Characterization of Cellulase Gene (MsGHF45) from Monochamus saltuarius Expressed in Yeast (효모에 발현된 북방수염하늘소(Monochamus saltuarius) Cellulase gene MsGHF45의 특성에 관한 연구)

  • In this study, the cellulase of Monochamus saltuarius (MsGHF45) gene was introduced in Kluyveromyces lactis, successfully. The molecular weight of recombinant enzyme was determined by SDS-PAGE and western blotting. The enzymatic activity was confirmed by native-PAGE containing carboxymethyl cellulose as a substrate. The optimul pH and temperature of recombinant MsGHF45 was pH5 and $40^{\circ}C$. The barium ($Ba^{2+}$) and ferrous ($Fe^{2+}$) enhanced enzyme activity, and the mercuty ($Hg^{2+}$) inhibited its activity.

  • To find antitumor components of Lyophyllum decastes, the mycelia of L. decastes were cultured in artificial media and a new antitumor component which showed potent antitumor activity against sarcoma 180 implanted in mice, was isolated and named 'Lyophyllan A'. Lyophyllan A was composed of a polysaccharide moiety (86%) and a protein moiety (2%). The polysaccharide moiety was found to be a heteroglycan which consisted of glucose (48.1%), mannose (30.8%), galactose, xylose and fucose. The protein moiety contained 18 amino acids. Cultural characteristics of L. decastes were investigated by shake culture method. The best result was obtained when L. decastes was cultured in medium glucose 50g, peptone 10g, corn steep liquor 30ml, $KH_2PO_4$ 0.87g, $MgSO_4$ $7H_2O$ 0.5g, $CaCl_2$ 0.3g, $FeSO_4{\cdot}7H_2O$ 10mg, $MnCl_2{\cdot}4H_2O$ 7mg, $ZnSO_4{\cdot}7H_2O$ 4mg and $CuSO_4{\cdot}5H_2O$ 1mg per one liter at $26^{circ}C$, 180rpm, for 9 days.

  • The Weolagsan area consists of four units; (1) Low grade meta-sediments of the upper members of Ogcheon age unknown group such as Changri (mainly black slate and phyllitic rock), Majeonri (mainly alternation of slate, limestone and chert) and Hwanggangri Formation (pebble bearing phyllitic sediments); (2) Samtaesan Formation of Chosun System of Ordovician; (3) So called meta-volcanics and (4) Weolagsan Granite and its associations which intruded above mentioned meta-sediments and meta-volcanics. This study was focused to know the Woelagsan granite and its metasomatic effects to the country rocks petrographically and petrochemically. According to the field survey, microscopic work and some chemical analysis, the granite is a "normal granite" based on the Streckeisen's classification and belongs to a mass of the Central-zone younger group in Ogcheon geosynclinal belt. The granite metasomatized the country rocks along its northern contact zone. Zone of calcareous and cherty rocks (Majeonri formation) was silicified partly and skarned locally at the contact with the granite. The chemical analysis of the zone show no difinite variations in contents of $SiO_2$ and CaO with the distance from the granite. It seems to be indicated that the silicification of this part was not so metasomatized by the granite body, but thermally affected as much as to be partially remelted in the specific parts of the formations. Meta-volcanic rock zone was slightly chloritized near contact with the granite. Limestone of Samtaesan Formation was silicified and skarned along the contact zone by the granite body. The chemical analysis of the zone show some noticiable changes in compositions of $SiO_2$ and CaO with distance from the granite boundary. It can be imagined that the silicification of this zone was metasomatically originated by Woelagsan Granite. According to chemical analysis on several trace elements, the ratio of Zn/Cr and Ni/Cr are relatively higher than that of Cu/Cr in the above mentioned silicified zones. Generally the variation of these metal elements in the zones tend to be regular with distance from the granite body.

