본 연구의 목적은 누에형질전환체를 이용하여 재조합단백질 대량생산 시스템을 개발하는 것으로서, 본 실험에서는 hSCF유전자를 이용하여 누에에서 재조합단백질을 생산하였다. 실험에 사용된 piggyBac 전이벡터는 hSCF 유전자의 발현 조절을 위해 초파리 유래의 dHsp70 promoter를 사용하였고, EGFP marker유전자는 3xP3 promoter로 발현을 조절하였다. 총 1,020 개의 누에알에 microinjection 하여 G1 세대에서 22 bloods의 형질전환체를 선발하였고, 선발된 누에형질전환체는 초기배 단계의 눈과 신경조직, 유충과 번데기 그리고 성충의 눈에서 GFP 형광을 관찰 할 수 있었다. hSCF 재조합단백질의 발현은 Western blot 분석으로 확인 할 수 있었고, inverse PCR 분석을 통해서 누에 게놈에 전이벡터가 삽입된 것을 확인할 수 있었다. 지금까지의 실험 결과에서 hSCF 재조합 단백질이 누에에서 생산될 수 있음을 확인 할 수 있었다. 비록 누에에서 생산된 hSCF 재조합단백질의 생리활성에 대한 실험이 추후에 요구되지만, 이러한 실험결과는 piggyBac 전이벡터와 microinjection 법으로 누에에서 고부가가치의 재조합단백질을 대량생산 할 수 있음을 보여 주었다고 할 수 있겠다. 따라서 누에를 유용물질 생산을 위한 생체반응기로서 활용할 수 있을 것으로 기대된다.
본 연구는 사이버나이프 Synchrony 호흡추적 장치를 이용하여 방사선 수술을 시행한 폐종양 환자 48명을 대상으로 전 치료기간 중 종양의 움직임과 방사선수술의 정확성을 평가하였다. 폐종양의 움직임은 종양이나 종양주변에 삽입된 금침의 좌표를 사이버나이프 영상유도 장치로 측정하였으며, 방사선수술의 정확성은 움직임 추적 컴퓨터(MTS)로 계산된 상관관계 오차로 평가하였다. 폐종양의 움직임은 두미방향으로 평균 $2.63{\pm}1.87\;mm$며, 좌우방향 $1.13{\pm}0.71\;mm$, 전후방향 $1.74{\pm}1.16\;mm$의 움직임을 보였으며, 회전 움직임 정도는 X축 $1.66{\pm}1.66^{\circ}$, Y축 $1.20{\pm}0.97^{\circ}$, Z축 $1.18{\pm}0.73^{\circ}$로 측정되었다. 직선 움직임의 벡터 값은 평균 $3.78{\pm}2.00\;mm$값을 나타냈다. 연구 결과에서 두미방향(p<0.001)과 전후방향(p<0.029), 3차원 벡터 값(p<0.002)들은 종양의 위치가 상부보다 하부의 움직임이 크게 나타나 통계적 유의성을 보였다. 사이버나이프 Synchrony 호흡추적 장치를 이용한 폐종양의 방사선 수술시 상관관계 오차는 전체 평균 $0.95{\pm}0.62\;mm$로 매우 정확한 조사로 종양의 움직임을 보상하여 방사선 수술이 이루어졌으며 그 유용성을 확인할 수 있었다.
