Gene Editing for Major Allergy Genes using Multiplex CRISPR-Cas9 System & Prime Editing in Peanuts (Arachis hypogaea L.)

  • Recently, food-induced allergies have emerged as major global concerns. In the past ten years, it has doubled in western nations, and it has also increased in Asia and Africa. In many cases of food allergy, peanut allergy is prevalent, typically permanent, and frequently life-threatening. Therefore, we utilized gene editing techniques on the three major allergen genes in peanuts, Ara h 1, Ara h 2, and Ara h 3. Using gibson assembly and golden gate assembly, we created two vectors, the gRNA-tRNA array CRISPR-Cas9 system and Prime-editing. Using LBA4404 strain and agrobacterium-mediated transformation, the vectors were transferred to two elite Korean peanut lines. After co-cultivation and tissue culture, we extracted the tissue cultured peanut DNA amplified the hygromycin resistance gene and Cas9 gene in the T-DNA region. The integration of the T-DNA region into the host genome was demonstrated by the presence of a specific band in some samples. There have only been a few reported peanut gene editing studies. So, this study will contribute to peanut allergy and gene editing research.

A new species of the genus Monoblastus Hartig (Hymenoptera: Ichneumonidae: Tryphoninae) with a key to species from South Korea

  • A new species, Monoblastus forsythia sp. nov. is described from South Korea. All specimens of this new species were collected on Forsythia koreana (Nakai). A taxonomic study of South Korean Monoblastus was initiated by Uchida in 1930, and until now four species have been recorded from South Korea. The genus Monoblastus is a Holarctic genus with 31 described species, 14 species are Nearctic, seven are Western Palaearctic region, and nine species occur in Eastern Palaearctic region. In the study, I provide description and photos of both sexes of this new species. Also, modified key to species of the South Korean Monoblastus is provided.

A Study on Costume of Arctic Circles in Pacific Coast (태평양 연안 지역 북극권 복식 특성 연구)

  • The North Pacific Arctic region has common factors such as climatic characteristics and similarity of animals inhabiting the region. But also there exists geographical barriers that separates the tribes, different languages between the tribes. Although there are such differences, the clothing and ornaments of the region have relatively similar design and style. And above all possess the ‘spirit’. The tribes find the motives of such ‘spirit’ in human, animal, and soul\`s adaptability to change and in grafting such changes of forms into clothing. Especially as means of pleasing the animal that they vitally rely on, the tribes made the clothing as beautiful as the nature itself and they tried to connect the humans and animals universally through such clothing that have social, artistic, and enchantic conditions. The supply of raw materials of animals has elevated the creativeness one step up and the precise knowledge about fur show their superior techniques in making fur clothing. The use of gutskin has is an excellent example of such knowledge, which is very unique of the region. The gutskin has moderate plasticity and thus can be cut into all sorts of pattern. It harmonizes the functionality and practicality. The worldwide fashion trend is dominated by Western style, but the clothing of this region is still keeping its distinctive folk identity. At the start of the research, Kayak and itelmen tribes of Asia, the tribes of Amur river and Aleut and Tlingit tribes of North America seems to be geographically too far from each other and therefore searching theoretical background for common cultural origins seems to be immoderate. But lighting the fact that geographical adjacency that can be perceived through costume cultural history, is the most important factor that gives mutual influences to costume culture between the neighboring tribes, cultural relative similarity of the costume is influenced by geographical location rather than physical distance between the tribes. Also humans\` adaptability to their environment is seriously contaminated with man-made products. This study on North Pacific Arctic region is telling us many things about our past, present and future.

