Real-time PCR Quantification of White Spot Syndrome Virus (WSSV) and Hepatopancreatic Parvovirus (HPV) Loads in Shrimp and Seawaters of Shrimp Ponds on the West Coast of South Korea

  • Viral diseases are major emerging problems of shrimp that have affected the production, and even complete losses for shrimp farms. In this study, we developed a sensitive TaqMan real-time PCR method to quantify white spot syndrome virus (WSSV) and hepatopancreatic parvovirus (HPV) in the shrimp and pond water in which fleshy shrimp, Fenneropenaeus chinensis, and Pacific white shrimp, Litopenaeus vannamei, are reared. WSSV and HPV in pond seawaters ranged from $1.65{\times}10^3$ to $2.43{\times}10^9$ and from 0 to $4.43{\times}10^5$ copies/L of seawater, respectively. Of 20 ponds analyzed, all pond water and shrimp were positive for WSSv. L. vannamei showed higher susceptibility to WSSV than F chinensis. HPV was detected only in the pond water for F chinensis. In shrimp tissue, however, HPV was found in both species, with 23-times higher infection rate in F chinensis than L. vannamei. The total bacterial counts in the pond water ranged from $2.23{\times}l0^3$ to $1.98{\times}l0^5\;CFU/mL$. The variations in total bacterial count for each pond appeared to correlate to the variations of the WSSV load. Statistical analysis indicated that there was no significant difference (P>0.05) between the WSSV load in pond water and shrimp, and there was no relationship between total bacterial load and viral load in the pond water. However, a significant difference (P<0.01) was found between HPV load and L. vannamei and F chinensis pond water.

Effects of Salinity and Standard Toxic Metals (Cu, Cd) on Fertilization and Embryo Development Rates in the Sea Urchin (Strongylocentrotus nudus) (염분과 표준 독성물질(Cu, Cd)이 둥근성게(Strongylocentrotus nudus)의 수정 및 배 발생률에 미치는 영향)

  • Effects of salinity and standard toxic metals on fertilization and embryo development rates were investigated in the sea urchin, Strongylocentrotus nudus. Spawning was induced by injecting 1ml of 0.5 M KCl into the coelomic cavity. The fertilization and embryo development rates were below 20% when salinity was 25 psu or lower, but were above 90% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition (not including Cu and Cd) were greater than 90%, but decreased with a high negative correlation (r) of 0.89 and 0.91 with the increasing of Cu and Cd concentrations, respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of S. nudus are between 30 and 35 psu, and the biological assays of fertilization and embryo development rates using S. nudus are useful methods for the ecological toxicity test of marine pollution elements.

Features of Malignancy Prevalence among Children in the Aral Sea Region

  • Objective: A study of primary cancer morbidity among children and subsequent calculation of average annual incidence were carried out for boys and girls, and young men and women in Kazakhstan. Methods: The investigated population lived in three areas of the Aral Sea region: designated catastrophe (Aral, Kazalt, Shalkar regions), crisis (Zhalagash, Karmakshy, Shiely regions), pre-crisis (Irgiz, Arys, Ulytau regions). Zhanaarka region of Karaganda oblast was applied as a control. Parameters were retrospective analyzed for the 10 years from 2004 to 2013. Result: The results indicate that indices of children cancer morbidity were slightly higher in the Aral Sea region than in the control district, but they were comparable with similar data from studies in other regions. In all areas of the Aral Sea region, except for Ulytau, primary cancer morbidity exceeded the control level by 1.3-2.7 times (4.7%000). Hematological malignancies, including solid tumors - tumors of musculoskeletal system and skin, digestive system, brain and central nervous system predominated. Stress levels in zones of the Aral Sea region were slightly higher in the crisis zone than in the catastrophe zone that can be explained by the phenomenon of wave-like dynamics of disease growth risk. Gender differences in characteristics of malignancy formation were not more pronounced in the studied region. Conclusion: Indices of children cancer are slightly higher in the Aral Sea region than in the control area of Kazakhstan, but they are comparable to results for other regions.