Background and Objectives: The Herpes Simplex type 2 Defective Infectious Single Cycle virus (DISC virus) is attenuated virus originally produced as viral vaccines but are also efficient gene transfer vehicle. The main goals of this study were to examine the efficiencies of the gene transfer using DISC vectors for various head and neck squamous cell carcinoma cell lines and to evaluate the efficacy of vaccination with DISC virus carrying a immunomodulatory genes (GM-CSF) as cancer therapy in a organotopic oral cavity squamous cell cancer model. Materials and Methods : We determinated the gene transfer efficiency of DISC virus by x-gal stain method and proved gene and protein expression of DISC-GMCSF transfected SCCVII cells by RT-PCR and ELISA method. Also we evaluated the ex vivo vaccination effects of SCCVII/GMCSF (DISC-GMCSF transfected SCCVII vaccine) vaccine on preventing the recurrence of micro-residual tumor. After the vaccination of SCCVII/GMCSF, specific cytotoxic T-cell responses was evaluated by CTL assay. Results: At an MOI of 10 DISC virus showed 64-88% of transfection rates in various head and neck squamous cancer cell lines. SCCVII cells transduced by DISC virus vector (MOI=10) carrying the GM-CSF gene, produced 4.5 nanogram quantities of GM-CSF per $10^6$ cells. In vivo vaccination using tumor cells transduced ex vivo with DISC-GMCSF resulted in better protection rate against subsequent tumor recurrence in organotopic oral cavity cancer model. Although tumor free survival rate was not statistically significantly increased in vaccination group (p=0.078), tumor specific cytotocic T-cell responses were significantly increased in SCCVII/GMCSF vaccination group. Conclusion: These data demonstrate that; 1) The DISC virus vector is capable of efficient gene transfer to various head and neck squamous cancer cell lines, 2) GM-CSF secreting genetically modified tumor vaccine (SCCVII/GMCSF) efficiently protected against tumor recurrence in organotopic micro-residual oral cavity cancer model and produced tumor specific cytotoxic T-cell response. DISC virus-mediated, cytokine gene transfer may prove to be useful as a clinical therapy for head and neck cancers.
In this study, we have cloned a novel cDNA encoding for a papain-family cysteine protease from the Uni-ZAP XR cDNA library of the polychaete, Periserrula leucophryna. This gene was expressed in Escherichia coli using the T7 promoter system, and the protease was characterized after partial purification. First, the partial DNA fragment (498 bp) was amplified from the total RNA via RT-PCR using degenerated primers derived from the conserved region of cysteine protease. The full-length cDNA of cysteine protease (PLCP) was prepared via the screening of the Uni-ZAP XR cDNA library using the $^{32}P-labeled$ partial DNA fragment. As a result, the PLCP gene was determined to consist of a 2591 bp nucleotide sequence (CDS: 173-1024 bp) which encodes for a 283-amino acid polypeptide, which is itself composed of an 59-residue signal sequence, a 6-residue propeptide, a 218-residue mature protein, and a long 3'-noncoding region encompassing 1564 bp. The predicted molecular weights of the preproprotein and the mature protein were calculated as 31.8 kDa and 25 kDa, respectively. The results of sequence analysis and alignment revealed a significant degree of sequence similarity with other eukaryotic cysteine proteases, including the conserved catalytic triad of the $Cys^{90},\;His^{226},\;and\;Asn^{250}$ residues which characterize the C1 family of papain-like cysteine protease. The nucleotide and amino acid sequences of the novel gene were deposited into the GenBank database under the accession numbers, AY390282 and AAR27011, respectively. The results of Northern blot analysis revealed the 2.5 kb size of the transcript and ubiquitous expression throughout the entirety of the body, head, gut, and skin, which suggested that the PLCP may be grouped within the cathepsin F-like proteases. The region encoding for the mature form of the protease was then subcloned into the pT7-7 expression vector following PCR amplification using the designed primers, including the initiation and termination codons. The recombinant cysteine proteases were generated in a range of 6.3 % to 12.5 % of the total cell proteins in the E. coli BL21(DE3) strain for 8 transformants. The results of SDS-PAGE and Western blot analysis indicated that a cysteine protease of approximately 25 kDa (mature form) was generated. The optimal pH and temperature of the enzyme were determined to be approximately 9.5 and $35^{\circ}C$, respectively, thereby indicating that the cysteine protease is a member of the alkaline protease group. The evaluation of substrate specificity indicated that the purified protease was more active towards Arg-X or Lys-X and did not efficiently cleave the substrates with non-polar amino acids at the P1 site. The PLCP evidenced fibrinolytic activity on the plasminogen-free fibrin plate test.