  • Chemical resistance of the cement mortar containing the Thermally Activated Diaomite(TAD) and H.M.(Heavy Metals) has been studied. The H.M.. extracted from EAF(Electrica1 Arc Furnace) Dust. were saturated with diatomite. The diatomite was then thermally activated at $750{\circ}C$ for 30minutes and powdeled. The powder was mixed with a portland cement on a weight basis from 0%. 2.5%. 5.0%. 10%. 20%. The optimum mixture. after those mixtures were subjected to compressive strength(7 and 28days) and leaching bchaviour of H.M.. was tested for its experiment on Wet/Dry cycles and chemical resistance(e.q. imrncrsion in 5%(Conc.) of H2S04, CaC12 and hlgSO4. It was shown that the cement, mortar containing 10% of' P.D. gave a rise to the remarkable increase in compressive strength. The compressive strength was generally decrease beyond the addition of 10% of P.D. The maximum $496kgf/cm^2$ of 28days compressive strength was acheiveti when 10% of P.D. was added to the cement mortar.

Feed Hygiene and Meat Safety of Cattle Fed Processed Rice Hulls-bedded Broiler Litter

  • A study was conducted to determine the safety of feeding processed broiler litter (BL) to beef cattle. The litter was processed by deepstacking, ensiling and composting. The health issues addressed relevant to the safety of feeding litter included pathogenic bacteria, mycotoxins, heavy metals, medicinal drugs and pesticide residues. Exp. 1 evaluated the feed hygiene of processed rice hulls-bedded BL. The presence of pathogenic bacteria in BL was determined before and after deepstacking. A total of 21 BL samples were collected over a 3-year period of commercial and experimental production of BL for beef cattle. Exp. 2 evaluated the safety of meat of cattle fed deepstacked BL. In Exp. 1, there were no pathogenic bacteria, such as coliform, E. coli, E. coli O157:H7, Salmonella, Listeria and Proteus, in deepstacked BL. Levels of heavy metals (Cu, Fe, Mn and Zn) and toxic heavy metals (As, Pb, Cd and Hg) were lower than the commercial feed tolerances. Aflatoxin, medicinal drug and pesticide residues were detected at extremely low levels. In Exp. 2, the meat of the BL-fed animals exhibited few differences in all analyzed items from that of the control group, showing safety from pathogenic microorganisms and heavy metals. When BL was withdrawn for 14 days prior to slaughtering the BLfed cattle, no medicinal drug residues were detected in the meat. Pesticides in the tissues of either group of animals were much lower than the tolerances. In conclusion, processed rice hulls-bedded BL and the meat of cattle fed BL were safe from the potential hazards of pathogenic bacteria, heavy metals, aflatoxin, medicinal drugs and pesticide residues.

Effects of Propolis oral administration according to mixture with Hovenia dulcis Thunb. and Artemisia capillaris Thunb. on D-galactosamine-induced liver injury in rats (지기자(枳期子) 및 인진호(茵蔯蒿) 배합(配合) Propolis의 구강투여(口腔投與)가 D-Galactosamine으로 유발(誘發)된 간손상(肝損傷)에 미치는 영향(影響))

  • Objectives : Propolis and Hovenia dulcis Thunb. has been used as treatment of diseases in the Korean medicine. In this study, we investigated that the hepatoprotective effects of Propolis oral administration according to mixture with Hovenia dulcis Thunb. on ${\gamma}-GTP$, GOT, GPT, Total bilirubin, LDH, ALP, Total cholesterol, Triglyceride, SOD, activity of catalase and Glutathione Peroxidase in galactosamine (GalN)-induced liver in rats. Methods : The animals were divided into 5 groups. Control, the liver injury-induced and not treated group. Pro1, liver injury and administrated propolis. Pro2, liver injury and administrated propolis capsule. Pro3, liver injury and administrated mixture of propolis capsule with Hovenia dulcis Thunb. Pro4, liver injury and administrated mixture of Propolis capsule with Hovenia dulcis Thunb. and Artemisia capillaris Thunb.. Animals were treated by Oral administration of Propolis, Hovenia dulcis Thunb., and Artemisia capillaris Thunb. mixture ltime 2 days for 14 days. Results : The Pro1 group was significantly increased on ${\gamma}-GTP$ and activity of Glutathione peroxidase but decreased on GOT in serum as compared with the control group. The Pro2 group was significantly increased on WBC, RBC, Hct, HGB in serum and activity of CuZnSOD as compared with the control group. The Pro 3 group was decreased on Total bilirubin, increased on LDH, WBC, RBC, Hct and HGB in serum as compared with the control group. The Pro 4 group was decreased on GOT in serum as compared with the control group. Conclusion : By evaluating the liver function and lipid metabolism, Pro3 had a hepatoprotective effect on the prevention of hepatotoxity.

