코로나 방전 플라즈마 처리수에 의한 어류 병원체 소독 효과 (Disinfection effect of corona discharged plasma water on fish pathogens)

  • Fish culture is constantly threatened by various infectious diseases which are largely transmitted by water. Plasma technology is being used to sterilize polluted water in many industries. In this study, two bacterial pathogens Aeromonas salmonicida and Streptococcus iniae, and a virus (viral hemorrhagic septicemia virus, VHSV) were subjected to plasma water that was produced by a corona discharge system. Growth of A. salmonicida was greatly inhibited from 105.61 CFU/ml in positive control to 103.51 CFU/ml in treated group by only 60 sec contact with plasma water. Similarly, S. iniae was inhibited from 105.85 CFU/ml to 103.40 CFU/ml. VHSV titer also decreased from 104.1 TCID50/ml to 101.45 TCID50/ml by the same treatment. Activation of water by the plasma was confirmed by the existence of ozone in the plasma water. These results suggest that plasma water could efficiently disinfect fish pathogens, possibly by the action of reactive oxygen species contained in the plasma water.

건강한 젊은 성인에서 액상 물질 한 모금 삼킴량의 측정 (Measurement of Volume of a Swallow for Liquid Swallowing in Healthy Young Adults)

  • Purpose: The aim of this study is to estimate one mouthful volume in a single swallow and average volume per swallow (AVS) in multiple swallows in the situation of toxic liquid poisoning. Methods: Thirty five men and 35 women were included in this study. Each subject was asked to drink one swallow and three consecutive swallows from bottle containing water and a bottle containing saline separately. We calculated one mouthful volume in a single swallow and AVS in three swallows. One mouthful volume and AVS were compared according to sex and content, respectively. One mouthful volume of water and saline was then compared with AVS of each. Results: Sixty seven adults(34 men; $26.9{\pm}3.2$ years, 33 women; $25.6{\pm}2.4$ years) completed the study. Men had larger one mouthful volume of water($49.1{\pm}19.9$ ml vs $39.7{\pm}10.2$ ml, p=0.02) and saline($20.7{\pm}10.9$ ml vs $14.0{\pm}4.6$ ml, p=0.004) and AVS of water($28.5{\pm}11.9$ ml vs $21.5{\pm}5.9$ ml, p=0.004) and saline($11.9{\pm}6.3$ ml vs $7.9{\pm}2.0$ ml, p=0.001) than women. One mouthful volume and AVS of saline swallow were lower than those of water swallow. AVS of three consecutive swallows was lower than one mouthful volume in water and saline swallow. Conclusion: We suggest that one mouthful volume in a single swallow is 21 ml in men and 14 ml in women and AVS in multiple swallows is 12 ml in men and 8 ml in women. AVS in multiple swallows is two-threefold lower than reference values(20~30 ml) commonly used in poisoning study.

한강유역의 수중미생물 오염도 조사 (Bacteriological Contamination of Water in Han River basin)