The Determining factors and Temporal and Spatial Characteristics of Chemical Oxygen Demand in Jinhae Bay (진해만에 있어서의 COD의 시공간적인 특성 및 결정인자)

  • Determining factors and temporal & spatial characteristics of COD(Chemical Oxygen Demand) at the sea surface in Jinhae bay have been examined by using seasonal data, taken at twenty six stations over the whole bay during 1989~1994 by NERDA. The data have been analyzed in terms of long term means, anomalously large values. Jinhae bay is divided into three regions based on the time mean : mouth of Jinhae bay, inner sea of Masan bay, western sea of Jinhae bay called region 1,2 and 3, respectively. The horizontal distribution of the long term mean of COD at each station is similar to those of nitrogen and phosphorus. Characteristics of whole mean variation in the year shows high range of variation in region 2. It was appear to decreases every year in whole trend. Factors determining seasonal variation in whole COD mean are relative to salinity and nutrient, affected by precipitation in summer. Spatial variation shows high range of fluctuation in region 2 compare to other region. Factors determining of spatial variation of COD was appear to nutrient, affected by pollutant load of land area and bottom sediment. The long term mean of COD at each station is closely related with thats of nutrients. The correlation coefficient between COD and nitrogen, phosphate phosphorus was found to be high as 0.75, 0.78, respectively. Anomalously large COD was observed 14 times at 6 stations. These stations are located in inner sea of Masan bay(Region 2) and Songjeong bay(Region 1). The seasonal frequency of the observed anomalous COD is large in April, and other seasons are much the same.

Functional Characterization of the Squid Calexcitin-2, a Calcium and GTP-binding Protein

  • Calexcitin, a calcium-binding protein, was previously cloned and functionally characterized in the squid Loligo pealei. We now report the cloning of a second form of Calexcitin, Calexcitin-2, found in the squid Todarodes pacificus optic lobe. Calexcitin-2 has a significantly different carboxyl terminal region than Calexcitin-1. It lacks the CAAX motif, which is a farnesylation site. The amino acid sequence of Calexcitin-2 shows an 84% identity with Calexcitin-1 and also displays a strong cross immunoreactivity. Western blotting shows that Calexcitin-2 was expressed exclusively in the optic lobe region of squid, but not in other body organs. Regardless of its lack of conserved regions for GTP-binding, Calexcitin-2 shows moderately low affinity GTP-binding and also shows dramatic conformational change induced by GTP-binding. Three possible GTP-binding region mutations, K142A, D144A, and K157A, did not change the G TP binding affinity. This raises the possibility that Calexcitin-2 may have a novel GTP-binding motif.

A Study on Upper-Class House in North Kyeung-Buk Region (경북북부지역의 상류주택에 관한 조사연구)

  • This paper was made to examine floor plan and block plan of traditional upper-class houses in the north Kyeungsangbukdo region with consideration into city and country each. The results of this study were as following; First, Munkyung area is latitudinally located in northern area along with the Yechon and Andong. However, Munkyung area has open plot system while Yechon and Andong has not. It is because even if Munkyung area is located in the western of Nak-Dong river and belongs to the northern region, it has belonged to the cultural area which is developed around Sangju area. Second, Youngduk area belongs to the east-coast but it had frequent cultural exchange and marriage with Andong which is located on the road along with Tae-Baek Mountain. Due to the cultural exchange, their housing type is very similar to those of Andong which is close plot plan of "ㅁ" type. Therefore, even if Youngduk is located in the east-coast area, it should belong to the cultural background of Andong area. Regionally, the culture of Kyongsangbukdo has been divided into northern area, southern area, and east-coast area. But, the findings of this study indicate that the analysis of location and floor plan of housing should not be based on the natural environment oand geographic characteristics. Hence, it should be based on the all aspects of environment, including social and cultural nature to find more reliable and accurate results.e results.

Detailed Analysis of the 5'-Coding Region of SCN5A Gene in Korean Genome

  • We have identified and analyzed the 5'-coding region of SCN5A gene in Korean genome. Although its sequence has already been reported in western countries it is still important to confirm our own sequence for the establishment of Korean-suitable diagnosis on genetic basis. Total RNAs were obtained from three healthy Korean adult hearts and reversely transcribed. RT products were then subjected to PCR reaction followed by DNA sequencing. Three different sets of SCN5A primers were designed and used for the amplification of 5'-coding region of SCN5A from Korean genome. Amplified sequence was roughly one-10th of the entire SCN5A mRNA in size and its detailed sequence was completely matched up to the previously reported sequence. There was no difference between three heart samples, either. So, SCN5A was concluded as the relatively stable gene comparing to other genes that are involved in long QT syndrome.

