In this study, total mercury and methyl mercury in whole blood of Korean was analyzed so as to investigate the correlation between total mercury (T-Hg) and methyl mercury (Me-Hg). 4000 whole blood samples were divided in four groups, according to T-Hg concentration in percentile: group I (p25-p50), group II (p50-p75), group III (p75-p95) and group IV (p95-p100). 100 samples were randomly selected from the each group, and Me-Hg concentration was measured. T-Hg concentration in whole blood was analyzed using a Direct Mercury Analyzer-80 and obtained limit of detection (LOD) was $0.2{\mu}gL^{-1}$. Me-Hg concentration was analyzed with ethylate derivatization using headspace-gas chromatography-mass spectrometry, and obtained LOD of methyl mercury was $0.5{\mu}gL^{-1}$. The geometric means of T-Hg and Me-Hg were $6.35{\mu}gL^{-1}$ and $4.44{\mu}gL^{-1}$, respectively, and 71.91% of T-Hg was presented as Me-Hg.
To investigate the effects of Siegesbeckiae Herba on the blood pressure, this study was carried out in the whole and the spinal rabbits, using its water and ethanol extracts. When Sigesbeckiae Herba water extract (SGWE) and ethanol extract (SGEE) were administered into the whole rabbits by route of e ear vein, both of them produced fall of blood pressure. The difference between these two extracts was that SGEE is more potent than SGWE. The depressor effects of SGWE and SGEE were not affected by vagoto minization but inhibited by pretreatment of atropine. The depressor responses of the whole rabbits to intravenous SGWE and SGEE were weakened by treatment of animals with bethanidine or phentolamine but not by propranolol. Pretreatment of the whole rabbits with diphenhydramine significantly weakened the d depressor effects of SGEE. Infusion of SGWE and SGEE in the whole rabbits did not influence the pressor effects caused by angiotensin, norepinephrine or carotid artery occlusion. SGEE, when given into the lateral ventricle of the whole rabbits or into the vein of the spinal rabbits, elicited fall of blood pressure, respectively.
This study presents an impedance method of in-vitro characterization of the thrombotic potential of whole blood. Whole blood samples of 0.2 cc were put into a micro-cell with embedded three electrodes immediately after venepuncture at $37^{\circ}C$. Anti-coagulated blood samples were also collected for hematocrit and blood viscosity analyses. The rate of change of electron flow was measured, which indicates the inverse of the thrombotic potential. A sudden decrease in the rate of change of electron flow was observed at a time equal to approximately 110 seconds. This sudden decrease was significantly delayed in anti-coagulated samples. After the sudden decrease, the rate continued to decrease, reaching a minimum value in unadulterated samples while the change in the rate in the anti-coagulated ones was found rather moderate. Based on these preliminary findings, the present method may be of used as a new tool for the diagnosis of the thrombotic potential of whole blood.
Effects of Astragali Radix Water Extract (ARWE) on the blood pressure were investigated in the whole rabbit and the spinalized rabbit. ARWE, when administered into the ear-vein or lateral ventricle, produced a fall in the blood pressure in the whole rabbit, but intravenous ARWE in the spinalized rabbit did not elicite the hypotensive action. Pretreatments with chlorisondamine, guanethidine, phentolamine and cyproheptadine in the whole rabbit weakened the depressor action of ARWE. The hypotensive action of the whole rabbit to ARWE was not influenced by the pretreatment of the animals with diphennylhydramine, propranolol, and vagotomization, whereas inhibited by atropine. ARWE did not affect the pressor response by angiotensin. However, it enhanced the hypertensive action by norepineprine and reduced the elevation in the blood pressure by carotid occlusion in the whole rabbit. These experimental observations suggest that ARWE may cause the depressor response via mechanisms of the central sympathetic blocking action, cholinergic action by peripheral origin and serotonin-like action.
Effects of Akebiae Lignum, whose scientific name is Akebia quinata Decaisne, on the blood pressure were investigated with EtOH extract in whole and spinal rabbits. Akebia Lignum EtOH extract (AEE), when given intravenously, produced a fall in blood pressure not only in whole rabbit but also in spinal rabbit and AEE administered into a lateral cerebral ventricle of whole rabbit did not elicit a fall in blood pressure. The depressor responses of the whole rabbit to intravenous AEE were weakened by treatment of the animals with atropine and chlorisondamine but not by vagotominijation, phentolamine avil, and then the depressor action causing by AEE in the whole rabbit was not affected by pretreatment of physostigmine which is cholinesterase inhibitor and of hemicholinium which blocks acetylcholine synthesis by interfering with choline uptake in nerves. These observations suggest that the hypotensive action of AEE of which component is not affected by cholinesterase is due to direct action at parasympathetic receptor.
