Aerosol Characterization Study for Individual Particle of PM10, PM2.5 Observed in Industrial Area (산업단지내 미세먼지 및 토양입자의 개별입자 분석)

  • Aerosol characterization study for individual particle in Busan metropolitan industrial complex was carried out from December 2010 to August 2011. SEM(scanning electron microscope)-EDX(energy dispersive x-ray) analysis was used for the analysis of 600 single particles during the sampling periods to identify non-metallic aerosol particle sources. Average $PM_{10}$ concentration was 65.5 ${\mu}g/m^3$ in summer, 104.1 ${\mu}g/m^3$ in winter during the sample periods. And Average $PM_{2.5}$ concentration was 24.5 ${\mu}g/m^3$ in summer, 64.5 ${\mu}g/m^3$ in winter individually. Particle density, enrichment factor, correlation analysis, principle component analysis were performed based on chemical composition data. Particle density distribution was measured to 2~4 $g/cm^3$, and the density of $PM_{2.5}$ was measured above 3 $g/cm^3$. In general, the elements Si, Ca, Fe and Al concentrations were higher in all samples of individual particles. The non-ferrous elements Zn, Br, Pb, Cu concentrations were higher in summer than in winter. The concentrations were not changed with the seasons because of non-ferrous industry emission pattern.

Characterization and Potent Application of Pleurotus floridanus Trypsin Inhibitor (PfTI)

  • Characterization and in vitro inhibition studies of protease inhibitor from the mushroom Pleurotus floridanus (PfTI) towards the pest Papilio demoleus is studied. The addition of 1 mM Mn2+, Na2+, Ba2+ and Ni 2+ enhanced the PfTI activity. The ICP-atomic emission spectrum showed the presence of Ca2+, Mg2+ and Zn2+ in the PfTI. Surfactants SDS and CTAB at a concentration of 1% reduced the PfTI activity whereas, the nonionic detergents Triton X and Tween 80 increased the activity. The inhibitory activity gradually decreased with increase in concentration of DMSO and H2O2. The activity was increased by dithiothreitol up to a concentration of 80 μM and inactivated at 140 μM. The activity of PMSF modified PfTI was drastically reduced to 0.234 U/mL at 4 mM concentration and similar results were obtained for modification of cysteine by N-Ethylmaleimide at slightly higher concentrations. The complex of trypsin and PfTI showed complete loss in fluorescence intensity at 343 nm compared with control. In vitro inhibition studies of PfTI with midgut proteases isolated from citrus pest P. demoleus with protease activity of 1.236 U was decreased to 0.613 U by 50 μL (0.1 mg/mL) of the inhibitor. Inhibitor was stable up to 0.04 M concentration of HCl.

Rapid, Sensitive, and Specific Detection of Salmonella Enteritidis in Contaminated Dairy Foods using Quantum Dot Biolabeling Coupled with Immunomagnetic Separation

  • Colloidal semiconductor CdSe-ZnS core-shell nanocrystal quantum dot (Qdot) are luminescent inorganic fluorophores that show potential to overcome some of the functional limitations encountered with organic dyes in fluorescence labeling applications. Salmonella Enteritidis has emerged as a major cause of human salmonellosis worldwide since the 1980s. A rapid, specific, and sensitive method for the detection of Salmonella Enteritidis was developed using Qdot as a fluorescence marker coupled with immunomagnetic separation. Magnetic beads coated with anti-Salmonella Enteritidis antibodies were employed to selectively capture the target bacteria, and biotin-conjugated anti-Salmonella antibodies were added to form sandwich immune complexes. After magnetic separation, the immune complexes were labeled with Qdot via biotin-streptavidin conjugation, and fluorescence measurement was carried out using a fluorescence measurement system. The detection limit of the Qdot method was a Salmonella Enteritidis concentration of $10^3$ colony-forming units (CFU)/mL, whereas the conventional fluorescein isothiocyanate-based method required over $10^5CFU/mL$. The total detection time was within 2 h. In addition to the potential for general nanotechnology development, these results suggest a new rapid detection method of various pathogenic bacteria from a complex food matrix.