  • In order to investigate the bacteriological contamination of water in Han river, the survey was carried out in eight reservoirs of Seoul water supply during the period from January to December in 1985. 1. The counts by means of total bacteria in eight reservoirs by standard plate count method were as follows: $7.7\times10^2$ per ml in Paldang reservior, $9.6\times10^3$ per ml in Gueiri, $8.4\times10^4$ per ml in Doogdo, $1.6\times10^6$ per ml in Bogwang, $2.5\times10^6$ per ml in Noryangjin, $2.2\times10^6$per ml in Seon yoo, $5.9\times10^6$ per ml in Yungdeungpo and $1.9\times10^7$per ml in Gayang. 2. The average counts of total coliform in eight reservoirs by MPN method were as follows : $2.4\times10$ per 100 ml in Paldang, $5.6\times10^2$ per 100 ml in Gueiri, $2.3\times10^3$ per 100 ml in Doogdo, $5.1\times10^4$ per 100 ml in Noryang-jin, $1.2\times10^5$ per 100 ml in Bogwang, $6.2\times10^4$ per 100 ml in Seonyoo, $1.1\times10^5$ per 100 ml in Yungdeungpo and $2.8\times10^5$ per 100 ml Gayang. 3. The counts by means of fecal coliform in eight reservoirs by MPN method were as follows : non detection per 100 ml in Paldang, 5.2 per 100 ml in Gueiri, $1.2\times10^2$ per 100 ml in Doogdo, $1.6\times10^3$ per 100ml in Bogwang, $2.0\times10^3$per 100ml in Noryangjin, $6.6\times10^2$ per 100ml in Seonyoo, $1.2\times10^3$ per 100 ml in Yungdeungpo and $2.5\times10^3$per 100 ml in Gayang. 4. The counts by means of fecal streptococci in eight reservoirs by MPN method were as follows: non detection per 100 ml in Paldang and Gueiri, $6.9\times10$ per 100 ml in Doogdo, $3.2\times10^2$ 102 per 100 ml in Bogwang, $2.9\times10^2$ per 100 ml in Noryangjin, $3.0\times10^2$ per 100 ml in Seonyoo, $4.0\times10^2$ per 100 ml in Yungdeungpo and $14\times10^3$ per 100 ml in Gayang. 5. The counts means of pseudomonas aeruginosa in eight reservoirs by MPN method were as follows; non detection per 100 ml in Paldang, 2.4 per 100 ml in Gueiri, $1.5\times10$ per 100 ml in Doogdo, $2.0\times10$ per 100ml in Bogwang,$6.2\times10$ per 100mI in Noryangjin, $2.1\times10$ per 100ml in Seonyoo, $6.4\times10$ per 100mI in Yungdeungpo and $7.1\times10$ per 100ml in Gaynag.

상수원 수질관리를 위한 분변오염 지표세균에 관한 연구 (A study on Investigation of Fecal Contamination Indicator Bacteria for Management of Source Water Quality)

  • Coliforms is currently being used as the standard of environmental water qualify to evaluate the level of source water quality especially condition of fecal contamination. However, not properly applied to water quality management. So in this study, in addition to Coliforms, fecal contamination indicator bacteria turk at Feral Coliforms(FC), E. coli, Fecal streptococci(FS), Clostridium and environmental parameters related with it's distribution were investigated on a monthly basis in 6 water intakes of Han River. The mean of BOD, DO, SS and pH, benchmarks of source water management were maintained the second grade of environmental water quality standard applied to Han River but Coliforms exceeded it. Distribution of Coliforms ranged from 1.0×10¹ to 2.7 10/sup 5/ CFU/ml, FC ranged from ND to 5.3×10¹ CFU/ml, E. coli ranged from ND to 9.2×10¹ CFU/ml, FS ranged from ND to 2.5×10¹CFU/ml, they were steepy rise on July and August in common when rainfalls was heavy and water temperature was high, but Clostridium perfringens ranged from 1.7×10¹to 1.7×10¹CFU/ml not fluctuate by month. Statistical analysis of sampling data showed that most significant correlations occurred among FC and Coliforms(r = 0.840), E. coli(r = 0.792), FS(r = 0.687) and environmental parameters(temperature, turbidity, SS, rotor were all r > 0.60) while no significant correlation was observed between ammonia generally recognized fecal contamination indicator and bacteria. Identification of the coliforms showed that Enterobacter, Klebsiella, Citrobacter were comprised of 32%, 24%, 16% respectively, and E. coli were 7% of it. while E. coli was made up 85.9% of FC. The mean value of FC/Coliforms ratio, 5.2(0.1-42) were higher in Amsa, Guui than Jayang. Fecal coliforms, as those are able to reflect more particularly the extent of the fecal contamination, were considered useful in deciding the level of water treatment while monitoring the fecal contamination from the source of water supply. Therefore, it is expected that the water quality is going to be managed more efficiently by using fecal coliforms supplementarily to total coliforms which are current standard item of water-quality environment.

