Chemical Modification of Extracellular Cytosine Deaminase from Chromobacterium violaceum YK 391

  • Essential amino acids involved in the catalytic role of the extracellular cytosine deaminase from Chromobacterium violaceum YK 391 were determined by chemical modification studies. The enzyme activity required the reduced form of Fe (II) ion, since the enzyme was inhibited by ο-phenanthroline. The enzyme activity was completely inhibited by the chemical modifiers, such as p-chloromercuribenzoate (p-CMB), p-hydroxymercuribenzoate, and chloramine-T at 1 mM each. The enzyme activity was also markedly inhibited by pyridoxal-5'-phosphate, diethyl pyrocarbonate, and phenylmethylsulfonyl fluroride at 1 mM each. The inactivation of the enzyme activity with p-CMB was reversed by a high concentration of cytosine. Furthermore, the inactivation of the enzyme activity with p-CMB was also reactivated by 1 mM dithiothreitol, 1 mM 2-mercaptoethanol, 1 mM cysteine-HCI, 10% ethyl alcohol, and 10% methyl alcohol. These results suggested that cysteine and methionine residues might be located in or near the active site of the enzyme, while lysine, histidine, and serine residues might be indirectly involved in the enzyme activity.

Purification and Properties of Extracellular Cytosine Deaminase from Chromobacterium violaceum YK 391

  • The extracellular cytosine deaminase (EC 3.5.4.1) from Chromobacterium violaceum YK 391 was purified 264.7-fold with an overall yield of 14.3%. The enzyme was for the first time homogeneous by the criteria of polyacrylamide gel electrophoresis performed in the absence and in the presence of sodium dodecyl sulfate. The molecular weight of the purified enzyme was estimated to be about 156 kDa. The enzyme consisted of two identical subunits of approximate molecular weight 78 kDa. The isoelectric point of the enzyme was pH 5.55. The enzyme had a pH optimum of 7.5 and a temperature optimum of around 40 to $45^{\circ}C$. Besides cytosine, the enzyme deaminated 5-fluorocytosine, cytidine, 5-methylcytosine, and 6-azacytosine, but not 5-azacytosine. The extracellular cytosine deaminase is believed to be unique because it was active not only on cytosine but also on cytidine. The apparent $K_m$ values for cytosine, 5-fluorocytosine, cytidine, and 5-methylcytosine were determined to be 1.55 mM, 5.52 mM, 10.4 mM, and 67.2 mM, respectively. The enzyme activity was strongly inhibited by heavy metal ions such as $Fe^{2+},Pb^{2+},Cd^{2+},Zn^{2+}, Hg^{2+}, and Cu^{2+}$ at 1 mM, and completely by $\alpha,\alpha$'-dipyridyl, and $\rho$-chloromercuribenzoate at 1 mM, and weakly inhibited by 1mM ο-phenanthroline. The enzyme activity was not affected by various nucleosides and nucleotides.

Transconjugation for Molecular Genetic Study of Streptomyces platensis Producing Transglutaminase (Transglutaminase를 생산하는 Streptomyces platensis의 분자생물학적인 연구를 위한 접합 전달법 확립)

  • Streptomyces platensis YK-2, newly isolated from forest soil, produces transglutaminase (TGase), which catalyses an acyl transfer reaction between the primary grade amine and protein or $\gamma$-carboxyamide group of peptide bound glutamine residues. For a molecular genetic study of S. platensis, an effective transformation method was established by using a conjugal transfer of DNA from Escherichia coli to spores of actinomycetes. The highest transconjugation frequency of S. platensis was obtained on an MS medium containing 50 mM $MgCl_2$, using $5{\times}10^7\;E$. coli as a DNA donor and $1{\times}10^8$ spores without heat treatment as a host. We also identified that S. platensis contains a single attB site within an ORF encoding a pirin-homolog, and that its attB site sequence shows high homology to that of S. logisporoflavus. In addition, it was confirmed by phenotypic analyses of exconjugants that the introduction of heterologous DNA into the attB site of the S. platensis chromosome does not affect its morphological differentiation and TGase production.

Characterization of Formaldehyde-degrading Bacteria Isolated from River Sediment (하천 저질에서 분리한 Formaldehyde 분해 미생물의 특징)

  • A bacterium growing on formaldehyde as a sole carbon source was isolated by the dilution method from an enrichment culture containing formaldehyde. The isolated strain, YK-32, was identified as Pseudomonas sp. by morphological, biochemical, and genetic analyses. Pseudomonas sp. YK-32 completely degraded 0.05% formaldehyde within 24 hrs. The isolated strain had a high level of formaldehyde dehydrogenase activity, which is thought to be one of the important factors for formaldehyde degradation, when cells were cultivated in the presence of formaldehyde.

