at higher temperature for both harvesting stages without significant differences. Firmness of immature fruits decreased below the initial level of mature fruit (8.1N) after 5, 8, and 19 days storage, when they were stored at 20, 12, and $8^{\circ}C$, respectively. Soluble solid contents of mature fruit increased at initial storage, and were higher as compared to immature fruits before deterioration at each storage temperature. Decrease in titratable acid of mature fruits depended on storage time and temperature. However, titratable acid of immature fruits showed little change during storage, and so it did not affect flavor. Hunter a value changed greatly in immature fruit stored at high temperature. Unlike ripe tomatoes, there was no significant difference in black tomato Hunter b values of immature and mature fruit at initial and 12 days storage. However, immature fruits stored at $8^{\circ}C$ did not reach full maturity and color development and ripening. High storage temperature increased lycopene production while low storage temperature blocked lycopene development. Shelf life of the immature fruits, which was evaluated by elapsed days to conventional mature stage, was 12 and 15 days when they were stored at 20 and $12^{\circ}C$, respectively. The optimum storage temperature to maintain the quality and lycopene content of mature fruits was $12^{\circ}C$. Moreover, the shelf life of mature fruits stored at $20^{\circ}C$ could reach up to 5 days.

Sitological Quality Evaluation of Cultured and Wild River Puffer, Takifugu obscurus (Abe) (양식산과 자연산 황복, Takifugu obscurus (Abe)의 식품학적 품질평가)

  • The present study was performed to evaluate sitological quality of the cultured and wild river puffer, Takifugu obscurus. Proximate composition, the content of extractive nitrogen, the content of nucleotides and their related compounds, total and free amino acid, and fatty acids were analysed and sensory evaluation in the muscle of the river puffer were compared. The cultured river puffer had a higher moisture content compared to the wild fish, while there was no significant difference in crude lipid, crude protein and ash contents. Nucleotides and their related compounds including ATP, ADP, AMP, IMP, HxR and Hx were detected. The result from analyzing ATP-related compound showed difference in total content by wild and cultured river puffer, and IMP content that had largest influence upon the savory taste of sliced raw fish, was higher in the cultured fishes than wild ones. Breaking strength level of the wild river puffer was higher than that of the cultured fish. High levels of C16:0 and C18:0 were shown in all samples and -3 polyunsaturated fatty acid content were not different between the cultured and wild river puffers (P>0.05). Total 17 amino acids were detected in the samples, and most of the samples had high contents of glutamic acid, aspartic acid, lysine and leucine and low contents of cystine, histidine, methionine and tyrosine. The result from surveying free amino acid content of wild and cultured river puffer showed difference in content, but generally taurine and lysine content for the whole free amino acid held the most part. There was no significant difference in texture, flavor and overall acceptance score between the cultured and wild fishes (P>0.05).

Assessment of Regional-Based Nitrogen Loading and Recycling Capacity of Livestock Manure (지역별 가축분뇨의 질소부하 및 자원화 용량 평가)

  • Reusing livestock manure have various advantages in securing soil organic resources. In this study, the N-loading and recycling capacity assessment of animal manure was analyzed by comparing between the cultivated areas of crops and the amount of manure units that are generated from livestock manure. From this assessment, the possibility of recycling resources of livestock manure was evaluated. The amount generated of livestock manure in Gyeonggi-do were evaluated by applying the manure units to the number of livestock manure. The analysis of manure unit per ha shows that the N loading by MU is quite different by region. When it comes to nitrogen loading, the MU per ha of cultivated land in excess of the N-amount was the highest in the Gyeonggi-do province with 2.70 MU/ha, which is higher than the appropriate level. The Chungcheongnam-do province came next with 2.31 MU. So the recycling capacity assessment was carried out mainly based on areas of forage crops, rice that can be provided by recycling livestock manure. The recycling capacity ware highest in Jeollanam-do and Jeollabuk-do. In order to properly apply the livestock manure into organic resources, the seasonal situation that effects the nitrogen demands of crops along with the regional effects crop cultivation should be seriously analyzed. In addition, a system that can effectively produce and manage fertilizer should be established.

Current State for Temperature Management of Cold and Frozen Food Transportation Vehicles in Jeonbuk Province (전북권내 냉장·냉동식품 유통차량 온도관리 현황)

  • To understand of the present state for temperature management of cold and frozen food transportation vehicles, we surveyed and measured the temperatures of eight transportation vehicles (including 3 small & medium and 5 large businesses) in Jeonbuk province, Korea. In the transportation vehicles of small & medium businesses, the mean temperature of cold and frozen foods was $8.35{\pm}5.72^{\circ}C$ and $-3.45{\pm}16.88^{\circ}C$; in large businesses, $3.92{\pm}1.44^{\circ}C$ and $-15.38{\pm}2.98^{\circ}C$, respectively. In the difference of temperature by the locations within transportation vehicles, the difference in each cold and frozen was $2.40{\pm}1.45^{\circ}C$ and $2.37{\pm}2.52^{\circ}C$, as a mean. But there was not statistically significant difference in locations between cold and frozen (p > 0.05). In the difference of surface temperatures on various foods before and after door opening during the loading, the temperatures of cold and frozen foods increased by $0.55^{\circ}C$ and $1.18^{\circ}C$, as means, respectively. The temperature of foods over time and placement of cold and frozen foods in transportation vehicles were not consistently maintained at optimal values in distribution. Therefore, the development of time-temperature history (TTH) system technology at the distribution level for cold and frozen foods is required.
