Distribution Characteristics Analysis of Pine Wilt Disease Using Time Series Hyperspectral Aerial Imagery (소나무재선충병 발생시기별 피해목 탐지를 위한 시계열 초분광 항공영상의 활용)

  • Pine wilt disease has greatly damaged pine forests not only in East Asia including South Korea and China, but also in European region. The damage caused by pine wood nematode (Bursaphelenchus xylophilus) is expressed in bundles within stands and rapidly spreading, however, present field survey methods have limitations to detecting damaged trees at regional level. This study extracted the damaged trees by pine wilt disease using time series hyperspectral aerial photographs, and analyzed their distribution characteristics. Hyperspectral aerial photographs of 1 meter spatial resolution were obtained in June, September, and October. Damaged trees by pine wilt disease were extracted using Normalized Difference Vegetation Index (NDVI) and Vegetation Index green (VIgreen) of the September photograph. Among extracted damaged trees, dead trees with leaves and without leaves were classified, and the spectral reflectance values from the photographs obtained in June, September, and October were compared to extract new outbreaks in September and October. Based on the time series dispersion of extracted damaged trees, nearest neighbor analysis was conducted to analyze distribution characteristics of the damaged trees within the region where hyperspectral aerial photographs were acquired. As a result, 2,262 damaged trees were extracted in the study area, and 604 dead trees (dead trees in last year) with leaves in relation to the damaged time and 300 and 101 newly damaged trees in September and October were classified. The result of nearest neighbor analysis using the data shows that aggregated distribution was the dominant pattern both previous and current year in the study area. Also, 80% of the damaged trees in current year were found within 60 m of dead trees in previous year.

Effect of Strength Increasing Sizes on the Quality of Fiberboard (섬유판(纖維板)의 증강(增强)사이즈제(齊)가 재질(材質)에 미치는 영향(影響))

  • The fiberboard and paper mills in this country are much affected by the price hikes and shortage of phenolic resins, since phenolic acid as a raw material depends on imported good. It is prerequisite to fiberboard industry to help replace with other sized and stabilize the prices and supply of them, improving the quality of boards. Thus, the present study was carried out to examine the effect of strength increasing sized such as urea formaldehyde resin (anion and cation type) and urea melamine copolymer resin, on the quality of the wet forming hardboard, and comparing them with two types of proprietary modified melamine resins, and ordinary size, phenol resin. The Asplund pulp was prepared from wood wastes mixed with 20 percent of lauan and 80 percent of pines as a fibrous material. After sizing agents were added at a pH of 4.5 for 10 minutes with alum in the beater, the stock was made in the form of wet sheet, prepared, and then performed by hot pressing cycle: $180^{\circ}C$, $50-6-5kg/cm^2$, 1-2-7 minutes. The properties of hardboard were examined after air conditioning. The results obtained are summarized as follows: 1. There is a significant difference in specific gravity among hardboards that were treated with strength increasing resins, but no difference is effected by the increase in the resin content. In the case of modified melamine resin, its specific gravity is highest. The middle group comprises cation type of urea resin, anion type of urea resin, and acid colloid of urea-melamine copolymer resin. The lowest is phenolic resin. 2. The difference of the moisture content of hardboard both by the resins and by the amount of each resin applied is significant. The moisture content of hardboard becomes lower along with the increase of each resin content, but there is no difference between 2 and 3 percent. 3. For water absorption, there is a significant difference both in the adhesives used and in the amount of paraffin wax emulsion. The water resistance becomes higher inn proportion to the content of the paraffin wax emulsion. To satisfy KS F standards of the water resistance, a proprietary modified melamine resin (p-6100) and modified cation type of urea resin (p-1500) do not require any paraffin wax emulsion, but in the case of anion type of urea resin, cation type of urea resin, and urea-melamine copolymer resin, 1 percent of paraffin wax emulsion is needed, and 2 percent of paraffin wax emulsion in the case of phenolic resin. 4. The difference of flexural strength of hardboard both by the resins and by the amount of each resin is significant. Modified melamine resin shows the highest degree of flexural strength. Among the middle group are urea-melamine copolymer resin, p-1500, anion type of urea resin, and cation type of urea resin. Phenolic resin is the lowest. The cause may be attributable to factors combined with the pressing temperature, sizing effect, and thermal efficiency of press platens heated electrically. 5. Considering the economic advantages and properties of hardboard, it is proposed that urea-melamine copolymer resin and cation type of urea resin be used for the development of the fiberboard industry. It is desirable to further develop the modified urea-melamine copolymer resin and cation type of urea resin through continuous study.

