Development of Simultaneous Analysis of Ferulic Acid, Caffeic Acid, Catechin and Taxifolin from Health Functional Food Pinus Pinaster Bark Extract by UPLC-MS/MS (건강기능식품 프랑스해안송껍질추출물 중 UPLC-MS/MS를 이용한 Ferulic acid, Caffeic acid, Catechin, Taxifolin 동시분석법 개발 연구)

  • This study was conducted to develop a simultaneous analysis method for ferulic acid, caffeic acid, catechin and taxifolin from Health Functional Food (HFF) Pinus Pinaster bark extract. The simultaneous analytical method for ferulic acid, caffeic acid, catechin and taxifolin is carried out using UPLC-MS/MS. The method validation was performed to determine selectivity, linearity, accuracy, limit of detection (LOD), limit of quantification (LOQ) and precision for ferulic acid, caffeic acid, catechin and taxifolin. LC-MS/MS method was established using an Acquity UPLC BEH $C_{18}$ Column and was applied for these 4 compounds. Product-ion traces, at m/z $194.2{\rightarrow}133$, $180.2{\rightarrow}135$, $290.3{\rightarrow}245$, $304.3{\rightarrow}248$, were used for quantitative analysis of ferulic acid, caffeic acid, catechin and taxifolin, respectively. Excellent linearity ($r^2=0.999$) was observed for ferulic acid, caffeic acid, catechin and taxifolin in the concentration range (50-2500 mg/L). The observed recoveries of these 4 compounds were found to be between 84.9 and 104.9%, while precision was between 1.20 and 4.43% relative standard deviation (% RSD).

Acoustic Analysis and Auditory-Perceptual Assessment for Diagnosis of Functional Dysphonia (기능성 음성장애의 진단을 위한 음향학적, 청지각적 평가)

  • Background and Objectives : The purpose of this study was to compare the measured values of acoustic and auditory perceptual assessments between normal and functional dysphonia (FD) groups. Materials and Methods : 102 subjects with FD and 59 normal voice groups were participated in this study. Mid-vowel portion of the sustained vowel /a/ and two sentences of 'Sanchaek' were edited, concatenated, and analyzed by Praat script. And then auditory-perceptual (AP) rating was completed by three listeners. Results : The FD group showed higher acoustic voice quality index version 2.02 and version 3.01 (AVQIv2 and AVQIv3), slope, Hammarberg index (HAM), grade (G) and overall severity (OS), values than normal group. Additionally, smoothed cepstral peak prominence in Praat (PraatCPPS), tilt, low-to high spectral band energies (L/H ratio), long-term average spectrum (LTAS) in FD group were lower than normal voice group. And the correlation among measured values ranged from -0.250 to 0.960. In ROC curve analysis, cutoff values of AVQIv2, AVQIv3, PraatCPPS, slope, tilt, L/H ratio, HAM, and LTAS were 3.270, 2.013, 13.838, -22.286, -9.754, 369.043, 27.912, and 34.523, respectively, and the AUC of each analysis was over .890 in AVQIv2, AVQIv3, and PraatCPPS, over 0.731 in HAM, tilt, and slope, over 0.605 in LTAS and L/H ratio. Conclusions : In conclusion, AVQI and CPPS showed the highest predictive power for distinguishing between normal and FD groups. Acoustic analyses and AP rating as noninvasive examination can reinforce the screening capability of FD and help to establish efficient diagnosis and treatment process plan for FD.

Push-pull Strategy for Control of Sweet-potato Whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae) in a Tomato Greenhouse (Push-pull 전략을 이용한 시설 토마토 담배가루이 방제효과)

  • This study was conducted to develop a technology for environmentally friendly control of sweet-potato whitefly, Bemisia tabaci, by controlling their behavior using a push-pull strategy in a tomato greenhouse. B. tabaci was attracted the most by yellow color, light source of 520 nm, whereas it avoided the complex light treatment of 450 + 660 nm. The two natural enemies of B. tabaci, Cyrtopeltis tenuis and Orius laevigatus, were attracted the most by 520 nm light source. B. tabaci was repelled by the volatile organic compounds ocimene and carvacrol and was the most attracted by methyl isonicotinate. When buckwheat was added into the tomato greenhouse, the density of C. tenuis was maintained at about 16 times higher than when untreated for 15 days. As a result of the combined treatment of push-pull strategy, the density per trap of B. tabaci was three times lower than when no treatment was applied, and the control of this pest increased with time and reached up to 68.7%.

