Protein Microarrays and Their Applications

  • In recent years, the importance of proteomic works, such as protein expression, detection and identification, has grown in the fields of proteomic and diagnostic research. This is because complete genome sequences of humans, and other organisms, progress as cellular processing and controlling are performed by proteins as well as DNA or RNA. However, conventional I protein analyses are time-consuming; therefore, high throughput protein analysis methods, which allow fast, direct and quantitative detection, are needed. These are so-called protein microarrays or protein chips, which have been developed to fulfill the need for high-throughput protein analyses. Although protein arrays are still in their infancy, technical development in immobilizing proteins in their native conformation on arrays, and the development of more sensitive detection methods, will facilitate the rapid deployment of protein arrays as high-throughput protein assay tools in proteomics and diagnostics. This review summarizes the basic technologies that are needed in the fabrication of protein arrays and their recent applications.

녹두 단백질을 첨가하여 조리한 Angel Parfiet의 특성에 관한 연구 (A Study on the Effect of Mungbean Protein on Quality Characteristics of Angel Parfiet)

  • This study was carried out in order to study the effect of mungbean protein on quality characteristics of angel parfait. The foaming properties of mungbean protein was tested and angel parfait was made with mungbean protein. The results were as follows: 1. Foam expansion values of mungbean protein were generally dependent on protein concentration to 3かio protein suspension. From 1% to 3% suspen-sion, foam expansion values increased. However, over 3% suspension, the values decreased. 2. The foaming stability appeared the greatest value as protein concentration increased. But it was not signifi-cantly different over than 5% concentration. 3. The overrun of angel parfait made with munbean protein was significantly higher than that of made with soybean protein and sensory evaluation data presented that angel parfait made with mungbean protein was significantly higher than that of soybean protein.

한국여성의 단백질 및 칼슘섭취가 칼슘대사에 미치는 영향 (The Effects of Dietary Protein and Calcium Levels on Calcium Metabolism in Young Korean Women)

  • This study was performed to investigate the effects of dietary protein and calcium levels on calcium metabolism in eight healthy Korean adult females. The 2-day metabolic study consisted of a 2 day adaptation period and three 6-day experimental periods. Three experimental diets were low protein low calcium(LPLCa : protein 44g, Ca 422mg), higher protein low calcium(HPLCa : protein 85g, Ca 365mg), and high protein high calcium (HPHCa : protein 84g, Ca 727mg). The apparent calcium absorption was likely to be affected by the calcium intake rather than by the protein intake. Average calcium absorption rate was about 23-29% of calcium intake. The calcium balance was -21.44mg for LPCa, -25.02mg for HPLCa, and -3.22mg for HPHCa. Avergae urinary calcium excretion was 127.7mg for LPLCa, 108.6mg for HPLCa, and 215.4mg for HPHCa. Urinary calcium excretion was more closely related to the changes of calcium intake rather than of protein intake. These results seemed to be due to the interactions between the high phosphours contained in the high protein diet and the little discrepancy of protein intake levels.

Salt-Induced Protein Precipitation in Aqueous Solution: Single and Binary Protein Systems

  • A molecular-thermodynamic model is developed for the salt-induced protein precipitation. The protein molecules interact through four intermolecular potentials. An equation of state is derived based on the statistical mechanical perturbation theory with the modified Chiew's equation for the fluid phase, Young's equation for the solid phase as the reference system and a perturbation based on the protein-protein effective two body potential. The equation of state provides an expression for the chemical potential of the protein. In a single protein system, the phase separation is represented by fluid-fluid equilibria. The precipitation behaviors are simulated with the partition coefficient at various salt concentrations and degree of pre-aggregation effect for the protein particles. In a binary protein system, we regard the system as a fluid-solid phase equilibrium. At equilibrium, we compute the reduced osmotic pressure-composition diagram in the diverse protein size difference and salt concentrations.

PERFORMANCE OF RAINBOW TROUT (Oncorhynchus mykiss) FED ON DIFFERENT DIETARY PROTEIN WITH FIXED ENERGY RATIO

  • An experiment was conducted on rainbow trout(Oncorhynclus mykiss) for eight weeks to investigate the growth performance of the fish fed with different dietary protein with constant diet energy of $20kJg^{-1}$. Four diets containing 25, 30, 35 and 40% crude protein were used. The highest mean final weight was obtained for the fish fed with diet having 35% protein. Growth performance in terms of Specific Growth Rate (SGR), Food Conversion Ratio (FCR) and Protein Efficiency Ratio (PER) were calculated for each diet. There were no significant differences in SGR but the highest value was exhibited by fish fed with 35% protein diet. Significant differences were found among FCR of different diets. Diets with 35 and 40% crude protein gave better FCR value than that of 25 and 30% crude protein. Although significant differences were not found between PER of different diets but PER of diet with 35% protein was found to be better than PER of both high and low protein diets (diets of 40 and 30% crude protein). It is concluded that diet having 35% protein with protein energy ratio of $17.53mgkJ^{-1}$ was suitable for rainbow trout (O. mykiss) among the protein spectrum used.

