In addition to its typical use for skin rejuvenation, fractional laser irradiation of early cancerous lesions may reduce the risk of tumor development as a byproduct of wound healing in the stroma after the controlled injury. While fractional ablative lasers are commonly used for cosmetic/aesthetic purposes (e.g., photorejuvenation, hair removal, and scar reduction), we propose a novel use of such laser treatments as a stromal treatment to delay tumorigenesis and suppress carcinogenesis. In this study, we found that non-ablative fractional laser (NAFL) irradiation may have a possible suppressive effect on early tumor growth in syngeneic mouse tumor models. We included two syngeneic mouse tumor models in irradiation groups and control groups. In the irradiation group, a thulium fiber based NAFL at 1927 nm was used to irradiate the skin area including the tumor injection region with 70 mJ/spot, while no laser irradiation was applied to the control group. Numerical simulation with the same experimental condition showed that thermal damage was confined only to the irradiation spots, sparing the adjacent tissue area. The irradiation groups of both tumor models showed smaller tumor volumes than the control group at an early tumor growth stage. We also detected elevated inflammatory cytokine levels a day after the NAFL irradiation. NAFL treatment of the stromal tissue could potentially be an alternative anticancer therapeutic modality for early tumorigenesis in a minimally invasive manner.
Ubiquitin D (UBD) is highly upregulated in many cancers, and plays a pivotal role in the pathophysiological processes of cancers. However, its roles and underlying mechanisms in oral squamous cell carcinoma (OSCC) are still unclear. In the present study, we investigated the role of UBD in patients with OSCC. Quantitative real-time polymerase chain reaction and Western blot were used to measure the expression of UBD in OSCC tissues. Immunohistochemistry assay was used to detect the differential expressions of UBD in 244 OSCC patients and 32 cases of normal oral mucosae. In addition, CCK-8, colony formation, wound healing and Transwell assays were performed to evaluate the effect of UBD on the cell proliferation, migration, and invasion in OSCC. Furthermore, a xenograft tumor model was established to verify the role of UBD on tumor formation in vivo. We found that UBD was upregulated in human OSCC tissues and cell lines and was associated with clinical and pathological features of patients. Moreover, the overexpression of UBD promoted the proliferation, migration and invasion of OSCC cells; however, the knockdown of UBD exerted the opposite effects. In this study, our results also suggested that UBD promoted OSCC progression through NF-κB signaling. Our findings indicated that UBD played a critical role in OSCC and may serve as a prognostic biomarker and potential therapeutic target for OSCC treatment.
Purpose: Aim of this study is to report real-life experience on the treatment of peripheral artery disease (PAD) with a specific drug-coated balloon (DCB), and to evaluate potential prognostic factors for outcomes. Materials and Methods: This is a retrospective study reporting outcomes in patients with PAD who were treated with the Lutonix DCB during a four-year period. Major outcomes included: all-cause mortality, amputation, clinical improvement, wound healing and target lesion revascularization (TLR). Mean follow-up was $24.2{\pm}2.3$ months. Results: Overall, 149 patients (mean age: $68.6{\pm}8.3$ years; 113 males) were treated, either for intermittent claudication (IC) (n=86) or critical limb ischemia (CLI) (n=63). More than half the target lesions (n=206 in total) were located in the superficial femoral artery and 18.0% were below-the-knee lesions. CLI patients presented more frequently with infrapopliteal (P=0.002) or multilevel disease (P=0.0004). Overall, all-cause mortality during follow-up was 10.7%, amputation-free survival was 81.2% and TLR-free survival was 96.6%. CLI patients showed higher all-cause mortality (P=0.007) and total amputation (P=0.0001) rates as well as lower clinical improvement (P=0.0002), compared to IC patients. Coronary artery disease (CAD), gangrene and infrapopliteal disease were found to be predictors for death whereas CLI and gangrene were found to be predictors for amputation, during follow-up. Conclusion: PAD treatment with Lutonix DCBs seems to be an efficient and safe endovascular strategy yielding promising results. However, CAD, gangrene, CLI and infrapopliteal lesions were found to be independent predictors for adverse outcomes. Larger series are needed to identify additional prognostic factors.
