Screening for Herbs Having Inhibitory Effect on Egg Hatching of Strongyloides venezuelensis

  • In vitro bioassays of aqueous extracts of 25 herbal species were performed by ovicidal activity against Strongvloides venuelensis. 500 mg of feces from Mongorian gerbil infected with S. venuelensis were placed into polyvinyl culture with extracts of herb and anthelmintic drugs in triplicates. Larval activity inside the polyvinyl bag was recorded microscopically at 12 hours, and every 24 hours thereafter for up to 72 hrs incubation. Eggs were not hatched within 72 hrs and degeneration was evident in the presence of extracts. The ovicidal activity characterized by the absence of L$_3$ in fecal cultures inside the polyvinyl bag. 1. Among 25 herbs, 12 herbs had ovicidal effects and caused degeneration of larvae in egg. S. angustifolia, I. helenium, A. korenum, P. tenuifolia, F. virridissima, A. asiatica, S. chinensis, A. tataricus, S. baicalensis, M. rimosa, S. glabra and Z. piperitum were the most ovicidal effect. However, P. thunbergiana, T. chinensis, C. flos, G. uralensis, C. obutusifolia, C. fortunei, A. koreana, P. grandiflorum, P. tenuifolia (Radix), P. mume and A. asphodelodes showed no ovicidal effects in vinyl bag culture. 2. Of 25 hrebs, 4 herbs had ovicidal and wormicidal effects, P. thunbergiana, C. flos, B. Striata and T. chinensis were characterized by degeneration of larvae.

Synthesis of the Anthelmintic Diaryl Butanes against Clonorchis sinensis (간흡충에 대하여 구충작용을 갖는 Diarylbutane류의 합성)

  • During investigation of the anthelmintic substances against Clonorchis sinensis which are based on the structure of meso-dihydroguairaretic acid, some non-mesoic diaryl butanes were synthesized by Grignard reaction and their anthelmintic activities were determined. In this reaction, an aryl butanone was reacted with benzylmagnesium chloride to produce the corresponding diaryl hydroxybutane which was converted to the corresponding diaryl butane by zinc and hydrogen chloride. The substituents in benzene ring of the diaryl butanes were changed by methylation or demethylation. Among the synthesized substances, 4-phenyl-1-(3, 4-dihydroxyphenyl)-2, 3-dimethylbutane(VII), 4-phenyl-1-(3-hydroxy-4-methoxyphenyl)-2, 3-dimethylbutane(IX) and 4-phenyl-1-(4-hydroxy-3-methoxyphenyl)-2, 3-dimethylbutane(VI) showed strong wormicidal effects against Clonorchis si-nensis in that order. Phenolic hydroxyl group seemed to play a certain role for the wormicidal activity of the diaryl butanes.

Studies on Chlorinated Tolyl Compounds. (Tolyl계 Chloro 화합물에 관한 연구)

  • P, P'-Hexachiorobitolyl, p, p-hexachlorobitolylketone and p, p' hexachlorobitolylthiourea were synthesized by the radical chlorination method of Hass and McBeer. Among these comppounds, l wormicidal activity of p.p'-hexachlorobitolythiourea was much most potent against Clonorchis sinensis than Hetol in vitro. In the process of the p, p'-biolyl chlorination, ${\alpha, \alpha, \alpha}$-trichlorobitolyl was not detected, but small amount of ${\alpha, \alpha, \alpha}$-dichlorobitolyl was detected. This compound was immediately converted bitolyl compounds were analyzed, linear relationship was observed between retention indices and number of chlorine in two series of odd and even numbered chlorine compounds.

The Wormicidal Substances of Fresh Water Fishes on Clonorchis sinensis VII. The Effect of Linolelc Acid and Ethyl Linoleate on Parasite Viability (간흡충에 대한 살충성 물질에 관한 연구)

  • In an attempt to analyze the clonorchicidal activity of linoleic acid and ethyl linoleate in vitro, the wormicidal effects on Clonorchis sinensis were chronologically monitored in dose titration experiments. Encysted metacercariae were killed within a period of 31, $0{\pm}4.0$ min, 149.3k4. 1 min and $207.0{\pm}13.5$ min with 100.0 mg, 0.1 mg and 0.001 mg linoleic acid, respectively. The time required for the linoleic acid to kill adult worms was 167, $0{\pm}0.8$ min with 100.0mg, $253.0{\pm}0.8$ min with 0.1mg, and $277.0{\pm}0.8$ min at 0.001mg titration. Clonorchicidal activity of ethyl linoleate was relatively delayed as death was observed within $263.3{\pm}2.9$ min, $286.0{\pm}0.5$ min, and $318.0{\pm}0.8$ min for 100.0 mg/ml, 0.1 mg/ml and 0.001 mg/ml concentrations, respectively. The wormicidal effects observed with these pure anti-clonorchal substances were found to be similar to the biological activity of native products derived from the mucus of the fresh water fish.

Electron Microscopic Study on the Effect of Albendazole Against Paragonimus Westermani (Albendazole의 폐흡충에 미치는 영향에 관한 전자현미경적 연구)

  • Albendazole is a broad spectrum anthelmintics with high activity against intestinal nematodes as well as tremtodes and cestodes infections. But so far wormicidal effect of albendazole against Paragonimus westermani is uncertain. The present study undertaken to observe the morphological changes of Paragonimus westermani which was obtained from experimentally infected with metacercariae isolated from Cambaroides similis and 9 of them were given albendazole 2 doses of 25mg/kg daily for the 2 days and 4 days from the day of 20 weeks after infection. The Paragonimus werms were collected from the lung of the cats which were autopsied at 24 hours after treatment. The fine structures of the collected worms were studied by means of scanning and transmission electron microscopy. The findings of the observations were compared with those of untreated worms. The results are as follows : 1) In the scanning electron microscopic(SEM) observations, the worms obtained from 2 days treated group showed many blebs which were formed on the surface of damaged tegument in between two suckers. Protrusion of oral sucker typically observed and surrounding tegumental ridges were damaged. 2) The worms obtained from 4 days treated group showed marked contraction revealing half size of normal worms. 3) The transmission electronmicroscopic(TEM) observations indicated that large blebs of irregular shapes were formed by the destruction of tegumental ridge of tegumental syncytium and also showed degeneration of mitochondriae. 4) Destruction of tegumental syncytium formation of blebs were usually observed in the ventral side of the worms. 5) Differentiation of tegumental layer and basement layer muscular layer and parenchymal layer around two suckers of worms obtained at 4 days treated group was difficult. 6) Many host cells invaded and destroyed the degenerated tegument by the occurrence of blebs or the exposed muscular layer of the worm after exfoliation of the tegument.