Wear Property of HVOF WC-CoCr Coating Manufactured by Optimal Coating Process (최적 고속화염용사코팅 공정기술에 의하여 제조된 WC-CoCr 코팅의 마모 특성)

  • Thermally sprayed tungsten carbide-based powder coatings are being widely used for a variety of wear resistance applications. The coating deposited by high velocity processes such as high velocity oxy-fuel (HVOF) thermal spraying is known to provide improved wear resistant property. In this study, optimal coating process (OCP) is obtained by the study of coating properties such as surface hardness, porosity, surface roughness and microstructure of 9 coatings prepared by Taguchi program for 3 levels of four spray parameters. The Friction and wear behaviors of HVOF WC-CoCr coating prepared by OCP, electrolytic hard chrome (EHC) plating and Inconel718 (In718) are investigated by reciprocating sliding wear test at $25^{\circ}C$, $450^{\circ}C$. Friction coefficients (FC) of all of the 3 samples are decreased as increasing sliding surface temperature from $25^{\circ}C$ to $450^{\circ}C$. FC of WC-CoCr decreases as increasing the surface temperature from $0.33{\pm}0.02$ at $25^{\circ}C$ to $0.26{\pm}0.02$ at $450^{\circ}C$, showing the lowest FC among the 3 samples. Wear trace (WT) and wear depth (WD) of WC-CoCr are smaller than those of EHC and In718 both at $25^{\circ}C$ and $450^{\circ}C$. These show that WC-CoCr is highly recommendable for protective coating on In718 and other metal components.

Trends in Heart Valve Surgery in Korea: A Report from the Heart Valve Surgery Registry Database

  • Background: In this study, we present recent trends in heart valve surgery in Korea through analyses of data from the Korea Heart Valve Surgery Registry (KHVSR). Methods: We enrolled 8,981 patients who were registered in the KHVSR from 2017 to 2020. Yearly trends in patients' baseline characteristics, surgical profiles, and early mortality rates were explored. The observed/expected mortality ratio (O/E ratio), calculated from the actual mortality in the KHVSR and the predicted mortality estimated using the EuroSCORE II, was also analyzed. Results: The proportion of aortic valve surgery significantly increased from 56.8% in 2017 to 60.3% in 2020. The proportion of all combined procedures and minimally invasive surgery significantly increased over the 4-year study period. The operative mortality rate was 2.9% in the entire cohort, while mitral valve repair showed the lowest mortality risk (0.9%). The mortality rates of isolated aortic valve replacement (AVR) significantly decreased from 2.1% in 2017 to 0.8% in 2020 (p=0.016). Overall, the O/E ratio was 0.784 (95% confidence interval [CI], 0.677-0.902) demonstrating significantly lower actual mortality risks than expected based on the EuroSCORE II. In particular, the O/E ratios were as low as 0.364 (95% CI, 0.208-0.591) for isolated AVR. Conclusion: The recent data from the KHVSR showed increasing trends for complex procedures and minimally invasive surgery in heart valve surgery in Korea, and demonstrated remarkably low risks of operative mortality.

Deep Learning Braille Block Recognition Method for Embedded Devices (임베디드 기기를 위한 딥러닝 점자블록 인식 방법)

  • In this paper, we propose a method to recognize the braille blocks for embedded devices in real time through deep learning. First, a deep learning model for braille block recognition is trained on a high-performance computer, and the learning model is applied to a lightweight tool to apply to an embedded device. To recognize the walking information of the braille block, an algorithm is used to determine the path using the distance from the braille block in the image. After detecting braille blocks, bollards, and crosswalks through the YOLOv8 model in the video captured by the embedded device, the walking information is recognized through the braille block path discrimination algorithm. We apply the model lightweight tool to YOLOv8 to detect braille blocks in real time. The precision of YOLOv8 model weights is lowered from the existing 32 bits to 8 bits, and the model is optimized by applying the TensorRT optimization engine. As the result of comparing the lightweight model through the proposed method with the existing model, the path recognition accuracy is 99.05%, which is almost the same as the existing model, but the recognition speed is reduced by 59% compared to the existing model, processing about 15 frames per second.

Evaluation of Therapeutic Monitoring of Prostate Cancer (PCa) using [18F]Florastamin, Diagnostic Radiopharmaceutical for PCa: Non-clinical Ex vivo Whole-body Autoradiographic Analysis

  • In this study, we evaluated the targeting of prostate cancer (PCa) using [18F]Florastamin in non-clinical study, for the purpose of therapeutic monitoring of [177Lu]Ludotadipep, a therapeutic radiopharmaceutical for PCa, [18F]Florastamin/[177Lu]Ludotadipep was co-administered to a single-individual prostate tumor bearing mouse model, mimicking clinical condition. Considering the difference in half-life of the two isotopes (18F or 177Lu), image scan of whole-body autoradiography was performed at 24 or 48 h after preparation of frozen section, respectively. Then, it was confirmed whether they showed the same targeting efficiency for the area of tumor. A tumor xenograft model was prepared using PSMA-overexpressing PC3-PIP prostate cancer cells. [18F]Florastamin [111 MBq (3 mCi) in 100 µL]/177Lu]Ludotadipep [3.7 MBq (100 µCi) in 100 µL] was co-administered through the tail vein, and 2 hours after administration, the mice were frozen, and after freezing for 24 hours, whole-body cryosection was performed at 24 h after freezing. Image scanning using cryosection was performed after 24 or 48 hours after freezing, respectively. In the scan image after 24 hours, tumor uptake of [18F] Florastamin/[177Lu]Ludotadipep were simultaneously observed specific uptake in the tumor. In the scan image after 48 hours in the same section, signal of 18F was lost by decay of radioisotope, and specific uptake image for [177Lu]Ludotadipep was observed in the tumor. Uptake of [177Lu]Ludotadipep was specific to the same tumor region where [18F]Florastamin/[177Lu]Ludotadipep was uptake. These results suggested that [18F]Florastamin showed the same tumor uptake efficiency to PCa as [177Lu]Ludotadipep, and effective therapeutic monitoring is expected to be enable using [18F]Florastamin during [177Lu]Ludotadipep therapy for PCa.