Pruritus (itching) is classically defined as an unpleasant cutaneous sensation that leads to scratching behavior. Although the scientific criteria of classification for pruritic diseases are not clear, it can be divided as acute or chronic by duration of symptoms. In this study, we investigated whether skin injury caused by chemical (contact hypersensitivity, CHS) or physical (skin-scratching stimulation, SSS) stimuli causes initial pruritus and analyzed gene expression profiles systemically to determine how changes in skin gene expression in the affected area are related to itching. In both CHS and SSS, we ranked the Gene Ontology Biological Process terms that are generally associated with changes. The factors associated with upregulation were keratinization, inflammatory response and neutrophil chemotaxis. The Kyoto Encyclopedia of Genes and Genomes pathway shows the difference of immune system, cell growth and death, signaling molecules and interactions, and signal transduction pathways. Il1a, Il1b and Il22 were upregulated in the CHS, and Tnf, Tnfrsf1b, Il1b, Il1r1 and Il6 were upregulated in the SSS. Trpc1 channel genes were observed in representative itching-related candidate genes. By comparing and analyzing RNA-sequencing data obtained from the skin tissue of each animal model in these characteristic stages, it is possible to find useful diagnostic markers for the treatment of itching, to diagnose itching causes and to apply customized treatment.
골격성 3급 부정교합의 악교정 수술로 구강기능과 안모를 개선할 수 있다. 악골부조화가 클수록 술전 교정치료에 의한 치아 이동량이 더욱 커지기 때문에 치주조직에도 큰 변화를 줄 수 있는데, 특히 하악 전치부 치주조직의 파괴가 주로 발생된다. 이 보고는 20년전 골격성 3급 부정교합과 하악전돌로 악교정수술을 받은 후 하악 전치부의 심한 동요 및 반대교합의 해결을 위해 내원한 49세 여성환자의 증례로, Face Hunter, Plane System 및 ARCUS digma II를 이용하여 Top-Down concept의 개인 맞춤형 분석으로 치료계획을 수립하였다. 하악 전치부 발치를 동반한 순측 치조골 성형술과 임플란트 식립 후 즉시임시치관에 의한 즉시부하를 시행하였다. 최종 보철물은 CAD/CAM에 의한 Zirconia 수복물로 제작 후 구강 내에서 나사유지형으로 장착하였다. 6개월이 경과한 현재까지 보철물의 파절과 동요 등은 관찰되지 않았으며, 기능 및 심미적으로 만족스러운 결과를 얻었기에 보고하고자 한다.
Photobiomodulation (PBM) 치료법은 특정 파장대역의 광원이 미토콘드리아에서 ATP 생성을 촉진하는 현상을 이용하는 치료법으로서 상처 치유, 염증 감소, 통증 완화 효과가 있는 것으로 알려져 생물 및 의학 분야에서 많은 관심을 받고 있다. PBM 치료법에 대한 연구는 주로 레이저, 발광다이오드(LED)를 광원으로 사용하였고, 유기발광다이오드(OLED)가 가지는 장점에도 불구하고 PBM 치료법에 사용된 사례는 제한적이다. 본 연구에서는 적색(λ = 620 nm), 녹색(λ = 525 nm), 청색(λ = 455 nm) OLED 조명모듈을 사용하여 PBM에 의한 피부관리 효과를 분석하고 LED에 의한 PBM 효과와 비교하였다. OLED 조명모듈의 PBM에 의한 피부미용효과는 적색 OLED 조명모듈에 의한 collagen type 1 합성량 증가, 녹색 OLED 조명모듈에 의한 melanin 합성 억제, 청색 OLED 조명모듈에 의한 nitric oxide 생성 억제를 각각 측정하여 입증되었다.
본 연구는 대두 단백질을 효소(pepsin) 분해 및 미생물(Lactobacillus Rhamnosus) 발효를 통해 생성된 저분자 생리활성 펩티드로 제조하여 인간 유래 피부세포에서 세포활성에 미치는 효과를 확인하고, 천연유래 기능성 미용소재로서의 이용 가능성을 확인하고자 하였다. 연구를 수행하기 위해 대두발효물을 제조하였고, 대두발효물에 대해 LC-MS 분석을 하였으며, WST-1 assay의 방법을 통해 cell viability를 측정하였다. 세포독성을 측정한 결과, 0~2,000 ㎍/mL의 모든 농도 구간에서 세포독성이 나타나지 않았다. 특히 800 ㎍/mL의 농도에서는 대조군에 비해 160~180 %, 양성대조군으로 사용한 EGF(epidermal growth factor)에 비해 120 %에서는 세포증식 효과가 나타나 피부노화 억제 및 피부세포 재생 소재로서의 뛰어난 가능성을 보여주었다. 또한 대두발효물과 그 분획물의 상처 치유 능력을 확인하기 위해 세포이동 실험을 수행한 결과, 대두발효물과 분핵물 중 F4와 F5가 대조군에 비해 우수한 세포이동능을 보여주었고, EGF와는 유사하거나 그 이상의 효과를 보여주었다. 결론적으로 대두발효물과 분핵물(F4 and F5)의 피부개선효과가 유사하게 나타나 피부미용 소재 및 피부 질환 치료의 소재로의 활용이 기대된다.
