Enhancing the Enzymatic Activity of the Multifunctional β-Glycosyl Hydrolase (Cel44C-Man26AP558) from Paenibacillus polymyxa GS01 Using DNA Shuffling (DNA Shuffling을 이용한 Paenibacillus polymyxa GS01의 다기능 β-Glycosyl Hydrolase (Cel44C-Man26AP558) 효소 활성 증가)

  • We previously reported that the truncated Cel44C-$Man26A_{P558}$ ${\beta}$-glycosyl hydrolase protein exhibits multifunctional activities, including cellulase, xylanase, and lichenase. DNA shuffling of the truncated Cel44C-$Man26A_{P558}$ enzyme was performed to enhance the enzymatic activity of the multifunctional ${\beta}$-glycosyl hydrolase. Two mutant enzymes, M2Cel44C-$Man26A_{P558}$ that carries one mutation (P438A) and M21Cel44C-$Man26A_{P558}$ that carries two mutations (A273T and P438A) were obtained. The enzymatic activity of the M21Cel44C-$Man26A_{P558}$ double mutant was lower than enzymatic activity of the single mutant (M2Cel44C-$Man26A_{P558}$). However, both mutants displayed the enhancements in their enzyme activities that were ${\approx}1.3$- to 2.2-fold higher than the original enzymatic activity in Cel44C-$Man26A_{P558}$. In particular, the mutant M2Cel44C-$Man26A_{P558}$ exhibited an approximate 1.5- to 2.2-fold increase in the cellulase, xylanase, and lichenase activities in comparison with the control (Cel44C-$Man26A_{P558}$). The optimum cellulase, linchenase, and xylanase activities of ${\beta}$-glycosyl hydrolase were observed at pH 7.0, pH 7.0 and pH 6.0, respectively. These results, therefore, suggest that the amino acid residue Ala438 plays important roles in the enhancement of the activity of multifunctional ${\beta}$-glycosyl hydrolase.

  • Sensory evaluation and Instron test were carried out to study me effect of different cooking methods on the quality of soybean jeolpyon. Cooking methods studied were rice cake steamer, steaming pot, pressure cooker and microwave oven. 1. There were no significant difference in color and flavor between samples cooked by various methods. Moistness of samples by steaming pot and rice cake steamer was similar. Tenderness of samples by rice caker and pressure cooker was better man others. 2. After 24 hours elapsed, me quality of soybean jeolpyon showed me order of by rice cake steamer, steaming pot, pressure cooker and microwave oven. 3. It showed mat soybean ieolpyon by microwave oven was harder man others through Instron test. 4. Hardness of jeolpyons were increased as storage time increased. The rate was higher by pressure cooker and microwave oven man by rice cake steamer and steaming pot.

As the growth of the old man over sixty-five years old, there needs a care Health specialist, care worker who helps different people in daily life. The old man welfare facilities are a medical doctor, nurse, physical therapy, occupational therapy, nutritionist, social worker, and care worker. They constitute teamwork and effort activities of daily living and rehabilitation. According to Japanese care worker low; a research worker makes the care welfare programs from Article 1 to 26 that agree with the real state of Korea.

  • A difference in a views of the Ong(癰) and juh(疽). In the Ong-juh(癰疽) of the Young-Chu(靈樞), if the Ki(氣) and Hul(血) are blocked in a man's body, the heat(熱) generated consists of Pus(膿). Pus(膿) which goes into the man's body and doesn't deteriorate the five viscera(五臟) is Ong(癰). Juh(疽) is pus(膿) which goes deeply into the man's body does hurt the muscle and bone. Tal-Juh(脫疽) is an ailment in the toe. It is a sort of the gangrene of the extremities. In a literature study oh the Tal- Juh(脫疽), it obtained some opinions, so it is report.

  • Railroad disasters are frequently occurred by man-made causes or natural causes. In general, man-made causes are illegal construction practices, deterioration with the lapse of time and railroad crossing accidents, and natural causes are rainfall. snow, wind, earthquake, etc. Of cause, railroad disasters by man-made causes are prevented from keeping the safety principle, constructing multi-level crossing, securing enough men of ability and financial resources and making a thorough check using equipments with high capacity. And railroad disasters by natural causes are also minimized by construction of disaster prevention facilities, introduction and operation of general disaster prevention system and reasonable train passage control. Therefore, to setup the criterion of train passage control for train safety at railroad bridge under heavy rainfall, risky factors, national and oversea criteria under such circumstances are reviewed and a scheme to setup the criterion is suggested.

  • This paper deals with the design problem of a man-on-board(MOB) storage and retrieval warehousing system which is suitable for storing items of small size and light weight. It is assumed that the operator carries out a sequence of retrieving(or storing) operations traveling on a specially designed truck. Considering the operating characteristics of the man-on-board system, an optimal design model is developed in which the investment and maintenance costs of the system are minimized over a time horizon satisfying a set of constructional restrictions. The model is formulated as a nonlinear integer program and a search algorithm is proposed to find an optimum solution.
