고려인삼 알카로이드 분획의 성분연구 (I) 인삼류별 TLC 비교 및 알카로이드 동정 (Chemical Studies on the Alkaloidal Fraction of Panax ginseng C.A. Meyer (I). TLC Analysis of Various Ginsengs and Identification of an Alkaloid Component)

  • 인삼에서 생리활성이 보고된 알카로이드 분획에 관한 성분연구의 일환으로 백삼 50kg으로부터 알카로이드 분획 2.6g(수율:$5{\times}10^{-3}$%)을 분리하였으며, $CHCl_3$/MeOH(10 : 1)의 전개용매로 TLC한 결과 약 13종 이상의 Dragendorff 시약에 양성인 spot들을 관찰 할 수 있었다. 인삼 종류별 비교에 있어서는 백삼, 홍삼 및 전칠인삼은 서로 별 차이가 없었지만 미국인삼에 있어 No.2(Rf 0.71), No.3(Rf, 0.68)등의 spot가 검출되지 않았다. 또한 백삼에서 1종의 알카로이드 성분을 순수분리하여 화학적, 분광학적인 방법으로 구조를 구명한바 $N_9$-formyl-1-methyl-$\beta$-carboline으로 동정하였다.

Investigation of Different Factors Affecting the Electron Spin Resomance-based Characterization of Gamma-irradiated Fresh, White, and Red Ginseng

  • Fresh (raw roots), white (dried), and red (steamed-drid) ginseng samples were gamma-irradiated at 0 to 7 kGy. Electron spin resonance (ESR) technique was used to characterize the irradiation status of the samples, targeting the radiation-induced cellulose radicals after different sample pretreatments. All non-irradiated samples exhibited a single central signal (g=2.006), whose intensity showed significant increase upon irradiation. The ESR spectra from the radiation-induced cellulose radicals, with two side peaks (g=2.0201 and g=1.9851) equally spaced (${\pm}3mT$) from the central signal, were also observed in the irradiated samples. The core sample analyzed after alcoholic-extraction produced the best results for irradiated fresh ginseng samples. In the case of irradiated white and red ginseng samples, the central (natural) and radiation-induced (two-side peaks corresponding to cellulose radical) signal intensities showed little improvement on alcoholic-extraction. The water-washing step minimized the effect of $Mn^{2+}$, but reduced the intensity of side peaks making them difficult to indentify. The effect of different origins was negligible, however harvesting year showed a clear effect on radiation-induced ESR signals.

Determination of 11 Ginsenosides in Black Ginseng Developed from Panax ginseng by High Performance Liquid Chromatography

  • A high performance liquid chromatography (HPLC) method has been developed for determination of 11 ginsenosides in black ginseng (BG, white ginseng that is subjected to 9 cycles of $95^{\circ}C$ for 3 hr). After eluted by gradient elution of water-acetonitrile without buffer in 70 min, 11 ginsenosides in BG were identified. The proposed method provided good linearity ($R^2$>0.9995), accuracy (92.2-106.6%), and intra- and interday precision (RSD<2.6%). In addition, ginsenosides compositions in white, red, and black ginsengs were investigated using this method, respectively. Interestingly, in BG, the content of ginsenoside $Rg_3$ which does not existed in white ginseng was 7.51 mg/g, approximately 20 times than that in red ginseng.

  • Korean ginseng (Panax ginseng Meyer) is a traditional herb used across the world to treat various diseases. Although, red ginseng is this herb's most famous product and has demonstrated diverse pharmacological activities, white ginseng (WG) is another ginseng product that is made fresh and individually regulated in Eastern Asia. Red and white ginseng show different characteristics due to distinct processing steps despite originating from the same plant, and the drug interactions induced by WG have not been well documented. Selegiline is a selective monoamine oxidase (MAO) inhibitor used as an antidyskinetic and antiparkinsonian agent. Here we developed a quantification method for selegiline in mouse plasma using a C8 stationary phase in triple quadrupole-mass spectrometry (LC-MS/MS) with multiple reaction monitoring (MRM). The validated LC-MS/MS method was successfully applied to determine the potential interaction with WG extract (0.1 g/kg/day) pre-administered for 4 weeks. The $AUC_{0-240min}$ of selegiline was altered due to a decrease in the absorption of selegiline with repeated administration of WG extract.