Background: Lung cancer has a high incidence worldwide, and most lung cancer-associated deaths are attributable to cancer metastasis. Although several medicinal properties of Panax ginseng Meyer have been reported, the effect of ginsenosides Rk1 and Rg5 on epithelial-mesenchymal transition (EMT) stimulated by transforming growth factor beta 1 (TGF-β1) and self-renewal in A549 cells is relatively unknown. Methods: We treated TGF-β1 or alternatively Rk1 and Rg5 in A549 cells. We used western blot analysis, real-time polymerase chain reaction (qPCR), wound healing assay, Matrigel invasion assay, and anoikis assays to determine the effect of Rk1 and Rg5 on TGF-mediated EMT in lung cancer cell. In addition, we performed tumorsphere formation assays and real-time PCR to evaluate the stem-like properties. Results: EMT is induced by TGF-β1 in A549 cells causing the development of cancer stem-like features. Expression of E-cadherin, an epithelial marker, decreased and an increase in vimentin expression was noted. Cell mobility, invasiveness, and anoikis resistance were enhanced with TGF-β1 treatment. In addition, the expression of stem cell markers, CD44, and CD133, was also increased. Treatment with Rk1 and Rg5 suppressed EMT by TGF-β1 and the development of stemness in a dose-dependent manner. Additionally, Rk1 and Rg5 markedly suppressed TGF-β1-induced metalloproteinase-2/9 (MMP2/9) activity, and activation of Smad2/3 and nuclear factor kappa B/extra-cellular signal regulated kinases (NF-kB/ERK) pathways in lung cancer cells. Conclusions: Rk1 and Rg5 regulate the EMT inducing TGF-β1 by suppressing the Smad and NF-κB/ERK pathways (non-Smad pathway).
본 연구에서는 경남 합천에서 수집한 병풀 12 자원을 대상으로 생육적 특성, 주요성분 함량, 항산화성분 및 활성 분석을 통해 우수한 특성을 지닌 병풀 자원의 확보 및 특성 분석을 위한 기초 연구로써 진행되었다. 수집 자원 중 생육적 특성이 우수한 품종은 CA10, CA9, CA1, CA4 및 CA12 였고, 그중에서도 특히 CA10이 건물중이 높았으며 지표성분인 asiaticoside 함량이 높은 자원은 CA5와 CA3 이었다. 또한, 항산화 성분인 총 폴리페놀과 총 플라보노이드를 가장 많이 함유하고 DPPH의 활성이 가장 높은 자원은 CA12으로 지표성분 함량은 적지만, 건물중도 높고 항산화 성분이 풍부한 자원으로 확인되었다. 그러나 수집 자원의 우수한 생육적 특성이 지표성분, 항산화 성분 및 활성이 반드시 일치하지 않는 것으로 확인되어 향후 수량성, asiaticoside, 항산화물질 및 활성 등 각 특성을 목적으로 하는 자원 선발이 필요할것으로 보여진다. 또한 병풀이 제한적인 자생 범위를 갖고 있는 만큼 추가적인 재래 자원 수집을 통한 다양한 특성 연구가 보완되어야 할 것이다.
본 연구에서는 천연 생리활성물질인 에코사펜타엔산을 다량 함유한 흑해삼의 항산화 효과를 조사하여 기능성 화장품 원료로서의 우수성을 제시하였다. 세포내 산화질소(NO) 생성 억제 효과에 대해 해삼 추출물의 활성산소 제거능을 평가하였다. 해삼의 생리활성물질은 활성산소를 제거하는 항산화 효과가 있으며, 평가 결과 우수한 NO 생성이 억제됨을 확인하였다. 연구에 따르면 생리활성 물질은 낮은 농도에서도 높은 항산화 효능을 보여 항산화 효과, 면역 및 염증 반응 조절에 관여하는 것으로 나타났습니다. 해삼의 항산화 제거능은 해삼 추출물의 EPA(eicosapent aenoic acid)를 함유하는 페놀 성분의 항산화 효능에 크게 기여하는 것으로 생각된다. 해삼의 항산화 성분이 작용한다는 보고와 유사한 것으로 밝혀졌다. 또한 해삼 추출물에 다량 함유된 에이코사펜타엔산(EPA) 성분이 염증 활성 억제 효과에 관여하는 것으로 입증돼 상처 치유에도 탁월하다. 본 연구의 결과, 해삼 추출물에 함유된 페놀 성분의 효과가 항산화 활성과 밀접한 인과관계가 있음을 확인하였다.