The aim of this study was to measure whole blood viscosity (WBV) and then to establish its reference values and identify correlation between WBV and hematology or serum chemistry in beagle. The experiment was made up of 82 healthy beagle dogs. Jugular vein blood samples (10 ml) were collected. WBV (cP) measured within 4 hours after collection by U-shaped scanning capillary-tube viscometer ($BVD-PRO1^{(R)}$) which is capable of measuring yield stress and viscosity of whole blood continuously over a wide range of shear rates from $1s^{-1}$ to $1000s^{-1}$ is new type of capillary tube viscometer and calculates viscosity using Casson fluid model. Measured values of WBV, Complete blood cell count, serum chemistry were analyzed by RM-ANOVA test. Mean diastolic and systolic WBV (cP) were $29.032{\pm}6.137$ and $4.528{\pm}0.865$. Bodyweight (BW), Red blood cell count (RBC), Hemoglobin (HGB), Hematocrit (HCT), Alkaline phosphatase (ALP), Cholestetol (CHOL), Total protein (TP), Globulin, Chloride (Cl), Fibrinogen, were statistically correlated with WBV over whole range of shear rates (p < 0.05). This study newly evaluated reference values of WBV by U-shaped viscometer in beagle. Correlation between WBV and BW, RBC, HGB, HCT, ALP, CHOL, TP, Globulin, Cl, Fibrinogen was presented.

This paper presents the characterization of a continuous magnetophoretic microseparator for separating white and red blood cells from peripheral whole blood cells based on their native magnetic properties. The magnetophoretic microseparator separated the blood cells using a high gradient magnetic separation (HGMS) method without the use of additives such as magnetic beads or probing materials. Experimental results show that the paramagnetic capture mode microseparator can continuously separate out 93.5% of red blood cells and 97.4% of white blood cells from diluted whole blood, and the diamagnetic capture mode microseparator can continuously separate out 89.7% of red blood cells and 72.7 % of white blood cells by using applying an external magnetic flux of 0.2 T using a permanent magnet.
This study was performed to evaluate chromium in air and chromium concentrations in whole blood and urine of workers at chrome plating factories, and to determine the correlation between environmental and biological chromium levels. This study involved 29 workers as study group and 24 undergraduate students as control group. The geometric means(GM) of airborne hexavalent chromium and total chromium concentrations in the plating factories were 3.4 $\mu\textrm{g}$/㎥ and 10.8 $\mu\textrm{g}$/㎥, respectively. Hexavalent chromium levels in two of total 29 measurements exceeded the korean occupational exposure limit and the American Conference of Governmental Industrial Hygienists Threshold Limit Value(ACGIH-TLV) of 50$\mu\textrm{g}$/㎥. Only one sample for total chromium exceeded the Korea occupational exposure limits, the ACGIH-TLV, and the National Institute for Occupational Safety and Health Recommended Exposure Limits(NIOSH-REL) of 500 $\mu\textrm{g}$/㎥. The GM of chromium concentrations in blood and urine of workers exposed to chromium were 8.4 $\mu\textrm{g}$/L and 11.9 $\mu\textrm{g}$/L. The GM of chromium concentrations in blood and urine of workers exposed to chromium were 8.4 $\mu\textrm{g}$/L and 11.9 $\mu\textrm{g}$/L, respectively, whereas the chromium concentrations in blood and urine of the controls were 1.6 $\mu\textrm{g}$/L and 3.8 $\mu\textrm{g}$/L, respectively. There were statistically significant differences of blood and urine concentrations between study group and control group (p<0.01). The chromium concentrations in urine were most highly related to hexavalent chromium, concentration in air(r=0.642, p<0.01). Also, there was a relatively high correlation between the hexavalent chromium concentrations in air and chromium concentrations in whole blood(r=0.557, p<0.05). These results indicate that whole-blood chromium with urinary chromium could be an indicator of chromium body burden caused by exposure to chromic acid mist in plating operation.
Reappraisal measurements of apparent half survival time of red cell by $^{51}Cr$ method was made and effects of blood-letting over red cell survival were observed. The study was performed on 53 normal male subjects under three different experimental conditions. 1. Group 1 Mean $^{51}Cr$ red cell half survival by ACD wash method was 29.7 days. $T\frac{1}{2}$ of Ascorbic acid method was 29.0 days in group with 100 mg dose and 29.1 days in group with 50 mg dose respectively. There was no difference between these two methods in regards to red cell half survival. No difference were noted in amount of ascorbic acid administered. 2. Group 2 As daily amount of blood loss is increased the shortening of red cell half survival was noted. Rapid phase was seen when blood loss ranged 10 to 25 ml per day, while slow phase noted when more loss amounted 25 ml or more daily. Thus, it was clear that there was more than an exponential relation between $T\frac{1}{2}$ and the amount of blood loss. 3. Group 3 $T\frac{1}{2}$ measured by cpm per whole blood was within normal range and $T\frac{1}{2}$ measured by cpm per red cell mass showed shortening tendency when compared with the former in the group measured after blood loss (from 25 ml daily up to 100 ml daily in 10 days). In the group with rather constant blood loss of 100 ml daily for 10 consecutive days revealed the significant difference in two measurements (P<0.01). 4. $T\frac{1}{2}$ in non-steady state When red cell production is increased compared with red cell destruction, $T\frac{1}{2}$ measured by cpm per red cell mass being shorter than that by cpm per whole blood. Shortening of $T\frac{1}{2}$ measured by cpm per whole blood is more prominent. if red cell destrction is enhanced and exceeds production. 5. It is clear that when expressing red cell destruction rate, $T\frac{1}{2}$ measured by cpm per whole blood is more adequate and production more consistent with cpm red cell mass. 6. $T\frac{1}{2}$ measured during blood-letting, when corrected by amount of blood loss, it remains normal. It is erroneous to use conventional equational when measuring $T\frac{1}{2}$ in non-steady. $T\frac{1}{2}$ measured by cpm per whole blood is considred more applicable in clinical evaluation.