본 연구는 Web기반의 CAD를 이용한 지리정보시스템으로 시스템 설계자, 운영자, 이용자에게 보다 편리하고 다양한 기능을 제공하기 위해 이루어 졌다. 시스템 구현은 공간자료를 이용하여 HTML, Java Script, ASP, Whip ActiveX Control로 사용자 인터페이스를 구현하였으며 다음과 같은 특징을 갖는다. 첫째, 시스템 설계자는 기존의 Web과 CAD에 대한 기초적인 지식만 있으면 구축이 가능하도록 하였다. 이를 위해 시스템구조는 2계층(2-Tier)으로 단순화하였다. 위치정보는 DWF를 이용한 파일시스템으로 구축하였으며, 속성정보는 확장성을 고려하여 DBMS를 이용하였다. 둘째, 시스템 운영자에게는 고가의 공간엔진을 구입하지 않고도 기존의 Web 방식만으로도 지리정보시스템을 독립적으로 구동할 수 있도록 함으로서 경제성을 갖도록 하였다. 셋째, 인터넷 이용자에게는 고차원적 GIS 기능은 없애고 표현과 검색 기능 중심으로 간략 명료하게 설계하였다. 주요기능은 정보검색, 지도조작, 인쇄 기능 등으로 하며, 필요에 따라 손쉽게 기능추가가 가능하도록 하였다. 이상과 같이 구현된 시스템은 기존 문자나 이미지 중심 방식보다 벡터 방식을 이용하므로 용량이 적을 뿐만 아니라 속도가 매우 빠르다. 또한, 이러한 시스템은 도시의 주요 상권 관광 홍보에 이용할 수 있을 뿐만 아니라 지자체에서 보유하고 있는 벡터지도를 이용할 경우 적은 비용으로 손쉽게 시스템을 구현할 수 있다. 또한, 민간 인터넷 시장에도 적용할 수 있을 것이다.
포노닉 크리스탈이란 기저물질 내에 주기적으로 배열된 산란체로 구성된 복합물질로서 포노닉 크리스탈에 입사된 음파가 특정 주파수 대역에서 차단되는 현상인 밴드 갭이라는 중요한 특성을 갖는다. 본 연구에서는 수중에서 산란체로서 1 mm의 직경을 갖는 원기둥 형태의 스테인리스 스틸 막대가 1.5 mm의 격자상수를 가지며 정방형으로 배열된 2차원 포노닉 크리스탈의 음향 밴드 구조를 이론 및 실험적으로 고찰하였다. 2차원 포노닉 크리스탈의 밴드 구조를 예측하기 위해 유한요소법을 이용하여 첫째 브릴루앙 영역의 ${\Gamma}X$ 방향에 대해 주파수와 파동벡터에 대한 분산관계를 계산하였다. 초음파가 입사되는 방향과 수직한 스테인리스 스틸 막대 층의 개수를 1, 3, 5, 7, 9개로 변화시켜가며 투과계수 및 반사계수를 측정하였다. 계산된 분산관계로부터 2 MHz 이하의 주파수 대역에서 5개의 밴드 갭이 존재하는 것으로 예측되었으며, 첫째 밴드 갭은 0.5 MHz를 중심으로 나타났다. 투과계수 및 반사계수로부터 실험적으로 확인된 밴드 갭은 분산관계로부터 예측된 밴드 갭과 잘 일치하는 것으로 나타났다.
Hyaluronidases are a family of enzymes that catalyse the breakdown of hyaluronic acid, which is abundant in the extracellular matrix and cumulus oocyte complex. To investigate the activity of recombinant bovine sperm hyaluronidase 1 (SPAM1) and determine the effect of the Asn-X-Ser/Thr motif on its activity, the bovine SPAM1 open reading frame was cloned into the mammalian expression vector pCXN2 and then transfected to the HEK293 cell line. Expression of recombinant bovine hyaluronidase was estimated using a hyaluronidase activity assay with gel electrophoresis. Recombinant hyaluronidase could resolve highly polymeric hyaluronic acid and also caused dispersal of the cumulus cell layer. Comparative analysis with respect to enzyme activity was carried out for the glycosylated and deglycosylated bovine sperm hyaluronidase by N-glycosidase F treatment. Finally, mutagenesis analysis revealed that among the five potential N-linked glycosylation sites, only three contributed to significant inhibition of hyaluronic activity. Recombinant bovine SPAM1 has hyaluronan degradation and cumulus oocyte complex dispersion ability, and the N-linked oligosaccharides are important for enzyme activity, providing a foundation for the commercialization of hyaluronidase.