  • This report is the result of the basic geologic investigation for the purpose of preparing the long-term development program of the U-jeon Consolidated Coal kline. The Consolidated Coal Mine is located at Gujeol-ri, Wangsan-myeon, Myeongju-gun, and Yucheon-ri, Bug-myeon, Jeongseon-gun, Gan-gweon Province (128$^{\circ}$ 43′10.4"-128$^{\circ}$ 46′10. 4"of east long-ititude, 37$^{\circ}$ 30′-37$^{\circ}$ 33′ of north latitude). This region, the western part of Taebaek mountain range, shows a ragged mountinous feature. Formations of the Pyeongan System of Paleozoic Era are distribu ted in the region with the surrounding Great Limestone Series of Joseon System which covers the south-eastern part of the region. The Pyeongan System is divided into four formations, namely, the Hongjeom, the Sadong, the Gobang and the Hongam, in ascending order. The sadong Formation intercalates several coal beds, and two coal beds out of them are minable. The coal beds are variable in thickness, having the repeated swelled or poket and the pinched parts, which suggest all intense disturbance caused by folding. The heat value of the coal is 5, 500cal. on the average. The total amount of coal reserves of the U-jeon consolidated Coal Mine is estimated at about thirty million metric tons.

Craftsmanship of Non-Educated Constructors in Korean-Style Secondary Station(Gong-So) of Naepo Region (충남 내포지방의 한옥 공소에 나타난 민간 기술자의 현장기법)

  • This study is aimed to look into the craftsmanship non-educated constructors in Korean-style secondary stations(Gong-So) of Naepo region which is located in the northwestern Chungcheongnam-do. Although the area was persecuted as one of three cradles of Korean Catholicism, Catholic activities flourished there. The 13 cases of Korean-style secondary stations were selected through a survey report list about modern cultural assets of Chungcheongnam-do published in december 2004. This study examined the methods of plan layout and framework of Gong-So to look into craftsmanship of non-educated constructors, the results are as follows; The methods of plan layout are divided into three types according to existence and arrangement of inner column(Nae-Ju); 'Layout with Nae-Ju' is considered as the primary form of Korean-style Gong-So with using regular Nae-Ju. 'Layout with the different position of the center between Nae-Ju and exterior column(Oe-Ju)' and 'layout without Nae-Ju' is appeared in Korean-style Gong-So which is important to assembly and alter function. The methods of framework are appeared variously according to different plan layout for securing worship space. The unusal beam(Chung-Ryang) is used to solve different spans of altar and narthex. When space was formed widely without Nae-Ju, diagonal members and M letter type rafters are used, they are influenced by the technique of western wooden roof truss.

  • Double-stranded RNA (dsRNA) induces gene silencing in a sequence-specific manner by a process known as RNA interference (RNAi). The RNA-induced silencing complex (RISC) is a multi-subunit ribonucleoprotein complex that plays a key role in RNAi. VIG (Vasa intronic gene) has been identified as a component of Drosophila RISC; however, the role VIG plays in regulating RNAi is poorly understood. Here, we examined the spatial and temporal expression patterns of VIG-1, the C. elegans ortholog of Drosophila VIG, using a vig-1::gfp fusion construct. This construct contains the 908-bp region immediately upstream of vig-1 gene translation initiation site. Analysis by confocal microscopy demonstrated GFP-VIG-1 expression in a number of tissues including the pharynx, body wall muscle, hypodermis, intestine, reproductive system, and nervous system at the larval and adult stages. Furthermore, western blot analysis showed that VIG-1 is present in each developmental stage examined. To investigate regulatory sequences for vig-1 gene expression, we generated constructs containing deletions in the upstream region. It was determined that the GFP expression pattern of a deletion construct (${\Delta}-908$ to -597) was generally similar to that of the non-deletion construct. In contrast, removal of a larger segment (${\Delta}-908$ to -191) resulted in the loss of GFP expression in most cell types. Collectively, these results indicate that the 406-bp upstream region (-596 to -191) contains essential regulatory sequences required for VIG-1 expression.