Quality of Steam Cooked Surimi Gel Prepared using Sandfish Arctoscopus japonicus Meat (도루묵(Arctoscopus japonicus) 어묵의 제조 및 품질특성)

  • This study examined the effect of pretreated sandfish Arctoscopus japonicus meat as a surimi complex for preparing sandfish flavored fish paste. To prepare the sandfish-flavored paste, fine chopped sandfish meat including backbone was added in a ratio of 0 to 50 wt.% to thawed Alaska pollock Theragra chalcogramma surimi to make a mixed surimi gel. To prepare the sandfish-flavored paste, the mixed surimi was ground with salt using a silent cutter, mixed with starch and stabilizers 0.2% transglutaminase and gluconolactone 0.3%, stuffed in a rectangular container, left for 3 h at $25^{\circ}C$, cooked in hot water for 30 min at $90^{\circ}C$, and finally chilled for 20 min at $4^{\circ}C$. The effects of the pretreatment of sandfish meat were investigated by analyzing the quality of the paste produced. The proximate composition of FP (fish paste containing 40% steam-cooked sandfish meat and 0.3% gluconolactone) was moisture 76.1%, crude protein 12.0%, crude fat 3.8%, carbohydrate 6.1%, and ash 2.0%. The major minerals in FP were Na (23.77 mg/L), Mg (1.46 mg/L), Zn (1.04 mg/L), and Fe (0.41 mg/L), and the major free amino acids were taurine, anserine, alanine, and glutamic acid. The monounsaturated fatty acid content of FP was 566.22 mg%, and the polyunsaturated fatty acid content was 498.43 mg%. The n-3 fatty acid content was 398.01 mg%, and C20:5n-3 (218.85 mg %) was a major component.

Geochemical Characteristics of Intertidal Sediment in the Semi-enclosed Bays of the Southern Region of Jeollanam Province (전남 남부 반폐쇄적인 내만 갯벌 퇴적물의 지화학적 특성)

  • To understand the geochemical characteristics of intertidal sediment in a semi-enclosed bay, we measured various geochemical parameters, including grain size, ignition loss (IL), chemical oxygen demand (COD), acid volatile sulfide (AVS), and trace metals (Al, Fe, Cu, Pb, Zn, Cd, Hg, and As), in intertidal sediment from three bays (Deukryang Bay, Yeoja Bay, and Gamak Bay) in the southern region of Jeollanam Province. The intertidal sediment in Deukryang Bay consisted of various sedimentary types, such as sand, gravelly muddy sand, mud, and silt, whereas the intertidal sediments in Yeoja and Gamak Bays were composed mainly of mud. The concentrations of IL, COD, AVS and trace metals in the intertidal sediments of the three study regions were relatively high near areas affected by input of stream waters and/or shellfish farming waste. The concentrations of organic matter and trace metals in Gamak Bay were much higher than those in Deukryang and Yeoja Bays, which appears to be due to the influence of anthropogenic pollutants, originating from the city and the industrial complex near Gamak Bay. The evaluation results of organic matter and metal pollution using the sediment quality guidelines showed that the intertidal sediments in the three study regions were not polluted in terms of organic matter and trace metals. In future, sustainable management for sources of organic matter and trace metal is necessary to conserve a healthy benthic ecosystem in intertidal sediments.

  • Separation of optical isomers of DNS derivatized amino acids by a reversed-phase high-performance liquid chromatography has been studied by adding a complex of an optically active amino acid (L-arginine) with the metal ion (Cu(II), Zn(II), Cd(II), Ni(II)) to the mobile phase. The separations are affected by the concentrations of acetonitrile, chelate and buffer. They are also affected by the pH and the kinds of metal and buffer. A separation mechanism, which is based on steric effect of the ligand exchange reaction for the formation of ternary complexes by the D,L-DNS-amino acids and the chiral additive associated with the stationary phase, is proposed to interpret the elution behaviors of D, L-dansyl-amino acids.

Assessment of Selected Heavy Metal Concentrations in Agricultural Soils around Industrial Complexes in Southwestern Areas of Korea