The Natural hazards and Drought Periodicity during the Medieval Times in Korea Based on the History of Goryeo(Goryeosa) (고려사를 통해 본 한국 중세의 자연재해와 가뭄주기)

  • The natural hazards such as droughts, floods and frost injuries, and their relationships with the social unrest phenomena such as years of famine, smallpox and revolutions during the Medieval times(AD 936~1391) in Korea are compared and analyzed based on the History of Goryeo containing astronomical weather and climate events during the Goryeo Dynasty. Among the natural hazards during the Goryeo Dynasty, the droughts with a period of 4~5 years on average are recorded most frequently. By time series analysis, the most frequency of droughts can be found in AD 286~335, AD 786~835 and AD 1261~1320 during the Ancient and Medieval times. It is suggested that three cycles of the droughts during approximately 1,500 years of the Ancient and Medieval times are recognized and they had happened with the time intervals of approximately 500 years. The frequency of droughts increased in the late Goryeo Dynasty is consistent with those of the social chaos factors such as years of famines, smallpox and revolutions. These records suggest that the natural hazard such as years of famines was one of the most important factors influencing on the collapse of the Goryeo Dynasty, agrarian country.

A Comparison Study on Interest of Dietary Life Behavior, Nutrient Intake and Health between Full-Time and Working Housewives (전업 주부와 직업주부의 식생활, 영양소 섭취상태 및 건강에 대한 관심도 비교 연구)

  • The purpose of this study was to compare the awareness of dietary patterns and health of full-time and employed housewives. The investigation gathered information on general characteristics, dietary lifestyle, nutritional status, and health by questionnaire as well as anthropometric measurements. The subjects included housewives living in Seoul and Gyeonggi Province (86 full-time and 127 working housewives) during November 2009 to January 2010. The average age, height, and weight of the housewives were $45.8{\pm}7.6\;yr$, $161.0{\pm}8.4\;cm$, and $55.7{\pm}6.3\;kg$, respectively. The average monthly income of the households was 3 million to 5 million won (31.4% full-time and 37.0% working housewives), and their average food expense was 200,000 to 500,000 won (46.5% full-time and 48.1% working housewives). More than 90% of the studied homes were nuclear families and the most common number of children was 2 to 3 (79.1% full-time and 76.4% working housewives). The ratio of employed housewives who ate regular meals was 35.4%, but that of the full-time housewives was 48.2% (p<0.05). Of the working housewives, 7.1% never ate breakfast and the primary reason for skipping breakfast was 'busy in the morning' (61.1%). The type of food that the housewives preferred when eating out was Korean food, both for the full-time and employed housewives (76.0%). All the housewives overate calories and the full-time housewives ate more protein, fat, vitamin A, vitamin B1, vitamin B6, niacin, vitamin C, vitamin E, folic acid, and cholesterol than the employed housewives (p<0.05). Both the full-time and employed housewives were more satisfied with supper than with other meals. About 30.6% of the full-time and 33.9% of the working housewives had no interest in health, but answered that regular exercise was very important for health (33.3% full-time and 39.7% working housewives). In conclusion, working housewives have poor dietary habits and nutrient intakes, thus substantial measures to improve these problems are urgently needed.

Holocene climate characteristics in Korean Peninsula with the special reference to sea level changes (해수면 변동으로 본 한반도 홀로세(Holocene) 기후변화)

  • Sea level fluctuations during the Holocene reconstructed by the results of age dating, microfossils researches and sedimentary facies from coastal alluvial plains contain the valuable informations on climatic changes. The sea level during 'maximum phase of transgression' during 6,000~5,000 yr BP was slightly higher than the present by approximately 0.8~1.0 m and the summer temperature conditions seemed to be higher than those of the present by 2~3℃ in the Central Europe when the period of 'Climatic Optimum' might be dominant. The sea level in Korean Peninsula was assumed by 0.8~1.0 m higher at that time compared to the present and climate seemed to be warmer. At 2,000~1,800 yr BP in Korean Peninsula, the sea level reached the higher stand than the present by approximately 1.1~1.3 m and the climatic conditions might be warm similar to the period of 'Climatic Optimum'. Although the temperature in the Central Europe during the period of 'Subboreal' was about 2~3℃ cooler, it is supposed that the sea level in Korean Peninsula was relatively higher than the present. The sea level at 2,300 yr BP might be similar to that of the present, which was the lowest level since the mid-Holocene. From the fact, climatic environment during the cold period might not be reflected exactly in the sea level.