본 연구의 목적은 누에형질전환 기술을 이용하여 녹색형광실크를 개발하는 것으로서, 본 실험에서는 피브로인 H-chain의 N-말단과 C-말단을 이용하여 피브로인 재조합 단백질 발현 시스템을 제작하였고, 종결코돈이 없는 EGFP 유전자를 위의 발현 시스템에 클로닝하여 녹색형광실크를 제작하였다. 누에형질전환체 선발을 위해서는 3xP3 promoter와 DsRed2를 이용하여 선발하였고, 1200 개의 누에알에 microinjection 하여 F1 세대에서 8 bloods의 형질전환체를 선발하였다. 선발된 누에형질전환체는 초기배 단계의 눈과 신경조직, 유충과 번데기 그리고 성충의 눈에서 DsRed2 형광단백질이 발현되는 것을 확인 할 수 있었다. 또한 실크의 피브로인에서 EGFP 단백질이 발현되는 것을 확인하기 위해, F2세대의 누에형질전환체 중에서 5령 3일 유충을 해부하여 견사선을 형광현미경으로 관찰하였고, 중부와 후부 견사선에서 형광단백질이 발현되는 것을 확인할 수 있었다. 그리고 F2 세대의 고치와 저온에서 정련한 실크에서도 녹색형광단백질의 발현을 확인할 수 있었고, Western blot 분석에서도 EGFP 재조합 단백질이 피브로인 H-chain과 융합된 형태로 존재하는 것이 확인되었다. 이상의 결과에서 녹색형광실크를 생산하는 누에형질전환체가 성공적으로 제작 되었음을 확인할 수 있었고 이러한 결과를 토대로 새로운 산업소재로서 실크를 활용할 수 있을 것으로 기대한다.

Anthocyanidin synthase (ANS, leucoanthocyanidin oxygenase), a 2-oxoglutarate iron-dependent oxygenase, catalyzed the penultimate step in the biosynthesis of the anthocyanin class of flavonoids, from the colorless leucoanthocyanidins to the colored anthocyanidins. The full-length cDNA and genomic DNA sequences of ANS gene (designated as GbANS) were isolated from Ginkgo biloba for the first time. The full-length cDNA of GbANS contained a 1062-bp open reading frame (ORF) encoding a 354-amino-acid protein. The genomic DNA analysis showed that GbANS gene had three exons and two introns. The deduced GbANS protein showed high identities to other plant ANSs. The conserved amino acids (H-X-D) ligating ferrous iron and residues (R-X-S) participating in 2-oxoglutarate binding were found in GbANS at the similar positions like other ANSs. Southern blot analysis indicated that GbANS belonged to a multi-gene family. The expression analysis by real-time PCR showed that GbANS expressed in a tissue-specific manner in G. biloba. GbANS was also found to be up-regulated by all of the six tested abiotic stresses, UV-B, abscisic acid, sucrose, salicylic acid, cold and ethylene, consistent with the promoter region analysis of GbANS. The recombinant protein was successfully expressed in E. coli strain with pET-28a vector. The in vitro enzyme activity assay by HPLC indicated that recombinant GbANS protein could catalyze the formation the cyanidin from leucocyanidin and conversion of dihydroquercetin to quercetin, suggesting GbANS is a bifunctional enzyme within the anthocyanidin and flavonol biosynthetic pathway.
c-myc proto-oncogene은 여러 세포들의 분화와 형질전화에 뿐만 아니라 정상세포의 분열조절에도 관여한다고 알려져왔다. 특히 생쥐의 초기배아에서 c-myc mRNA가 발현되고 antisense c-myc oligomer의 미세주입에 의해 배발생이 억제된다는 연구결과는 c-myc이 초기배아의 발생 및 분열에 관여하는 것을 시사한다. 그러나 최근까지 초기배아에 존재하는 c-myc promoter의 기능적 활성화에 관한 연구는 미진하였다. 이를 위하여, c-myc promoter와 대장균의 lacZ 유전자를 결합시킨 두 종류의 vector(pcmyc-Gall, pcmyc-Ga12)를 만들어 수정란의 전핵에 미세주입한 후, 배 발생에 따른 c-myc promoter의 활성화를 lacZ 유전자의 산물인 $\beta$-galactosidase 에 의한 X-gal 염색으로 조사하였다. 미세주입된 초기 배아는 2세포기 배아를 포함하는 여러 발생단계에서 $\beta$-galactosidase 의 활성을 보였다. 이는 c-myc 유전자가 배아의 게놈유전자로부터 발현되며, 또한 궁극적으로 초기 배아의 발생과정에 중요한 역할을 하고 있음을 시사하고 있다.

① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