Multiphasic Analysis of Growth Curve of Body Weight in Mice

  • The present study describes the analysis of the multiphasic growth function (MGF) to body weight in laboratory and wild mice. Three genetic groups of laboratory mice (Mus musculus domesticus) designated $CF_{{\sharp}1}$, C3H/HeNCrj and C57BL/6NCrj, and a genetic group of Yonakuni wild mice (Mus musculus molossinus yonakuni, Yk) were used. Mean body weights of each genetic group-sex subclass from birth to 69 days of age taken at 3-day intervals were analyzed by a monophasic, diphasic and triphasic functions for describing growth patterns. A comparison among the three functions of the MGF was based on the goodness-of-fit criteria: residual standard deviation (RSD), adjusted R-square (Adj $R^2$) and Akaike's information criterion (AIC). Result of this study indicated that body weight averaged heavier for males than for females. Among the four genetic groups within both sexes, $CF_{{\sharp}1}$ showed the highest, subsequent followed by C3H/HeNCrj, C57BL/6NCrj and Yk. Comparison among the three functions revealed that the triphasic function was the best fit to growth data, with the lowest RSD, the highest Adj $R^2$ and the lowest AIC, for the four genetic groups. For the triphasic function, RSD within each genetic group-sex subclass was similar for males and females. Adj $R^2$ was 0.999 for all genetic group-sex subclasses. AIC for laboratory mice males and females ranged from -70.48 to 66.50 and from -92.81 to -68.64, respectively; whereas for Yk wild mice males was -74.29 and females -78.42.

  • In this study, Multiple Factor Analysis was undertaken for the purpose of substituting General Vocational Aptitude tester for paper tests according to the standardized and partially modified norm, and compared and analyzed these aptitude tests YK Type Working Character test for a test battery. In this analysis, four basis aptitude cluster of AQE was utilized as aptitude cluster, the study for skill was carried out by the method of sampling electronic aptitude cluster in four basis ones, and the parts needed in the process of its analysis were investigated by means of Video-Tape Recording. This paper was performed with sample test by application of the inverse variation curve from learning theory and induced learning rate as a measure of the degree of proficient of technicians, and from the obtained results illustrated optimum newly-production plan of ability program and load program by the use of computer program.

Analysis of Growth in Intersubspecific Crossing of Mice Using Gompertz Model

  • The aim of this study was to describe growth patterns of mice using Gompertz model. Two distinct types of mice, laboratory mouse $CF_{\sharp1}$ (Mus musculus domesticus) and Yonakuni wild mouse (Yk, Mus musculus molossinus yonakuni) were used. From all possible crosses, there were two parental types and two reciprocal $F_1$ crosses obtained. Individual body weights were measured weekly from birth to ten weeks of age on 321 mice. Standardization to six mice was conducted and only first litters were used. Growth curve parameters were estimated to fit growth data. The results showed that growth among genetic groups were significantly different (p < 0.05) for both sexes, in which parental type of $CF_{\sharp1}$ and Yk had the highest and the smallest values, respectively. Meanwhile, reciprocal $F_1$ crosses were intermediate between parental types. It was concluded that Gompertz model provided and excellent fit for the growth data with a high coefficient determination $(R^2 = 0.999)$.

Quality Characteristics of Adzuki Beans Sediment According to Variety (품종에 따른 팥 앙금의 품질 특성)

  • We evaluated the quality characteristics of adzuki bean sediment according to variety. The moisture, crude protein, and crude ash contents of the various adzuki bean varieties were 8.2~11.1, 15.4~20.6 and 3.3~3.6 g/100 g, respectively. The potassium contents of Chilbo-pat (CB) and Hongeon-pat (HE) were 875.1 and 873.1 mg/100 g, respectively. The calcium contents of Jungbu-pat (JB) and Kumsil-pat (KS)were 73.6 and 73.2 mg/100 g, respectively. A high level of magnesium (131.4 mg/100 g) was found in Yeonkeum-pat (YK). The yields of adzuki bean sediment according to variety were no different either wet (188.3~204.7%) or dry (62.1~66.0%). The L-values on sediment of YK and KS were 67.0 and 68.0, respectively; however, the CB L-value was low at 54.0. A high level of a- (6.6) and b-value (12.8) was found in YK; however, the values for CB were much lower at 3.8 and 5.9, respectively. There was no difference in particle-size distribution, water binding capacity, and solubility of adzuki bean sediment according to variety. High levels of peak (3.79 RVU), trough (3.75 RVU), final (7.33 RVU), and setback viscosity (3.54 RVU) were found in JB. The sensory properties of products in food processing are important, and the variety of adzuki bean sediment should be chosen depending on desired product characteristics.