  • Agricultural soils near or around industrial complexes can contain a certain amount of heavy metals that readily enter the food chain and negatively affect human health. Therefore, we conducted the study to investigate the distribution of selected heavy metals, including arsenic (As), cadmium (Cd), chromium (Cr), nickel (Ni), lead (Pb), mercury (Hg), and zinc (Zn), in farm-land soils around fifteen industrial complexes in the southwestern provinces, Korea. The concentrations of heavy metals in the soil samples were determined by the pseudo-total aqua regia (3 HCl : $1HNO_3$) digestion procedure. The heavy metal concentrations in most soils examined did not exceed the levels of Soil Contamination Warning Standard (SCWS) for agricultural lands (Region 1) presented in Soil Environment Conservation Law (SECL) established by Ministry of Environment (MOE), Korea. However, only one sampling site showed higher As amount ($27.1mg\;kg^{-1}$) than the SCWS level of As ($25mg\;kg^{-1}$). Pollution index (PI) for heavy metals did not exceed 1.0. The PI values were significantly positively correlated (p < 0.01) with the heavy metal concentrations. In particular, the values of correlation coefficient between the Cd and Pb concentrations and the PI values were higher than those estimated from other combinations, and thus the amounts of Cd and Pb in the agricultural soils highly affected the PI values for the heavy metals.

  • Mg-and Zn-ferrites having spinel structure, a kind of complex oxides showing the advantageous properties of constituently single metal oxides, were selected to find a relationship between their catalytic activities in the dehydrogenation of ethylbenzene to styrene and the catalytic properties. For the structural and physical analyses of ferrites, XRD, BET, DTA, XPS, TEM and TPD methods were employed. Potassium added to the catalyst played a role of bifunctional promoter which brought the electronic effect as well as the structural one for the increment of particle dispersion. K-addition decreased acid strength of the catalyst by neutralization and increased its acidity. In the dehydrogenation of ethylbenzene, K-addition let the selectivity to styrene be constant throughout the reaction by the proper acid strength of the ferrite for the reaction, which could be obtained from the neutralization of strong acid sites by potassium.

Changes in Nutritional Composition and Physicochemical Properties of Mustard Leaf (Brassica juncea) Kimchi during Fermentation (갓김치(Brassica juncea) 숙성 중 영양성분 및 이화학적 특성 변화)

  • Nutritional composition and physicochemical properties changes in mustard leaf kimchi were investigated during fermentation of up to 3 months. The pH decreased, and the titratable acidity gradually increased according to increase of fermentation periods. Fructose and glucose were the major free sugars in mustard leaf kimchi, and their amounts were significantly decreased with fermentation periods (p<0.05). Lactic acid content showed a significant increase with maximum increase at 3 months. All types of kimchi contained 20 amino acids, but the content of most amino acid fluctuated during fermentation. Except for K and Zn, the content of other ingredients including Ca, Fe, Mg, Na, Se were the highest in kimchi fermented for 2 months. The unsaturated fatty acid of mustard leaf kimchi was higher than that of saturated fatty acid, and total fatty acid of kimchi significantly decreased after 2 months (p<0.05). Most vitamin contents showed a tendency to decrease with fermentation, in particular, vitamin B complex except for $B_2$ significantly decreased after 3 months (p<0.05). The results provide fundamental data for determining the appropriate fermentation period to improve the quality of kimchi.

Gadolinium Complexes of Bifunctional Diethylenetriaminepentaacetic Acid (DTPA)-bis(amides) as Copper Responsive Smart Magnetic Resonance Imaging Contrast Agents (MRI CAs)

  • We present the synthesis and characterization of DTPA-bis(histidylamide) (1a), DTPA-bis(aspartamide) (1b), and their gadolinium complexes of the type $[Gd(L)(H_2O)]$ (2a:L = 1a; 2b:L = 1b). Thermodynamic stabilities and $R_1$ relaxivities of 2a-b compare well with Omniscan$^{(R)}$, a well-known commercial, extracellular (ECF) MRI CA which adopts the DTPA-bis(amide) framework for the chelate: $R_1$ = 5.5 and 5.1 $mM^{-1}$ for 2a and 2b, respectively. Addition of the Cu(II) ion to a solution containing 2b triggers relaxivity enhancement to raise $R_1$ as high as 15.3 $mM^{-1}$, which corresponds to a 300% enhancement. Such an increase levels off at the concentration beyond two equiv. of Cu(II), suggesting the formation of a trimetallic ($Gd/Cu_2$) complex in situ. Such a relaxivity increase is almost negligible with Zn(II) and other endogenous ions such as Na(I), K(I), Mg(II), and Ca(II). In vivo MR images and the signal-to-noise ratio (SNR) obtained with an aqueous mixture of 2b and Cu(II) ion in an 1:2 ratio demonstrate the potentiality of 2 as a copper responsive MRI CA.