공침-부선기술에 의한 수용액 시료 중 흔적량 수은 [Hg(II)]의 분리 및 농축 (Separation and Concentration of Trace Mercury [Hg(II)] in Water Sample by Coprecipitation Flotation Technique)

  • 수용액 시료 중 흔적량 수은[Hg(II)]을 공침-부선기술에 의하여 분리-농축하는 방법에 관하여 연구하였다. 물시료 1000ml에 0.1M Ce$^{3+}$ 용액 3.0 ml를 가하고, 1.0M NaOH 용액으로 pH를 11.0으로 되게 조절하여 Hg(II)를 Ce(OH)$_3$와 함께 공침시켰다. 0.1${\%}$ sodium oleate 용액 2.0 ml를 가하여 만든 소수성 침전을 질소기체로 bubbling하여 용액 표면으로 띄웠다. 뜬 침전을 감압 플라스크에 모으고 2.0M HNO$_3$ 5.0 ml 로 녹였다. 탈이온수로 용액을 묽혀 25.00 ml로 정확히 만들었다. Hg(II)의 함량을 냉증기-원자흡수 분광법으로 정량하였다. 정량과정에서 Ag$^+$, Br$^-$, I$^- $ 등의 간섭은 관찰되지 않았다. 고려대학교 서창캠퍼스의 폐수에서 Hg(II)의 분석결과는 1.98 ng/ml 이었고, 이 값에 대한 상대표준편차는 3.6${\%}$이었다. 또 이 폐수에 1.0과 2.0ng/ml의 Hg(II)를 첨가하여 분석한 회수율은 각각 95와 91${\%}$이었다. 이런 결과로부터 본 분석법이 물시료 중 흔적량 Hg(II)의 정량에 꽤 정확하고 재현성이 있음을 확인할 수 있었다.

인삼식초에 관한연구 (Studies on Ginseng Vinegar)

  • Ginseng-vinegars were produced by the fermentation of 5% ethanol solution contained ginseng, red ginseng, ginseng marc and red ginseng marc using Acetobacter aceti 3281 for 26 days at 35$^{\circ}C$. The ginseng and red ginseng vinegar contained 0.236mg/ml of total sugar 0.236mg/ml of reducing sugar and 0.05% of ethanol and 1.005 of specific gravity 8,58CFU of viable cell count 3,24 of pH and 5.11% of acidity. Whereas the vinegars produced using the water-extracted red ginseng marc and the ethanol-extracted red ginseng marc were consisted of total sugar was 1.27mg/ml and 1.60mg/ml reducing sugar was 0.077mg/ml and 0.725mg/ml specific gravity was 1.001 and 1.004 the number of viable cells was 8.51CFU/ml and 8.1CFU/ml pH was 2.81 and 2.89 acidity was 5.18% and 5.32% respectvely ethanol concentration was 0.05% in both cases. In five-grade scoring test of sensory evaluation, it was estimated favorable that each vinegar made by were-extracted red ginseng marc, ethanol-extracted red ginseng marc ginseng and red ginseng ginseng from 0.5 to 32% of water-and ethanol-extract red ginseng was extracted with 10% white vinegar for 30 days. The best sensory vinegars were obtained that ginseng of 0.4~1.6% above red glnsend of 0.8% water-extracted red ginseng marc of 0.8~1.6% and ethanol-extracted red ginseng marc of 0.4~1.6% added in 10% white vinegar respectively.

  • 본 연구는 배지에 첨가된 목초액 및 코코넛액이 석곡 유묘의 증식 및 기내 개화에 미치는 영향을 조사하기 위하여 실시되었다. 목초액이 석곡 유묘의 증식에 미치는 영향은 NAA 0.1 mg/L와 kinetin 1.0 mg/L 가 첨가된 $H_{3} P_{4}$ 배지에 목초액 1.0 ml/L를 처리했을 때 유묘의 증식 및 생장이 양호 하였다. 코코넛액은 30 ml/L가 첨가되었을 때 유묘의 증식 및 생장이 가장 양호하였다. 기내 개화 역시 NAA 0.1 mg/L 와 kinetin 1.0 mg/L 가 첨가된 $H_{3} P_{4}$ 배지에 목초액 1.0 ml/L 또는 코코넛액 30 ml/L가 첨가되었을 때 전체 개체의 $ 20\% $에서 기내개화가 일어났다