식이내 Cadmium과 단백질 수준이 흰쥐의 체내 단백질 대사 및 Cadmium 중독에 미치는 영향 (Effects of dietary Cadmium and Protein Levels on the Body Protein Metabolism and Cadmium Toxicity in Growing Rats)

  • This study were performed to investigate effect of dietary cadmium(Cd) and protein levels on growth, body protein metabolism and Cd toxicity in growing rats. Forty eight male rats of Sprague-Dawley weighing 113$\pm$2g were blocked into 6 groups accoridng to body weight. Dietary protein were given at the levels of 7, 15 and 40% of diet and Cd (200ppm)were either added or not. The result obtained were summerized as follow; 1) Food intake, weight gain, FER PER, liver and kidney weight, weight and length of bones, hematocrit, and hemoglobin content in Cd-added groups were low than those in Cd-free groups. 2) Serum total protein showed no significant difference with Cd addition, but it was significantly lower in low protein diet groups. Liver protein in Cd-added groups was lower than Cd-free groups, and was tend to be increased with increasing dietary protein level. 3) Daily urinary and fecal nitrogen excretions in Cd-added groups were lower than Cd-free groups, and were increased with increasing dietary protein level. 4) Cadmium contents in blood, liver, kidney, and femur were tend to be decreased with increasing dietary protein level. Especially, Cd content in kidney of Cd-added groups was significantly decreased with increasing dietary protein level. 5) Daily urinary and fecal Cd excretions were tend to be increased with increasing dietary protein level, and Cd-added-high protein diet group showed the highest Cd excretion among the Cd-added groups, Cd absorption ration and Cd retention ratio were tend to be decreased with increasing dietary protein level.

EFFECTS OF DIETARY CELLULOSE AND PROTEIN LEVELS ON NUTRIENT UTILIZATION IN CHICKENS

  • Effects of dietary cellulose and protein levels on nutrient utilization in chickens were investigated. Four experimental diets containing 5% (low cellulose) or 20% (high cellulose) cellulose in combination with 10% (low protein) or 20% (high protein) protein of 70 g/day were alternatively forced-fed to eight colostomized White Leghorn cockerels once a day to make $4{\times}4$ Latin-square design. The digestibilities of DM and energy decreased with the increase in cellulose level, but not affected by dietary protein level. Ether extract digestibility was higher in the high cellulose diets than in the low cellulose protein level. Ether extract digestibility was higher in the high cellulose diets than in the low cellulose diets. The digestibility of nitrogen free extract had the same trend with the digestibility of DM and energy. The digestibility of acid detergent fiber was not so much different among the diets, but the NDF digestibility was lower in the high cellulose diets than in the low cellulose diets, due to the low hemicellulose digestibility. The true digestibility of protein was influenced by both of the dietary protein and cellulose levels, and their interaction was found. The dietary protein level affected the biological value of protein but the dietary cellulose level did not, and consequently the biological value of protein in the low protein diets was lower than in the high protein diets.

EC-4 세포에 있어서 c-raf Protein Kinase의 면역세포화학적 위치 (Immunocytochemical Localization of c-raf Protein Kinase in EC-4 Cell)

  • Onocogene의 일종인 c-raf protein kinase는 세포질 속에 존재하는 serine / threonine-speccific protein이며, 이것은 mitogene signal에 의해 활성화된다. c-raf protein kinase의 구조와 기능은 protein kinase C와 매우 유사한 것으로 생각된다. 면역세포화학적으로 c-raf protein kinase의 signal transduction을 조사하기 위하여 EC-4 세포에 tumor promotor인 12-0-tet-radecanoylphorbol-13-acetae와 mitogenic gactor인 platelet-derived growth factor로 time-course에 따라서 처리하였다. Translocotion되는 c-raf는 먼저 perinuclear membrane에 모이고 그 후에 핵내로 이동되었다. 그런데 TPA와 PDGF로 처리한 c-raf의 translocotion은 각각의 다른 경로를 가짐을 알 수 있었다. TPA와 PAGF을 장기간 처리하였을 때, c-raf protein kinase의 down regulation이 유도됨을 알 수 있었다.

돼지에서 분리한 Staphylococcus hyicus subsp hyicus의 protein A (Protein A of Staphylococcus hyicus subsp hyicus isolated from pigs)

  • 돼지로부터 분리한 Staphylococcus hyicus subsp hyicus 489주의 protein A 존재여부와 함량을 indirect hemagglutination 및 enzyme-linked immunosorbent assay(ELISA)법으로 조사하였다. Indirect hemagglutination text에 의하여 cell-bound protein A 및 extracellular protein A 보유균은 489주 중 각각 87.7% 및 36.0%로 나타났다. ELISA법에 의한 이들 균의 protein A함량 측정에서 전균주의 extracellular protein A는 1ng/ml미만이었으며, cell-bound protein A함량은 대부분의 균주에서 1ng/ml 미만이었고 11주가 25~108ng/ml 수준이었다.

한국여성의 단백질 섭취수준과 동.식물성 급원이 체내질소 대사에 미치는 영향 (The Effect of Dietary protein Levels and Sources from Animals or Plants on Nitrogen metabolism of Korean Women)

  • To evaluate the differences of the levels and sources of protein intake human protein metabolism, an 26-day metabolic balance study was conducted in 10 healthy Korean adult females. In the pre-study, the subjects recorded their own diets for 3 days. The metabolic balance study consisted of 6-day adaptation period, 10-day moderate protein period(60-65g/d) and 10-day high protein period(90-95g/d). During the moderate and high protein period, 5 subjects were fed the higher animal protein meals and the other 5 subjects were fed the high plant protein meals. Body weight, nitrogen balance and blood chemistries were monitored through out the study. The urine volume were sighificantly larger in the animal protein group and, the dietary fiber and fecal weights were significantly heavier in the plant protein diet group. But no statistically significant differences were found between the two dietary groups in apparent nitrogen digestability, urinary nitrogen excretion and nitrogen balance. Body weight, serum protein, albumin and HDL-cholesterol levels were not changed, but serum total cholesterol level in the animal protein diet group was elevated significantly from 143.8mg/dl on moderate potein diet to 173.0mg/dl on high proetin diet. In conclusion, from the observation of this short-term N balance study, plant diet on the adequate level of calorie and protein intake had almost the same effect of animal protein diet for protein maintenace in adults.
