Acute Oral, Pulmonary and Intravenous Toxicity/Pathogenicity Testing of Burkholderia pyrrocinia CAB08106-4 of in Rats (랫드를 이용한 Burkholderia pyrrocinia CAB08106-4의 급성경구, 호흡기, 정맥독성/병원성시험)

  • Burkholderia pyrrocinia CAB08106-4 has an anti-bacterial effect on Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp.. It is an environmentally friendly microbial product that prevents and controls a variety of phytopathogens involving Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp.. The aim of this study was to assess and to compare the pathogenicity of Burkholderia pyrrocinia CAB08106-4 by single exposure of rats through several routes such as oral, intranasal and intravenous. For the acute toxicity / pathogenicity study, the animals were sacrificed on days 3, 7, 14 and 21, and macroscopically observed their organs to examine the numbers of internally-retained pesticidal microbes. Clinical examinations were performed daily during administration period, and body weight gain was evaluated. In the study, no clinical sign, weight gain and mortality were observed in relation to the administration of test article. The significant changes of internal/external microbes by test article were not detected. The pathological findings in relation to the administration of the test article in the necropsy were not observed. It could be concluded that the microorganism was not toxic or pathogenic in rats via oral, intranasal and intravenous route.

New method for sclerotial isolation of Sclerotium spp. from infested soil

  • White rot on Allium species recently had a high incidence as increased cultivating areas of tropical garlic types in Korea. Two types of Sclerotium have been known as causal agents producing different size and shapes of sclerotia in infested fields. We developed a new method for isolation of two types of sclerotia from infested field soils that can be used for ecological study of sclerotium spp. and establishment of control strategy. Soil samples collected from heavily infested fields were evenly mixed and placed on a automatic sieve shaker connected with tap water. After 10 min. of shaking, residues on 0.5mm and 0.25mm sieve were separately collected and suspended with 70% sugar solution, which method floats sclerotia in aqueous layer. Then, floated fraction was carefully separated and mixed with a same volume of 1% sodium hypochlorite solution to differentiate with organic materials. This method provides direct count of sclerotia under dissecting microscopy.

Review of Disease Incidence of Major Crops in 2001 (2001년 농작물 병해 발생개황)

  • Climate in the year of 2001 could be summarized as severe drought from March to May, unusually high temperature in whole plant growth periods and clear weather condition especially in harvesting time of September and October without any typoons. In rice, major diseases such as leaf and panicle blast, bacterial blight, sheath blight and bacterial grain rot developed little due to unfavorable weather conditions, however, brown spot occurred severely due partly to the reduced use of N-fertilizer, Rice stripe virus infection was unusually high mainly in the west coast areas with four times more diseaseD area campared to the previous year, In pepper phytophthora blight was extremely severe in Cheonnam and Cheonbuk provinces, where had frequent rainfalls during growing period. Incidence of major diseases of tomato and cucumber in 2001 was relatively mild. In watermelon, penicillium fruit rot, one of the peculiar disease, spread over major production areas, whereas CGMMV, usually severe in every years was much reduced. Watermelon plants growing in open-fields were more severely diseased than those in plastic houses. Major diseases of chinese melon and strawberry occurred slightly and in particular, sudden wilt syndrome of chinese melon which was severe in 2000 showed mild infections in 2001. Incidence of white rot of garlic and onion was much lower unlike other years due mainly to spring drought and high temperature in the growing period. Infected area of potato bacterial wilt tended to increase in 2001 while fusarium wilt of sweetpotato was decreased. Apple diseases were generally mild, but powdery mildew of pear increased sharply. Barley scab was not severe as seen in other years.

Present Status of Soilborne Disease Incidence and Scheme for Its Integrated Management in Korea (국내 토양병해 발생현황과 종합 관리방안)

  • Incidence of soilborne diseases, as a major cause of failure of continuous monocropping becomes severe in recent years. For examples, recent epidemics of club root of chinese cabbage, white rot of garlic, bacterial wilt of potato, pepper phytophthora blight, tomato fusarium wilt and CGMMV of watermelon are the diseases that require urgent control measures. Reasons for the severe incidence of soilborne diseases are the simplified cropping system or continuous monocropping associated with allocation of major production areas of certain crop and year-round cultivation system that results in rapid degradation of soil environment. Neglect of breeding for disease resistance relative to giving much emphasis on high yield and good quality, and cultural methods putting first on the use of chemical fertilizers are thought to be the reason. Counter-measures against soilborne disease epidemics would become most effective when the remedies are seeded for individual causes. As long-term strategies, development of rational cropping system which fits local cropping and economic condition, development and supply of cultivars resistant to multiple diseases, and improvement of soil environment by soil conditioning are suggested. In short-term strategies, simple and economical soil-disinfestation technology, and quick and accurate forecasting methods for soilborne diseases are urgent matter far development. for these, extensive supports are required in governmental level for rearing soilborne disease specialists and activation of collaborating researches to solve encountering problems of soilborne diseases.