부산시내 약수터의 세균학적 수질 (Bacteriological quality of ground water in Pusan area)

  • 부산시내에 산재하고 있는 70여개소의 약수터중 11개소를 선정하여 이들의 수질관리에 필요한 기초자료를 얻고자 1981년 10월부터 1982년 9월까지 132개 시료를 취하여 대장균군, 분편계대장균, 일반생균수 및 대장균군의 조성을 시험한 결과를 요약하면 다음과 같다. 1. 11개소의 대장균군 최확수의 중앙치는 구포가 93/100ml로 제일 높았고, 수영, 연산동, 성지곡이 68/100ml 우동과 대신동이 59/100ml, 전포동, 망미동은 43/100ml 영도, 동래가 33/100ml그리고 감만동이 13.3/100ml로 제일 낮았으며 이들 중 6개소가 50/100ml 이상이었다. 2. 분편계대장균 최확수의 중앙치는 영도가 12.1/100ml으로 제일 불결하였고, 수영을 비롯한 8개소는 9.1/100ml, 우동은 6.4/100ml이었으며 감만동이 제일 깨끗하였다. 3. IMViC 반응에 의한 대장균군의 조성은 Escherichia coli group이 $7.64\%$, Klebsiella aerogenes group이 $16.78\%$, Citrobacter freundii group이 $42.86\%$, 기타가 $32.72\%$로 나타났었다. 4. 일반생균수의 중앙치는 $48{\sim}4.4{\times10}^2/ml$의 넓은 분포로서 보사부(1963)의 수질기준에 절반 지역이 초과되었다.

정수용 입상활성탄상의 세균 (Bacteria on Granular Activated Carbon for Tap Water Purifier)

  • 정수용 입상활성탄(Granular Activated Carbon, GAC)에서 발생하는 세균 생장 현상을 파악하기 위해 활성탄 칼럼을 제작하고, 수돗물을 고정식 상향류 방식으로 (EBCT = 0.92~1분) 통수시켰다. 봄에는 14일 동안 세균수의 파과(breakthrough) 현상이 없었다. 여름에는 야자계와 석탄계 활성탄 칼럼의 유출 세균수는 비슷하여 파과전에 $10^2$ CFU/ ml수준으로 유입수보다 낮았지만, 10일 이후 유출수가 $10^3$ CFU/ml 로 유입수의 $10^2$ CFU/ml보다 높은 값을 보였다. 칼럼 내부 4지점의 활성탄을 깊이에 따라 분석했을 때 유입수 유입부에 가까울수록 많은 세균수가 관찰되었고 세균군집의 활성은 유사하였다 성장하는 세균은 야자계와 석탄계 활성탄이 서로 달랐지만 Acinetobacter 속이 우점을 이루고 있는 것으로 나타났다.

청량음료의 미생물 오염에 대하여 (On the Microbial Contamination in Softdrink Manufacturing Process)

  • The authors have investigated about the microbial contamination of goods which is a criterion of hygienic control. Conducting on investigation, a special attention has been paid on the rate of microbial contamination in the goods, especially in manufacturing process of the softdrink. The authors also made an experiment on total microbes which is the criterion of contamination in each step of the process and in raw materials together with materials to be used for subdividing. Results obtained were as follows : 1) The orgin of microbial contamination was found in bottle cap and in tap water, that is, there appeared 9 colony per ml in bottle cap and 31-74 colony per ml in tap water, respectively. 2) It was found that microbial contamination are 4 colony per ml in average through year. However, it appeared 1 colony per ml in winter and 8 colony per ml in summer. 3) Coliform groups are not detected in goods through a year. 4) There was no variation in number of total microbes after ion exchange resin passage in purification process of tap water. 5) The number of microbes in goods are decreased when the raw materials are treated in high temperature short time (HTST) sterilization.
