High Level Expression of XMP Aminase Gene in Esherichia coli (Esherichia coli XMP Aminase 유전자의 발현 증대)

  • In order to increase the expression of XMP aminase [EC 6.3.4.1], which catalizes the conversion of 5'-XMP to the DNA fragment containing gua A gene coding for XMP aminase from pLC 34-10 plasmid was subcloned into pBR 322, and 1.7 kb gua A gene fragment was recloned under the control of trp promoter of pDR 720, E. coli expression vector. XMP aminase activity had increased by about 17 times when compared with that of the strain earring pLC 34-10.

  • For the enzymatic conversion of 5'-XMP to 5'-GMP, partially purified XMP aminase from Escherichia coli was coupled with the yeast, Saccharomycrs cerevisiae, capable of ATP regeneration through glycolytic pathway. In order to elevate the level of XMP aminase in E. coli, $guaB^{-}(IMP\;dehydrogenase-less)$ mutant were introduced, and the yeast used as ATP supplier was treated by some method to increase its membrane permeability. The optimum conditions for efficient conversion reaction by energy-coupled system were investigated. As the results, a CH 41, $guaB^-$ mutant of E. coli K-12, showed 2.75 fold increase in the level of XMP aminase, compared with its parent cell. And the lyophylized yeast was the most effective at the ATP supplier. The optimum temperature and pH of conversion reaction were $40{\circ]C$ and pH 7.4, and the highest conversion ratio was shown under the reaction condition of 100 mM glucose, 100 mM inorganic phosphate and 6 mM AMP. When 36 units/ml XMP aminase used under the above conditions, the amount of 60 mg/ml yeast was sufficient to be used. Under the optimum condition, 71% of 1.8 mM(65.6 mg/100 ml) 5'-XMP was converted to 5'-GMP within 8 hr.

Accumulation of Xanthosine-5'-monophosphate by Adenine-Guanine Double Auxotroph of Brevibacterium ammoniagenes (Brevibacterium ammoniagenes 의 아데닌-구아닌 복영양요구주에(複營養要求株)에 의한 5'-크산틸산(酸)의 축적(蓄積))

  • An adenine-guanine doubless and $\beta$-alanine requiring mutant, D-1550-40, which had been derived from Brevibacterium ammoniagenes ATCC 6872, produced a copious amount of xanthosine-5'-monophosphate (XMP). The optimum concentration of adenine and guanine for maximal accumulation of XMP was about 75 ml/l and 100 ml/l for growth. Concentrations higher than 100 mg/l of adenine and guanine inhibited cell growth and XMP accumulation strongly. The inhibition, however, could be recovered by adding $100{\mu}g$ of biotin per liter or 0.3% of casamino acids to the culture solution. High concentrations of phosphate and magnesium salts (1.0 to 1.5%(w/v) in media) were found to be indispensable for XMP accumulation, and the presence of manganese in the culture medium stimulated both growth of cells and accumulation of XMP leaving 5'-inosinic acid unaffected. The maximal accumulation of XMP reached to 60.5 mg/l after 4 days of fermentation which had been started with a medium containing 100 mg of adenine-guanine, 5 mg of $MnSO_4{\cdot}H_2O$ and $100{\mu}g$ of biotin per liter. The specific XMP synthesis(mg of XMP/mg of cells) was increased with the increase of the cell growth rate.

  • By the treatment of several mutagens, a number of 5'-guanylic acid producing mutants from 5'-xan-thylic acid were obtained from Brevibacterium ammoniagenes ATCC 6871. The indispensensable genetic-characters of the mutants were adenine requirement, lack of GMP-reductase and mutation to adenosine resistance from adenosine sensitiveness. Main product from 5'-xanthylic acirl by strain BA-17-2 was 5'-guanulic acid, and was isolated in a crystalline form by the use of anion exchange resin, Duolite 102 D. The isolated crystalline was identified as 5'-guanylic acid by means of paper chromatography, ultrav-iolet absorption spectra, and infrared absorption spectrum.

  • Ribonucleic acid contents and mononucleotides distribution from the mycelium and fruit bodies of Ganoderma lucidum were studied. P.E.I. cellulose TLC and HPLC were applied in this study. The obtained results are as follows; The levels of ribonucleic acids from the young basidiocarp mycelium were higher than those of mature basidiocarp. Guanosine 5'-monophosphate and xanthosine 5'-monophosphate were found in both young basidiocarp mycelium and mature basidiocarp. The levels of guanosine 5'-monophosphate and xanthosine 5'-monophosphate from the young basidiocarp were higher than those of the mature basidiocarp. However, inosine 5'-mono­phosphate was not detected.

  • 사용자 참여, 개방, 공유를 기반으로 하는 웹2.0 환경 하에서 보다 효율적이고 합법적으로 UCC(User Created Contents)가 제작, 공유, 유통되기 위한 저작권 시스템으로서, CCL(Creative Commons License)이 빠르게 확산되고 있다. 그러나, UCC에 CCL메타데이터를 직접 첨부하지 않는 기존의 CCL어노테이션 방식은 의도적으로 또는 비의도적으로 UCC와 CCL메타데이터가 분리될 수 있는 문제점을 가지고 있다. 그 결과, UCC로부터 분리되어진 저작권관련정보는 변경 또는 분실되어 추후에 저작권분쟁의 원인이 될 수 있다. 본 연구에서는 RDF와 XMP를 기반으로 CCL메타데이터를 이미지 컨텐츠에 직접 부착하는 방법을 제안하고 이를 지원하기 위한 어노테이션 지원도구(CCL-MASTer)를 개발하였다. 본 연구결과는, 기존의 어노테이션 기법의 문제점을 해결하고 다양하고 풍부한 CCL메타데이터의 효율적인 어노테이션을 가능케 하여 UCC 저작권분쟁 해결방안 및 UCC와 UCC관련 CCL메타데이터의 생성과 검색 등의 토대가 된다.