Spawning was induced by injecting $1{\sim}2$ mL of 0.5 M KCl into the coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. The fertilization rate was below 30% when salinity was 20 psu and lower, but was almost above 90% when salinity was 25 psu and higher. The embryo development rate was below 60% when salinity was 25 psu and lower, but was above 80% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition(not including Cu and Cd) were greater than 90%, but decreased a high negative correlation with the increasing of Cu(r=-0.80, r=-0.78) and Cd(r=-0.90, r=-0.82) concentrations, respectively. The fertilization and embryo development rates were significantly inhibited in the addition of Cu($EC_{50}$=17.30 ppb, $EC_{50}$=10.32 ppb) and Cd($EC_{50}$=364.57 ppb, $EC_{50}$=244.04 ppb), respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of H. pulcherrimus are above 25 psu and 30 psu, respectively, and the biological assays of fertilization and embryo development rates using H. pulcherrimus are useful methods for the ecological toxicity test of marine pollution elements.

  • The purpose of this study is to present the direction of development of our navy's response strategy through analysis at the ends, ways, and means level of the Chinese navy's offensive strategy for the West Sea. As a result of the analysis, at the ends level, the Chinese Navy's offensive strategy for the West Sea strategy is being linked to a grand strategy to protect maritime rights and achieve maritime power between the U.S. and China competition, at the ways level, the Chinese Navy is expected to create a foundation for the international community to recognize the West Sea as China's inland sea through "routine entry" and "exercise authority", and in case of emergency, it will try to secure sea control in the West Sea in a short period of time by blocking Korea's maritime transportation route based on the overwhelming preemptive attack capability of aircraft carriers. At the means level, it is accelerating the construction of aircraft carrier warfare units and improving its ability to engage long-range missiles. As a direction of development of the Korean Navy's response strategy in response to this, first, Establishment and Development of National Maritime Security Strategy in conjunction with the Korean Indo-Pacific Strategy. Second, it proposes the development of the concept of effective security operations for the east sea area of the West Sea intermediate line, and third, the development of the concept of combat performance and capacity building to strengthen survival and lethality.

Virulence Factors and Antimicrobial Susceptibility of Vibrio parahaemolyticus Isolated from the Oyster Crassostrea gigas (양식 굴(Crassostrea gigas)에서 분리된 장염비브리오균의 독소 유전자 보유 및 항균제 감수성)

  • This study investigated the prevalence of Vibrio parahaemolyticus in the oyster Crassostrea gigas, which is commonly consumed raw. The presence of virulence factors and the antimicrobial susceptibility of isolates were also investigated. The overall prevalence rate of V. parahaemolyticus in oysters was 37.5% (36/96) and the range of concentrations was 30-11,000 MPN/100 g. PCR-based assays indicated that 9.6% (11/115) of the isolates were positive for the thermostable direct hemolysin-related hemolysin gene (trh), while none of the isolates were positive for the thermostable direct hemolysin gene (tdh). The Multiple Antibiotics Resistance (MAR) index was measured for 16 common antimicrobial agents and 46.1% (53/115) of the isolates had a MAR index > 0.2. The MAR index ranged from 0.07 to 0.73. The highest MAR index was observed with strain s150608, isolated in June 2015, which exhibited resistance to 11 antimicrobial agents. Our results demonstrate that oysters are high-risk sources of V. parahaemolyticus, although no antimicrobial agent was being used to promote growth or to treat bacterial infections in the sampled oyster-growing areas.

Analysis for gillnet loss in the West Sea using numerical modeling (수치 모델링을 이용한 홑자망 어구의 유실 원인 분석)

  • The Fishing gear loss has been repeated every year in the West Sea; however, there has been no solution. So fisher men have undergone economic loss every year. Thus it is required to reduce the loss of fishing gear. In this study to find out the reason that the fishing gear is lost in the Sea, 10 years data of wave and current for 6 locations in the West Sea were investigated and a numerical modelling were conducted into the behaviour of a gillnet in wave and current. The fishing gear was modelled with the mass spring model. As a result, it came out into the open that the location where fishing gear loss occurred most frequently was Choongnam province. The height of the maximum significant wave in this province was 6.7 m and the period of that was 4.4 second. The maximum current speed was 0.7 m/s. As a result of simulation with these data, it was revealed that the buoy is one of the reasons to decrease the holding power of the gillnet. For example, the tension of anchor rope was decreased to 50% while the drag coefficient or volume of buoy was decreased to 25%. So it is predicted that an improvement of the buoy contributes to the reduction of the gillnet loss.