비진공법을 이용한 CIGS광흡수층의 합성과 특성평가

  • Chalcopyrite계 화합물 반도체인 $Cu(InGa)Se_2$ (CIGS)는 직접천이형 에너지 밴드갭과 전파장 영역에 대하여 높은 광흡수계수($1{\times}$[10]^5/cm)를 가지므로 두께 $1{\sim}2{\mu}m$인 박막형태으로 고효율의 태양전지 제조가 가능하다. 또한, 박막공정의 저가 가능성을 나타내면서 전세계적으로 많은 연구와 관심을 받고 있고, 현재 상용화되어 있는 결정질실리콘 태양전지를 대체할만한 재료로 주목 받고 있다. 일반적으로, CIGS박막형 태양전지 구성은는 유리를 기판으로 하여 5개의 단위 박막인 Mo 후면전극, p형 반도체 CIGS 광흡수층, n형 반도체 CdS 버퍼층, doped-ZnO 상부 투명전극, $MgF_2$ 반사방지막으로 이루어진다. 이들 중에서 태양전지의 에너지 변환효율에 결정적인 영향을 미치는 구성된다. CIGS 광흡수층의 제조는 크게 진공법과 비진공방법으로 나뉜다. 현재까지 보고된 문헌에 따르면 CIGS 박막형 태양전지의 경우에 동시증발법으로 20.3%의 에너지 변환효율을 보였지만,는데, 이는 진공장비 특성상 공정단가가 높고 대면적화가 어렵다는 단점을 가진다. 따라서, 비진공법을 이용하여 광흡수층 제작하는 것이 기술적으로 진보할 여지가 크다고 볼 수 있다. 반면 현재 상용화되어 있는 결정질실리콘 태양전지를 대체할만한 방법으로 주목 받고 있는 비진공을 이용한 저가공정은 최근 15.5%의 에너지 변환효율이 보고 되었다. 비진공법에는 전계를 이용한 증착법 및 스프레이법으로 나뉘며, 이들 광흡수층 재료의 화학적 합성은 III족 원소인 In, Ga의 함량비에 따라 광흡수층의 에너지 밴드갭(1.04~1.5 eV) 조절이 가능하다. 따라서, 본 연구에서는 비진공법에 사용되는 CIGS재료의 화학적 합성조건을 변화시켜 III족 원소의 조성비 조절을 시도하였다. CIGS 분말 시료의 입자 형태와 크기를 FE-SEM을 이용하여 관찰하였고, 화합물의 성분비를 EDX 및 XRD 분석을 통해 Ga 함량에 따른 구조적 차이를 비교해 보았다.

Mineralogy and Chemical Composition of the Residual Soils (Hwangto) from South Korea (우리 나라 황토(풍화토)의 구성광물 및 화학성분)

  • The mineralogy and chemical composition of reddish to brownish yellow residual soils, so called "Hwangto" have been examined according to representative host rocks. The result of the study indicates that Hwangto consists of 40-80% clay minerals and various minerals such as quartz, feldspar, hornblende, goethite, and gibbsite. Clay minerals include kaolinite, halloysite, illite, hydroxy interlayered vermiculite (HIV), mica/vermiculite interstratifield mineral and chlorite. The mineralogical constituents and contents of Hwangto were different depending on the types of host rocks. Moreover, the Jurassic granitic rocks contain relatively more kaolin minerals, whereas the Cretaceous granitic rocks contain more HIV and illite. In addition, reddish Hwangto contains relatively more kaolinite and HIV, and yellowish Hwangto contains more illite and halloysite. It is suggested that feldspars and micas of host rocks were chemically weathered into illite, halloysite, illite/vermiculite interstratified minerals, and HIV, and finally into kaolinite. Compared with their host rocks, the major chemical compositions of Hwangto tend to contain more $Al_2O_3,\;Fe_2O_3,\;H_2O$ in amount and less Ca, Mg, and Na. Hwangto contains relatively high amount of trace elements, P, S, Zr, Sr, Ba, Rb, and Ce including considerable amount of Li, V, Cr, Zn, Co, Ni, Cu, Y, Nb, La, Nd, Pb, Th in excess of 10 ppm. Relatively high amount of most trace elements were detected in the Hwangto. The major and minor chemical compositions of the Hwangto were different depending on the types of host rocks. However, their difference was in the similar range compared with the compositions of host rocks.