고려인삼의 식품 안전성에 관한 임상 연구 (Clinical Study on Food Safety evaluation of Panax ginseng)

  • In this study, the clinical safety and toxicology of oral ingestion of supplement capsules containing ginseng radix was investigated in healthy young volunteers. This study was a pilot randomized, double blinded, placebo controlled trial. The healthy volunteers were divided into 6 groups of 20 each (10 males and 10 females). They took the ginseng powder for 35 days (3g/day) for safety evaluation. There were measured general healthy levels such as hematological, biochemical and electrocardiographic parameters. After the first week, besides Korean white ginseng the other treatments led to an significant increase of white blood cells. Korean red ginseng increased UREA (blood urea nitrogen) in healthy volunteers, but it didn't exceed the range of normal values, and in the subsequent process of treatment there is no effect of elevating UREA. After the three weeks, Korean white ginseng showed relatively low the content of blood glucose and low-density lipoprotein cholesterol. After the five weeks, compared with the other treatments, Korean red ginseng increased white blood cells, platelet distribution width and average volume of platelet. Korean white ginseng decreased low-density lipoprotein cholesterol. American ginseng decreased blood creatinine in healthy volunteers. In conclusion, through test the blood routine, urine routine, liver function, renal function, blood glucose, blood lipid and electrocardiogram, the healthy volunteers continuous taking ginseng for 35 days (3 g/day) is safe and reliable, and have no obvious adverse reactions and side effects.

고려인삼과 미국삼의 수삼 및 홍삼품질 비교 (Comparison of Quality on the Raw and Red Ginseng in Korean and American Ginseng)

  • 고려인삼과 미국삼을 공시하여 Panax속 종간의 원료수삼 및 홍삼 등급에 기초하여 품질을 평가분석하였다. 1. 미국삼은 고려인삼에 비하여 근중이 적고 부정형 지근이 동체에서 많이 발생하는 관계로 체형이 불량하였다. 2. 홍삼수율은 고려인삼이 30.4%,미국삼이 33.8%로 미국삼이 높았고 홍삼의 본삼수율은 고려인삼이 80.4%, 미국삼이 72.2로 고려인삼이 높았다. 3. 원료수삼 1등급은 고려인삼이 미국삼에 비하여 2배정도 많았으나 2등급은 미국삼이 고려인삼에 비하여 많았다. 한편 고급홍삼인 천지삼율은 고려인삼이 미국삼에 비하여 높았다. 4. 홍삼 품질의 저하 원인은 고려인삼 미국삼 모두 내공과 내백 발생에 있었고 특히 미국삼은 내공에 비해 내백이 3배 가량 많이 발생하였다. 5. 홍삼의 지별 분포에서 고려인삼은 20지 이상의 대편급이 많았고 미국삼은 20지 이하의 소편급이 많았다.

Simultaneous Quantification of 13 Ginsenosides by LC-MS/MS and its Application in Diverse Ginseng Extracts

  • Ginseng (Panax ginseng Meyer) has been used as traditional herbal drug in Asian countries. Ginsenosides are major components having pharmacological and biological efficacy like anti-inflammatory, anti-diabetic and anti-tumor effects. To control the quality of the components in diverse ginseng products, we developed a new quantitative method using LC-MS/MS for 13 ginsenosides; Rb1, Rb2, Rc, Rd, Re, Rf, 20(S)-Rh1, 20(S)-Rh2, Rg1, 20(S)-Rg3, F1, F2, and compound K. This method was successfully validated for linearity, precision, and accuracy. This quantification method applied in four representative ginseng products; fresh ginseng powder, white ginseng powder, red ginseng extract powder, and red ginseng extract. Here the amounts of the 13 ginsenosides in the various type of ginseng samples could be analyzed simultaneously and expected to be suitable for quality control of ginseng products.