Characteristics of Biodegradation of Geosmin using BAC Attached Bacteria in Batch Bioreactor (정수처리용 생물활성탄(BAC) 부착 박테리아를 이용한 회분식 반응기에서의 Geosmin 생분해 특성)

  • In this study, three different biological activated carbons (BACs) were prepared from activated carbons made of each coal (F400, Calgon), coconut (Samchully) and wood(Pica, Picabiol) which were run for two and half years in the pilot plant. The attached bio-film microorganisms in and on the BACs were isolated and identified. The results showed that nine different bacteria species (Chryseomonas luteola, Stenotrophomonas maltophilia, Pseudomonas vesicularis, Aeromonas hydrophila, Spingomonas paucimobilis, Agrobacterium radiobacter, Pseudomonas fluorescens, Spirillum spp., and Pasteurella haemolytica) were isolated and identified, the dominant species was Pseudomonas sp. that had occupied 56.5%. More specifically, it was observed that the populations of the microorganisms deceased in the order: Pasteurella haemolytica (18.9%) > Chryseomonas luteola (4.0%) > Agrobacterium radiobacter (3.5%) > Aeromonas hydrophila (2.0%) in and on the BACs. After isolating of 9 species of biofilm microorganisms, the growth curve for the biomass was investigated. During 24~96 hours, the biomass has the highest concentration, and activity of the biomass was the best to uptake geosmin as carbon resources. The operation temperatures for investigating the biodegradation of geosmin were set at $4^{\circ}C$ and $25^{\circ}C$. Pseudomonas vesicularis, Pseudomonas fluorescens, Agrobacterium radiobacter and Stenotrophomonas maltophilia played a maior role in removing the target compound as geosmin. However, geosmin was not biodegraded well by Chryseomonas luteola, Spingomonas paucimobilis, and Spirillum spp.. It is also interesting to evaluate kinetics of biodegradability of geosmin. The first-order rate constants for biodegradability of geosmin at $4^{\circ}C$ and $25^{\circ}C$ were $0.00006{\sim}0.0002\;hr^{-1}$ and $0.0043{\sim}0.0046\;hr^{-1}$ respectively. Higher water temperature produced better geosmin removal rates. When concentrations of geosmin increased from 10 to 10,000 ng/L, the rate constants for biodegradability of geosmin increased from 0.0003 to $0.0882\;hr^{-1}$. As described earlier, higher geosmin concentration in the reactor produced higher rate constant.

Characteristic of mycelial growth of cauliflower mushroom (Sparassis latifolia) using replacement culture with Trichoderma and rDNA analysis in genealogy of crossbreeding strain (푸른곰팡이 대치배양에 의한 꽃송이버섯 균사 생장 특성 및 계통간 교잡균주의 rDNA 분석)