Characterization of D-Xylose Isomerase from Streptomyces albus (Stleptomyces albus의 D-Xylose Isomerase의 성질에 관하여)

  • Strptomyces albus T-12 which ahd been isolated and identified in the laboratory, was selected for the studies on the cultural conditions on the production of D-xylose iosmerase and the enzymological characteristics using the partially purified enzyme. The best results in the enzyme production came from D-xylose medium than wheat bran. The divalent metla ions as $Co^{2+},\;Fe^{2+},\;Zn^{2+}\;and\;Cu^{2+}$ retard or inhibit the cell-growth at the early stages of mycelia propagations, and T-12 strain is especially sensitive to $Co^{2+}$. After 60 hours of shaking cultivation at $30^{\circ}C$ and 200 rpm, a maximum enzyme activitz, 0.49 enzyme units, was obtained. Cell-free enzyme obtained from mycelia heat-treated in the prescence of 0.5mM $Co^{2+}$, showed a 2.4-fold increase in specific than the enzyme from untreated mycelia. The specific activity of the purified enzyme through Sephadex G-150 columm showed 180 fold to the crude enzyme. The effective activators of the enzyme appeared to be $Mg^{2+}\;and\;Co^{2+}$ ions, and it exhibited the maximal enzyme activity showed at pH 7.0 and at tempersture around $80^{\circ}C$ when $Mg^{2+}\;and\;Co^{2+}$ ions were added. The enzyme isomerized D-glucose, D-xylose, D-ribose, L-arabinose, D-mannose, and L-rhamnose in the present of $Mg^{2+}\;and\;Co^{2+}$ ions as an activatiors. $Mg^{2+}\;and\;Co^{2+}$ ions were non-competitively bound at different allosterix sites of enzyme molecule. $Mg^{2+}(5mM)\;or\;Co^{2+}(1.0mM)$ protected against the thermal denaturations of the enzyme activities. The michelis constant(Km) and $V_{max}$ values of the emzyme for D-glucose and D-xylose were 0.52M, $2.12{\mu}moles/ml{\cdot}min.\;and\;0.28M,\;0.65moles/ml{\cdot}min.$, respectively.

Corrosion Charateristics of PEO-treated Ti-6Al-4V Alloy in Solution Containing Si and Mg Ions

  • The application of the coating supports the mechanical characteristics of the implant, and various materials and coatings are currently being used in the implant in a way to accelerate adhesion. Especially, plasma electrolytic oxidation (PEO) coating has been proposed continually with good surface treatment of titanium alloys. Also, the PEO process can incorporate Ca and P ions on the titanium surface through variables varied factor. PEO process for bioactive surface has carried out in electrolytes containing Ca and P ions. Natural bone is composed of mineral elements such as Mg, Si, Zn, Sr, and Mn, etc. Especially, Mg and Si of these elements play role in bone formation and growth after clinical implantation of bio-implants. In this study, corrosion charateristics of PEO-treated Ti-6Al-4V alloy in solution containing Si and Mg ions has been investigated using several experimental techniques. The PEO-treated surfaces were identified by X-ray diffraction, using a diffractometer (XRD, Philips X' pert PRO, Netherlands) with Cu $K{\alpha}$ radiation. The morphology was observed by field-emission scanning electron microscopy (FE-SEM, Hitachi 4800, Japan) and energy-dispersive X-ray spectroscopy (EDX, Oxford ISIS 310, England). The potentiodynamic polarization and AC impedance tests for electrochemical degradations were carried out in 0.9% NaCl solution at similar body temperature using a potentiostat with a scan rate of 1.67mV/s and potential range from -1500mV to + 2000mV.