Ecotoxicological Evaluation of Sewage Sludge Using Bioluminescent Marine Bacteria and Rotifer

  • Bioassay using the marine bacteria, Vibrio fischeri and rotifer, Brachionus plicatilis, and chemical analyses were conducted to assess the toxicity of the various sewage sludges, one of the major ocean dumped materials in the Yellow Sea of Korea. Sludge elutriates extracted by filtered seawater were used to estimate the ecotoxicity of the sludge. Chemical characterization included the analyses of organic contents, heavy metals, and persistent organic pollutants in sludge. Bacterial bioluminescent inhibition (15 min), rotifer mortality (24 hr) and rotifer population growth inhibition (48 hr) assay were conducted to estimate the sludge toxicity. EC50 15 min (inhibition concentration of bioluminescence after 15 minutes exposed) values by Microtox(R) bioassay clearly revealed different toxicity levels depending on the sludge sources. Highest toxicity for the bacteria was found with the sludge extract from dyeing waste and followed by industrial waste, livestock waste, and leather processing waste. Clear toxic effects on the bacteria were not found in the sludge extract from filtration bed sludge and rural sewage sludge. Consistent with Microtox(R) results, rotifer neonate mortality and population growth inhibition test also showed highest toxicity in dyeing waste and low in filtration bed and rural sewage sludge. High concentrations of persistent organic pollutants (POPs) and heavy metals were measured in the samples from the industrial wastes, leather processing plant waste sludge, and urban sewage sludge. However, there was no significant correlation between pollutant concentration levels and the toxicity values of the sludge. This suggests that the ecotoxicity in addition to the chemical analyses of various sludge samples must be estimated before release of potential harmful waste in the natural environment as part of an ecological risk assessment.

The Characteristic of the Disasters caused by Typhoons passing through the Sea Area around the Korean Peninsula (한반도 주변 해역을 통과한 태풍의 재해특성)

  • The purpose of this study is to find out the characteristics of disasters caused by typhoons passing through the sea area around the Korean Peninsula. It analyzed two cases, that is, in WEST and EAST cases. These include the typhoons passing through the Yellow Sea, west of the Peninsula and East Sea, east of the Peninsula without landing on the Peninsula. FCM (Fuzzy Clustering Method) analysis was performed on typhoons affecting the Korean Peninsula from 1951 to 2006. The analysis shows that WEST case's cluster has the curved track of NE-S, and EAST case's cluster has the straight track of NE-SW. Typhoons that pass through the Yellow Sea have little change in frequency and the weak intensity. On the other hand, the frequency and the intensity of typhoons passing through the East Sea show the increasing trend. The characteristic of disasters by typhoons affecting the Korean Peninsula from 1973 to 2006 appears differently for each case: EAST cases caused significant damage in flooding, while WEST cases did damage in houses, ships, roads, and bridges. Rainfall amount and maximum wind speed data are analyzed in order to understand the impact of the typhoons, and the result indicates that the WEST cases are influenced by the wind, and East cases by precipitation. The result of this study indicates that the characteristic of disasters is distinctive according to the Typhoon's track. If applied to establish the disaster prevention plan, this result could make a contribution to the damage reduction.

Feeding Habits of Pacific cod (Gadus macrocephalus) in the West Coast of Yellow Sea of Korea (우리나라 서해에서 출현하는 대구 (Gadus macrocephalus)의 식성)

  • The feeding habits of pacific cod (Gadus macrocephalus) were studied by analyses of stomach contents. Specimens of G. macrocephalus (n=407) were collected from the western coastal waters of Korea, monthly from July 2017 to June 2018. The size of G. macrocephalus ranged from 30.4 to 86.0 cm in total length (TL). G. macrocephalus was consumed mainly Pisces (71.5%) and Caridea (24.2%) based on % IRI, whereas other prey items including, Paguroidea, Euphausiacea and Cephalopods showed a low contribution to the diet. The feeding ratio of Caridea decreased with increasing fish size. While this decrease paralleled the increased consumption of fish preys. The feeding ratios of Pisces were the highest during spring, autumn and winter, whereas the proportion of Caridea was higher in summer.