  • Growth responses of Brevibadterium ammoniagenes BA 17-2, which had been obtained by the treatment of several mutagens in our previous report, were investigated to select the preliminary optimal concentrations of phosphate, $Mg^{++}$, $Mn^{++}$ and thiamine for the production of 5'-XMP aminase. In this experiment it was shown that the concentration of phosphate in the medium has an important effect on the growth of microorganism. Using the medium containing 0.2% of $MgSO_4{\cdot}7H_2O$, 3mg/l of $MnSO_4{\cdot}H_2O$and $1\;mg/l$ of thiamine-HCl, the maximum cell mass was obtained at the concentration of 0.4% of $KH_2PO_4$ and $K_2HPO_4$, respectively. Above the concentration of these phosphates, cell growth was inhibited as the phosphate concentration increased to 1%, but the inhibition was overcome by the addition of 1% of $MgSO_4{\cdot}7H_2O$ and 3mg/l of thiamine-HCl. The 5'-XMP aminase activity was also influenced by the concentration of phosphate, $Mg^{++}$, $Mn^{++}$, and thiamine. In addition, the optimal culture pH and temperature for the enzyme activity were found to be 6.8 and $32^{\circ}C$, respectively.

On Developing a Semantic Annotation Tool for Managing Metadata of Web Documents based on XMP and Ontology (웹 문서의 메타데이터 관리를 위한 XMP 및 온톨로지 기반의 시맨틱 어노테이션 지원도구 개발)

  • The goal of Semantic Web is to provide efficient and effective semantic search and web services based on the machine-processable semantic information of web resources. Therefore, the process of creating and adding computer-understandable metadata for a variety of web contents, namely, semantic annotation is one of the fundamental technologies for the semantic web. Recently, in order to manage annotation metadata, direct approach for embedding metadata into the document is mainly used in semantic annotation. However, many semantic annotation tools for web documents have been mainly worked with HTML documents, and most of these tools do not support semantic search functionalities using the metadata. In this paper, based on these problems and previous works, we propose the Ontology-based Semantic Annotation tool(OSA) to efficiently support semantic annotation for web documents(such as HTML, PDF). We define a semantic annotation model that represents ontological-semantic information by using RDFS(RDF Schema). Based on XMP(eXtensible Metadata Platform) standard, the model is encoded directly into the document. By using OSA with XMP, user can perform semantic annotation on web documents which are able to keep compatibility for managing annotation metadata. Eventually, the integrated semantic annotation metadata can be used effectively in semantic search for a variety of web contents.

Design of XMP-Based Electronic Document Architecture for Electronic Circulation of Litigation Documents (소송문서의 전자적 유통을 위한 XMP 기반 전자문서 구조 설계)

  • It has become commonplace to use web-based business process systems in a variety of fields, and electronic litigation is not an exception. In electronic procedures where court records are at the core of the system, the electronic document architecture should be designed in a way that electronic documents are safely circulated and utilized on the web with a consideration of the authentication of records, particularity of cases and document security such as prevention of forgery or falsification. Based on a study of electronic formats suitable for court records, the Extensible Markup Language (XMP) for management of special case information and security requirements for circulation of electronic documents, this paper suggests an adequate architecture for electronic documents designed for electronic litigation involving constitutional matters and looks into cases where such architectures are applied. The studies in this paper will serve as a useful reference for those planning to realize web-based business process that enables exchanges of electronic documents.

Quantitative determination of inosine 5'-monophosphate dehydrogenase activity in human peripheral blood mononuclear cells by ion-pair reversed-phase high-performance liquid chromatography (이온쌍 역상 HPLC를 이용한 인체 말초혈액단핵구에서 이노신 5'-일인산 탈수소효소 활성의 정량적 측정)

  • A quantitative analytical method has been established for the measurement of inosine 5'-monophosphate dehydrogenase (IMPDH) activity in human peripheral blood mononuclear cells (PBMCs) by ion-pair reversed-phase high performance liquid chromatography equipped with ultraviolet detection (HPLC/UV). IMPDH is a ${\beta}$-nicotinamide adenine dinucleotide hydrate (NAD+)-dependent dehydrogenase in which the enzyme converts inosine 5'-monophosphate (IMP) into xanthosine 5'-monophosphate (XMP). Its activity was measured by quantifying a HPLC chromatogram corresponding to XMP produced during the incubation of lysed PBMCs with IMP as a substrate and $NAD^+$ as a coenzyme. XMP produced was detected at a wavelength of 260 nm. The mobile phase was composed of a mixture of 37 mM potassium dihydrogen phosphate containing 7 mM tetra-n-butylammonium hydrogen sulfate adjusted to pH 5.5 and methanol (85:15, v/v) with a flow rate of 1 mL/min. The calibration curve was linear ($r^2$=0.999999) in the range of $0.2-50.0\;{\mu}M$ and the limit of quantification (LOQ) was $0.2\;{\mu}M$. The intra- and inter-day precisions were between 0.88-1.47% and 0.85-5.24%, respectively. The intra- and inter-day accuracies were between 98.74-99.99% and 99.95-101.65%, respectively. IMPDH activity in 11 Korean healthy volunteers ranged from 18.29 to 36.60 nmol/h/mg protein (mean = $27.70{\pm}6.28\;nmol/h/mg$ protein).