  • Cauliflower mushroom widely known high concent of ${\beta}$-glucan for farm cultivation invigoration verified characteristics of mycelia growth, genetic diversity, resistance to Trichoderma by replacement culture with Trichoderma and growth characteristics of new variety crossbleeding strain. The result of replacement culture with Trichoderma for verification resistance about Trichoderma, 6951 (T. viride) strain did not show special change after formation of confrontation line and 6952 (T. spp.) strain was showed more formation of spore after formation of confrontation line. But 6426 (T. harzianum) strain found to encroach part of growth area of cauliflower mushroom mycelia. Among 10 kinds cauliflower mushroom strain, JF02-06 strain collected by Gurye, found did not spore of Trichoderma and thought to be resistant to Trichoderma. The result of crossbleeding after selected that mother strain good growth and formation of fruit body, verified good mycelia growth at JF02-47, 49 and 50 strain in Korean pine of wood-chip media. The result of gene sequence about ITS1, 5.8S and ITS4 for analysis of genetic diversity at crossbleeding strain, found high significance to other cauliflower mushroom in registered Genebank. The result of growth characteristic of spore and mycelia of cauliflower mushroom by observation microscope, size of spore showed water drop shape to major axis $6{\mu}m$ and minor axis $5{\mu}m$ and clamp showed 3 types in mycelia. The wide of mycelia was $3{\mu}m$. The characteristic of mycelia of cauliflower mushroom found to grow mycelia in clamp at approximately 50%. The growth speed of mycelia was $0.507{\mu}m/min$ and 2nd mycelia grown similar speed to mother mycelia at parallel with mother mycelia after growth speed at $0.082{\mu}m/min$. The formation of clamp made small clamp for 5 hours after shown transfer of electrolyte in mycelia inside. The septum formation started after 3 hours and then finally completed after 2 hours. In this study, strain of cauliflower mushroom verified resistance of Trichoderma, genetic diversity and characteristic of mycelia growth. Therefore, basic knowledge of cauliflower mushroom will improve and further contribute to development of mushroom industry.

Isolation and Characterization of Tartaric Acid-Degrading Bacteria from Korean Grape Wine Pomace (국산 포도주 주박으로부터 주석산 분해 세균의 분리 및 특성)

  • Several tartaric acid-degrading bacteria were isolated from Korean grape wine pomace after enrichment culture at $30^{\circ}C$ for 10 days in liquid media containing tartaric acid Among them, strains KMBL 5777 and KMBL 5778 exhibited the highest level in the growth and tartaric acid degradability in a medium containing 0.2%(w/v) tartaric acid as a sole carbon source. They were identified as Acetobacter tropicalis based on their morphological and physiological characteristics as well as their 16S rDNA sequences. Blast search of the 16S rDNA sequences revealed that the isolated strains are closest to Acetobacter tropicalis. Homologies of the sequences of KMBL 5777 and KMBL 5778 were 96.0 and 98.9%, respectively with those of A. tropicalis LMG 1663. Both the two bacteria showed higher tartaric acid degradation at $25^{\circ}C$ that those at 20 and $30^{\circ}C$. They could degrade tartaric acid at a wide range of pH between 4.0 and 7.0 with the most rapid degradability at pH 7.0. However, when the bacteria were grown for 8 days, the same level of tartaric acid degradation was observed at pH 4.0, 5.0, 6.0 and 7.0, which was 90.0% of degradation of the acid.

A Study on the Origin of Human Governance Periods in the Hidden Stems (인원용사(人元用事)의 연원에 관한 연구)

  • The purpose of this study is to examine the validity of Hidden Stems (支藏干) in the Four Pillars of Destiny with regard to the use of human governance periods in the hidden stems (人元用事). First, there is a theory of assigning period of governance for designated constituents (司令論) in the Hidden Stems of the Earthly Branch. Second, there is a theory that determines the structure of the Four Pillars by the Exposed Constituent from the Hidden Stems (透出論) in the Month Earthly Branch. Since these two theories conflict with each other and cause confusions, this study examined the theory of Hidden Stems in the Four Pillars Classics and examined the historical development of governance period for constituent hidden stems and their validity. The results of the study are as follows: Firstly, the number of dates assigned to respective constituents does not correspond to the calendarical principle, and the assignment of the governance dates for each constituent does not correspond to the principles proposed in ancient books of Four Pillars. Second, though it is said in the Classics that 72 days are equally assigned to each of the Five Elements, actual distributed days for the five elements was 65 days for Wood, 55 days for Fire, 100 days for Earth, 65 days for Metal, and 65 days for Water. Third, though it is said that 7 days should be designated to Yang Earth Mu for the months of Tiger 寅, Monkey, Snake, and Pig, it is logically more legitimate to assign those days to Yin Earth Ki since the month before Tiger is Ox, and the month before Monkey is Goat. Lastly, rationale behind assigning Ki Earth only to Horse Oh as constituting Hidden Stem while disregarding months of Rat, Rabbit, and Rooster is considered not reasonable. Looking at these results comprehensively, it is concluded that the Exposed Constituent theory is logically more appropriate than Assigned Governance theory.
