Crystallization and Preliminary Crystallographic Study of 26kDa Clonorchis sinensis glutathione S-transferase Complexed with Inhibitors

  • A helminth glutathione S-transfease, 26 kDa isozyme from Clonorchis sinensis was cocrystallized with inhibitors by the hanging-drop method with monomethylether 550 as a precipitant. The crystals, grown in the presence of S-methylglutathione or trans-4-phenyl-3-buten-2-one, suitable for X-ray analysis, belong to the hexagonal space group P6₂(or P6₄), with unit cell parameters a= b= 97.6Å, c=117.4Å. X-ray diffraction data were collected with 2.5Å resolution.

  • Metal oxide nanostructures have been applied to various fields such as energy, catalysts and electronics. We have freely designed one and two-dimensional (1 and 2-D) metal (transition metals and lanthanides) oxide nanostructures, characterized them using various techniques including scanning electron microscopy, transmission electron microscopy, X-ray diffraction crystallography, thermogravimetric analysis, FT-IR, UV-visible-NIR absorption, Raman, photoluminescence, X-ray photoelectron spectroscopy, and temperature-programmed thermal desorption (reaction) mass spectrometry. In addition, Ag- and Au-doped metal oxides will be discussed in this talk.

  • Reaction between 2-pyridinecarboxylic acid (Hpic) and $K_3[Cr(O_2)_4]$ give complex $[Cr(pic)_3].H_2O$ (1) which is characterized by elemental analysis and spectroscopic methods (FT-IR, Raman) and X-ray crystallography. In the crystal structure of 1, chromium atom with coordinated by three nitrogen and three oxygen atoms has a distorted octahedral geometry. Also a water molecule is incorporated in crystal network. Each water molecule acts as hydrogen bond bridging and connects two adjacent complexes by two $O-H{\cdots}O$ hydrogen bonds.

Crystal structure analysis of orthohombic $Sr_{0.6}Ca_{0.4}CuO_2$ compound (사방정계 $Sr_{0.6}Ca_{0.4}CuO_2$ 화합물의 결정구조해석)

  • Sr0.6Ca0.4CuO2 single crystal has been synthesized by flux method and characterized by the single crystal X-ray diffraction. The compound has the orthorhombic system and the space group is Cmcm(63), lattice parameters are a=3.4645Å, b=16.1417Å, c=3.8727Å. In the (Sr1-xCax)CuO2 compound the limit of Ca from substitution for Sr was determined by the change of bond length. For this, X-ray diffraction, scanning electron microscopy (SEM), energy dispersive X-ray fluorescence (EDAX) and electron probe micro-analysis (EPMA) were used. From the change of Cu-O bond length as the Ca substitution, we concluded the limit of Ca incorporation Xca≒0.73.

  • Reduced glutathione (GSH) plays a vital role in the metabolism of all cells. Glutathions, a tripeptide cowfosed of glutamic acid, cysteane, and gtycina is synthesized by two synthesized reutions. The first is catalyzed by Y-glutamylcysteine synthetase (GSH-I) and the second by glutathione synthetase (GSH-ll). The glutathione biosynthetic pathway of E. coziis mainly controlled by nonallosteric feedback inhibition of GHS-I by GSH. Determination of the three-dimensional structure of GSH-I by X-ray crystallography is necessary in order to understand the structure-function relationship at the molecular level. As the (irst step toward its structure determination, crystallization of 5. coli V-glutamylcystfine synthetase (GSH-I) has been achived using the hanging drop vapor diffusion method and capillaw method. Crystals of GSH-I have been grown from ammonium sulfate solution. The crystals grew at room temperature within 10 days to dimensions of 0.2 m x 0.2 m x 0.2 ml by hanging drop vapor diffusion method and diffracted to about 4 A resolution using synchrotron X-rays. Another crystal, grown by the capillary method to dimensions of 0.25 mm x 0.25 mm x 0.3 mm within 40 days, diffracted to about 4 A resolution using X-rays from a rotating anode.

  • We have investigated bulk and hetero-epitaxial growth of GaAs single crystal. Various growth techniques such as HB, HZM, and VGF for high quality bulk GaAs were successfully developed by appling the specially designed DM(direct monitoring) furnace. Al GaAs/GaAs superlattice structure and In(x)Ga(1-x) As/GaAs epilayers were also grown by MOCVD and VPE, respectively. The characterization of GaAs single crystals and epilayers was made by X-ray diffraction, Hall effect, PL, chemical etching and angle lapping technique.

  • A platinum(II) pincer complex composed of two six-membered fused metallacycles was directly synthesized using 1,3-bis(2-pyridyloxy)benzene and $K_2PtCl_4$. The structure of the complex was elucidated via NMR and X-ray crystallography analysis. The stable complex was formed due to the six-membered fused cycle structure around the Pt(II) center which reduced the bond angle strain. The complex was applied to the silylcyanation reaction of aldehydes and imines and showed an efficient catalytic activity with 99% yield.

  • Since pathogens resistant against vancomycin occur rapidly, the development of a new drug is needed. To make a new drug based on a rational drug design, the structural study of vancomycin is necessary. Accordingly, this study reports on a comparison of the solution structure of vancomycin determined by NMR spectroscopy, which was performed in the present work, with the X-ray crystallographic structure previously deposited in the Protein Data Bank (PDB).

  • The knowledge on the number of necessary and collectable reflections for the crystal structure analysis is greatly helpful in choosing the conditions for X-ray intensity data collection using automatic four circle diffractometer. In this paper, we represent a method to calculate the total unmber of collectable intensity date in an asymmetric unit.

  • The three stereoisomers of the 6,13-bis(thymidinyl)dioxocyclam 6 were synthesized through photoreaction of the chromium alkoxycarbene complex 2 and 1-(benzyloxycarbonyl)-4,4-dimethyl-D 2 -imidazoline. The molecular structure of (R,S)-6 was elucidated by X-ray crystallography.