고려인삼 추출물이 고온환경에 노출된 흰쥐에 미치는 영향 (Effects of Korean Ginseng(Panax Ginseng, C. A. Meyer) Extracts on Rat Exposed to Heat Environment)

  • 주기적인 고온환경 부가와 백삼 및 홍삼 추출물의 투여가 흰쥐의 체중, 식이섭취량, 혈액성분, 장기중량 및 간의 지질과 산화에 미치는 영향을 살펴보았다. 흰쥐를 이용한 동물모델 실험결과, 매일 $38^{\circ}C$에서 5시간씩 7일간 고온환경을 부가한 경우 주당 평균체중 증가률이 감소하는 현상이 관찰되었으나 뚜렷한 식이섭취량 변화는 없었다. 반면 고온환경 부가전후에 백삼과 홍삼 추출물을 투여한 경우에는 고온환경 부가에 따른 평균 체중증가률 감소가 억제되었다. 고온환경 부가에 의한 혈액성분 변화를 살펴본 결과에서는 적혈구수와 혈색소량은 큰 차이를 나타내지 않았으나 백혈구수는 다소 증가하는 경향을 나타내었으며 인삼 추출물 투여는 백혈구수 증가도 억제하는 효과를 나타내었다. 고온환경 부가에 따라 장기중 간장의 중량비가 감소하였고 간의 MDA 함량이 크게 증가하였다. 인삼 추출물 투여시에 고온환경부가에 따른 간의 중량감소 및 MDA 함량 증가 등의 변화도 억제되는 효과가 있었다. 특히 간의 중량변화 및 간의 지질과산화 억제효과에 있어서는 백삼보다 홍삼 추출물의 효과가 다소 우수하였으며 통계학적으로 유의적인 수준은 아니었다. 향후 인삼중의 유효성분 구명이나 관련 물질들의 활성 변화에 대한 연구가 필요할 것으로 판단되지만 고온 등 급격한 주위환경가 변화에 대한 인삼의 생체항상성 유지효과를 일부 증명할 수 있는 결과인 것으로 판단되었다.

인삼박으로부터 수용성 산성다당체의 추출 조건 분석 (Analysis of the Extraction Condition of Soluble Acidic Polysaccharides from Ginseng Marc)

  • This study was carried out to investigate the optimum conditions for extraction of soluble acidic polysaccharides from ginseng marc. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in ginseng marc. The amounts of soluble acidic polysaccharides in water extract of ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite the extraction time increasing from 0.5 hours to 6 hours. To estimate the rehydration rate of the freeze dried polysaccharide, the extracted acidic polysaccharide fraction powder was determined the amount of soluble acidic polysaccharides by carbazole-sulfuric acid method again. The rehydration rate of acidic polysaccharides from water-extract of red ginseng marc at room temperature was 100%. On the other hand, the rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 50%. The rehydration rate of acidic polysaccharides from water-extract of white ginseng marc at room temperature was 50%. The rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 40%. The rate of soluble acidic polysaccharide of Red Ginseng is higher than that of White Ginseng. We can find out the maximum extraction method of soluble acidic polysaccharide from ginseng marc.

감마선 조사선양이 인삼의 지방산 함량에 미치는 영향 (Influences of Gamma Ray Radiation Doses on Fatty Acid Contents in Ginseng)

  • In order to investigate the influence of gamma-irradiation on fatty acids in ginseng, ginseng were irradiated with various radiation doses(0-50 Mrads). The results are as follows. 1. With increasing dose, standard fatty acids were decreased in the order of linolenic acid, linoleic acid and oleic acid, and their contents in the presence of oxygen were decreased more t than that in the absence of oxygen. 2. Changes of lipid content in white ginseng by gamma irradiation were varied with the samples, but the changes of crude lipid and purified lipid in a sample were showed same fashion. 3. The fatty acid contents in white ginseng were decreased by gamma irradiation in the order of linolenic acid, linoleic acid, oleic acid, stearic acid and palmitic acid.
